[Study of treating experimental ulcerative colitis of spleen deficiency type with "guben yichang tablet" in guinea pigs].

Guinea pigs were perfused with the extract of senna (20%). 3 days later glacial acetic acid (5%) was given intra-anally so as to replicate the animal model of Spleen Deficiency (SD) and ulcerative colitis which is the disease of Western medicine combined with the Syndrome in TCM. The model animals showed the symptoms of SD such as loose stool, anorexia, wasting, aversion of cold, laziness and loss of hair luster and the symptoms of ulcerative colitis such as abdominal distension and mucous bloody stool. The colonic lesion were observed by eyes that the mucosa were edematous, congestive with ulcerative foci. The pathological examinations showed edema and congestion in submucous layers; large amount of inflammatory cell infiltration; and the scaling and ulcer formation of epithelium mucosae. The above-mentioned symptoms and pathological changes were the same as SD and ulcerative colitis in clinical practice. The new medicine "Guben Yichang Tablet" against ulcerative colitis was given to guinea pigs for 7 days could abate their symptoms, increase their body weights and decrease the size of colonic ulcer and edema.

[Graduated quantitative diagnosis of spleen deficiency syndrome].

90 Cases with Spleen Deficiency Syndrome (SDS) in various diseases of different body system were studied to obtain a quantitative measurement of the syndrome for more accurate diagnosis. Rates of symptom manifestation of SDS, absorptivity of D-xylose, activity of salivary amylase, hemoglobin level, RBC count, plasma albumin, cardiac function as well as some parameters of hemorheology and lymphocyte transformation rate were observed. The results clearly showed that the D-xylose absorptivity was much lower in patients with higher appearance rate of SDS symptoms, which was dose-dependent. Along with the lowering of D-xylose absorptivity, the values of above-mentioned parameters dropped were positively correlated to it. On the contrary, the peripheral vascular resistance elevated as D-xylose absorptivity dropped. The author divided the SDS into three stages as a graduated quantitative diagnosis based on the drop of D-xylose absorptivity, the increase of SDS symptom and other changes of laboratory findings.

[Effects of Chinese herbs xianzhen tablet on the deformability of erythrocyte in non-insulin-dependent diabetes mellitus patients with deficiency of both qi and yin and deficiency of kidney with blood stasis].

OBJECTIVE: To explore the effects of Xianzhen Tablet (XZT) on the hemorheology of type 2 diabetes (NIDDM) on the basis of previous studies in XZT, that have indicated its ability to deplete fasting blood glucose, to reduce the injury of free radical to the body, to improve the Na(+)-K(+)-ATPase in the erythrocyte membrane of NIDDM. METHODS: Erythrocyte deformability and aggregation were studied with Ektacytometer in a total of 60 type 2 diabetes who were randomly divided into two groups. One was ZXT treated group, another was placebo group. RESULTS: (1) The erythrocyte deformability of type 2 diabetes was lower than that in normal subjects (P < 0.001), but the erythrocyte aggregation increased abnormally than that of health people (P < 0.001). The correlation test indicated there were negative correlation with plasma fibronogen and positive correlation with blood cholesterol (P < 0.05). (2) The erythrocyte deformability in the ZXT treated group with 30 patients improved after 8 weeks treatment (P < 0.01), with the decreased fasting blood glucose and plasma fibrinogen (P < 0.05). Compared with placebo group, there was significant statistical difference (P < 0.01). CONCLUSIONS: XZT could improve the erythrocyte deformability in the type 2 diabetes.

[Preliminary study on pathophysiology of liver-qi deficiency syndrome].

The authors advocate a program for diagnosing Liver-Qi Deficiency Syndrome (LQDS) based on the TCM theory and clinical practice. Through investigation, LQDS was found to be widely existed as latent period or as external manifestation, which occupied 18.85% in Qi Deficiency Syndrome. The subjects were divided into four groups including normal group, Spleen-Qi Deficiency Syndrome (SQDS) group, LQDS with Liver diseases (LD) and LQDS with non-Liver diseases (NLD) group. In order to explore the essence, the simultaneous determinations were done on lactate dehydrogenase (LDH) and its isoenzyme, dopamine beta-hydroxylase (D beta H), trace element Zn and Cu, and other serological indexes such as GPT, TP, Alb, A/G, etc. The results were, the content of LDH and trace element Zn in both LQDS-NLD and SQDS were significantly lower than that of normal group. D beta H reflecting sympathetic nerve function in LQDS-NLD was significantly higher than that of both normal and SQDS group; comparing LQDS-LD and LQDS-NLD group, the content of GPT, LDH, LDH5 and trace element Cu in former was significantly higher than that of the latter, but the content of TP, Alb, A/G, D beta H and Zn in former was remarkably lower than that of the latter. In order to avoid confusion, in studying this syndrome, one should distinguish LD and NLD.

[Comparative study on changes of digestive and absorptive functions in three different models of Pi-deficiency syndrome].

