Ultrastructural localization of phosphatases in the testes of the domestic fowl: acid phosphatase and thiamine pyrophosphatase.

ACPase and TPPase activity has been examined in the germinal epithelium of the testes in the domestic fowl. ACPase activity in spermatogonia and spermatocytes was confined to the Golgi complex. In spermatids ACPase activity was seen in the endoplasmic reticulum and nuclear envelope in the phase I and especially in the phase II (the elongating phase). This activity gradually decreased during the next phase III (the elongating phase). This activity gradually decreased during the next phase III, and had disappeared in the final phase IV. The membrane body showed ACPase reaction in the small peripheral vacuoles and cisternal structures surrounding large central vacuoles. ACPase was also present in vesicles surrounding the developing tail. Late spermatids showed an abundance of autophagic vacuoles which had a complex array of ACPase positive delimiting membranes. In Sertoli cells ACPase activity was predominant in the lysosomes. TPPase activity was seen in the cisternae of the Golgi complex in spermatogonia and spermatocytes. In spermatids activity was present in the endoplasmic reticulum during the phase II, but it is lost in later stages. The smaller vacuoles and cisternal structures in the membrane body also showed reaction products. According to the present results it is thought likely that the smaller vacuoles and cisternal structures of the membrane body are of endoplasmic reticulum origin. The autophagic vacuoles in spermatids and the lysosomes of Sertoli cells are considered responsible for the degradation of residual bodies cast off by spermatids.

Behavioral and hormonal correlates of social dominance in stable and disrupted groups of male domestic fowl.

An experiment was conducted to determine the behavioral and hormonal correlates of social dominance in stable and disrupted groups of male domestic fowl (Gallus domesticus). In addition, the relative importance of visual and auditory cues in the maintenance of dominance status in this species was assessed. Alpha roosters were removed from their six-bird home pens for 1 week, and placed in either (1) and adjacent pen in visual contact with penmates, (2) and adjacent pen where visual, but not auditory, contact was prevented, or (3) a different building where they were in visual and auditory isolation. Dominance rank, aggressive activity, and crowing frequency were intercorrelated during the premove period. There was a significant increase in both aggression and crowing among the males remaining in the home pen following removal of the alpha male, except in the treatment in which males were still in visual contact with the removed alpha, in which only aggression increased. Increases in crowing frequency were also noted in alpha males. High levels of aggression occurred when alpha males were returned except in those pens in which visual contact had been maintained. Visual cues thus appear to be more important than auditory cues alone with respect to the maintenance of dominant social status in roosters. There were no clear correlations between behavioral variables and plasma androgen or corticosterone levels in either the stable or disrupted flocks, and no consistent changes in the levels of these hormones as a result of experimental manipulation; several possible explanations for this finding are discussed.

The efficacy of praziquantel (Droncit R) against Raillietina tetragona (Molin, 1958) in domestic fowl.

The efficacy of different dose rates of Praziquantel (Droncit R) in tablet and liquid (injectable) form against Raillietina tetragona in the domestic fowl was studied. A dose of 10 mg kg-1 of Praziquantel in tablet form and 0.15 ml kg-1 in liquid form was found to be effective against R. tetragona infection irrespective of age, sex and intensity of infestation of the host. Intramuscular (IM) administration of liquid Praziquantel was found to be more effective than subcutaneous (SC) administration. Susceptibility of the parasite to this drug increased with age.

Comparison of running energetics in male and female domestic fowl.

Carbon dioxide production (VCO2) and end-exercise venous lactate concentration were compared in intensively-trained, mature male and female domestic fowl running on a treadmill for 10-min periods at maximal and sub-maximal work loads. The relationship between VCO2 and running speed was linear and had the same slope in male and female birds. However the maximum sustainable running speed of the males was approximately twice that in the females and the maximum weight-specific VCO2 was 60% higher in males. End-exercise venous lactate concentration rose significantly above resting at work loads above 75% maximum in females, but only at work loads above 95% maximum in fully-trained males. The results indicate a greater work capability in male than in female birds in the conditions of the experiments.

