Aflatoxin B1 and reproduction. I. Reproductive performance in female rats.

An experimental group of female rats was exposed to aflatoxin B1 at a dose of 7.5 mg/kg body weight daily for 14 days through oral intubation. A deleterious effect on the gonads and embryotoxicity in the experimental animals were observed. There were reductions in ovarian and uterine sizes, increases in fetal resorption, implantation loss, and intra-uterine death. The data showed significant disturbances in oestrus cycle, inhibition of lordosis, and reductions in the conception rates and litter sizes, suggesting severe impairment of fertility in the aflatoxin exposed rats. An aflatoxin blood level of 86.2 +/- 1.3 ppb was recovered in the experimental animals.

Reproduction of the normal sexual cycle and reproductive ability in castrated guinea pigs by implantation of the ovary by a new method.

Hormonal and generative activity of the "free" ovarian graft in the uterine wall was studied in 43 sexually mature guinea pigs. Ovaries were grafted by a personal method and by the method of Bland and Donovan. In the author's method, halves of the ovaries were implanted in the uterine wall in such a way that the graft communicated with the uterine cavity and received supplementary vascularization from the connective tissue of the parametrium. Morphologic and histochemical (PAS-positive substances) changes in the free ovarian grafts in the uterus were studied. Hormonal activity of grafts was assessed on the basis of vaginal smears. It was concluded that hormonal activity of the ovary engrafted in the uterine wall depends on the method of grafting. By the author's method, in which blood vessels from the parametrial connective tissue penetrate into the ovary, hormonal activity is the same as before operation and pregnancy is possible. In the method of Bland and Donovan, the sexual cycle and time of opening of the vagina are considerably shortened.

[The treatment of assisted reproduction--clinical and preclinical aspects, functioning of the Unit of Human Reproduction at the Central Hospital of Luxembourg].

Since a few years the OBGYN department at the Centre Hospitalier in Luxembourg has intensified its practice of ART with the introduction of a multidisciplinary team and a decentralized IVF program. While clinical investigations, cycle-monitoring and surgery, oocyte pick-up are commonly performed at the CHL, the biological supplement is transferred to next situated Belgian laboratories. As Luxembourg has no national bio-ethical law, we apply partly the French law of 1994.
