[Effect of whole-body therapeutic gamma irradiation of patients on molecular characteristics of blood plasma from NMR data].

The changes in molecular characteristics of patients' blood after a totally therapeutic irradiation were studied by the NMR method. The changes in the intensity and shape of peaks attributed to methylene and methyl protons of fatty acids and lipoproteins were found in the NMR spectra of the blood plasma of the irradiated subjects. The data on the role of peroxide lipid oxidation in changes of lipid concentration in blood plasma were obtained. The changes in the composition of metabolites in the blood and their relative equilibrium level were found.

Substance P in the blood plasma in hypophysial portal vessels.

The aim of the present study was to check if Substance P (SP) is released from the hypothalamus into the hypophysial portal vessels and by this route exerts its direct influence on the adenohypophysis. For this purpose SP radioimmunoactivity was assayed in the blood plasma collected from hypophysial portal vessels and from the cephalic end of the external jugular vein. The SP levels in blood plasma collected from hypophysial portal vessels and from the jugular vein do not differ significantly. Neither does application of a noxious factor, such as bilateral femoral bone fracture, change significantly the SP level in the blood plasma from portal vessels and from the jugular vein. Hypoxia seems to increase the SP level in portal blood plasma and may be followed by its decrease. It is concluded that hypothalamic SP is not released into the hypophysial portal vessels under normal conditions and its direct influence on the adenohypophysis is not mediated this way.

Evaluation of different treatment methods in patients with psoriasis and content of kallikrein and kallikreinogen in blood plasma.

158 patients with different forms of psoriasis were examined for the content of kallikrein and kallikreinogen in blood plasma and treated by different preparations including andecalin and dilminal-D. Changes in the kallikrein-kinin system may play a role in the development of psoriasis. The aim of this study was to investigate the relationship of changes in the kallikrein-kinin system induced by andecalin and dilminal-D and the clinical course of psoriasis. Determination of kallikrein and kallikreinogen content in blood plasma was made according to the methods of Paskhina and Krynskaya. Almost all psoriatic patients showed high kallikrein and kallikreinogen levels in blood plasma. We conclude that changes in the kallikrein-kinin system are involved in the pathogenesis of psoriasis. Andecalin and dilminal-D preparations resulted in a marked amelioration of psoriasis.

[Total antioxidant capacity of blood plasma in healthy men and in men with coronary heart disease].

BACKGROUND: The aim of the study was to determine the level of total antioxidant capacity (TAC) of blood plasma in healthy men and in men with coronary heart disease differentiated by their level of physical activity. MATERIAL AND METHODS: The study was carried out in the two groups of age-adjusted men. Group I comprised 100 healthy men aged 56.6 +/- 8.5 years and group II involved 100 men with identified coronary heart disease aged 56.6 +/- 7.2 years. BMI was determined in every participant. Level of energy expediture was determined by calculating the amount of hours per week spent for different recreational sports activities. Spectrophotometric method FRAP (Ferric Reducing Ability of Plasma) was applied to estimate total antioxidant capacity of blood plasma. RESULTS: Values of absorbance describing level of total antioxidant capacity of blood plasma in 1', 3' and 10' minut of reaction time window differed between groups. Men with coronary heart disease were characterised by higher antioxidant defense system than healthy ones. Energy expediture expressed in kcal per week was statistically higher in healthy subjects. In healthy men positive correlation was found between level of total antioxidant capacity (in 1', 3' and 10' minut of reaction time window) and BMI (p< or = 0.05). Statistically significant negative correlation was found for TAC and level of energy expediture. Among men with coronary heart disease there was statistically significant positive correlation between the values of total antioxidant capacity (in 1', 3' and 10' minut of reaction time window) and BMI. There was no statistically significant correlation neither between age and TAC, nor between energy expediture and TAC. CONCLUSIONS: The level of total antioxidant capacity estimated with FRAP method was higher in men with coronary heart disease in comparison with healthy men. Antioxidant potential was higher along with the increase of BMI in both groups. Increased energy expenditure was related to decreased total antioxidant capacity, but only in healthy men.

Rapid HPLC method for measurement of vitamin D3 and 25(OH)D3 in blood plasma.

The present work describes a novel, simplified high-performance liquid chromatography (HPLC) method for evaluation of vitamin D3 and its 25-hydroxy metabolite in blood plasma. The retrieval of the analytes from the blood plasma matrix is based on a single-step extraction using acetonitrile. The method is specific, sensitive, and ensures good reproducibility. The recovery of the analytes, precision, and reproducibility obtained using the present approach gave results comparable to or better than more complex, laborious, and time-consuming procedures. This method is suitable for evaluation of the host's vitamin D physiological status, as well as for rapid analysis of blood plasma samples in suspected cholecalciferol toxicity. With a significantly shortened time of analysis (10 minutes), the present method allows the possibility for processing of a large number of samples rapidly, efficiently, and at a low cost.

