[Changes in the number of Ec cells in the small intestine and the serotonin level in the blood plasma of fasting rats].

A study was made of the amount of Ec-cells in the small intestine mucosa and blood plasma serotonin of rats in health and fasting. It was established that 24 hours after food deprivation the amount of Ec-cells rises approximately 2-fold as compared with control. The cells demonstrate the intensification of the argentaffin reaction. The content of serotonin in blood plasma increases 2-fold accordingly. On day 3 of fasting the amount of Ec-cells and intensity of the argentaffin reaction decrease to normal, whereas the content of blood plasma serotonin does not change essentially. This may be linked with a massive release of serotonin to blood and depletion of Ec-cells because of which the threshold of their histochemical demonstration is reduced. On day 7 of food deprivation the amount of Ec-cells and the intensity of the argentaffin reaction increase again but the serotonin content dramatically falls down. This phenomenon may be related to the derangement of serotonin release to blood or to the transformation from the synthesis of serotonin to the synthesis of some other hormone, most likely melatonin.

Dry drop of blood plasma--a new approach in the diagnosis of neoplastic diseases.

The dried drop of blood plasma and serum was first described by Białowas in 1967. The dry drop as a method for diagnosis of neoplasms was introduced in 1984 by Hungarian authors from Cancer Research Group. The aim of our investigations was to estimate the usefulness of this method in diagnostic of central nervous system's tumours and lung cancers. We worked out our own modification of dry drop test, i.e., we used the blood plasma instead the blood serum. In 93% of neoplasms the result of test was positive (in 92% of neoplasms of central nervous system and in 83% of lung cancers). In the control group occurred the large number of falsely positive results, as well as in group of patients with lung tuberculosis (70%), multiple sclerosis (63%) and myasthenia (45%). The results of our investigations showed the usefulness of this method in the screening diagnostic of neoplasia. The further investigations are necessary, especially correlation of dry drop test results with erythrocyte sedimentation test and antibodies and fibrinogen's level in the blood.

[Cationic composition of pancreatic juice and blood plasma during functional changes in the pancreas].

In chronic experiments on dogs with Pavlov's fistula, potassium and calcium concentration in the blood plasma was increased after feeding; their pancreatic concentration was lower, respectively, by 0.2 and 0.3 units/ml prior to feeding and 0.3 and 0.6 after the feeding. Sodium concentration in the pancreatic juice corresponded to its concentration in the blood plasma and did not change after feeding. Changes of sodium concentration in the blood plasma or pancreatic juice indicate disturbances in pancreatic function.

[Effect of the reparative therapy on indices of lipid and phospholipid spectra of the blood plasma during neonathal enteropathology in calves].

The paper deals with dynamics peculiarities of quantitave changes of the same classes of lipid and phospholipid spectra of blood plasma of calves which have recovered from dyspepsia. The obtained reliable changes of blood plasma lipidogrammas testify to development of dyslipidhaemia, hypercholesterolhaemia and hyperthreacylglycerolhaemia as well as the deficiency of choline fractions of phosphatide--main structural components of cell membranes. It was established that changes of lipid and phospholipid spectrum of the blood plasma caused by enteropathology could be corrected by the inclusion of reparative therapy remedies to dyspepsia treatment plan, in particular, experimental phospholipids-containing remedy, which is prepared on the basis of milk phospholipids--their natural source for newborn calves.

[Isolation and properties of superoxide dismutase from human blood plasma].

A procedure for purification of superoxide dismutase (SOD) from human blood plasma has been developed, which includes gel filtration on Ultrogels AcA-34 and AcA-44 (LKB, Sweden). The protein purified from blood plasma is a glycoprotein which is thermostable at 70-80 degrees C. The molecular mass of the protein determined immediately after gel filtration is approximately 147,000 daltons. A comparative analysis of effects on the SOD activity of plasma and erythrocytes of compounds capable of forming chelating complexes with metals within the enzyme active center has been carried out. The purified enzyme differs by its physico-chemical characteristics from cytosolic Cu,Zn-SOD and pertains to a new class of SOD, the so-called extracellular SOD, detected in some biological fluids.

