[Sporulation in Bacillus licheniformis strains that do not synthesize bacitracin and serine exo- and endoproteases].

Sporulation and synthesis of bacitracin, a peptide antibiotic and serine exoproteases and endoproteases were studied in 6 strains of Bacillus licheniformis at various periods of cultivation in sporulation media. Thermostable spores in all the strains were shown to form in the absence of both the exogenic and endogenic antibiotic and proteases. It was suggested that bacitracin and the serine proteases were not specific regulators of sporogenesis in B. licheniformis and their biological function under the natural conditions included, respectively, the antibiotic action and hydrolysis of proteins and peptides of the environment and cells.

[Changes in the level of bacitracin synthesis in autolysin-resistant strains of Bacillus licheniformis].

Production of bacitracin by Bac. licheniformis 1001 and its spontaneous autolysin resistant variants was studied. It was found that the antibiotic activity of some variants was 1.5--2 times higher than that of the initial strain. No differences in the activity of serine exoprotease in the initial strain and resistant variants were observed. The latter variants lost their resistance to autolysis in 2--3 subcultures on solid and liquid nutrient media. their antibiotic activity in these cases decreased to the control level. The study indicates that there is a phenomenologic relation between the autolytic and antibiotic activities of Bac. licheniformis. The nature of the relation is not known yet. Possibly, it is due to changes in the specific metabolic steps connected with regulation of bacitracin synthesis.

[Sporulation in Bacillus licheniformis during altered bacitracin synthesis].

Sporulation in different strains of Bacillus licheniformis, 10716 and 1001 in connection with changes in synthesis of bacitracin was studied. It was shown that the sporulation efficiency did not depend on the synthesis of the antibiotic: in some strains with low potency for the antibiotic production, the sporulation level was lowered, while in the others, it was not lowered. Moreover, normal sporulation was also observed, when the synthesis of bacitracin was inhibited. Therefore, it is suggested that there is no correlation between the sporulation and antibiotic production.

Inhibition by concanavalin A, dansylcadaverine and bacitracin of muscarinic receptor-mediated desensitization in guinea-pig Ileal smooth muscle.

The muscarinic agonist, cis-2-methyl-4-dimethylaminomethyl-1,3-dioxolane methiodide (CD), and histamine desensitize the responses of guinea-pig ileal longitudinal smooth muscle to subsequently administered CD or histamine, respectively. Desensitization by CD, but not by histamine, was inhibited by concanavalin A (Con A) and this protection was prevented by co-administration of succinyl-Con A or by cytochalasin B or colchicine at concentrations which alone did not affect desensitization. Dansylcadaverine and bacitracin, inhibitors of transglutaminase, both protected against CD desensitization but were without effect on desensitization mediated by histamine. Benzilylcholine mustard, which reduced muscarinic receptor density, significantly reduced CD-induced desensitization. The effects of Con A and the transglutaminase inhibitors suggest that agonist-induced changes in muscarinic receptor distribution or receptor-effector coupling, via cytoskeletal and transglutaminase-sensitive mechanisms, are a component of CD-induced desensitization. These processes do not appear to be involved in histamine receptor-mediated desensitization. Although desensitization by CD or histamine is nonspecific in this preparation, both agonists cross-desensitizing, the effects of Con A, dansylcadaverine and bacitracin clearly indicate differences in the desensitization process elicited by these two stimulants.

[Ureolytic activity in the intestinal tract of layers, effects of bacitracin and flavomycin (author's transl)].

Counting of bacteria, detection of urease-positive bacteria and estimation of bacterial ureolysis influenced by Bacitracin and Flavomycin was done on intestinal contents from different segments of the digestive tract of laying hens. It was concluded that: - the intestinal flora was numerically most important in the caecum and was composed principally of lactobacilli in the crop, gizzard, duodenum, jejunum and ileum, and of lactobacilli and anaerobes in the caecum and colon. - 17% of the total number of intestinal bacteria were urease-positive. - the ureolytic activity was most important in the caecum: 24 mg of urea hydrolysed/g of contents during 4 hours of incubation. The range is between 2 and 4 mg in the other segments. - until the level of 500 ppm, Bacitracin and Flavomycin did not influence the in vitro ureolytic activity of fecal and cecal material.

