Effect of silk fibroin interpenetrating networks on swelling/deswelling kinetics and rheological properties of poly(N-isopropylacrylamide) hydrogels.

Novel protein/synthetic polymer hybrid interpenetrating polymer networks (IPNs) of poly(N-isopropylacrylamide) (PNIPAAm) with Bombyx mori silk fibroin (SF) have been prepared by using methanol to postinduce SF crystallization. Those IPNs having the beta sheet crystalline structure of SF show improved storage and loss moduli. The IPN hydrogels show the same volume phase transition temperature and NaCl concentration as pure PNIPAAm hydrogels. The PNIPAAm/SF IPNs keep the swelling kinetics of PNIPAAm, while showing increased deswelling kinetics. The IPNs with SF beta sheet structure should decrease the formation of the skin layer observed in conventional PNIPAAm hydrogels. Therefore, the proposed IPN hydrogels composed of protein/polymer provide fast deswelling rates as well as improved mechanical properties over pure PNIPAAm hydrogels. The effect of SF beta sheet networks on the IPNs copolymerized with acrylic acid (AAc) (P(NIPAAm-co-AAc)/SF IPNs) is compared with that on the PNIPAAm/SF IPNs, and the parameters controlling the deswelling kinetics of the IPNs are investigated. Three parameters, (1) the skin layer formation, (2) the restriction of SF beta sheet networks, and (3) the aggregation force of NIPAAm chains, are cooperatively involved in the deswelling process of IPN hydrogels according to the SF content and the presence of the AAc moiety.

NMR chemical shift powder pattern recoupling at high spinning speed and theoretical tensor evaluation applied to silk fibroin.

The NMR pulse sequence RAI (recoupling of anisotropy information) has been improved to obtain powder patterns at high MAS spinning speeds. The 2D iso-aniso experiment displays the static chemical shift spectra on the indirect dimension and the MAS spectra on the direct dimension; hence overlapping chemical shift tensor patterns can be well resolved. This efficient technique is applicable to compounds containing (13)C sp(3) (C(alpha), C(beta)) and sp(2) (C=O) sites with higher chemical shift (CS) anisotropy (CSA), and the reliability of the method was tested here on the (13)C chemical shift tensors of polycrystalline glycine, alanine, and serine. Subsequently, the same experiment was applied to the native silk protein fibroin from Bombyx mori, which consists mainly of these three amino acids. Molecular dynamics (MD) simulations of the silk II crystal structure of Takahashi et al. (Takahashi et al. Int. J. Biol. Macromol. 1999, 24, 127-138) were carried out to study the influence of motions on the chemical shift tensors. The (13)C chemical shift tensors were calculated using the bond polarization theory BPT on 200 structures created by an MD simulation. Very good agreement of the theoretical chemical shift anisotropy values with the experimental NMR results was obtained. The tensor orientations in the protein structure could thus be reliably derived.

Nano-scaled hydroxyapatite/polymer composite I. Coating of sintered hydroxyapatite particles on poly(gamma-methacryloxypropyl trimethoxysilane)grafted silk fibroin fibers through chemical bonding.

The inorganic-organic composite consisting of nano-scaled hydroxyapatite (HAp) and silk fibroin (SF) fibers was prepared through covalent linkage to develop a novel biomaterial for a soft-tissue-compatible material. The preparation of the composite was conducted through the three-step procedure consisting of chemical modification using 2-methacryloxyethyl isocyanate (MOI) monomer to introduce vinyl groups on SF, poly(gamma-methacryloxypropyl trimethoxysilane) (MPTS) graft-polymerization on SF, and coupling process between the surface of polyMPTS-grafted SF and HAp nano-particles. The amount of the graft-polymerization of polyMPTS through vinyl groups was well controlled by the reaction time. The nano-crystals were subsequently coated on the grafted fibers by heating at 120 degrees C for 2 h in a vacuum. The crystalline structure of the SF substrate did not change in the procedure. In the SEM observation of the composite surface, it was found that the bonded nano-crystals were separated and partially aggregated with several crystals attached on the SF fiber surface. The HAp particles adhered more strongly on the SF surface with separation or aggregation of several crystals than on the surface of the original SF after ultrasonic treatment.

Silk fibroin modified porous poly(epsilon-caprolactone) scaffold for human fibroblast culture in vitro.

