Parathyroid hormone secretion and serum calcium concentration--a deterministic view of its regulation.

In seven normal men we investigated the basal secretion of PTH by blood sampling in 30 sec intervals for estimation of intact PTH and at 2-min intervals for measurement of ionized calcium. In 4 of these subjects we also investigated the PTH-secretion under conditions of intermittent hypercalcemia and hypocalcemia. Our measurements demonstrate the existence of three time scales in the secretion of PTH, viz. short-term pulses (faster than 2 min), an intermediate pattern of adjusting PTH levels to changed Ca2+ averages, and finally a long-term coupling between PTH and Ca2+ averages after 20 min. Ionized calcium controls the long-term regulation and intermediate adaptation mechanisms, but the short-term fluctuations seem to be due to spontaneous secretion.

A breakthrough in neuroscience needs a "Nebulous Cartesian System" Oscillations, quantum dynamics and chaos in the brain and vegetative system.

The Cartesian System is a fundamental conceptual and analytical framework related and interwoven with the concept and applications of Newtonian Dynamics. In order to analyze quantum processes physicist moved to a Probabilistic Cartesian System in which the causality principle became a probabilistic one. This means the trajectories of particles (obeying quantum rules) can be described only with the concept of cloudy wave packets. The approach to the brain-body-mind problem requires more than the prerequisite of modern physics and quantum dynamics. In the analysis of the brain-body-mind construct we have to include uncertain causalities and consequently multiple uncertain causalities. These multiple causalities originate from (1) nonlinear properties of the vegetative system (e.g. irregularities in biochemical transmitters, cardiac output, turbulences in the vascular system, respiratory apnea, nonlinear oscillatory interactions in peristalsis); (2) nonlinear behavior of the neuronal electricity (e.g. chaotic behavior measured by EEG), (3) genetic modulations, and (4) additional to these physiological entities nonlinear properties of physical processes in the body. The brain shows deterministic chaos with a correlation dimension of approx. D(2)=6, the smooth muscles approx. D(2)=3. According to these facts we propose a hyper-probabilistic approach or a hyper-probabilistic Cartesian System to describe and analyze the processes in the brain-body-mind system. If we add aspects as our sentiments, emotions and creativity to this construct, better said to this already hyper-probabilistic construct, this "New Cartesian System" is more than hyper-probabilistic, it is a nebulous system, we can predict the future only in a nebulous way; however, despite this chain of reasoning we can still provide predictions on brain-body-mind incorporations. We tentatively assume that the processes or mechanisms of the brain-body-mind system can be analyzed and predicted similar to the metaphor of "finding the walking path in a cloudy or foggy day". This is meant by stating "The Nebulous Cartesian System" (NCS). Descartes, at his time undertaking his genius step, did not possess the knowledge of today's physiology and modern physics; we think that the time has come to consider such a New Cartesian System. To deal with this, we propose the utilization of the Heisenberg S-Matrix and a modified version of the Feynman Diagrams which we call "Brain Feynman Diagrams". Another metaphor to consider within the oscillatory approach of the NCS is the "string theory". We also emphasize that fundamental steps should be undertaken in order to create the own dynamical framework of the brain-body-mind incorporation; suggestions or metaphors from physics and mathematics are useful; however, the grammar of the brains intrinsic language must be understood with the help of a new biologically founded, adaptive-probabilistic Cartesian system. This new Cartesian System will undergo mutations and transcend to the philosophy of Henri Bergson in parallel to the Evolution theory of Charles Darwin to open gateways for approaching the brain-body-mind problem.

Analysing an accident related to orphan sources including dose assessment.

