[Studies on fluoride concentration and cholinesterase activity in rat hippocampus].

OBJECTIVE: To study the accumulation of fluoride in rat hippocampus and its effect on cholinesterase activity. METHODS: Rats were subchronically exposed to NaF, and fluoride concentration and cholinesterase activity in rat hippocampus were determined. RESULTS: Fluoride concentration in rat hippocampus was significantly correlated with the dosage of fluoride, and there were significant differences among high dosage group [(13.03 +/- 1.79) micro g/g], low dosage group [(9.83 +/- 0.92) micro g/g] and control [(8.27 +/- 1.11) micro g/g], P < 0.01. Acetylcholinesterase activities among three groups [(0.111 +/- 0.031) micro mol/mg, (0.143 +/- 0.025) micro mol/mg, (0.183 +/- 0.027) micro mol/mg] were also significantly different (P < 0.01), which was negatively correlated with fluoride concentration in rat hippocampus (r = -0.700, P < 0.01). The activity of butylcholinesterase in high dosage group [(0.041 +/- 0.010) micro mol/mg] was different from that of control [(0.067 +/- 0.025) micro mol/mg, P < 0.05], but the activity was not significantly related with fluoride concentration in rat hippocampus (r = -0.317, P = 0.094). CONCLUSION: Fluoride may go through the blood-brain barrier and accumulate in rat hippocampus, and inhibit the activity of cholinesterase.

[Study on the defluoridation of drinking water with model-La cation exchange resin].

Model-La cation exchange resin was transformed from strong acid cation exchange resin, which was used for the defluoridation of drinking water. The 001 x 7 strong acid cation exchange resin was transformed into model-La cation exchange by soaking in the La(NO3)3 solution. The F- in the water was removed by model-La cation exchange resin under the optimum condition because it was combined into fluoride with the La3+ in the model-La cation exchange resin. The used resin was regenerated with the La(NO3)3 solution and was used to defluoridate many times. The optimum conditions of transform and defluoridation and regeneration were d(rasin) = 0.315-0.600 mm, V(rasin):V(regeneration) = 1:6, t(transform) = 48 h, T = 298.16 K(25 degrees C). The defluoridation capacity was 5.60 mg/g in column test, and 4.08 mg/g in batch test. The model-La cation exchange resin could be used for defluoridation for 8 times. The results suggest that the model-La cation exchange resin is a novel material of defluoridation. This method is easy to master and the pH value of the solution doesn't need to be regenerated that the cost of defluoridation can be reduced significantly and it can be used for a long time.

A case study of domestic defluoridation of ground water in Jaipur, India.

This study was undertaken to assess the success of the intervention programmes of Centre for Community Economics and Developmental Consultants Society (CECOEDECON) initiated in the rural areas of Jaipur district of Rajasthan, India. The programme focussed on capacity building among villagers to create awareness about fluorosis and used Nalgonda technique for removal of excess florides from ground water in selected areas. The data and information were collected from respondents in six village related to aworeness and incidence of fluorosis, awareness regarding the CECOEDECON project and the use and effectiveness of domestic defluoridation technology. Analysis of collected data revealed that level of awareness was highest (68%) among the respondents of Barkheda and Yarlipura villages where the project was withdrawn in 1998. It was observed that the level of awareness decreased over time and it was found lowest in the villages where project was withdrawn in 1996. About 87% of the respondents reported that most of the women are members of the Mahila Mandal started under CECOEDECON project. Some of the respondents (38%) mentioned that they were experiencing problems after the withdrawal of project teams. Thus, it is evident that follow up programmes play an important role in sustenance of the use of defluoridation techniques in villages along with the infrastructure to take care of minor repairs and supply of chemicals.

[Features of occupational fluorosis course].

Analysis of X-ray signs in 397 patients suffering from fluorosis revealed degenerative and dystrophic lumbar disorders in 94%, calcified anterior longitudinal ligament in 65%, various changes (epicondylosis, periarthrosis and arthrosis deformans) in 91%. Locomotory disorders appeared systemic, multiple and symmetrical.

[Fluorosis on expression of nicotinic acetylcholine receptors in protein and gene levels in human SH-SY5Y neuroblastoma cells].

OBJECTIVE: To investigate the influence of fluorosis on nicotinic acetylcholine receptors (nAChRs) in protein and gene levels in SH-SY5Y cells and the mechanism of the receptor modification. METHODS: SH-SY5Y cells, a human neuroblastoma cell line, were incubated with different concentrations of fluoride or with antioxidant for 48 hours. The functions of cells were measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) method, and protein oxidation detected by carbonyl content; the alpha3 and alpha7 nAChR subunits in protein level were measured by Western blotting and in mRNA level by RT-polymerase chain reaction (RT-PCR). RESULTS: In high-dose group as compared to the control, the decreased MTT (49%), increased protein oxidation (72%), and lower expression of alpha3 (51%) and alpha7 (47%) nAChR subunit proteins were obviously observed in SH-SY5Y cells. There were no changes in expression of nAChR subunit mRNAs between the cells treated with fluoride and those un-treated in controls. Prior treatment with antioxidant resulted in preventing the decrease of nAChR protein in cells exposed to the high doses of fluoride. CONCLUSION: Fluorosis should result in damage of cells and the declined expression of nAChRs in protein levels, but no influences on gene expression of the receptors in human neuroblastoma neurons. The decreased nAChR proteins might be involved in the mechanism of oxidative stress induced by fluorosis.

[Experimental studies of pathogenesis of chronic fluoride intoxication].

The article presents the results of studies of occupational fluorosis pathogenesis on experimental model of chronic fluoride intoxication (CFI). In early fluoride intoxication, fluoride and calcium in the body are in compensatory relations. Later, they are disturbed. High reaction ability of fluoride in CFI is associated with hypocalciemia which triggers parathyroid hyperactivity. This results in hyperproduction of PHH which is unrelated with development of secondary hyperparathyrosis. CFI is accompanied also with elevated calcitonin having a hypocalciemic and hypophosphatemic action. The experiments showed double content of collagen fragments of bone tissue in urine of animals with fluoride intoxication which reflects resorption of bone tissue. Serum osteocalcin rose three-fold as it was unable to include in the bone tissue. There was also activation of lipid peroxidation in subnormal activity of respiratory enzymes.

[Fluorides in urine of the population living in polluted environment].

The increased excretion of fluorides in urine was shown by ion-selective method, in three different groups of workers, inhabitants of Chorzów--the most polluted town in the Upper Silesia. In nearly all workers (90%) of Nitrogen Chemical Plant and Slaughterhouse (groups A and M), the urine fluoride exceeded three to four times the upper limit of the normal range, while only two times in slightly less number of workers (77%) of the Steelworks (group S). The subjective complaints and bone pains were correlated with excessive urine fluoride concentrations in about 40% of workers studied in groups A and M, and in 18% of workers in group S. These results point to an excessive exposition of people to fluorides from the polluted environment, as well as to a high risk of fluorine retention in bones and osteofluorosis.