In order to develop scaffolds with improved biocompatibility for cell culture, hybrid scaffolds were fabricated by modifying poly(epsilon-caprolactone) (PCL) with silk fibroin (SF) in a porous structure. Scanning electronic microscopy revealed that the morphology of the PCL-SF hybrid scaffold was affected by the concentration of the SF solution. Availability of SF on the surface and the conformational transition induced by methanol treatment were proved by attenuated total reflection Fourier transformed infrared spectroscopy (ATR-FTIR), and wettability of the hybrid scaffold was greatly improved. To evaluate scaffold biocompatibility, human fibroblasts were cultured on the hybrid scaffold with the unmodified PCL scaffold as control. An MTT assay indicated that although fewer cells were initially held on the hybrid scaffold after one day of culture, comparable cell numbers were achieved after four days and significantly more cells proliferated on the hybrid after seven days. The cell morphology also indicated that the PCL-SF hybrid scaffold was favorable for cell culture. This study suggests that surface modification with SF would be an effective way to improve the biocompatibility of PCL, facilitating its application in practical tissue engineering.

Fractionation of the chymotryptic precipitate of Bombyx mori silk fibroin.

1. A solution of Bombyx mori silk fibroin was digested with chymotrypsin. Amino acid analyses of the chymotryptic precipitate showed in addition to the main constituents Gly, Ala, Ser and Tyr, very small amounts of Lys, His, Arg, Asp, Thr, Glu, Pro, Cys, Val, Met, Ile, Leu, Phe and Trp. 2. A stable solution of the chymotryptic precipitate in 6m-urea was obtained by dialysing a solution in 50% (w/v) lithium thiocyanate against 6m-urea. 3. The dinitrophenylated chymotryptic precipitate in 6m-urea was fractionated by gel filtration and by ion-exchange chromatography. On Dowex 1 (X2), a main fraction I(d) and three further fractions with different amino acid compositions and molecular weights were obtained. 4. Specific rearrangement and fission of the bonds involving the serine nitrogen atoms of fraction I(d) and fractionation of the resulting mixture by gel filtration yielded five fractions. Two of these fractions had the compositions DNP-Ser-(Gly(6),Ala(4),Ser) and DNP-Ser-(Gly(4),Ala(2) or Ala(3),Ser) and are presumably double repeating units according to the proposed formula of Lucas, Shaw & Smith (1957), namely [Ser-Gly-(Ala-Gly)(n)](2), for n values of 2 and 1 respectively.

Mass-spectrometric determination of the amino acid sequences in peptides isolated from the protein silk fibroin of Bombyx mori.

Several peptides were isolated from the protein silk fibroin of Bombyx mori by means of ion-exchange chromatography of a chymotryptic digest. The sequences of three of the peptides, Gly-Ala-Gly-Tyr, Gly-Val-Gly-Tyr and Gly-Ala-Gly-Ala-Gly-Ala-Gly-Tyr, were known from previous chemical work, but the sequence of the fourth, Gly-Ala-Gly-Val-Gly-Ala-Gly-Tyr, was previously only partially known. The necessary volatility for mass-spectrometric examination of the peptides was achieved by permethylation of the N-acetyl-peptide methyl ester derivatives. From the mass spectra it was possible to confirm the known sequences and to establish that of the partially known one. In one instance it was possible to deduce from the same mass spectrum the sequence of a main peptide component and that of a small amount of contaminating peptide. These results demonstrate for the first time the use of mass spectrometry in the determination of the amino acid sequences in peptides from a protein hydrolysate.

In vitro characterization of the fibroin gene promoter by the use of single-base substitution mutants.

A highly efficient method for segment-directed mutagenesis has been developed. The method relies on the deamination by sodium nitrite of the bases in the separated strands of a small DNA restriction fragment. The mutagen-treated strands produce transition mutations by the following sequence: (i) hybridization with the complementary strand of the wild-type DNA that had been cloned into a phage fl vector, (ii) repair synthesis in vitro, and (iii) transfection of Escherichia coli. Using this method, we have isolated 14 single-point mutants within a 31-base-pair stretch of the fibroin gene (from the T-A-T-A box at the nucleotide position -30 to the cap site at +1). In vitro transcription experiments with the HeLa cell or the silk gland cell extract show that single-base transitions at the T-A-T-A box (T to C at -30, A to G at -29, and T to C at -28) and at the -20 region (G to A at -21, T to C at -20, and A to G at -17) result in decreased promoter activities, whereas those at the cap site and the -10 regions have no effect. The initiation site of transcription is the same for five "down" (reduced activity) mutants (T to C at -30, T to C at -28, G to A at -21, T to C at -20, and A to G at -17), the cap site mutant (A to G at +1), and the wild-type genes--position +1. However, the A-to-G transition at -29 (the second base of the T-A-T-A box) induces an additional transcription start from position +4. Functions of the T-A-T-A box and the -20 regions are discussed.

Transcription modulation in vitro of the fibroin gene exerted by a 200-base-pair region upstream from the "TATA" box.

