[Interactions of vanadate with the contractile system of the rabbit psoas muscle in active, relaxed and rigor states].

Interaction between 0.1-0.3 mM vanadate (Vi) and muscle fibres was studied by measuring their force and stiffness. Only interaction with the fibres having cycling cross-bridges caused a strong decrease of these parameters. There was no vanadate effect on the rigor fibres and its complexes with Mg.AdP or Mg.beta-gamma-imido-ATP. Judging by the following transient tension in Vi-containing solutions Vi was slowly incorporated into relaxed fibres, Vi complex with muscle was stable under relaxation conditions, but it was splitted when actomyosin was formed. Formation of M.ADP.Vi+, the stable biochemical analogue of M.ADP.Pi ( Goodno C. C., Taylor E. W. Proc. Natl. Ac. Sci. USA, 1982, 79, p. 21) is assumed to be formed in muscle. On the basis of the difference between rigor contraction of ATP- and Vi-relaxed fibres a conclusion can be drawn that the Vi complex in muscle is not a mechanochemical analogue of the myosin intermediate developing force when interacting with actin.

[Effect of phallotoxins on the mechanism of Ca2+-activation of glycerinated fibers of the rabbit psoas muscle].

The effect of phalloidin (Ph) and some of its derivatives on isometric tension and spectrum of mechanical relaxation times of single fibres was under investigation. It was shown that Ph had a great effect only at low levels of Ca2+-activation, evoking multi-phase changes of isometric tension and a sharp rise of the rate constant of delayed tension development up to value close to that for control fibres at high [Ca2+]. As to their efficiency to change Ca2+-sensitive parameters of the fibres, phallotoxins can be arranged as follows: Ph approximately equal to Ph-sulfone greater than Ph-sulfoxide B approximately equal to dethio-Ph greater than Ph-sulfoxide A greater than or equal to seco-Ph. There is a qualitative correlation of this sequence with the actin stabilising properties for all these substances with the exception of dethio-Ph. These findings are discussed from the viewpoint of two different "on" actin states, only one of them being Ph-sensitive.

The polymerase chain reaction and post-mortem forensic identity testing: application of amplified D1S80 and HLA-DQ alpha loci to the identification of fire victims.

The application of DNA typing methods after amplification by the polymerase chain reaction (PCR) of DNA derived from body tissues from charred fire victims was investigated. A total of 26 different tissue specimens from ten extensively burned individuals were analyzed. The samples included femoral muscle, psoas muscle, bone marrow and blood. The post-mortem period varied from 38 to 183 h. After amplifying the DNA by PCR from the various tissues, the D1S80 locus was analyzed with a high resolution polyacrylamide gel electrophoresis technique followed by silver staining and the alleles of the HLA-DQ alpha locus were detected by using a reverse dot blot format. All samples could be typed for both loci and the genotypes were consistent in the various tissues from each individual. A parentage test was performed in two cases and Mendelian inheritance of the alleles for both loci was observed.

Comparison between duplex scanning and angiographic findings in the evaluation of functional iliac obstruction in top endurance athletes.

OBJECTIVE: Review of a 10 year-experience, to evaluate the efficacy of pre-operative investigations in the detection of external iliac artery (EIA) endofibrosis in top endurance athletes. DESIGN: Retrospective study. MATERIALS: From September 1995 to March 2004, 13 highly-trained athletes (all men, mean age 32.3 years) underwent surgery for disease involving 14 lower limbs (11 left, one right, one bilateral). METHODS: We compared ultrasound scan (US) and digital subtraction angiography (DSA) data, at rest and at hip flexion with intra-operative findings for all 14 lower limbs. We analyzed the presence of stenosis in the external and common iliac arteries, the presence of psoas muscle arteries and the presence of excessive EIA length. RESULTS: In the affected limbs, before treatment, the mean ankle brachial index (ABI) at rest was 0.98 compared with 0.56 after exercise, p=0.0001. The sensitivities of the US vs DSA examination in the detection of external and common iliac artery stenosis were, respectively, 84.6 and 53.8% vs 53.8 and 12.5%. The muscle psoas artery was detected by DSA with a sensitivity of 57.1 and 100% specificity. For the detection of excessive EIA length, the sensitivity of US was 85.7% with 57.1% specificity. CONCLUSIONS: A fall of ABI after exercise proves the presence of a significant stenosis in symptomatic athletes. Color coded duplex ultrasonography is recommended for non-invasive imaging of suspected endofibrotic stenosis in young athletes, since it detects reliably both stenosis and elongation of iliacal arteries.

Primary structure of the human laminin A chain. Limited expression in human tissues.

cDNA clones for the human laminin A chain were isolated from libraries prepared from human gestational choriocarcinoma cell line (JAR) RNA. They cover approx. 8 kb from the 5'-end of the 9.5 kb mRNA coding for this protein. Our clones contain 94 nucleotide residues for the 5'-end untranslated region and 7885 nucleotide residues of coding sequence. The complete human laminin A chain contains a 17-amino acid-residue signal peptide and a 3058-residue A chain proper. The human laminin A chain has a distinct domain structure with numerous internal cysteine-rich repeats. The large globular domain G has five repeats, which have several conserved glycine and cysteine residues. Furthermore the A chain contains 20 internal cysteine-rich repeats present in tandem arrays in three separate clusters (domains IIIa, IIIb and V). Domain I + II has a predicted continuous alpha-helical structure characterized by heptad repeats and three domains (IVa, IVb and VI) are predicted to contain a number of beta-sheets and coiled-coil structures. Northern-blot analysis was used to study the laminin A chain expression in the JAR cell line, full-term placenta and newborn-human tissues (kidney, spleen, lung, heart muscle, psoas muscle and diaphragm muscle). The expression was detectable in newborn-human kidney and JAR cell line only. The overall amino acid sequence identity between human and mouse is 76%. The human chain has only one Arg-Gly-Asp (RGD) sequence, which is located in the long arm within domain G, whereas the single RGD sequence in the mouse chain is located in the short arm in domain IIIb. The degree of identity between the human laminin A chain sequence and the sequence available for merosin [Ehrig, Leivo, Argraves, Ruoslahti & Engvall (1990) Proc. Natl. Acad. Sci. U.S.A. 87, 3264-3268] is about 41% and when conservative substitutions are included the degree of similarity is 54%.
