On the basis of functional morphology, biomechanical properties and topographic relations of the cells of the Corti organ, a physical model was designed for the study of mechanical action in the cochlear partition during sound stimulation. The topographic relations between the supporting and the sensory cells of the organ of Corti transform the basilar membrane deviation into a complex movement of the reticular lamina, which enhances the differences in the deviation of stereocilia of the inner and outer hair cells.

With the scanning electron microscope we have examined normal cochlear sensory epithelium of the guinea pig and cat and that damaged by noise. The studies demonstrate how the regular surface architecture of the organ of Corti is altered after exposure to noise. The changes include loss of sensory hairs, formation of giant hairs, and complete degeneration of circumscribed areas of the organ of Corti. Our method greatly reduces the artifacts.
Inner ear tissue of the normal guinea pig was conductively stained (OTOTO-method) for SEM investigations. The Hensen's cells of the organ of Corti were removed using a micromanipulator inside the SEM. By this method atypical bodies of sensory and supporting cells were revealed in the apical turns of the cochlea. Atypical sensory cells showed great variations in size and shape. Several had no contact to Deiter's cells and no or only one nerve supply at their basal end. Atypical Deiter's cells showed alterations in shape and in the form of their phalangeal processes. Additionally altered parts of the organ of Corti were isolated by micromanipulation and embedded for correlative TEM-investigations.
A new technique of examining the organ of Corti in the perfused human cochlea is described. The fixed cochlea is microsliced and surface preparations are prepared and then stained for light microscopy. A method of assessing the distance/length ratio of samples is also described. Counts of inner and outer hair cells in such stained surface preparations were performed on elderly subjects and compared with those of infants. A marked loss of outer and a mild loss of inner hair cells was present in the elderly subjects. Complete atrophy of variable lengths of the terminal basal coil cells was also found in all elderly cochleas. Giant stereociliary degeneration takes place in hair cells (except basal coil outer hair cells) before they disappear.