We have previously reported that the 5'-flanking sequence upstream from the "TATA" box modulates the faithful transcription initiation of the fibroin gene in a homologous whole cell extract prepared from the silk glands, whereas such a modulating effect is not observed in a HeLa cell extract. Subsequently we have determined that major signals responsible for the modulating effect are located within a 200-base-pair region upstream from the TATA box, mainly in a distal region between nucleotide positions -238 and -116 and in a proximal one between -73 and -53. Inversion of the sequence element -234 to -66 did not alter its modulating effect. A similar modulating effect by the upstream region of the sericin gene was also observed in the silk gland extract but not in the HeLa cell extract. In contrast, a modulating effect by the upstream region of the adenovirus 2 major late gene was observed in the HeLa cell extract but not in the silk gland extract. Thus, the modulating effect by the upstream regions of these genes is exerted only in their own homologous extracts.

Contact points between transcription machinery and the fibroin gene promoter deduced by functional tests of single-base substitution mutants.

An efficient method for oligonucleotide-directed mutagenesis was developed to construct a set of site-specific mutations by using a mixture of oligodeoxyribonucleotides. With this method, as high as 40% of the tested clones turned out to be desired mutants. Seven single-point mutants were isolated in the "TATA" box region of the fibroin gene. In vitro transcription experiments showed that single-base transversions at the TATA box (A----T at position -29, T----A or G at -28, A----T at -27, and A----T at -26) resulted in decreased promoter activities, whereas A----G transitions at positions -24 and -23 had no effect. The initiation site of transcription was normal in three down-promoter mutants at positions -28 and -26, but the A----T transversions at positions -29 and -27 induced an additional transcription start from position +4. Using 10 single-point mutants obtained as above or by nitrous acid-induced mutagenesis, we have prepared a pair of heteroduplex DNAs consisting of a mutant strand and the wild-type one. The molecules heterozygous at positions -30, -21, and -20 showed reduced transcription activities when the noncoding strand bears the mutation, whereas that at position -26 gave a low activity when the coding strand carries the mutation. Both types of heteroduplex at positions -29, -28, -27, and -17 exhibited decreased activities. These results suggest a transcription machinery contact to a major groove of the DNA helix at the TATA box and region of position -20.

Negative supercoiling of DNA facilitates an interaction between transcription factor IID and the fibroin gene promoter.

Transcription of the fibroin gene can be reconstituted with partially purified components from HeLa cells. Transcription factors IIB, IID, and IIE and RNA polymerase II are required for accurate initiation of transcription. Linear and relaxed closed circular DNA show a similar level of template activity. However, transcription of closed circular DNA is stimulated when negative supercoils are introduced by the addition of DNA topoisomerase II and supercoiling factor purified from the posterior silk gland of Bombyx mori. Dissection of transcription into pre- and postinitiation steps by the use of Sarkosyl reveals that DNA supercoiling promotes formation of a preinitiation complex. Furthermore, order of addition experiments suggest that DNA supercoiling facilitates a functional binding of transcription factor IID to the promoter.

New oral dosage form for elderly patients. III. Stability of trichlormethiazide in silk fibroin gel and various sugar solutions.

The hydrolysis of trichlormethiazide (TCM) in silk fibroin gel (SFG) prepared in various sugar solutions (such as ribose, fructose, mannose, and glucose solutions) was determined. The hydrolysis rate of TCM differed with the variety of sugars utilized in this study; that is, it decreased in the following order: ribose > fructose > mannose > glucose. To investigate the relationship between the hydrolysis rate of TCM and the physicochemical properties of the sugar molecule, the amount of unfrozen water of sugar molecules was calculated from differential scanning calorimetry (DSC). The amount of unfrozen water increased with an increase in the number of the equatorial OH groups n(e-OH) per sugar molecule that are able to hydrate favorably to the surrounding water molecules. The hydrolysis rate constant decreased with increase in n(e-OH); glucose, having a large n(e-OH) in this study could effectively stabilize TCM.

Silk fibroin-coated three-dimensional polyurethane scaffolds for tissue engineering: interactions with normal human fibroblasts.