This paper discusses an accident, which occurred in one of the radiation application centres in Iran, follow-up investigations as well as lessons learnt. In January 2004 the Regulatory Authority was informed through a university radiation protection officer of an accident regarding orphan sources. Investigations revealed that one Am-Be and three (137)Cs sources in the container were subject to extensive heat due to burning of the container and melting of the paraffin content of the container; consequently, sources were stuck to the side wall of the container, but they were still undamaged and no radioactive leaking had occurred. Further investigations showed that the container had been given to the above mentioned centre a long time before by a foreign well-logging company without notifying the Regulatory Authority. Follow-up measurements and assessments indicated that the collective effective dose due to the accident is unlikely to be more than 21 mSv; consequently, no severe deterministic effect to individuals was expected. The findings showed that the main reasons for the accident were as follows: (1) violation of obligation under radiation protection regulations by the owner of the sources; (2) leaving the sources in an improper storage condition; (3) unauthorised access to the radiation sources at the owner centre; and (4) lack of an integrated national registration system in the Regulatory Authority.

Classification of chronic radiation sickness cases using neural networks and classification trees.

Chronic radiation sickness is a deterministic radiation health effect observed among the Mayak Production Association workers in Russia. In this study, unsupervised neural networks were used to cluster hematological measurements in a subset (n = 88) of the Mayak Production Association population while excluding from the analysis the radiation dose and the historical clinical diagnosis. Clusters of observations that had lower average leukocyte and thrombocyte counts were labeled "affected" and those having higher average blood cell counts were labeled "unaffected." The class (cluster) membership for each individual was used subsequently as a dependent variable in a classification tree model in order to identify significant features of the underlying classification model. After re-classification of cases using this method, the results showed a better data separation between the blood cell counts for affected vs. unaffected groups compared to those based on historical classification, and a greater difference between group means for differential blood counts was observed than for the historical diagnosis. The reclassification of diagnostic groups changed the group mean radiation doses. The geometric means (and 95% CL) of cumulative radiation dose equivalent from external exposures, based on the historical diagnosis, are 0.31 (0.0035, 3.4) vs. 1.7 (0.0007, 18) Sv. After clustering and classification tree analyses, the group geometric means were 0.78 (0.0014, 8.6) vs. 1.5 (0.0007, 17) and 0.82 (0.0013, 9.0) vs. 1.4 (0.0008, 16) Sv, using (respectively) whole blood cell counts or differential counts as the independent variables. The approach presented here is useful as a diagnostic aid for both retrospective analyses and in the event of future radiation accidents.

Finite statistical complexity for sofic systems.

We propose a measure of complexity for symbolic sequences, which is based on conditional probabilities, and captures computational aspects of complexity without the explicit construction of minimal deterministic finite automata (DFA). Moreover, if the sequence is obtained from a dynamical system through a suitable encoding and its equations of motion are known, we show how to estimate the regions of phase space that correspond to computational states with statistically equivalent futures (causal states).

Nutrient flows in agriculture in The Netherlands with special emphasis on pig production.

Annual nitrogen (N), phosphorus (P), and potassium (K) flows in agriculture in The Netherlands were identified and quantified in 1990, with special emphasis on pig production. Also, the effects that various management strategies in pig production have on NPK emission in 1990 were compared using a static deterministic simulation model. Ammonia emission from pig production in 1990 (60.9 Gg N) exceeded the defined target for the year 2000 (12.7 Gg N). Measures that affect volatilization of ammonia directly (i.e., introduction of low-emission stables, manure storage facilities, or manure application techniques) reduced ammonia emission most effectively. These measures, however, should be combined with a reduction in application of artificial N fertilizer to avoid an increase in N losses through leaching, run-off, or denitrification. Targets for ammonia emission in the year 2010 require a reduction in the pig population of 24 to 62%, in addition to implications of measures described in this article. National NPK losses in 1990 through leaching, run-off, or denitrification, predicted at 223.5 kg/ha for N, 32.7 kg/ha for P, and 67 kg/ha for K, exceeded government targets for the year 2010 (185 kg N/ha; 8.7 kg P/ha; norm not set for K). Reducing application of artificial NPK fertilizer reduced national NPK losses most effectively. For P, use of phytase and feeding pigs in accordance with their P requirements is required, in addition to limited use of artificial P fertilizer to meet targets for the year 2010. Hence, from an environmental point of view, pig production in The Netherlands is limited primarily by ammonia emission targets for the year 2010.

