Extrahepatic organs metabolism of inhaled trichloroethylene.

An extrahepatic circulation system for dogs was developed using a portal vein to right femoral vein bypass procedure. This system maintained nearly normal biochemical and physiological parameters, i.e. arterial blood pressure, heart rate, electrocardiogram, leukocyte and erythrocyte count, hematocrit, alkaline phosphatase, blood urea nitrogen, ammonia and creatinine, for 2 h. Thus, the system appears to be a valid technique for investigating extrahepatic metabolism. Dogs were exposed for 1 h to 500, 700 and 1500 ppm of trichloroethylene. Free-trichloroethanol, trichloroacetic acid and conjugated-trichloroethanol appeared in the blood and urine after 30 min of exposure. The amounts of metabolite formed by dogs with hepatic bypass were less than by similarly exposed dogs without hepatic bypasses, specifically 50-80%, 10% and 10-20% for free-trichloroethanol, trichloroacetic acid and conjugated-trichloroethanol, respectively. In addition, trichloroethylene exposure produced a smaller decrease in leukocyte counts in the hepatic bypass dogs than in the non-bypass dogs. This observation may indicate that the liver itself played some role in the elimination or increment of leukocyte counts in the blood.

Short-term toxicity of grass carp bile powder, 5alpha-cyprinol and 5alpha-cyprinol sulfate in rats.

We compared the short-term toxicity of toxic components of grass carp bile juice (GCBJ) in rats. Twenty-four male Wistar rats were divided into four groups and treated orally every 3 days with 40 mg each of freeze-dried GCBJ powder, 5alpha-cyprinol and 5alpha-cyprinol sulfate for 19 days. After treatment, the relative ratio of liver and kidney weight to body weight, the concentrations of RBC, hemoglobin and hematocrit in the blood, the levels of aspartate amino transferase, alanine amino transferase, alkaline phosphatase, blood urea nitrogen and creatinine in the plasma, and the levels of urinary urea nitrogen and creatinine in the urine were significantly increased. Body weight of rats and the levels of Na+, K+, Ca2+ in the urine were significantly decreased, especially for groups treated with GCBJ powder and 5alpha-cyprinol sulfate. Pathological examination of liver and kidney also showed cell enlargement and lesions, especially for groups treated with GCBJ powder and 5alpha-cyprinol sulfate. The grass carp bile juice exhibited significant toxicity, and the short-term toxicity of 5alpha-cyprinol sulfate and GCBJ powder was similar to each other. Renal but not hepatic failure induced by grass carp bile juice could be prevented by pentoxifylline.

Evaluation of triflupromazine as a sedative in camels (Camelus dromedarius).

Effects of administration of triflupromazine were evaluated in 11 adult domesticated camels (Camelus dromedarius) weighing 403 +/- 29.5 kg (Mean +/- SE). Six camels were used to evaluate sedative properties of the drug and its effects on haematological and blood biochemical parameters. In the remaining 5 camels, effects on haemodynamics, acid base status and blood gases were studied. In all the animals triflupromazine was administered intramuscularly in the gluteal region at the rate of 2 mg/kg. Camels voluntarily sat down 48.9 +/- 5.4 min after administration of the drug but stood up again if disturbed. Drowsiness, drooping of lower lip and salivation were evident. The animals stood on their own and started walking with ataxia after 159 +/- 7 min and recovered completely from the effect of drug within 259 +/- 23 min. The drug caused a significant tachycardia and a moderate hypotension. The decrease in central venous pressure was also significant. Rectal temperature, respiratory rate, acid base status, blood gases, haemoglobin concentration, packed cell volume, total erythrocyte count, total leucocyte count, differential leukocyte count, blood urea nitrogen, plasma alanine aminotransferase, aspartate aminotransferase, lactate dehydrogenase, alkaline phosphatase, blood glucose and plasma concentrations of sodium, potassium, chloride and inorganic phosphate were not significantly affected by triflupromazine.

[Relationship between the activity of alanine aminopeptidase and alkaline phosphatase in the blood of pregnant women].

The determination of the activity of alkaline phosphatase (AP)--orthophosphate-monoesterphosphohydrolase, EC.3.1.3.1.--and alanine-aminopeptidase (ANA)--alpha-aminoacyl-peptide hydrolase (microsomal), EC.3.4.11.2--in the serum of non-gravid and gravid women has shown that in non-gravid women normal ANA values range from 17.0 to 32.0 I. U. and normal AP values from 14.4 to 26.0 I. U., the ANA/AP quotient amounting to 1.28 (S = +/- 0.301 I. U., KV = 23.5%, n = 29). The determination of the activity of the above quoted enzymes has shown that in the course of pregnancy the values of both enzymes increased by the exponential curve which allowed the calculation of the ANA/AP quotient for each month of pregnancy. The ANA/AP quotient determined in this way is proposed to serve as a diagnostic parameter in the routine control of pregnant women.

[Kinetic method for determination of isoenzymes of alkaline phosphatase in human blood serum].

