Hemoblastoses in mice contaminated with low activities of 239Pu.

Female ICR mice were injected intravenously with low activities of 239Pu (3.0 kBq, 6.0 kBq, 12.3 kBq/kg). In these mice with high spontaneous incidence of hemoblastoses the occurrence of myeloid leukemia, lymphocytic leukemia, lymphosarcoma, reticulum-cell sarcomas and osteosarcoma was studied. Hemoblastoses, on the whole, remained in their numbers radiation-independent, nevertheless, the distribution into specific types changed, with moderate prevalence of myeloid and lymphocytic leukemia and lymphosarcoma. After plutonium injection the mean survival time of mice bearing myeloid and lymphocytic neoplasias was significantly shorter than the survival of mice that died of retothelosarcoma and from other causes. These contamination-dependent differences could not be observed in matched controls. As expected, 239Pu activities used in this experiment induced osteosarcomas. Whereas in leukemogenesis alpha-radiation appeared as a factor promoting and modifying the leukemogenic process, in osteosarcoma the alpha-particles acted rather as an initiator, the effect of which was dependent on the dose to the endosteal progenitor cells.

Critical assessment of epidemiologic studies on the human carcinogenicity of 1,3-butadiene.

1,3-Butadiene, a major ingredient of synthetic rubber, has been shown to be carcinogenic in two animal species. To assess the possible human carcinogenicity of 1,3-butadiene, a critical review was undertaken of the epidemiologic literature. An early retrospective study of 8017 males employed in tire manufacturing found excess mortality for lymphatic and hematopoietic neoplasms in production workers (standardized mortality ratio, SMR = 560); these workers were exposed to 1,3-butadiene as well as to styrene and possibly to benzene. A recently updated epidemiologic study of 2568 workers at a butadiene manufacturing plant in Texas reported low mortality overall (SMR = 84) but found excess deaths for lymphosarcoma and reticulum cell sarcoma (SMR = 229). A retrospective study of workers employed at two synthetic rubber plants in Texas found excess mortality for lymphatic and hematopoietic malignancies in the older of these facilities; the excesses for lymphosarcoma (SMR = 224) and leukemia (SMR = 278) were most significant in wartime workers. A large, recently updated retrospective study of 12,113 workers employed in eight synthetic rubber manufacturing plants in the United States and Canada found excess mortality for lymphatic and hematopoietic cancer in production workers; the SMR for other lymphatic cancers in white production workers was 230, and the SMR for all lymphatic malignancies in black production workers was 507. These updated epidemiologic results strongly suggest an etiologic association between occupational exposure to 1,3-butadiene and human cancer. It is reasonable, therefore, to conclude that there now exists at least limited evidence for the human carcinogenicity of 1,3-butadiene.

The treatment of mycosis fungoides and related conditions with particular emphasis on electron therapy.

The difficulties created by the lack of generally accepted views on classification, diagnostic criteria and natural history of the cutaneous lymphomata are discussed. Only the lymphomata which remain confined to the skin throughout the major part of their development are considered. It is emphasized that these are extremely radiosensitive and that there must be some factors as yet unknown which play a part in their unique natural history and response to treatment. The cutaneous lymphomata are considered under three headings: (1) lymphocytoma, (2) lymphosarcoma and reticulum cell sarcoma, (3) mycosis fungoides. The characteristics of the first two are discussed briefly and illustrative examples are given. The relatively benign nature of the lymphocytoma is stressed. It is suggested that the solitary lymphosarcoma of the skin should be treated by radical radiotherapy since long-term survival or cure may be achieved. The treatment of mycosis fungoides is considered in greater detail. The physical advantages of electron therapy are described and details given of whole body surface treatment with electrons produced by a 6 MeV linear accelerator and slowed down with carbon decelerators of appropriate thickness. The results in 73 patients treated by electrons are analysed. The early and late side-effects are mentioned and the necessity for investigating other forms of treatment is stressed. It is concluded that at the present time electron therapy is the most satisfactory form of treatment for mycosis fungoides.

[Synthesis and antitumor properties of 3'-desamino-3'-dimethylformamidinorubomycin].

Interaction of rubomycin (daunorubicin) chlorhydrate with dimethylformamidine diethyl acetal yielded 3'-desamino-3'dimethylformamidinorubomycin chlorhydrate (DFR). Comparative antitumor activity of DFR and rubomycin was studied on mice with respect to ascitic lymphadenosis NK/Ly and Ehrlich carcinoma, hemocytoblastosis La, leukemia P-388 and two solid tumors i. e. lymphosarcoma LIO-I and sarcoma 180. The highest antitumor effect of DFR was observed in the mice with Ehrlich carcinoma and lymphadenosis NK/Ly after the drug intravenous administration for 4 times. By selectivity of the antitumor effect DFR was inferior to rubomycin with respect to lymphosarcoma LIO-I and sarcoma 180. It was shown that the antileukemic activity of DFR and rubomycin with respect to hemocytoblastosis La was practically the same. In the experiments with leukemia P-388 DFR was inferior to rubomycin.

