In inflammatory macrophages, plasminogen activator exists in two active forms, a soluble form released into the extracellular medium and a cell-associated form. This communication describes some properties of the cellular form of plasminogen activator, in intact macrophages and in cell lysates. Cellular plasminogen activator is a membrane protein, associated with the outer face of the plasma membrane; in intact macrophages, it participates in the activation of exogenous plasminogen and, thus, has to be considered as an ectoenzyme. A plasminogen activator activity can be detected in cell lysates (macrophage monolayers lysed in 0.1% Triton X-100) only when plasmin production is followed by the use of small synthetic substrates because a soluble inhibitor, released during extraction, blocks plasmin fibrinolytic activity. In these lysates, plasminogen activator molecules exist as high molecular weight unstable complexes exhibiting a high affinity for plasminogen.

The surface charge of semiconductor nanoparticles, Q, is an important parameter which determines their electrokinetic behavior, stability in water and polar solvents, functions of optical and electronic devices, self-assembly properties, and interactions with cell membranes. We have developed a simple method for quantitative determination of Q in their native aqueous environment. The method does not require the knowledge of exact atomic structure or make assumptions about effects of drying on charge distribution. The method is based on titration of nanoparticle dispersion with a solution of oppositely charged polyelectrolyte. The point of complete neutralization is recognized as an inflection point on the dependence of fluorescence intensity on the amount of polyelectrolyte added. Thioglycolic acid-stabilized CdTe nanoparticles 2 nm in diameter were found to carry an average Q from -2.6 to -5.5 for pH 7.5 to 10, respectively. This charge is found to be smaller than that calculated theoretically for an analogous structure (i.e., Q = -8), presumably due to adsorption of Cd(2+) ions on the stabilizer shell and on Te atoms with unsaturated valence located on the side planes of CdTe tetrahedrons.
Vertically aligned ZnO nanorods with uniform diameter and length have been synthesized on a zinc foil substrate with ammonium persulfate as oxidant via a facile, larger scale production and inexpensively synthesized method without any templates or additives. SEM and XRD studies indicate that ZnO nanorods are well-oriented along the c-axis. The PL spectrum indicates that our as-synthesized ZnO nanorods with a stronger and wider green emission are promising candidates as electron nanoconductors in nano-optoelectronic devices. Furthermore, by an effective thioglycolic acid-assisted solution route, well-aligned ZnO/ZnS nanocable and ZnS nanotube arrays have been successfully synthesized. ZnS nanotubes show a perfect hexagonal and obvious tubular shape. Our present strategy shows mild growth conditions and good reproducibility.
A novel and facile preparation method for layer-by-layer (LbL) self-assembled films incorporating quantum dots (QDs) and having intense photoluminescence (PL) from blue to red is presented. Functional sol-gel-derived glass layers prepared by the hydrolysis of 3-aminopropyltrimethoxysilane (APS) or 3-mercaptopropyltrimethoxysilane (MPS) have been used as a linkage between QD layers. Absorption, PL spectroscopy, transmission electron microscopy, and atomic force microscopy were employed for characterization, which revealed that the QDs in the prepared films had a nearly close-packed coverage and were not aggregated. The PL efficiencies of the QDs (CdTe or ZnSe, both are thioglycolic acid-stabilized) dispersed in the films were roughly half that of the initial colloidal solutions but reached 24% before a refractive index correction. The thickness of the red-emitting film with 10 CdTe QD layers was approximately 50 nm. The concentration of QDs in the film derived from the first absorption peak was approximately 0.01 M. Because the PL starts to show a red shift, the obtained concentration is practically the ultimate one in the glass matrix. The mercapto, amino, and carboxyl groups play important roles in LbL self-assembling processes.
A full-length C-reactive protein (CRP) cDNA clone has been isolated from a CBA/J-strain-mouse acute-phase liver library. The 1614-nucleotide cDNA specifies mRNA 5' and 3' untranslated regions of 81 and 858 bases respectively that flank 675 bases encoding mouse pre-CRP. The derived amino acid sequence predicts a 19-residue leader peptide followed by a 206-residue mature mouse CRP that shows considerable sequence identity with both human and rabbit CRP. Northern-blot analysis of mouse liver CRP mRNA concentrations after inflammatory stimuli and comparison with hepatic induction of mRNA for the major mouse acute-phase protein serum amyloid P component established that CRP, a major acute-phase reactant in human and rabbit, is a minor acute-phase reactant in mouse. The size and organization of the mouse CRP mRNA 5' and 3' untranslated regions are significantly different from those of human and rabbit CRP mRNA and may have implications for its anomalous minimal induction during acute inflammation.

Partial characterization of Fusobacterium necrophorum protease was investigated. The protease was partially purified by gel filtration with Toyopearl HW 55. The final preparation was inactivated completely by heating at 60 degrees C for 30 min and inhibited by ascorbic acid, sodium thioglycollate and p-hydromercuribenzoate. Antibody response to the protease was demonstrated in mice receiving 10(4) CFU of F. necrophorum.
