A flexible instrument control and image acquisition system for a scanning electron microscope.

We have developed an instrument control and image acquisition system for use with scanning electron microscopes. By making the system flexible over a wide range of operating voltages, scan generation and image acquisition modes can be easily accommodated to a wide range of instruments. We show the implementation of this system for use with a custom-built low-voltage scanning electron microscope. We then explore the simple modifications that are required for control of two instruments intended for use as free electron lasers.

Deposits on soft contact lenses. Electrophoresis and scanning electron microscopic examinations.

Deposits on soft contact lenses were studied by several methods, microscopic examination, scanning electron microscopy, electron microprobe and electrophoresis on polyacrylamide gels of removed proteins. The first two approaches permitted classification of deposits into globular deposits, "Mulberry-like growths", opaque deposits, protein films and red spots. The electron microprobe indicated specific compositions of each. The electrophoretic technique demonstrated the presence of lysozyme in all the deposit types, with other unidentified proteins or degradation peptides. Whatever the deposit type, heat disinfection is not advisable since proteins are always present. Combination of chelating agents and oxidizing products are logically to be preferred for cleaning lenses.

Early tissue infiltrate in porous polyethylene implants into bone: a scanning electron microscope study.

The results of the present study demonstrate the utility of the scanning electron microscope for characterizing the ultrastructure of the initial tissue infiltrate in porous polyethylene implants. Shortly after implantation a thin noncellular fibrous-like coating was observed to form on the pore surface. The cells observed in the polyethylene pellets 3 days after implantation were generally consistent with what one would expect to see in a hematoma. As early as 14 days after implantation much of the blood clot was replaced by newly formed bone spicules. Tissue shrinkage accompanying dehydration of the specimen for scanning electron microscope study although a disadvantage occasionally proved useful in that it provided the opportunity to study the internal surface of the fibrous coating when separated from the surface of the implant. Less shrinkage was observed in implants whose pores were filled with bone spicules.

A method of dynamic chromatic aberration correction in low-voltage scanning electron microscopes.

A time-of-flight concept that dynamically corrects for chromatic aberration effects in scanning electron microscopes (SEMs) is presented. The method is predicted to reduce the microscope's chromatic aberration by an order of magnitude. The scheme should significantly improve the spatial resolution of low-voltage scanning electron microscopes (LVSEMs). The dynamic means of correcting for chromatic aberration also allows for the possibility of obtaining high image resolution from electron guns that have relatively large energy spreads.

Observation of the freeze-drying process of biological materials with a scanning electron microscope.

Over the past few decades, numerous studies have been done on the freeze-drying of biological materials from a physical, chemical and biological point of view. Morphological observation of the freeze-drying process of specimens, however, has been tried by only a few investigators. In those studies, thin-layered aqueous specimens, which were sandwiched between two cover slips, were mostly observed with an optical microscope. For ultrastructural and stereoscopic observation, the scanning electron microscope has a great advantage, unlike that of the optical microscope. A specially designed cryo-scanning electron microscope, employed in the present study, made it possible to observe the freezing patterns of the specimens and also the sublimation process of ice in frozen specimens under vacuum. With this specially designed microscope, shrinkage of some specimens due to dehydration during the freeze-drying process was revealed and the extent of such shrinkage was quantitatively determined.

Ochronotic arthropathy. A comparative scanning electron microscopic and light microscopic study of the synovium in ochronosis.

The synovium in two cases of ochronosis was studied by scanning electron microscopy and light microscopy. Several features not yet described were seen by scanning electron microscopic examination and correlated with the light microscopic findings. This is the first report in the English language literature comparing scanning electron microscopic findings with the light microscopic findings in ochronosis.

Structural changes in glutaraldehyde-treated porcine heterografts used as substitute cardiac valves. Transmission and scanning electron microscopic observations in 12 patients.

Scanning and transmission electron microscopic studies were made of (1) 12 glutaraldehyde-treated porcine valvular heterografts that had been implanted in patients for 2 days to 76 months; (2) 3 unimplanted commercially processed porcine aortic valves; and (3) 1 unprocessed porcine aortic valve. Comparison of unprocessed porcine valves and unimplanted commercially processed valves showed loss of endothelium and acid mucopolysaccharides during preimplantation processing. Short-term (less than 2 months) changes after implantation consisted of insudation of plasma proteins, penetration of erythrocytes into surface crevices, formation of a thin surface layer of fibrin, and deposition of macrophages, giant cells and a few platelets. Longer-term (more than 2 months) changes were proportional to the time interval after implantation and consisted of progressive disruption of collagen, erosion of the valve surfaces, formation of aggregates of platelets and accumulation of lipid. The surfaces of the leaflets did not become covered with endothelium or with a fibrous sheath. Calcific deposits were found in one valve and bacterial organisms in another. Thus, progressive breakdown of collagen appears to be a critical factor in determining the long-term durability of glutaraldehyde-treated porcine valvular heterografts.