On the holistic approach in cellular and cancer biology: nonlinearity, complexity, and quasi-determinism of the dynamic cellular network.

A keystone of the molecular reductionist approach to cellular biology is a specific deductive strategy relating genotype to phenotype-two distinct categories. This relationship is based on the assumption that the intermediary cellular network of actively transcribed genes and their regulatory elements is deterministic (i.e., a link between expression of a gene and a phenotypic trait can always be identified, and evolution of the network in time is predetermined). However, experimental data suggest that the relationship between genotype and phenotype is nonbijective (i.e., a gene can contribute to the emergence of more than just one phenotypic trait or a phenotypic trait can be determined by expression of several genes). This implies nonlinearity (i.e., lack of the proportional relationship between input and the outcome), complexity (i.e. emergence of the hierarchical network of multiple cross-interacting elements that is sensitive to initial conditions, possesses multiple equilibria, organizes spontaneously into different morphological patterns, and is controlled in dispersed rather than centralized manner), and quasi-determinism (i.e., coexistence of deterministic and nondeterministic events) of the network. Nonlinearity within the space of the cellular molecular events underlies the existence of a fractal structure within a number of metabolic processes, and patterns of tissue growth, which is measured experimentally as a fractal dimension. Because of its complexity, the same phenotype can be associated with a number of alternative sequences of cellular events. Moreover, the primary cause initiating phenotypic evolution of cells such as malignant transformation can be favored probabilistically, but not identified unequivocally. Thermodynamic fluctuations of energy rather than gene mutations, the material traits of the fluctuations alter both the molecular and informational structure of the network. Then, the interplay between deterministic chaos, complexity, self-organization, and natural selection drives formation of malignant phenotype. This concept offers a novel perspective for investigation of tumorigenesis without invalidating current molecular findings. The essay integrates the ideas of the sciences of complexity in a biological context.

Markedness and the grammar in lexical diffusion of fricatives.

This paper examines the contributions of markedness and a child's grammar to the process of lexical diffusion in phonological acquisition. Archival data from 19 preschoolers with functional phonological delays were submitted to descriptive analyses of productive sound change in fricatives. Children's presenting fricative inventory, the fricatives newly learned, and their position of occurrence were varied, with word frequency and neighbourhood density measured. Results indicated that lexical diffusion of fricatives occurred differentially by word position. Positional, featural and structural markedness further converged such that change in unmarked structure of any type was implemented in low frequency words. A child's presenting fricative inventory was not directly affiliated with systematic patterns of diffusion. These results have clinical applications for the evaluation of productive sound change and theoretical implications for deterministic models of lexical diffusion and processing models of word recognition.

Cytophotometric analysis of basic nuclear proteins during gametogenesis in the free-living nematode Panagrellus silusiae.

The cytochemical staining characteristics of basic nuclear proteins during gametogenesis in both sexes of the free-living nematode Panagrellus silusiae were monitored by absorption microspectrophotometry. During spermatogenesis, the levels of both of the nuclear-bound Sakaguchi and 1-fluoro-2, 4-dinitrobenzene (FDNB) reagents per Feulgen-DNA content increase prior to meiosis and subsequently drop off during spermiogenesis. During oogenesis, the amount of Sakaguchi-protein to DNA ratio rises slightly whereas the FDNB-protein to DNA ratio undergoes a meagre decline. Since the oocyte of Panagrellus accumulates DNA in excess of a 4C DNA equivalent, the relative constancy of the ratio of basic nuclear protein content to DNA amount during oogenesis signifies that these proteins are being continously produced during oocyte maturation. After treatment with alkaline fast green (pH 8.1) the cytoplasmic and nuclear regions of the gonocytes of both sexes yield such an intense reaction that microspectrophotometric measurements could not be taken. The basis of the cytoplasmic staining reaction to the fast green (pH 8.1) dye was not determined. The rapid, highly deterministic programme of embryogenesis in Panagrellus may require a substantial stockpiling of basic nuclear proteins during oogenesis.

