The effect of spironolactone on the elemental composition of the intraluminal fluids of the seminiferous tubules, rete testis and epididymis of the rat.

Embryologically, the epididymis and renal collecting tubules develop from the mesonephric duct. Aldosterone enhances the reabsorption of sodium from the collecting tubule, and there is evidence that sodium is removed from the epididymal lumen against a concentration gradient. Rats were treated with spironolactone, an aldosterone antagonist, and the concentrations of seven elements, including sodium, were measured in intraluminal fluids from the testis and epididymis. The sodium concentration in caput epididymidal fluid rose (102.5 +/- 4.4 to 128.0 +/- 6.8 mmol./l., p less than 0.01) after 14 days of spironolactone treatment. This finding is consistent with the hypothesis that aldosterone acts on the caput epididymidis, like the collecting tubule, to augment the removal of sodium from luminal fluid.

[Scanning electron and light microscopic studies of the surface epithelium of the rete testis and epididymis in the boar. II. Ductus epididymis].

The entire ductus epididymidis is characterized by pseudostratified columnar epithelium which contain principal and basal cells. Apical cells are found in the proximal segments. Long stereocilia projected from the apical surface of the principal cells. Signs of adjacent apocrine secretory activity are shown. The various segments of the ductus epididymidis present differences in the height of the epithelium, the development of the stereocilia, the appearance of granules in the cytoplasm, the luminal sperm concentration and the height of the peritubular layer.

Content of K+ and Na+ in seminiferous tubule and rete testis fluids from Sertoli cell-enriched testes.

Seminiferous tubules of rats exposed to x-irradiation before birth were subjected to micropuncture in situ at 50 days of age to obtain samples of fluid 4 h after ligation of efferent ducts. The concentrations of cations in this fluid were: potassium, 39.7 +/- 1.2 mM, and sodium, 136.3 +/- 1.2 mM (means and standard errors, n = 5). Histologic examination revealed that germ cells constitute less than 1% of the cell population within the seminiferous tubules of these rats; the remaining cells were all Sertoli cells. Sertoli cells showed efflux of 86Rb+ with t1/2 of approximately 11 min and an active ATPase in plasma membranes. These activities were similar to those of Sertoli cells from normal rats. Germ cells from normal rats showed less rapid efflux of 86Rb+ (t1/2 greater than 60 min) and less active Na+/K+ ATPase in plasma membranes. It is concluded that Sertoli cells are responsible for the high concentration of potassium in seminiferous tubule fluid and that plasma membranes of these cells contain an active K+ pump that is not inhibited by ouabain (1 mM).

The characterization of a UDP-N-acetylglucosamine: Gal beta 1-4Glc(NAc) beta 1-3 N-acetylglucosaminyltransferase in fluids from rat rete testis.

Using Gal beta 1-4GlcNAc or Gal beta 1-4Glc as substrates, the UDP-N-acetylglucosamine: Gal beta 1-4Glc(NAc) beta 1-3-N-acetylglucosaminyltransferase (GlcNAcTase) activity of rat rete testicular fluid was investigated with respect to the effects of pH and of divalent metal ions, apparent Km value, acceptor specificity and identity of products. The enzyme, whose activity was dependent absolutely on the presence of Mn2+, had an optimum pH of 7.5. It was completely inhibited by EDTA, Zn2+ and gossypol, and partially by Co2+ and Ca2+. Triton X-100 (1%) had no effect. When several oligosaccharides were tested as acceptors, the highest affinity was found with Gal beta 1-3[Gal beta 1-4GlcNAc beta 1-6]Gal beta 1-4Glc. With Gal beta 1-4Glc as acceptor, the product was identified from its 1H-NMR spectrum as GlcNAc beta 1-3Gal beta 1-4Glc (lacto-N-triose II) by reference to the spectrum of authentic lacto-N-triose II. Both testicular and epididymal fluids showed GlcNAcTase activity. In epididymis the specific activity increased gradually from the proximal caput to the proximal cauda.

Evidence of interepithelial seminoma spread into the rete testis by immunostaining of paraffin sections with antibodies against cytokeratin and vimentin.

Seminomas are tumors of high proliferative activity and show a marked tendency towards local invasion with the capacity for interepithelial spread within the seminiferous tubules as well as into the rete ductules. Immunohistologic investigations were carried out on paraffin sections of 47 typical seminomas. Immunostaining with antibodies against cytokeratin and vimentin allows the convenient detection of even small rete residuals in cases of subtotal rete destruction as well as the identification of discrete interepithelial seminoma spread within the rete ductules, thus facilitating seminoma staging.