Thrombus formation on the balloon of heparin-bonded pulmonary artery catheters: an ultrastructural scanning electron microscope study.

OBJECTIVE: To investigate heparin-bonded pulmonary artery catheters with respect to thrombus formation and platelet aggregation at the balloon and the shaft using a scanning electron microscope in critically ill patients. DESIGN: Prospective study. SETTINGS: Critical care unit and research laboratories. PATIENTS: Pulmonary artery catheters were inserted in critically ill patients (n = 10). INTERVENTIONS: Pulmonary artery catheters were removed after 24, 48, 72, or 120 hrs, and the ultrastructure was investigated in specialized research laboratories. MEASUREMENTS AND MAIN RESULTS: Balloon and shaft were investigated using a scanning electron microscopic technique. Area of thrombus formation was quantified using image analysis. Heparin release of the catheters was measured. The frequency of balloon inflations was investigated in in vitro experiments by inflating catheters different times (0, 10, 20, and 30 times). Twenty-four hours after catheter insertion, scanning electron microscopic images showed thrombus formation and platelet aggregation at the site of the balloon. Seventy-two hours after catheter insertion, a thrombus started to detach. The areas of thrombus formation did not differ, but thrombus organization changed dramatically 72 and 120 hrs after catheter insertion. The shaft was colonized by single cells only. Cracks of the balloon could be observed after 72 hrs, whereas no cracks could be found in in vitro controls. In vitro, heparin release of the pulmonary artery catheters decreased significantly after 24 hrs. CONCLUSIONS: Scanning electron microscopic images of heparin-bonded pulmonary artery catheters demonstrate thrombus formation on the balloon 24 hrs after pulmonary artery catheter insertion, increasing dramatically at 72 and 120 hrs. The shaft was colonized by single cells only. The thrombus size is not significantly different during the observation time, but the grade and quality of thrombus formation differ.

Choroidal melanoma with retinal perforation and vitreous hemorrhage: a scanning electron microscopic study.

Extensive retinal perforation and subsequent growth within the vitreous cavity permitted the scanning electron microscopic examination of a primary, mixed cell type malignant melanoma of the choroid. The surface of the tumor was covered by fusiform cells oriented in a longitudinal, parallel fashion. Plumper globoid, presumed epitheloid forms were interspersed among the spindle cells. In some areas, the spindle cells had long tapering processes. At the margin of the tumor, slender fascicles of parallelly oriented, proliferating spindle cells were identified. This fascicular growth pattern has been observed in previous tissue culture and scanning electron microscopic studies. Vitreous hemorrhage which occurred in our case may herald a malignant melanoma that has perforated the retina. Additionally, retinal perforation may signify an aggressively growing tumor which should be treated.

[Ixodes ricinus under the scanning electron microscope].

The tick Ixodes ricinus was examined with a scanning electron microscope. The morphology of the mouth parts explains the well known tight attachment of the head of the tick in the skin. Diseases following tick bites and the possibilities of removing the ticks are discussed.

Varied light and scanning electron microscopic appearances of barium sulphate in smears and histological sections.

The various light and scanning electron microscopic appearances of barium sulphate in smears and in histopathological lesions in sections are illustrated. One type of barium sulphate (E-Z-HD) includes much larger, bright birefringent particles and has very different appearances from other widely used types. Such larger particles in sections were not originally suspected to be barium sulphate and were identified only by energy dispersive x ray analysis in a scanning electron microscope. This form of barium sulphate is used for double contrast upper gastrointestinal radiography, and is not thought to have been responsible for the lesions in which it has been observed by us.

Shaping and cleaning the root canal system: a scanning electron microscopic evaluation of a new instrumentation and irrigation technique.