Discriminating deterministic versus stochastic dynamics in neuronal activity.

An approach to discriminating deterministic versus stochastic dynamics from neuronal data is presented. Direct tests for determinism are emphasized, as well as using time series with clear physical correlates measured from small ensembles of neurons. Surrogate data are used to provide null hypotheses that the dynamics in our data could be accounted for by linear stochastic systems. Algorithms are given in full, and the analysis of an experimental example is given.

Assessment of historical exposures to talc at a mining and milling facility.

The purpose of this study was to develop an estimate of exposure to respirable dust for all job categories and all years in a retrospective follow-up study of worker mortality in a talc mining and milling facility. All jobs were assigned to work areas that were considered to have similar exposure profiles. Uniform exposure time periods during which non-random, deterministic variables were thought to be constant were identified and an experienced rater assigned categorical exposure scores to each work area/time period. These scores and measured baseline respirable dust concentrations were used to calculate the estimated job area/year concentrations for each work area/time period. Estimates were compared to available historical measurements. The estimated exposures ranged from 1.7 to 0.1 mg/m(3) and displayed a decreasing trend over time. When compared with measured exposures, the estimated exposures had a correlation coefficient of 0.55 with an average difference of 0.01 mg/m(3) and a range of 0.60 to -0.48 mg/m(3). The estimates were considered to be acceptable for determining relative ranking of subjects according to cumulative exposure.

Dose conversion coefficients for interventional procedures.

Effective dose (E) is a convenient quantity to estimate the stochastic risk of radiation applied to patients in interventional procedures and can be used for optimisation. Relatively long exposure times may cause deterministic effects. Hence it is necessary to know the (maximum local) doses in organs owing to the interventional procedure. In practice, organ doses cannot be measured directly. They are derived by applying a conversion coefficient to a measurable quantity, e.g. dose-area product (DAP) or entrance skin dose. For a number of interventional procedures, dose conversion coefficients (DCCs) can be found in the literature. Various DCCs are stated for nominally equal procedures, e.g. for percutaneous transluminal coronary angioplasty both 0.18 and 0.27 mSv Gy(-1) cm(-2) were reported to convert DAP to effective dose. Dependence of DCC on protocol and equipment parameters, as demonstrated through Monte Carlo simulation in this paper, makes it hazardous to simply adopt a literature value.

A survey of films for use as dosimeters in interventional radiology.

Analysis of radiation doses in interventional radiological procedures that can lead to deterministic radiation effects such as erythema and epilation would assist physicians in planning patients care after exposure and in reducing doses. Photographic films used to measure skin exposure in the past are too sensitive for the high doses involved in interventional procedures. Seventeen different types of films, many of which are generally available in hospitals, were surveyed to see if any would meet the demands of interventional radiology. Sensitometric curves obtained demonstrate that most films are inappropriate for high dose procedures. Using Kodak Fine Grain Positive and Dupont duplicating films and automatic processing, doses as high as 2.8 Gy could be measured with reasonable accuracy. Similar results can be obtained by manually processing Kodak XV-2 verification film at room temperature.

Stochastic dynamics of nanoscale mechanical oscillators immersed in a viscous fluid.

The stochastic response of nanoscale oscillators of arbitrary geometry immersed in a viscous fluid is studied. Using the fluctuation-dissipation theorem, it is shown that deterministic calculations of the governing fluid and solid equations can be used in a straightforward manner to directly calculate the stochastic response that would be measured in experiment. We use this approach to investigate the fluid coupled motion of single and multiple cantilevers with experimentally motivated geometries.

EEG dipole localization bounds and MAP algorithms for head models with parameter uncertainties.