[Quantitative changes in glucocorticoid binding capacity of blood plasma, liver and small intestine mucosa following consumption of various dietary proteins].

Studies on corticosteroid binding proteins in blood plasma, cytoplasm, and nuclei of cells from liver and mucosa of the small intestine of growing rats were carried out under the influence of different protein diets. Increasing protein quality induced higher growth rates and an increase of the glucocorticoid binding capacity as well as the concentration of total and free corticosterone in blood plasma and increasing contents of specific corticosteroid binding receptors in the cytoplasm and nuclei of cells from liver and mucosa of the small intestine. In the cytoplasm the bound fraction takes only 5 to 10% of the total hormone concentration, compared to 80 to 90% in blood plasma. These findings suggest that the changes in the concentration of specific hormone binding proteins may be a general principle of the effect of dietary protein in the regulation of metabolism.

Ultrastructure of circulating immune complexes isolated from the blood plasma of patients suffering from infectious diseases.

An original procedure which permits to isolate circulating immune complexes (CIC) from the blood plasma in a form of a dense pellet was developed. This procedure was applied for the ultrastructural analysis of CIC isolated from blood of healthy blood donors and patients suffering from Yersinia enterocolitica (YE) and Yersinia pseudotuberculosis (YP) infections. The here reported method of CIC isolation from blood plasma permitted to visualize CIC electronmicroscopically as amorphous masses of low, middle, and high electron density with inclusions of cell debris. In contrast to CIC of healthy blood donors, CIC of infected patients contained various bacteria and fungiformic structures. For the first time, this method made possible an ultrastructural demonstration of bacterial destruction outside of phagocytes in vivo. This method also permits to visualize and identify bacteria in cases of lingering forms of infection when hemoculture tests fail. Therefore, electronmicroscopic examination of CIC preparations from the blood plasma might be a very informative indicator of bacteriemia in the course of an infection process and serve as an indicator of therapeutic effects. In lingering forms of an infection process, ultrastructural analysis of CIC preparations can be of prognostic value and serve as an indicator of therapeutic effects. This method might be also advantageous as an additional test for the exposure of latent bacterial persistence in diagnostically complicated cases.

Variations in concentration of fluoride in blood plasma of pregnant women and their possible consequences for amelogenesis in a fetus.

The aim of the study was to assess the fluoride exposure of pregnant women living in Poznan (Poland) by examination of fluoride levels in blood plasma. The subjects of the study were 31 pregnant women aged 22-34 years in the course of regular pregnancy. Data concerning the sources of fluoride exposure such as diet, oral hygiene measures and topical application of fluoride procedures, were collected from each individual with a questionnaire. Samples of blood plasma were drawn in the 28th, 33rd week of pregnancy and during delivery. The analysis evaluating the fluoride concentration in the samples was carried out with the use of fluoride electrode ORION (model 96-09). The data were statistically analyzed using the program Statistica for Windows. Mean value of fluoride concentration in the samples of blood plasma from the 28th week of pregnancy was lower than the mean concentration detected in the 33rd week of pregnancy (3.29 and 3.73mumol/l, respectively). These values suggest that apart from drinking water, there were other important sources of fluoride in the examined sample. The results indicate that a reliable assessment of fluoride exposure in a given population cannot be based solely on the concentration of fluoride in drinking water. Relatively high levels of fluoride in blood plasma of examined women suggest that there is no need for fluoride supplementation in this group of patients.

[The effect of the use of milk-based feed mixtures and whole milk on the development of selected indicators in the blood plasma in calves].

Two milk feeding systems were investigated as influencing the health and development of calves. After the termination of colostrum feeding, the ten animals of the experimental group were given whole milk whereas the control group (also ten calves) was given the Laktosan milk replacer. By the age of three months, blood was collected from the calves for biochemical examination in weekly intervals, later once a month. The content of urea, determined in the blood plasma of the calves of the experimental group was significantly lower in the fourth to seventh week. The plasma levels of nitrogen, potassium, calcium and magnesium were about the same in the experimental and control groups, being within the limits of the reference values. At the age of six to nine weeks, the content of inorganic phosphorus in the blood plasma of the tested animals was statistically significantly higher. Vitamin A concentration in the blood plasma was about the same in both groups. The content of vitamin E in the blood plasma of the calves of the experimental group was statistically significantly higher in the fourth to eight week of age. No significant differences between the two groups were observed in the plasmatic activities of aspartate aminotransferase (AST) and gamma-glutamyl transferase (GMT). The activities of alkaline phosphatase (ALP) were significantly higher in the third to fifth week of life. From the fifth to eighteenth week of age, the average daily weight gains were significantly higher in the calves given whole milk.