The purpose of the present scanning electron microscopic study was to investigate the efficacy of a combination of EDTA, NaOCl, and surface-active irrigating solutions during and after root canal preparation with ProFile nickel-titanium rotary instruments. Thirty canals were divided into three groups, instrumented and irrigated as follows: 5% NaOCl and a final flush with 17% EDTA were used for group A; group B specimens were irrigated using 17% EDTA, followed 15 s later by 1% TRITON X-100 (tensioactive agent) and then by 5% NaOCl; and group C specimens were irrigated with the same combination, but once shaping procedures were completed the irrigating sequence was repeated three times. After scanning electron microscopic evaluation, group C specimens exhibited the most effective debridement of the root canals. Results showed that tensioactive agent contributed to enhanced debridement. Cleaning was significantly improved once shaping procedures were completed.

Scanning electron microscopic findings in interstitial cystitis.

Scanning electron microscopy (SEM) was performed on transurethral resection biopsies from 13 patients with classic interstitial cystitis. Biopsies from 9 patients with stress incontinence served as controls. The SEM appearance of the bladder surface in interstitial cystitis varied considerably, exhibiting small round and large polygonal cells. The proportion of cells displaying round uniform and pleomorphic microvilli was high and sometimes dominated the area examined. SEM characteristics earlier assigned to bladder tumours were detected in patients with interstitial cystitis and, at a lower frequency, also in control patients. The mucin layer covering the urothelial cells seemed reduced in interstitial cystitis compared with control specimens. Surface characteristics specific for interstitial cystitis were, however, not detected by SEM.

The insulo-acinar portal and insulo-venous drainage systems in the pancreas of the mouse, dog, monkey and certain other animals: a scanning electron microscopic study of corrosion casts.

Scanning electron microscopy of vascular casts of the pancreas from monkies, cattle, pigs, dogs, cats, rabbits, guinea pigs and mice showed certain species differences in the occurrence of intralobular and interlobular islets and in the microcirculatory pattern of these islets. Interlobularly located islets were frequently found in the mouse and guinea pig, as has been previously established in the rat (Murakami and Fujita, 1992); they emitted insulo-venous efferent vessels directly draining into veins. In contrast, the intralobular islets in the guinea pig usually issued insulo-acinar portal vessels continuous with the lobular capillary network. In the mouse, they usually emitted both the insulo-acinar portal and insulo-venous efferent vessels. The insulo-venous efferent vessels, including those of the interlobular islets, could partly be portal in nature since they occasionally issued portal branches directed to the lobular capillary network. In rabbits, cats, dogs, pigs, cattle and monkies, as in men (Murakami et al., 1992), essentially all islets in the pancreas were intralobular in location and usually emitted the portal vessels only. In the mouse and rabbit, as in the rat (Murakami and Fujita, 1992), the islet received afferent vessels in its superficial aspect and issued efferent vessels from its deep aspect. In the Formosan monkey, as previously reported in the rhesus monkey (Fujita and murakami, 1973), the afferent vessels usually ran deep into the islet which emitted vessels from its superficial aspect. In other animals examined in this study, as in humans (Murakami et al., 1992), no consistent rule concerning the microcirculatory pattern within the islet could be determined.

Patterns of tissue damage produced by repeated use of a corneal trephine: a scanning electron microscopic study.

We studied by scanning electron microscopy the effects of the repeated use of a newly-designed, disposable, suction trephine (Microsurgical, Ltd, Bedfordshire, England) on the nature of the cut of successive corneal buttons from sheep eyes. After the trephine was used more than nine times, the cut edge became more irregular as successive buttons were produced. Also, progressively greater degrees of endothelial cells adjacent to the cut edge were lost. Although the corneal buttons initially were circular, those produced after the 20th cut became more oval. These results suggest that a disposable corneal trephine should be reused no more than nine times.

Scanning electron microscopic study of prostatic cancer.

Scanning electron microscopy is applied to the study of 3-dimensional surface and organization of a normal prostate gland and of samples of prostatic disease, including five cases of benign hyperplasia and 15 cases of prostatic adenocarcinoma of different degrees of differentiation. The study provided further evidence regarding the nature and origin of prostatic cancer at a cytologic level and indicated alternate pathways to the neoplastic spread within the prostate gland.

[Structural changes in the endothelium of the major arteries under conditions of longitudinal stretching in vivo (based on scanning electron microscopic data)].

The method of scanning electron microscopy was used for studying three-dimensional organization of the endothelium of major vessels under conditions of distraction in vivo. The distraction resulted in an enlargement of the spaces between endotheliocytes without formation of considerable areas of deendothelialized surface. The main mechanism of induction of flattening and division of endotheliocytes in the zone of distraction seems to be the elimination of the contact inhibition resulting from a mechanical distraction.