The Cramer-Rao bound for unbiased dipole location estimation is derived under the assumption of a general head model parameterized by deterministic and stochastic parameters. The expression thus characterizes fundamental limits on EEG dipole localization performance due to the effects of both model uncertainty and statistical measurements noise. Expressions are derived for the cases of multivariate Gaussian and gamma distribution priors, and examples are given to illustrate the derived bounds when the radii and conductivities of a four-concentric sphere head model are allowed to be random. The joint MAP estimate of location/model parameters is then examined as a means of achieving robustness to deviations from an ideal head model. Random variations in both the multiple sphere radii and the layer conductivities are shown, via the stochastic Cramer-Rao bounds and Monte Carlo simulation of the MAP estimator, to have the most impact on localization performance in high SNR regions, where finite sample effects are not the limiting factors. This corresponds most often to spatial regions that are close to the scalp electrodes.

Dynamics of ongoing activity: explanation of the large variability in evoked cortical responses.

Evoked activity in the mammalian cortex and the resulting behavioral responses exhibit a large variability to repeated presentations of the same stimulus. This study examined whether the variability can be attributed to ongoing activity. Ongoing and evoked spatiotemporal activity patterns in the cat visual cortex were measured with real-time optical imaging; local field potentials and discharges of single neurons were recorded simultaneously, by electrophysiological techniques. The evoked activity appeared deterministic, and the variability resulted from the dynamics of ongoing activity, presumably reflecting the instantaneous state of cortical networks. In spite of the large variability, evoked responses in single trials could be predicted by linear summation of the deterministic response and the preceding ongoing activity. Ongoing activity must play an important role in cortical function and cannot be ignored in exploration of cognitive processes.

Can deterministic penalty terms model the effects of synaptic weight noise on network fault-tolerance?

This paper investigates fault tolerance in feedforward neural networks, for a realistic fault model based on analog hardware. In our previous work with synaptic weight noise we showed significant fault tolerance enhancement over standard training algorithms. We proposed that when introduced into training, weight noise distributes the network computation more evenly across the weights and thus enhances fault tolerance. Here we compare those results with an approximation to the mechanisms induced by stochastic weight noise, incorporated into training deterministically via penalty terms. The penalty terms are an approximation to weight saliency and therefore, in addition, we assess a number of other weight saliency measures and perform comparison experiments. The results show that the first term approximation is an incomplete model of weight noise in terms of fault tolerance. Also the error Hessian is shown to be the most accurate measure of weight saliency.

Cost-effectiveness of alternative treatments for women with osteoporosis in Canada.

BACKGROUND: During the years following menopause, estrogen levels decline leading to accelerated bone loss and an increased risk of osteoporosis and osteoporosis-related fractures. METHODS: Using a Markov model and decision analytic techniques, the long-term costs and outcomes of five treatment and secondary prevention strategies for osteoporosis were compared: 'no intervention', alendronate, etidronate, risedronate, and raloxifene. The base case analysis examined postmenopausal (65 year old) osteoporotic women without prior fracture. Probabilistic sensitivity analysis (PSA) was used to incorporate the impact of parameter uncertainty, and deterministic sensitivity analysis (DSA) was used to compare alternative patient populations and modeling assumptions. Life years and Quality Adjusted Life Years (QALYs) were used as measures of effectiveness. RESULTS: In the base case analysis, risedronate was dominated by etidronate and alendronate. Alendronate and etidronate were projected to have similar costs and QALYs, and the efficiency frontier was represented by 'no intervention', etidronate, alendronate, and raloxifene (Can$32 571, Can$38 623 and Can$114 070 per QALY respectively). Alternative assumptions of raloxifene's impact on CHD and breast cancer, alternative discount rates and alternative patient risk factors (e.g., starting age of therapy, CHD risk, and prior fracture risk) had significant impacts on the overall cost-effectiveness results for both the bisphosphonates and raloxifene. DISCUSSION: Using conventionally quoted benchmarks and compared to no therapy, alendronate, etidronate, and raloxifene would all be considered cost-effective alternatives for treating women with osteoporosis. Potential limitations of this study include the usual caveats and cautions associated with long-term projection models and the fact that not all inputs into the model are Canadian data sources.