OBJECTIVE: To compare the changes of digestive and absorptive functions in three different models of Pi-deficiency syndrome (PDS). METHODS: Experimental mice were divided into four groups, the control group(CG), the rhubarb group (RG), the exhaustion group (EG) and the over-exertion group(OG). Criteria including general physical signs, D-xylose excretion rate, serum amylase activity, velocity of stomach emptying and enterokinesia, serum gastrin content and indexes of organs were determined before and after treatment. RESULTS: Decrease of D-xylose excretion rate and increase of stomach emptying and enterokinesia velocity appeared in all the three PDS models. As compared with CG, changes of all indices in OG were significant, while the decreasing of spleen index and serum amylase activity in EG, and the changes of serum gastrin content and thymus index in RG were insignificantly different. CONCLUSION: All the changes in various criteria showed that PDS mice model established by over-exertion was superior to that established by frequently used methods as purging with rhubarb and exhausting by swimming.

[Clinical and experimental study on separately decocted and mingly decocted jianweishu granule].

OBJECTIVE: To observe the difference in therapeutic effect of Chinese herbal granule decocted separately and mingly. METHODS: One hundred patients of functional dyspepsia of Spleen deficiency and Liver stagnancy type were treated with Jianweishu Granule (JWSG, a self-formulated recipe by the author). Half of the patients received JWSG decocted separately, half of them received that decocted mingly. The therapeutic effects between the two groups were compared. Experimental observation on the effects of differently decocted remedies in rats was also conducted. RESULTS: The therapeutic effects obtained in the two groups were similar, with respective cure rate of 72% and 70% and the total effective rate 96% in both group. There was insignificant difference between the two groups (P > 0.05). Experimental study all showed no difference between the differently decocted remedies in inhibiting gastric acid, pepsin activity, gastric function regulation, small intestine movement improvement and pain alleviation in rats. CONCLUSION: The therapeutic effects of JWSG decocted separately or mingly are the same. This fact provides a scientific basis for clinical use of granule form of single Chinese herbs.

[Effect of tonifying kidney on compliability of the aged].

UNLABELLED: The clinical study of 80 aged individuals divided into treated group (Group I) and normal control (Group II) was carried out. According to the symptoms of Kidney Deficiency the aged was subdivided into Kidney Deficiency group (KDG) and non-Kidney Deficiency group. A semi-quantitative method and some indexes about compliability were used to evaluate the symptoms. RESULTS: After treatment the main symptoms of Kidney Deficiency such as lumbago, cold limbs, fatigue, cold aversion, feverish sensation in chest, palms and soles, etc. were significantly alleviated (P < 0.05) and the speed of reaction (SR) that reflects NS function in Group I and its KDG was also significantly improved. After exercise test, the ultrasono-cardiogram showed that in KDG of Group I, the ascending range of heart rate (HR) lessened during exercise and recovered more rapid than that before treatment. The difference was significant (P < 0.05). The HR was also significant different between the KDG groups of Group I and II during resting, exercise and recovering status (P < 0.05). Furthermore, after treatment the level of salival progesterone of the aged female was significantly. The determination of bone mineral content showed that in KDG after treatment, the density of ulnar line was remarkably elevated. These results showed that through tonifying Kidney and replenishing Qi could improve the functions of heart, brain, bone and endocrine systems of the aged persons, therefore improve their compliability.

[Clinical study on relationship among Heart-Qi Deficiency, left ventricular function and myocardial ischemia in patients with coronary heart disease].

OBJECTIVE: To study the relationship among Heart-Qi Deficiency (HQD), left ventricular function and myocardial ischemia in patients with coronary heart disease (CHD). METHODS: Doppler echocardiogram, electrocardiogram were used to observe 103 patients of CHD clinically. Patients of stable angina pectoris treated with Radix Codonopsitis Pilosulae (50 cases) or Nifedipine (20 cases) were observed as well. RESULTS: Occurrence of left ventricular function insufficiency, both systolic and diastolic, was higher in CHD patients of HQD type as compared with that of non-HQD type. The left ventricular diastolic function showed high susceptibility (87%) and the left ventricular systolic function showed high specificity (88%) for HQD evaluation. Radix Codonopsitis Pilosulae could protect the left ventricular function and improve myocardial ischemia. CONCLUSIONS: Myocardial ischemia and blood stasis cause HQD, while the Chinese herbal medicine treatment on HQD could improve the blood stasis to alleviate myocardial ischemia effectively. The mechanism of which might be to enhance the positive diastolic and systolic function of cardiac muscles, reduce the resistance pressure and keep pressure of coronary perfusion so as to improve the blood supply of heart.

[Expression of NF-kappaB and COX-2 mRNA in rats with phlegm obstruction due to lung-deficiency].