The occurrence of lipid in the oxyntico-peptic cells of the proventriculus of the fasting domestic fowl.

A histological study has shown that considerable amounts of lipid accumulate in the oxyntico-peptic cells of the proventriculus of domestic fowls deprived of food. Lipid droplets were first seen in the cells 6 hours after commencing the fast and they reached a maximum in 24 hours; a decrease was detected histologically 2 hours after re-feeding and all lipid had disappeared 8 hours after access to food. Lipid was observed in fasted chickens aged from 1 to 27 weeks, in four different breeds, and in certain naturally occurring diseases. Small amounts of lipid were also seen in the proximal convoluted tubules of the kidneys of chickens which had been fasted. The relationship of the lipid to the metabolism of the oxyntico-peptic cells and to certain disease states is discussed.

Enzyme variants of Eimeria parasitizing the domestic fowl and possibilities of species diagnostics.

Electrophoretic variation of the enzymes lactate dehydrogenase (LDH) and glucosephosphate isomerase (GPI) of Eimeria parasitizing the domestic fowl in Czechoslovakia is summarized and the differentiation of species of poultry coccidia is discussed. A new method for evaluation of zymograms of coccidial enzymes is presented. This method enables the results of different experiments to be compared by calculating standardized rates of mobility of each enzyme band relative to the positions of reference variants coded LDH-8 or GPI-9.

Long-term vasoligation in the domestic fowl (Gallus domesticus).

The effects of long-term (14 months) unilateral vasoligation on the tests and their excurrent ducts of the domestic fowl were studied histologically and ultrastructurally. Severe testicular degeneration and epididymal atrophy were observed in the ipsilateral organs while the contralateral control organs were normal. Massive macrophage activity was observed in the epididymal regions and to a lesser extent in some severely degenerated seminiferous tubules. Several granulomata occurred in the epididymal regions and the ducti deferentes, and most were resorbed or were undergoing resorption at the time of examination.

The passage of spermatozoa through the vitelline membrane in the domestic fowl, Gallus gallus.

The developing outer layer of the vitelline membrane of the ovum in the posterior part of the infundibulum of the domestic fowl contains many spermatozoa in nearly parallel orientation with its inner layer. When the acrosomal region of a spermatozoon approaches or contacts the inner layer, promptly undergoes the acrosome reaction. The outer acrosomal membrane and overlying plasma membrane fuse together and the apical region of the acrosome opens, so that the acrosomal contents are released. Meanwhile the spermatozoon remains a time in contact with the surface of the inner layer, and the network of the inner layer just under the tip of the sperm head begins to be dissolved. This dissolution extends downward forming a tunnel, approximately 9 micrometer in diameter. The spermatozoon then passes through the inner layer obliquely via the central region of the tunnel and arrives at the perivitelline space.

Effects of corticosteroids on short-circuit current across the cecum of the domestic fowl, Gallus domesticus.

Both avian corticosteroid hormones, aldosterone and corticosterone, increased short-circuit current across the wall of the ceca of the domestic fowl (Gallus domesticus) in vitro. About 80% of this short-circuit current was inhibited by the Na-channel blocking drug amiloride. Corticosterone was about ten times less potent than aldosterone in increasing short-circuit current and it exerted a similar maximal effect. Cortisol (an endogenous corticosteroid hormone in mammals but not birds) was about ten times less potent than corticosterone and this difference appeared to reflect the presence of the 17 alpha-OH group in cortisol. Carbenoxolene, which inhibits 11 beta-hydroxysteroid dehydrogenase, increased the effect of corticosterone. This effect is consistent with inhibition of the metabolism of corticosterone to 11-dehydrocorticosterone. The latter was found to be about 100 times less potent than corticosterone. The effects of both aldosterone and corticosterone (also dexamethasone) were abolished by the mineralocorticoid receptor antagonist spironolactone. The results suggest that corticosterone has an effect similar to aldosterone but in vivo its action may be depressed by the activity of 11 beta-hydroxysteroid dehydrogenase. The sensitivity of the cecal preparations to corticosterone indicates that this hormone could contribute to the regulation of transcecal Na transport (absorption) in vivo.

