[Cell culture and its application. Culture systems of normal human epithelial cells and their potential applications in cancer research].

In cancer research, it is essential to understand the differences of normal and carcinoma cells. To date there are not many studies of normal human epithelial cells because of difficulties in obtaining human materials and culturing them. After Peehl and Ham developed a low calcium medium which does not require feeder cells, the culture of normal human epithelial cells became relatively easy. In this paper, I will describe and compare different culture methods for normal human epithelial cells (mainly keratinocyte culture systems). We also present establishment processes of normal human ectocervical epithelial cell culture system that is dramatically improved by a combination of serum supplemented high-calcium and serum-free low-calcium media according to changes of a physiological state of the cells by passages. This suggest that we have to be aware of changes of cells in responses and use them in developing culture systems. I will also describe a study of mouse epidermal keratinocyte culture system and discuss the use of normal human epithelial cell system in screening potential chemotherapy reagents. In the study normal epithelial cells inhibit growth of initiated cells and this inhibition can be modified by tumor promoters and their inhibitors.

[Application of hydroxyapatite in micro-otosurgery: experiment with guinea pigs and clinical application].

OBJECTIVE: To investigate the feasibility of hydroxyapatite (HA) middle ear prostheses in otosurgery. METHODS: In the animal experiment HA prostheses were put in the bilateral bullae of the ears of 12 healthy white guinea pigs (totally 24 ears). Three guinea pigs were killed every month 1-4 months later. Morphological examination and electron microscopy were conducted to observe grafts. In the clinical experiment HA prostheses was used in ossiculoplasty in 31 cases, reconstruction of posterior wall of external auditory canal in 5 cases, and mastoid obliteration in 11 cases. RESULTS: The HA prostheses taken out from 23 ears of the guinea pigs were with normal and complete structure, and clear margin in gross anatomy and microscopically. Only one ear suffered infection. Follow-up or the patients for 1-4 years showed excellent healing of the operated parts and an improvement of hearing threshold by about 16 dBHL. CONCLUSION: Hydroxyapatite with excellent physical and chemical properties and biological tolerance HA is a good material in ossiculoplasty, canaloplasty, mastoid obliteration, and other otological operations.

Remineralisation of carious enamel lesions after application of a CHX/F-mouthrinse compared with sole CHX- and placebo-application.

PURPOSE: The purpose of this cross-over, double-blind, placebo-controlled, randomised in situ study was to evaluate the remineralisation of demineralised enamel specimens using a CHX/NaF mouthrinse in comparison with a CHX- and a placebo-mouthrinse. MATERIALS AND METHODS: Twenty-four volunteers received intraoral appliances with mounted demineralised enamel specimens. They rinsed their mouths twice a day (10 ml for 1 minute) with the respective preparation. After 14 days, the in situ mineral gain and fluoride uptake were determined. RESULTS: Mineral gain was significantly higher after using CHX/F mouthrinse (358.4 +/- 372.1 Vol.% X microm) compared with CHX (95.6+/-192.2 Vol.% X microm) and placebo treatment (80.8 +/- 159.6 Vol.% x microm). Moreover, CHX/F treatment (2751.1+/-1494.9 microg/cm(3)) resulted in a significantly higher fluoride uptake compared with CHX- (83.8+/-94.0 microg/cm(3)) and placebo- (136.6+/-137.3 microg/cm(3)) mouthrinses. CONCLUSION: The in situ study approves that using a CHX/F mouthrinse supports the remineralisation of initial carious lesions.

[Application of occlusal splint for the patient with temporomandibular joint dysfunction. An application of silane coupling treatment: a case report].

The bite plane therapy has been one of the most useful treatment methods for patients with temporomandibular joint (TMJ) dysfunction. But the bite plane usually causes esthetic and articulation disturbances and it is occasionally difficult to use during mastication. The authors have fabricated a new type of bite plane, named occlusal splints, which covers each side of the premolars and molars. The occlusal splint was constructed by a hard-resin for the occlusal part and cast clasps for the retentive part and these parts were adhered after a pre-treatment of metal by the use of a silane coupling agent (Silicoater system). The patient was a 35 year-old man, with a chief complaint of left side reciprocal TMJ clicking. The anterior repositioning type of bite plane was applied for the treatment with the diagnosis of anterior disk displacement with reduction. But he complained of esthetic and articulation disturbances especially during working and eating. After four months we fabricated an occlusal splint that can be used for a long period of time. The patient was satisfied to use this occlusal splint and no symptoms of TMJ dysfunction were found with this appliance. It is suggested that the occlusal splint consisting of a hard-resin and cast clasps is a useful appliance especially for the purpose of use during working and eating, and for evaluating the properness of mandibular position.

[Ethically chosen studies with early-weaned piglets during their raising in pens with different applications of straw. 1. The effects of different applications of straw and different floor conditions in areas of uniform size].

In extension of earlier experiments in housing systems with perforated floors, utilization rates of solid floor with litter, straw in a rack and deep litter, as well as the behaviour and physical condition, especially of claws, were investigated. Each pen had an area of 0.45 m2/piglet. In further experiments the influences of race (DL instead of DL x Piétrain), kind of rearing (without straw instead of rearing with straw), lowering temperature and the effect of a preferred perforated floor in comparison with solid floor were investigated. Deep litter was preferred for activity and lying behaviour only in the case of low temperature (14-18 degrees C) whereas solid floor with litter was preferred in the case of higher temperature (19-25 degrees C). The perforated floor was equivalent to solid floor on the condition that the area dimension was sufficient, temperature 3 degrees C higher than in the other experiments and straw in a rack was offered. However, the utilization rates of solid floor increased in the case of lowering temperature by 4 degrees C. There was no significant influences of the different race or kind of rearing. In the experiments there were significant signs of a more harmonious condition of the animals and a shorter, respectively no period where the animals had to adapt to the housing conditions. These findings were in contrast to those of earlier investigations. The possibility of acting with straw led to an undisturbed circadian rhythm and to a higher rate of standing within the total activity which was as high as in earlier studies. Further social companions and environmental objects lost attractiveness.(ABSTRACT TRUNCATED AT 250 WORDS)