[Transfer of vitamin A to the egg influenced by bacitracin or flavomycin (author's transl)].

During two experiments hens are fed with diet supplemented with or without 10 ppm of Bacitracin or 0,5 ppm of Flavomycin. Both increase retinol transfer from diet to yolk. Differences are not always significant. They are concerning almost a more important percentage of eggs whose yolks contain between 500 and more than 700 IU of vitamin A (66,6 p. 100 for Bacitracin supplement; 80,9 p. 100 for Flavomycin supplement and 45 p. 100 for control). Each additive increases hepatic storage and serum level of retinol but not significantly. Transfer of vitamin A from diet to yolk is between 12,1 p. 100 and 24,0 p. 100 regarding individual variations. There is no influence from the strain. Warren or Dekalb, used.

Turbidimetric and diffusion assay of bacitracin in feeds.

A manual turbidimetric method for determining zinc or methylene disalicylate bacitracin in feeds was developed. When zinc ions (10-4M) were added to the medium and the phosphate ion concentration in standard solutions was decreased to 1%, the median response of Streptococcus faecalis was about 0.038 unit. Feeds analyzed by the turbidimetric assay should be prewashed with petroleum ether and extracted with pyridine as in 42.204. Zinc bacitracin standards added to swine and broiler rations were recovered at 93.1-102.6% by the turbidimetric method. Excellent agreement between the manual turbidimetric method and the plate assay was also obtained for finished feeds containing the zinc or methylene disalicylate salt of the antibiotic. The turbidimetric method appears to have high accuracy and precision. It is more rapid and less costly than the plate assay.

[Purification of proteinases on bacitracin-modified silochromes with differing concentration of ligands and fragment lengths].

The binding of subtilisin, bovine pancreatic trypsin, and human blood thrombin to bacitracin-modified silochromes differing in the ligand concentrations and arm lengths were studied. The optimal conditions for trypsin isolation and purification were found. Methods for isolation and purification of subtilisin and thrombin were developed that provided the extents of enzyme purification of 20-80. The specific activities of thrombin trypsin, and subtilisin were 1200-2625 NIH units per mg protein and 7-18 and 19-34 units per mg protein, respectively. The possibility of isolation and purification of trypsin, thrombin, and subtilisin on bacitracin-modified silochromes was demonstrated. The sorbents studied were suggested for industrial application.

Improved microbiological procedure for determining bacitracin in premixes and mixed feeds.

An improved microbiological procedure is presented for determining bacitracin in premixes and mixed feeds. Premixes are extracted with acidic 50% aqueous methanol, and mixed feeds are extracted with 50% dimethylformamide after washing with acetone to remove fats and pigments. Extracts are diluted with pH 6.0 phosphate buffer and assayed by using a 2-point assay system with Micrococcus flavus as the test organism. The simpler extraction coupled with the 2-point assay system measured bacitracin levels as low as 10 g/ton. Accuracy and repeatability of the 2-point system were at least equivalent to those for official AOAC methods.

The selective inhibition of beta 1 and beta 7 integrin-mediated lymphocyte adhesion by bacitracin.

Integrins play an important role in lymphocyte adhesion to cellular and extracellular components of their microenvironment. The regulation of such adhesion often involves changes in the functional state of the integrins rather than alterations in their expression levels. Although the functional basis for such transitions is unknown, a possible role for disulfide exchange might be postulated based on the observations that integrin function can be activated by bifunctional reducing agents or by Abs that react with areas adjacent to predicted long-range disulfide bonds in integrins. Recently, it has been reported that enzymes that catalyze disulfide exchanges such as protein disulfide isomerase (PDI) are present on the surface of lymphoid cells, raising the possibility that such enzymes might be involved in the control of lymphocyte adhesion. A number of inhibitors of PDI function were examined for their effects on integrin-mediated adherence of T cells. The results did not support role for PDI in the regulation of integrin function, as the inhibitors somatostatin A, tocinoic acid, dithiobisnitrobenzoic acid, and anti-PDI mAb did not interfere with adherence. However, one of the PDI inhibitors, bacitracin, selectively interfered with the beta1 integrin-mediated adherence of lymphoid cells to collagen, fibronectin, laminin, and VCAM-1, and with alpha4beta7-dependent adherence to fibronectin and to VCAM-1. In contrast, alpha(v)beta3- and alpha(L)beta2-mediated adherence were not inhibited. Thus, it appears that bacitracin may be a selective inhibitor of beta1 and beta7 integrin functions by an as yet unknown mechanism.