Silk fibroin (SF)-based or -coated biomaterials hold structural and surface properties that render them suitable for biomedical applications. In this work, we investigated the behavior of four strains of normal human adult fibroblasts (HAFs) seeded onto polyurethane foam, uncoated (PUF) or SF coated (PUF/SF). HAF adhesion within 3 h to PUF/SF was 2-fold that of adhesion to PUF. After 30 days of incubation in vitro, 37% more HAFs had grown on PUF/SF than on PUF. Taking 10(5) cells as a basis for comparisons, HAFs on PUF/SF exhibited initially higher glucose consumption rates, but persistently lower glutamine uptake rates than on PUF, whereas the rates of lactate and interleukin 6 release and of extracellular assembly of type I collagen fibers were alike on either substrate. Moreover, HAFs on both PUF/SF and PUF never secreted any ELISA-assayable amounts of interleukin 1beta, tumor necrosis factor alpha, and transforming growth factor beta(1). Hence, PUF/SF scaffolds embody a novel class of biomaterials favoring the adhesion, proliferation, and performance of specific metabolic tasks by HAFs without eliciting any concurrent secretion of the chief proinflammatory cytokines.

Loss of posterior silk gland transcription specificity of fibroin light chain promoter due to absence of 41 bp sequence containing possible inhibitor binding sites.

The gene encoding fibroin light chain protein (FibL) is specifically expressed in the posterior silk gland of silkworm and repressed in other tissues. The binding sites of several transcription factors involved in the silk gland transcription specificity of fibl promoter have been recognized, including SGFB, PSGF and BMFA. Here we report the leak expression of the enhanced green fluorescent protein (EGFP) reporter gene in tissues other than the posterior silk gland in vivo when under the control of a shortened fibl promoter with deletion of the 5' terminal 41 bp sequence, which is located at -650 nt to -610 nt upstream of the fibl transcription starting site. Assay of silk gland specificity of the promoters was performed by observation of green fluorescence in tissues of silkworm larvae following inter-haemocoelic injection of recombinant Autographa californica multiple nuclear polyhedrosis virus carrying the EGFP reporter gene controlled by different lengths of fibl promoters. Our results indicated that availability of the binding sites of several known factors, including SGFB, PSGF and BMFA, is not sufficient for intact silk gland transcription specificity of fibl promoter, and there are possible inhibitor binding sites in the 41 bp sequence (-650 nt to -610 nt) upstream of the transcription starting site which may be required to repress the activity of fibl promoter in other tissues.

Structural analysis of fibroin heavy chain signal peptide of silkworm Bombyx mori.

To study the minimal length required for the secretion of recombinant proteins and silk proteins in posterior silk gland, the signal peptide (SP) of the fibroin heavy chain (FibH) of silkworm Bombyx mori was systematically shortened from the C-terminal. Its effect on the secretion of protein was observed using enhanced green fluorescent protein (EGFP) as a reporter. Secretion of EGFP fusion proteins was examined under fluorescence microscope. FibH SPs with lengths of 20, 18, 16 and 12 a.a. can direct the secretion of the reporter, yet those with lengths of 11, 10, 9, 8 and 1 a.a. can not. When the FibH SP was shortened to 12 a.a., the secretion efficiency was decreased slightly and cleavage occurred within EGFP. When 16 a.a. of the FibH SP were used, the secretion of fusion protein was normal and the cleavage site was between the Gly-Ser linker and Met, the starting amino acid of EGFP. These findings are applicable for the expression of foreign proteins in silkworm silk gland. The cleavage site of the SP is discussed and compared with the predictive results of the SignalP 3.0 online prediction program.

Proteomic studies of isoforms of the P25 component of Bombyx mori fibroin.

It is recognized that P25 is one of three polypeptide components of the fibroin synthesized in the larval silk gland (SG) of silkworm, having two glycosylated isoforms. In the present study, however, eight P25 isoforms were separated by proteomics, including two-dimensional gel electrophoresis of whole SG proteins, and were identified by the peptide mass fingerprinting method. Four of the eight isoforms were identified as Bombyx mandarina P25s, although the SG of Bombyx mori has never been considered to contain the P25 from B. mandarina. It is suggested that this diversity of P25 isoforms depends on phosphorylation modification in addition to glycosylation.

[Study of enzyme immunosensor immobilized by regenerated silk fibroin].

The author studies the enzyme immunosensor made up of graphite electrode and protein film of regenerated silk fibroin which is employed to immobilized antigen (rabbit IgG). IgG will be recognized and combined by antibody (goat-anti-rabbit IgG-HRP). After enlarging the signal of the combination of antigen and antibody by H2O2, the concentration of IgG is able to be measured by testing the electrode potential directly. The Enzyme Immunosensor is sensitive when measuring IgG and the detection limit of 1.2 x 10(-10) mol/L is found. It shows linear response over the concentration range of 4.1 x 10(-7)-1.2 x 10(-10) mol/L. Electrophoresis cuts down the time of antibody-antigen interaction from 90 min to 30 min. The response time is 15 s. Enzyme immunosensor with better stability and higher sensitivity can be used extensively in clinical diagnosis, medical and environmental studies, HLA molecular diagnosis and so on.