Three-year study on trace mineral concentration in the blood plasma of Shetland pony mares.

Changes in some trace minerals concentrations (calcium, inorganic phosphorus, magnesium, zinc, iron, copper, sodium and potassium) in blood plasma were investigated during a three-year period in Shetland pony mares. Blood plasma mineral concentrations were determined by the atomic absorption method and colorimetric method. The three-year averages were in micrograms/ml; Na 4630 +/- 168; K 277 +/- 3.8; Ca 171 +/- 3.8; P 31.5 +/- 0.74; Fe 1.92 +/- 0.14; Zn 1.07 +/- 0.04 and Cu 1.06 +/- 0.02. Two trace minerals (magnesium in inorganic phosphorus) showed only long-term tendency changes--upward or downward trends were not connected with the season. Most of the investigated minerals showed both a long-term change tendency and more or less marked seasonally related concentration changes, being higher during autumn or winter (zinc, sodium), or during spring and summer (calcium, copper, iron and potassium). Some interrelationships between particular trace mineral concentrations in blood plasma were also observed.

Proton nuclear magnetic resonance lineshape studies on human blood plasma lipids from newborn infants, healthy adults, and adults with tumors.

The usefulness of proton NMR spectroscopy of human blood plasma for cancer research has been extensively studied in recent years. Two main starting points have been offered by Fossel et al. (N. Engl. J. Med. 315, 1369 (1986)) and Mountford et al. (FEBS Lett. 203, 164 (1986)). In this work the experimental proton NMR spectra of blood plasma were analyzed with the aid of the multivariate lineshape fitting method. An appropriate model structure, in terms of the various lipoprotein (VLDL, LDL, and HDL) signals, for the methylene region was used. Neonates, healthy adults, and adults with nonmalignant and malignant tumors were studied. The linewidth of the methylene region was found to be linearly dependent on the relative concentrations of the lipoproteins. The correlation coefficient was -0.89 (P less than 0.001) for VLDL and 0.88 (P less than 0.001) for HDL. A correlation between VLDL concentration and age, 0.76 (P less than 0.001), was also established. VLDL was modeled using two components. The half-linewidth of the lower field component was slightly elevated for the adults with large metastases. This might be in association with the fucose-containing proteolipid complex detected earlier in cancer cells or in sera of cancer patients. Some signals of this complex may fall in the same region of the spectra. The spectra for the neonates were indicated to be totally different from the adults. This and other related questions were explained by means of the model parameters and the relative concentrations of the lipoproteins VLDL, LDL, and HDL. The presented technique can be used as a rapid research tool for figuring out the relative concentrations of the lipoproteins in blood plasma and explaining the reasons behind the changes in the spectra.

The content of unbound polyamines in blood plasma and leukocytes of patients with polycythemia vera.

In the blood plasma and isolated leukocytes of 11 patients with polycythemia vera and 3 healthy subjects, the polyamines putrescine, spermidine and spermine were determined. The average values in the leukocytes of the healthy volunteers were found to be 1.8 +/- 1.4 nmol putrescine/10(8) cells, 3.0 +/- 0.9 nmol spermidine/10(8) cells and 12.9 +/- 3.8 nmol spermine/10(8) cells. In the plasma of healthy persons the amounts of the polyamines were below the sensitivity level of the method employed. In 4 patients with polycythemia vera no polyamines were detected. In contrast, in 7 cases 0.1 to 3 nmol polyamines/ml were found. The level of polyamines in the leukocytes of 6 of these patients was decreased and in one patient corresponded to the values found in the normal range (17.7 +/- 6.0 nmol polyamines/10(8) cells). Continued blood-letting therapy on 3 patients led to values approaching the concentrations found in normal subjects in both blood plasma and leukocytes. A decreased amount of these diamines in the leukocytes of the patients was seen to correlate with an elevated concentration in the plasma.

Determination of copper and zinc in blood plasma by ion chromatography using a cobalt internal standard.

Ion chromatography was used to detect levels of copper and zinc in blood plasma from renal dialysis patients on continuous ambulatory peritoneal dialysis (CAPD) and haemodialysis (HD). The developed method used cobalt as an internal standard and when combined with the standard additions method improved the overall precision of the results by between -0.3 and 6.0% and -0.8 and 5.7% for copper and zinc, respectively. The method was compared with inductively coupled plasma optical emission spectrometry (ICP-OES) and the results indicated no significant difference between the two methods with or without an internal standard. Without an internal standard, tcalc was 0.869 with a tcrit of 2.201 (n = 12, P = 0.05) and with an internal standard, tcalc was 0.189 compared to a tcrit of 2.201 (n = 12, P = 0.05). The copper and zinc levels in blood plasma in both dialysis groups were not significantly different to the copper and zinc levels in blood plasma of the control patients. A convenient method of analysis of trace elements in blood such as ion chromatography with UV/VIS detection is useful for determining whether inorganic elements may be disrupted in the body due to changes in the state of health.