Carcinogenicity of toxaphene: a review.

Toxaphene is highly carcinogenic in rats and mice. Toxaphene induced malignant neoplasms of the liver in rats. Neoplasms at all sites, as well as malignant neoplasms, were increased in male and female rats ingesting toxaphene. Sarcomas were found more often in male rats and carcinomas in female rats. Neoplasms of the endocrine organs were also increased in male and female toxaphene-treated rats. The incidence of neoplasms of the reproductive system was increased in female rats, as was the incidence of mammary gland neoplasms in male rats. Toxic changes in male rats given toxaphene included interstitial fibrosis of the kidney and atrophy of the testes. Toxaphene induced malignant neoplasms of the liver in male and female mice. The incidence of malignant neoplasms at all sites was also increased. In addition to hepatic neoplasms, male mice had leukemia or lymphosarcoma and females had sarcomas of the uterus.

Comparative studies on the structural phosphoproteins of mammalian type C viruses.

The major phosphoprotein common to woolly monkey sarcoma virus, gibbon ape lymphosarcoma virus, and type C viruses of the lower mammalian species (mouse, rat, cat), with the exception of the endogenous cat virus (RD-114), is the polypeptide of about 12,000 molecular weight. The protein-phosphate bond in this polypeptide of several viruses is of the phosphoserine variety excepting gibbon ape virus, which contains both phosphoserine and phosphothreonine. The primary phosphoprotein of RD-114 virus and the endogenous baboon type C virus, on the other hand, is the polypeptide of about 15,000 molecular weight which contains phosphothreonine as its phosphoamino acid. A second major phosphoprotein of molecular weight of 10,000 is detected only in viruses genetically related to rat species including those derived from the RPL cell line, from Sprague-Dawley rat embryo cells, and the Kirsten mouse sarcoma virus which was recovered from a mouse erythroblastosis virus after in vivo propagation through rat. These phosphorylated polypeptides of molecular weight 15,000, 12,000, or 10,000 are present in the virion structure in several different but nonrandom phosphorylated states.

Mortality among workers at a butadiene facility.

Several studies of styrene-butadiene rubber (SBR) workers have reported excess cancers at various sites; however, little could be concluded concerning specific etiologic agents because of the multiple exposures encountered by these groups. The current study examined cause-specific mortality in a cohort of 2,586 male workers employed for at least 6 months between 1943 and 1979 in a butadiene manufacturing plant that supplied butadiene to two SBR plants. Standardized mortality ratios were calculated using national (NSMR) and local (LSMR) comparison populations. The all-cause NSMR was 80 (p less than 0.05) and the all-cancer NSMR was 84; the corresponding LSMRs were 96 and 76 (p less than 0.05). No significant excesses were observed for any cause of death except lymphosarcoma and reticulum cell sarcoma (NSMR = 235). When the cohort was subdivided into routine, nonroutine, and low-exposure groups, the SMRs were consistently elevated for this cause of death in all three groups. However, direct comparisons between each of the two exposure groups and the low-exposure group were inconsistent. This suggests butadiene may not be responsible for the excess, but the association deserves close attention in future studies.

An ultrastructural analysis of lympho-reticular cell interactions in primary cultures of human non-lymphoid neoplasms and lymphomas.

The morphology and interactions of lymphoreticular cells in cultures of human tumour tissue was studied by transmission and surface scanning electron microscopy. Macrophages are very common in non-lymphoid neoplasms and vary considerably in shape and surface morphology, with features of mature stimulated cells. Lymphocytes adhere to macrophages more often than to tumour cells. Close contact between macrophages and tumour cells also occurs, but there is no evidence that the infiltrating macrophages or lymphocytes have an overt cytotoxic effect under these cultural conditions. A variety of interactions are seen in cultures of Hodgkin's lymphoma, lymphosarcoma and reticulum cell sarcoma. The only cultural characteristics that may be specific for lymphoma tissue are large intravacuolar collections of lymphocytes within macrophages or giant cells and the rare close spatial relationship between lymphocytes and degenerate macrophages, the latter particularly in Hodgkin's lymphoma. These observations demonstrate the disturbed cellular interrelationships that exist in lymphoma tissue.

Monocyte production in Hodgkin's disease and non-Hodgkin's lymphoma.

Monocytopoietic proliferation activity was investigated in patients with untreated Hodgkin's disease, Hodgkin's disease in long-term complete remission, and untreated non-Hodgkin's lymphoma of the lymphosarcoma and reticulosarcoma type. Untreated Hodgkin's disease was found to be associated with a rise in medullary monocyte production which returned to normal during long-term complete remissions. In contrast, monocyte production was increased in only 5 out of 14 patients with lymphosarcoma and reticulum cell sarcoma, normal in 3, and reduced in 6. In neither of these lymphomas was any relation between monocyte production and stage or histology of the disease detectable.