[Investigation of chlorfenvinphos influence on levels of free tryptophan in blood plasma of the rat].

The authors studied the effect of chlorfenvinphos on the free tryptophan level in the plasma of blood of male Wistar rats. The animals were intoxicated with pesticide in a single oral dose of 2.6 mg/kg (0.2 LD50). The tryptophan was assayed spectrophotometrically after 2, 4, 24 and 72 hours following administration of the insecticide. The obtained results indicate that after intoxication with chlorfenvinphos there are no changes in the free tryptophan level after 2, 4 and 24 hours. After 72 hours a significant increase of this amino acid level was observed. Inhibition of the tryptophan elimination by kidney or disturbance of dynamic equilibrium in the distribution between plasma and erythrocytes, probably is the cause of this change in concentration.

High-performance liquid chromatographic determination of penicillamine in whole blood, plasma, and urine.

A high-performance liquid chromatographic method for the determination of penicillamine in plasma, whole blood, and urine samples is described. The method uses a commercially available electrochemical detector at a potential of +0.1 V versus the Ag/AgCl reference electrode. This method is selective and sensitive for sulfhydryl compounds. The chromatography separates penicillamine from other endogenous sulfhydryl compounds with a limit of detection for penicillamine in biological samples of ca. 10(07) M.

Effect of volatile fatty acid infusion on blood plasma free amino acid profiles in growing lambs.

Five ram lambs (average body mass: 25 kg) were given, through a catheter inserted into the left ruminal vein, a total of 28.8 mM sodium acetate, 14.4 mM sodium propionate and 4.8 mM sodium butyrate per kg body mass as a 2-hour infusion. During and at 0, 1, 2, 4, 6, 10 and 24 h after the infusion blood samples were taken from the jugular vein and the blood plasma was assayed for free amino acid (FAA) and immunoreactive insulin (IRI) concentrations. Volatile fatty acid (VFA) infusion significantly decreased the blood plasma concentrations of all FAA but cystine. The lowest FAA concentrations were measured in plasma samples taken at the end of the 2-h infusion. Subsequently the level of all amino acids rose and by 24 h after the infusion the blood plasma concentration of all FAA came close to the preinfusion value. The largest differences were observed in the concentration of glutamate, glycine, leucine and isoleucine. In contrast to FAA, IRI concentration was increased significantly (almost fivefold) by VFA infusion. By 10 h after the infusion IRI concentration returned to the initial level. The results reported here indicate that energy supply given in the form of VFA infusion significantly affects blood plasma FAA profiles, supposedly as a result of changes induced in protein synthesis in tissues. Insulin presumably plays a role in the regulation of these changes.

T cell depletion using anti-CD2 coated magnetic beads simplifies the detection of peripheral blood plasma cells.

The detection of peripheral blood plasma cells is clinically important, but difficult to perform by use of routine smears. To simplify the detection process, the peripheral blood T cells from ten patients with known active multiple myeloma were depleted using anti-CD2 coated magnetic beads. In all cases, there was enrichment of the immunoglobulin (Ig) positive cells after T cell depletion (mean enrichment factor, 3.4; median, 3.2; range, 1.2-5.1) with a mean pre-bead %Ig+ cells of 7.3 compared to 20.4 for the post-bead sample (p = 0.005). The monoclonal plasma cells were similarly enriched and more easily enumerated on the microscope slides prepared from the T cell depleted sample.

Determination of formaldehyde in blood plasma by high-performance liquid chromatography with fluorescence detection.

An HPLC method was developed for the determination of formaldehyde in human blood plasma. The method was based on the determination of the fluorescent product of the chemical reaction between formaldehyde and ampicillin. A 0.2-ml aliquot of blood plasma was reacted directly with ampicillin under acidic and heating conditions. The reaction product was extracted from the matrix with diethyl ether and analyzed by reversed-phase HPLC with fluorescence detection. Recoveries of spiked formaldehyde at the low ppm (microg/ml) level were between 93% and 102% with relative standard deviations less than 8%. The limits of detection and quantitation of formaldehyde in blood plasma samples were 0.46 microg/ml and 0.87 microg/ml, respectively.