One-step affinity purification of Bacillus neutral proteases using bacitracin-silica.

A purification procedure for neutral proteases from bacilli is described, in which bacitracin-silica was used as affinity medium. This enabled a one-step purification of the proteases directly from culture supernatant. Since neutral proteases are extremely sensitive towards autodigestion, conditions were chosen such, that autodigestion was largely prevented. Isopropanol appeared to be useful in both eluting the enzymes from the affinity medium, and inhibiting enzymatic activity during this step. The bacitracin-silica medium allowed high flow rates: with columns prepared for use in an FPLC system flow rates up to one column volume per minute were feasible, and still gave satisfactory results. The neutral proteases purified by this method were found to be homogeneous both by SDS-PAGE and analytical gel filtration.

Genetic mapping of the bacitracin synthetase gene(s) in Bacillus licheniformis.

The map position of a mutation in the bacitracin synthetase gene(s) in Bacillus licheniformis ATCC 10716 was determined by transduction with phage SP-15. Results indicate that it is linked to the lys and trp loci and is distinct from the known sporulation loci on the chromosome of Bacillus licheniformis. The defect(s) of the enzyme complex were analysed in terms of its ability to bind covalently 14C-labelled amino acid precursors of the bacitracin molecule.

Micellar electrokinetic capillary chromatography determination of zinc bacitracin and nystatin in animal feed.

An MEKC procedure was developed for the separation of zinc bacitracin (Zn-BC) and nystatin (NYS) in mixtures and in animal feedstuff. The running buffer was 15 mM borate/19 mM phosphate, pH 8.2, containing 20 mM SDS and 10% v/v methanol. Samples were run at 25 degrees C, the applied voltage was 25 kV, and an additional pressure of 5 mbar was applied. Both analytes were detected by UV simultaneously at 215 nm, Zn-BC alone at 192 and 254 nm, and NYS alone at 305 nm. The method was shown to be specific, accurate (recoveries were 100.0 +/- 0.6% and 100.1 +/- 0.6% for Zn-BC and NYS, respectively), linear over the tested range (correlation coefficients 0.9991 and 0.9994), and precise (RSD below 1.3% for both analytes). The method was applied to determine Zn-BC and NYS as additives in animal feed.

Bacitracin production by a new strain of Bacillus subtilis. Extraction, purification, and characterization.

A new strain of Bacillus subtilis C 126 was isolated from sugar cane fermentation and produced an antibiotic that inhibited the growth of Micrococcus flavus. The production of the antibiotic in culture medium followed to extraction with n-butanol, thin layer chromatography, and microbiological tests indicated that a polypeptide antibiotic was produced. The fraction obtained by Sephadex G-25 column and analyzed by HPLC indicated that bacitracin complex was produced.

Determination of hydrocortisone, polymyxin B and Zn-bacitracin in pharmaceutical preparations by micellar electrokinetic chromatography.

A new, simple and accurate micellar electrokinetic chromatography (MEKC) method is established for quantification of hydrocortisone, polymyxin B and Zn-bacitracin in local pharmaceutical preparations. The separation was carried out at 25 degrees C and 25 kV, using a 15 mmol L(-1) phosphate-15 mmol L(-1) borate buffer (pH 8.2), 60 mmol L(-1) sodium dodecylsulfate (SDS), and 10% methanol-water (v/v) as background electrolyte. Under these conditions the analysis takes about 23 min. The method has been applied for quantifying these compounds in two different commercial pharmaceutical products and the method gave good results when compared with a reference spectrophotometric multivariate calibration method.