Resistance of liver, bone, intestinal and placental human alkaline phosphatase isozymes to the effect of L-phenylalanine and urea was studied. The combined treatment with urea and L-phenylalanine increased an accuracy of estimation due to inhibition of bone and intestinal isozymes. A special equation enabled to simplify the calculations. As shown by laboratory and clinical tests, the method developed is suitable for quantitative estimation of alkaline phosphatase isoenzymes.

[Effect of anticoagulants on the change of blood parameters in diestric gilts].

The above problem was studied by furnishing four groups of six gilts each with ear catheters for the following intravenous catheter treatment: 2,500 IU heparin per die over ten days, supported by two daily oral applications of 1 g Falithrom, or 2,500 IU heparin plus two daily applications of 1 g Falithrom, all intravenously. The last group remained untreated for control. One day of the dioestrus was chosen for catheter bleeding of all animals at 6 a.m., 8 a.m., noon, and 4 p.m. and subsequent determination from the plasma of free fatty acids, copper, inorganic phosphorus, total protein, albumin, globulin, chloride, urea, cholesterol, alkaline phosphatase, blood sugar, and beta-lipoproteides. Significant differences regarding these parameters between the various groups were not even established, if alpha = 0.25. The anticoagulants used in the study may be used without any reservation for catheter rinsing and clearing and will not cause any significant change in the levels of the blood parameters.

Metabolic findings after colocystoplasty in children.

OBJECTIVES: To investigate whether colocystoplasty has resulted in metabolic changes in the growing child during long-term follow-up according to whether cecum with ascending or sigmoid colon was used. METHODS: Twenty-eight patients (mean age at surgery 11 years) were included in the study and divided into two groups: group 1, cystoplasty with cecum and ascending colon (12 patients) and group 2, sigmoid cystoplasty (16 patients). Patients' linear growth, body mass index, and the following parameters were estimated before surgery and at 3, 6, and 12 months, and then yearly after surgery: blood and urine electrolytes (sodium, potassium, chloride, calcium, phosphorus, magnesium), creatinine, urea, blood gases, blood pH, urine pH, and blood alkaline phosphatase (ALP). All the data were statistically analyzed. RESULTS: In group 1, the blood ALP increased significantly (P = 0.026) during follow-up. Severe metabolic acidosis with or without hyperchloremia was found in 7 patients. In group 2, the serum sodium and serum calcium levels decreased significantly (P = 0.014 and P = 0.003, respectively); however, the blood ALP, urine sodium, and urine phosphorus levels increased significantly (P = 0.033, P = 0.027, and P = 0.026, respectively) during follow-up. A statistically significant decrease in blood pH (P = 0.022) was found after surgery. Severe metabolic acidosis with or without hyperchloremia was detected in 5 patients. The average linear growth decreased significantly (P = 0.001 and P = 0.016, respectively) 1 and 2 years postoperatively. CONCLUSIONS: The statistically significant increase in blood ALP and decrease in serum calcium indicate bone demineralization after colocystoplasty. Our investigations in children suggest that bone demineralization is more frequent after sigmoid cystoplasty than after the use of cecum and ascending colon.

Effects of vanadyl sulfate on liver of streptozotocin-induced diabetic rats.

The aim of this study was to investigate the microscopic and biochemical effects of vanadyl sulfate on liver tissue of normal and streptozotocin (65 mg/kg) diabetic rats. Vanadyl sulfate was administered by gavage at a dose of 100 mg/kg. Degenerative changes were observed in diabetic animals by light and transmission electron microscopes. Although there were individual differences in diabetic animals to which vanadium was given, some reduction of degenerative changes were detected. After 60 d of treatment, serum aspartate and alanine transaminase, alkaline phosphatase, blood glucose levels, liver lipid peroxidation, and nonenzymatic glycosylation significantly increased, but liver glutathione levels significantly decreased in the diabetic group. On the other hand, treatment with vanadyl sulfate reversed these effects. As a result, it might be concluded that vanadyl sulfate has a protective effect on damage of liver of streptozotocin-induced diabetic rats.

Vitamin D deficiency in veiled Kuwaiti women.

OBJECTIVE: To study vitamin D deficiency in veiled Kuwaiti women. DESIGN: Prospective on cases and volunteers. SETTING: The medical department in a general hospital in Kuwait serving about 300000 persons (Adan Hospital). SUBJECTS: The authors saw four cases of unexplained vitamin D deficiency osteomalacia in otherwise healthy veiled Kuwaiti women. This stimulated us to study 50 veiled volunteer Kuwaiti women aged between 14 and 45 years old who had three children or less, and 22 unveiled volunteer Kuwaiti women with matched age and number of children as control. INTERVENTIONS: Blood samples were taken to check levels of 25-hydroxy cholecalciferol, calcium, phosphorus and alkaline phosphatase. Blood urea, serum creatinine, serum bilirubin, serum alanine transaminase, serum aspartate transaminase, serum lactic dehydroginase, prothrombin time, urine for proteins and pH were checked to exclude renal and hepatic disease. RESULTS: showed subclinical vitamin D deficiency among veiled women and two cases of clinically overt osteomalacia were discovered among the veiled volunteers suggesting a high prevalence of the disease among veiled Kuwaiti women.