[Transcortin binding capacity in the blood plasma of rats with a growing sarcoma 45].

It was shown that at the 10th day of sarcoma 45 growth the content of transcortine bound and free 11-oxycorticosteroids (11-OCS) in plasma of rat peripheral blood was increased. At the 30th day of the tumor growth the content of total and protein bound 11-OCS was decreased, accordingly by 57 and 61%. Following single hydrocortisone injections in intact rats and rats with sarcoma 45, on the 10th day of its growth, a marked increase in the content of bound 11-OCS was noted, as compared with animals receiving saline solution. On the 20-30th day of the tumor growth such effect was not observed. On the 20-30th day of sarcoma 45 growth 8-9% of the value of exogenic hydrocortisone, bound to blood plasma transcortins in intact rats, were found to be tracortin bound.

[Comparison of the blood plasma depressor prostaglandin content and the indicators of hemodynamics in dogs with experimental myocardial infarct and cardiogenic shock].

The quantitative composition of depressor prostaglandins PGE1 and PGA1 in the plasma of arterial and venous blood and blood from the great cardiac vein was studied in comparison with the values of hemodynamics in dogs with experimental myocardial infarction and cardiogenic shock. The content of prostaglandins in blood plasma was determined by the radioimmune method. The development of acute coronary insufficiency was attended with changes in the quantitative composition of the depressor PGE1 and PGA1 in the plasma of arterial and venous blood and blood from the great cardiac vein. In myocardial infarction and particularly in cardiogenic shock, intensification of PGE1 and PGA1 synthesis and an increase in the consumption of these PG by the myocardium occurred beginning with the first hours following occlusion of the coronary arteries, which suggests that PGE1 and PGA1 may probably take part in the normalization of the activity of the affected heart.

[Content of components of the renin-angiotensin-aldosterone system of blood plasma of healthy children in the early neonatal period].

The dynamics of the renin and angiotensin converting ferment activity and of the aldosterone concentration in the plasma of blood by healthy newborn full-term in the first week of the life after the birth was determined. The normative values of the studied indexes were obtained. The researches have shown the considerable functional tension of the renin-angiotensin-aldosterone system, that has a very important signification for optimal course of adaptation and anabolic processes by the children at the very beginning of the postnatal development.

Effect of dietary zinc on the levels and distribution of Fatty acids and vitamin A in blood plasma chylomicrons.

The aim of this work was to explore the effects of a low- and high-zinc diet and vitamin Aon the distribution of fatty acids in chylomicrons. Mongolian Gerbils were fed a basal diet (for 3 wk) containing 8 or 38 mg zinc/kg of feed (low-zinc group [termed LZ group] and saturated zinc group [termed SZ group], respectively). The following day, the animals were given sunflower oil containing 50 nmol vitamin A. The results showed that the concentration of zinc in blood plasma was similar in both groups. The amount of plasma chylomicrons was lower in the LZ group than in the SZ group (p < 0.001). The concentration of retinol in blood plasma was lower in the LZ group than in the SZ group (p < 0.01). However, the results demonstrated an increase in the blood plasma retinol concentration in the LZ group compared to the SZ group when calculated per milligram of plasma chylomicrons (p < 0.01). In plasma chylomicrons, fatty acids corresponding to 16:0, 16:1, 17:0, 17:1, 18:0, 18:1, 18:2, 18:3, 20:0, 21:0, and 20:4 were detected. The fatty acid distribution was similar in both groups. There was no major difference in the concentration of fatty acids in plasma chylomicrons between both experimental groups, except for 20:4 (a lower amount was found in the SZ group). Our results show that dietary zinc influences both the amount of chylomicrons in blood plasma and the concentrations of retinol and arachidonic acid in chylomicrons.