Effects of age, sex and housing on the trabecular bone of laying strain domestic fowl.

To determine the effects of age, sex and housing on trabecular bone volume, samples were collected from groups of male and female domestic fowl housed in cages or floor pens from four to 60 weeks old. Between 25 and 60 weeks old, trabecular bone volume decreased by 25 per cent in sections of free thoracic vertebrae (T5) from female birds, the loss occurring at an earlier age in caged birds. Over the sample period, TBV in male caged birds diminished by 35 percent, but male floor birds showed no reduction in trabecular bone volume. At 60 weeks, trabecular bone volume was 30 per cent greater in male caged birds and 40 per cent greater in male floor birds than in the corresponding females. In reproductively active females, no trabecular osteoid was observed, indicating no new trabecular bone formation. However, trabecular osteoid was present in two birds aged 60 weeks which had regressed ovaries. Osteomalacia was not seen in any of the bone samples.

Creatine kinase isoenzyme profiles in the plasma of the domestic fowl (Gallus domesticus): effects of acute heat stress.

Creatine kinase isoenzyme activities in extracts of plasma, skeletal muscle, heart and brain tissue of domestic fowls were separated by anion exchange chromatography and tissue specific distributions of the isoenzyme designated MM-CK, BB-CK1 and BB-CK2 were demonstrated. The muscle isoenzyme (MM-CK) was the predominant form in plasma (99 per cent) and its activity increased in response to an episode of acute heat stress.

Calbindin, tyrosine hydroxylase and opioid-like immunoreactivity in the intestinal nerve of Remak of the domestic fowl.

Four classes of neurons were identified in both juxta-jejunal and juxta-rectal ganglia of Remak's nerve of the domestic fowl using double-labeling immunohistochemistry. Neurons immunoreactive (IR) for tyrosine hydroxylase (TH) formed a mutually exclusive subpopulation from neurons displaying calbindin (CaBP)-IR. Between 48-72% of juxta-jejunal neurons labeled for TH whereas 36-57% of juxta-rectal neurons displayed TH-IR. CaBP-IR was present in 18-40% of juxta-jejunal neurons; this increased to 31-46% in juxta-rectal neurons. The majority of CaBP-IR neurons (78-85%) also displayed opioid (beta-EP)-IR. Within each ganglion a small percentage of neurons (4-18%) were non-IR with any of the three antibodies. This is the first report of an immunohistochemically identified subpopulation of non-catecholaminergic neurons within the juxta-jejunal ganglia of Remak's nerve. It is proposed that these perikarya are a major source of the CaBP-IR and opioid-IR nerve fibers found in the chicken gut.

Location of motoneurons innervating the musculus sphincter cloacae in the domestic fowl.

Horseradish peroxidase was injected into the musculus (m.) sphincter cloacae through the mucosa from the lumen side of the cloaca in the domestic fowl. Labeled neurons were found in the ventrolateral area of the ventral horn from the lumbosacral segments 7-9. The craniocaudal distribution of labeled neurons overlapped with that of hindlimb motoneurons. The m. sphincter cloacae is innervated by motoneurons sending axons to the sphincter cloacae either through the connexus caudalis branching from the caudal coxal nerve of the sacral plexus or through the lateral caudal nerve arising from the pudendal plexus.

Relationship between thinning of eggshells and reduction in number of eggs produced after administration of some saluretic drugs in domestic fowls.