[Thrombomodulin, von Willebrand factor and tissue plasminogen activator in the blood plasma of obese women and men].

Disturbances of lipids metabolism described in obese persons are important factor damaging vascular endothelium. Known markers of endothelium impairment are: von Willebrand factor (vWf), tissue plasminogen activator (t-PA:Ag) and thrombomodulin (TM). The aim of the work was to evaluate markers of the endothelial disturbance in the blood plasma of persons with obesity. The study was performed in the group of 50 obese persons (39 W, 11 M) aged 35-65 (means 48.8) years with abdominal obesity. The control group consisted of 30 healthy volunteers aged 25-56 (means 41.0) years. In the poor platelet plasma obtained from venous citric blood concentrations of TM, von Willebrand factor antigen (vWf:Ag) and tissue plasminogen activator antigen (t-PA:Ag) were determined using immunoenzyme-linked assay (ELISA). In the obese persons significantly higher concentration of vWf:Ag and t-PA:Ag in comparison to control group. Analysis of results obtained according sex showed that in the blood plasma of obese women TM concentration was significantly higher than in healthy women. Our study proved that in the blood plasma of obese men there are evidences of impairment of endothelial function as higher concentration of vWf:Ag and t-PA:Ag, but in the group of obese women as the increased TM concentration.

Amine oxidase enzymes of sheep blood vessels and blood plasma: a comparison of their properties.

1. A comparison between the biochemical properties of semicarbazide-sensitive amine oxidase (SSAO) activities has been made in sheep blood plasma and arterial wall. 2. The metabolism of benzylamine (BZ) by blood plasma was resolved into high affinity (Km 2.76 +/- 0.24 microM) and low affinity (Km 743 +/- 49 microM) activities. Spermidine metabolism was by a single component (Km 174 +/- 22 microM) and this amine reduced the metabolism of high concentrations of BZ. 3. A single component metabolised BZ in arterial homogenates (Km 11.3 +/- 1.3 microM) and which metabolised spermidine at a very slow rate. 4. Dopamine was deaminated by plasma SSAO and competed with both low and high concentrations of BZ. Dopamine also interfered with arterial metabolism of BZ. 5. These results suggest that there are two SSAO enzymes in sheep blood plasma, with one having similar properties to SSAO in the arterial wall.

[The development of transaminase activity (SGOT and SGPT), lactate dehydrogenase and alkaline phosphatase in the blood plasma of calves up to the age of 4 months].

A trial was performed in 204 healthy calves (heifers) of the Bohemian Spotted breed in the post-natal period from birth to the age of four months. The activities of the following enzymes in blood plasma were determined: L-aspartate: 2-oxoglutarate: aminotransferase, EC.2.6.1.1. (GOT), L-alanine: oxoglutarate: aminotransferase, EC.2.6.1.2. (GPT), L-lactate: NAD oxidoreductase, EC.1.1.1.27 (LDH), and orthophosphoric acid monoester phosphohydrolase, EC.3.1.3.1. (alkaline phosphatase). The calves were divided into age categories according to the date of birth with an interval of one week. GOT activity in blood plasma increased significantly until the age of eight weeks (from the original value of 1.1708 +/- 0.2598 micronmol ml-1 to 1.8150 +/- 0.6362 micronmol ml-1, with the maximum of 2.0317 +/- 0.7777 micronmol ml-1 of plasma in the sixth week). In the subsequent period the GOT curve has not a characteristic course. While the activity of GOT increased in the first weeks after birth, the activity of GPT showed a significant drop (from the original level of 0.9000 +/- 0.3364 micronmol ml-1 to the minimum of 0.3675 +/- 0.1901 micronmol ml-1 of plasma in the seventh week); from the 10th week on the values rise so that at the end of the period of study they reach almost the same levels as in calves in the first postnatal week. The activity of LDH in blood plasma remains at almost the same level in the first five weeks after birth (between 43.4025 +/- 8.4893 micronmol ml-1 and 46.3792 +/- 14.8952 micronmol ml-1 of plasma); it was at a statistically significantly higher level only in a short period between the 7th and 10th week after birth. The highest values of alkaline phosphatase in blood plasma were recorded at the age of two or three weeks (maximum in the second week 23.9833 +/- 9.0945 micronmol ml-1 of plasma); from the fourth week on, the values of alkaline phosphatase are significantly lower until the end of the test period, ranging betweek 5.3133 +/- 1.6017 micronmol ml-1 and 7.5425 +/- 2.2437 micronmol ml-1 of plasma. Changes conditioned by postnatal development were observed in the development of all the enzymatic activities under study, the greatest changes being observed in alkaline phosphatase.