OBJECTIVE: To study the role of nuclear factor-kappa B (NF-kappaB) and cycloxygenase-2 (COX-2) in the onset of phlegm obstruction due to lung-deficiency in rats and the therapeutic mechanism of Huatan Recipe. METHODS: Twenty-four SD rats were randomly divided into normal group, model group and treatment group, with 8 rats in each group. The rats in the model group and treatment group were exposed to sulfur dioxide and cold wind to establish the rat model of phlegm obstruction due to lung-deficiency, and the rats in the treatment group were also treated with Huatan Recipe, a compound traditional Chinese medicine. The expression of NF-kappaB in the bronchial epithelial cells of the rats was tested with the method of immunohistochemistry, and the COX-2 mRNA in the lung tissues of the rats was measured by using reverse transcription-polymerase chain reaction. RESULTS: The expressions of NF-kappaB and COX-2 mRNA in rats of the model group were higher than those of the normal group (P<0.01), and the expressions of NF-kappaB and COX-2 mRNA in rats of the treatment group were obviously lower than those of the model group (P<0.01). CONCLUSION: The NF-kappaB and COX-2 play an important role in the onset of phlegm obstruction in rats. Huatan Recipe may prevent the development of phlegm obstruction by down-regulating the expressions of NF-kappaB and COX-2 mRNA.

[Study on erythrocytic immune function in diarrhea patients with spleen deficiency].

The erythrocytic immune function of 40 cases of patients with Spleen Deficiency (SD) and 20 cases of the normal control group were observed, and the former were subdivide into diarrhea group and non-diarrhea group. The results showed that the rosette rate of red cell C3b receptor (C3b-RR) in the SD patients was significantly lower than that of normal group and the rosette rate of red cell immune complex (IC-R) was significantly higher than that of normal group (P < 0.01); the C3b-RR in the diarrhea group was markedly lower than the non-diarrhea group, tce IC-R in the diarrhea group was markedly higher than the non-diarrhea group (P < 0.01), C3b -RR and IC-R correlated positively or negatively to the absorptivity of D-xylose hemoglobin, red blood cell and plasma albumin (P < 0.01). The results pointed out that the mechanism for the reduction of erythrocytic immune function was that the malnutrition in SD patients lowered the C3b receptors or damaged them or reduced their activity. The degree of malabsorption and systemic dysfunction in the diarrhea group was more serious, the erythrocytic immune function in the SD patient was lower.

[Effect of yishen jianpi drugs on T lymphocyte subsets, soluble interleukin-2 receptor and red blood cell immunity of senile deficiency syndrome patients].

UNLABELLED: To evaluate the effect of Yishen Jianpi drugs (YJD) on immunological function of the Senile Deficiency Syndrome (SDS) patients, T lymphocyte subsets, soluble interleukin-2 receptor (SIL-2 R), the red blood cell immunity, the rate of red blood cell C 3b receptor rosette (RBC-C 3 bRR) and red blood cell immune complex rosette (RBC-ICR) were dynamically investigated in 31 SDS patients as well as in normal controls. RESULTS: the percentage of CD3, CD4 and CD4/CD8 ratio and the rate of RBC-C 3 bRR in SDS were all significantly lower than that of control (P < 0.01). The further analysis also revealed that between the immunological indices of CD4/CD8 ratio and SIL-2 R in SDS patients were markedly negative correlated (r = -0.572, P < 0.01) while positive correlated (r = 0.435, P < 0.02) markedly between CD4/CD8 and RBC-C 3 bRR. After treatment with YJD, in comparing pretreatmentally, all the above-mentioned immunological indices, following the relief of clinical manifestation of SDS, they were corresponding negatively changed, and there were marked significance (P < 0.01) except red blood cell immunity. It suggested that YJD would markedly improve and/or regulate immunological function in SDS.

[Effect of modified xiaoyao powder on electrogastrographic picture and pharmacokinetic characteristics of ferulic acid in patients with functional dyspepsia of gan-qi stagnation with pi-deficiency syndrome type].

OBJECTIVE: To investigate the mechanism of Modified Xiaoyao Powder (MXP) on electrogastrogram (EGG) and pharmacokinetic characteristics of ferulic acid (FA) in patients with functional dyspepsia (FD) of Gan-qi stagnation with Pi-deficiency syndrome type (FD-GP). METHODS: The study was conducted following method of pharmacodynamics and pharmacokinetics synchronous determination. The 20 patients with FD-GP and 21 healthy volunteers enrolled were orally administrated with MXP to observe the changes in symptom scores, EGG and pharmacokinetic characteristics of FA in patients using gastrointestinal analyzer and high performance liquid chromatography (HPLC) before and after treatment. RESULTS: After treatment, the symptom scores of patients significantly decreased (P < 0.01). EGG showed that the average amplitude of peak and dominant frequency, etc. at the monitoring points, including gastric body, lesser curvature and the near gastric antrum part of greater curvature, etc. were lower than the normal range, the changes were especially obvious and would be worsened after meals (P < 0.01). After treatment by MXP, all the above indexes were improved either before or after meals. The pharmacokinetic parameters of FA in patients of FD-GP before treatment were lower than those in the healthy control group (P < 0.05) and they were insignificant different by MXP comparing with those in the healthy control group. CONCLUSION: MXP is effective not only in improving clinical symptoms and EGG, but also on pharmacokinetic parameters of FD-GP patients.