1. The effect of some diuretic drugs (acetazolamide, hydrochlorothiazide, chlorthalidone, furosemide, ethacrynic acid and amiloride) on eggshell formation and egg production in domestic fowls were investigated. All of the tested compounds significantly inhibited eggshell formation, furosemide being the most potent drug. 2. The number of laid eggs was also reduced by several of the diuretics. There was a correlation between reduction of shell thickness and number of eggs (r = 0.77; P less than 0.01). 3. If the tested diuretics were subdivided into two groups according to their different modes of action, where furosemide, ethacrynic acid and amiloride represent sodium transport inhibitors, and acetazolamide, hydrochlorothiazide and chlorthalidone (contributory action of the two latter) are inhibitors of carbanhydrase, the highest correlation coefficient (r = 0.96) between reduction of shell thickness and egg production was found for sodium transport inhibitors. The corresponding correlation coefficient for carbanhydrase inhibitors (r = 0.44) was not significant. 4. The probable mechanisms of action of the diuretics on eggshell formation and ovulation are discussed.

Changes in somatomedin-like activity and growth hormone concentrations in the plasma of the domestic fowl (Gallus domesticus).

Variations to immunoreactive growth hormone (GH) and bioassayable somatomedin-like activity (SLA) were determined in the plasma of the domestic fowl. The plasma concentrations of SLA (potency relative to a pooled serum standard) and GH were both inversely age-related in growing cockerels (Thornber 909) between 3 and 20 weeks of age (SLA, r = 0.454; P less than 0.01, GH, r = 0.548; P less than 0.01) and directly related to each other (r = 0.54; P less than 0.01). In immature birds maintained on a photoperiod of 16 h light, 8 h darkness, plasma concentrations of SLA were highest during the first hour of darkness (1.19 +/- 0.18 units/ ml (u/ml) mean +/- S.E.M., n = 6) and significantly lower 1 h after dawn (0.78 +/- 0.07 u/ml n = 6). No diurnal variations in plasma concentrations of GH were observed. Deprivation of food in two strains of chickens (Thornber 909 and broiler) significantly reduced plasma concentrations of SLA in both strains and increased plasma concentrations of GH in the Thornber strain. Re-feeding fasted Thornber chickens caused the plasma concentrations of SLA and GH to return to normal within 24 h but had no effect in broilers. Deprivation of water, which also reduces food intake, for 12, 18 and 24 h markedly reduced plasma concentrations of SLA but had no effect on plasma GH concentration in Thornber cockerels.

Effects of anti-inflammatory steroid drugs on pain coping behaviours in a model of articular pain in the domestic fowl.

The optimum dose rates of betamethasone, dexamethasone (sodium phosphate) and methylprednisolone (acetate salt) were determined for the treatment of inflammatory pain in domestic fowl using the microcrystalline sodium urate model of articular pain. The response criteria were the changes in behavioural profiles and pain-related activity over 60 minutes commencing 1 hour after an intra-articular injection of sodium urate and a course of intramuscular injections of a range of doses of each drug on the previous 2 days. The optimum dose rates for betamethasone, dexamethasone and methylprednisolone respectively were 0.04, 0.06 and 2 mg kg(-1)and are in the range of the recommended doses for these drugs in mammals.

Immunohistochemical demonstration of myoid cells in the testis and its excurrent ducts in the domestic fowl.

(1) Immunohistochemical methods and three antibodies (against actin, desmin and smooth muscle actin) were used to demonstrate the myoid cells in the domestic fowl testis and its excurrent ducts. (2) A positive reaction to actin, smooth muscle actin and desmin was found in the myoid cells of peritubular tissue of the testis and in rete testis, ductuli efferentes and ductus epididymidis. (3) In the testis myoid-reactive cells form a single layer. In the rete testis, ductuli efferentes and the ductus epididymidis reactive myoid cells form a main component of the stroma. (4) Positive reaction to actin, smooth muscle actin and desmin was also observed in the myoid cells of the tunica albuginea and in the wall of blood vessels in the testis and epididymis, indicating a contractile function for the testicular capsule.
