[The wasp/mosquito syndrome].

The authors describe for the first time the Wasp/Mosquito syndrome. The allergenic cross-reactivity between Vespula wasp venom and mosquito extract and the reverse rest objectively on several criteria. The frequency of the association between sensibilization to Vespula wasp venom and mosquito: In 10 subjects who had specific IgE to Vespula wasp venom 3 also had mosquito-specific IgE (30%), but out of 11 subjects with positive mosquito-specific IgE 10 also were positive with Vespula wasp venom specific IgE (about 91%). Three observations were documented showing a clinical and biological relationship between sensitisation to mosquito and wasp venom. A first patient had an anaphylactic reaction after a dozen mosquito bites. The allergy assessment confirmed sensitisation to the mosquitos, but also to Vespides (wasp, hornet) venoms. A second patient had an anaphylactic reaction after a wasp sting in 1997. From that date, he had local reactions, indeed loco-regional, to mosquito bites. The allergy assessment confirmed the double sensibilization to mosquito and wasp venom. Finally, a third patient at first had local reactions, also loco-regional, to mosquito bites, then two systemic reactions after mosquito bites. A first allergy assessment, made exclusively to hymenoptera venoms (not to mosquitos), showed a sensitivity to wasp venom and the patient, by mistake, was desensitised to wasp venom, which did not prevent recurrence after mosquito bites. After a second time, the allergy assessment for hymenoptera and mosquito, confirmed a double sensitisation (mosquito and Vespula wasp). Indirect criteria such as parallel increase in specific IgEs to Vespula wasp venom and mosquito in a patient who was desensitised only to mosquito and who had not been re-stung by wasps. Finally on direct criteria, such as: The inhibition technique by CAP RAST, which was positive in two ways: Vespula wasp--specific IgE was inhibited by mosquito extract, and vice versa. Electrophoresis, which showed a protein common to Vespula wasp venom and mosquito extract of M.W. of the order or 42KD and isoelectric point between 4.5 and 5. An immunoblot study identified this common protein with a M.W. of 44KD which is the same as hyaluronidase.

[Antiallergic effects of tranilast in rats and guinea pigs].

Tranilast is an anti-allergic drug. In this study, we made a comparision between the Tranilast synthized by School of Pharmacy WCUMS using new technical and the Tranilast produced by Kissei pharmaceutical Co. LTD, Japan on their antiallergic effects. We found that the two tranilasts had the same antiallergic effects: (1) they inhibit the passive cutaneous anaphylaxis in sensitized rats with the dose of 100, 200 mg/kg(P < 0.01); (2) they inhibit degranulation of mast cells in sensitized rats (10(-5) and 10(-4) mol/L) (P < 0.05); (3) they inhibit schultz-Dale response in sensitized guinea pigs (10(-3) and 10(-4) (mol/L); (4) the inhibit SRS-A release from the lung of sensitized guinea pigs(10(-3), 10(-4) and 10(-5) mol/L) (P < 0.05); and (5) they inhibit the contraction of ileum of normal guinea pigs induced by SRS-A(10(-4) and 10(-3) mol/L).

Inhibition of degranulation and interleukin-6 production in mast cells derived from mice deficient in protein kinase Cbeta.

The antigen-mediated activation of mast cells by means of IgE antibodies bound to the cell surface leads to direct interactions between FcepsilonRI receptor cytoplasmic domains and various intracellular proteins. These interactions initiate diverse signal-transduction pathways, and the activation of these pathways results in the immediate release of proinflammatory agents. A delayed response also occurs and includes the release of various cytokines. It is clear that the activation of kinases is a requirement for the exocytosis observed in mast cells. In addition to the tyrosine phosphorylation of the affected system by soluble tyrosine kinases, activity of protein kinase C (PKC) results in serine or threonine phosphorylation of multiple protein substrates. In this study, we found that mast cells derived from PKCbeta-deficient mice produce less interleukin 6 in response to IgE-Ag. The inhibition of exocytosis in the PKCbeta-deficient mast cells occurred whether the stimuli were due to the aggregation of the mast cell surface FcepsilonRI or to the calcium ionophore, ionomycin. However, no significant changes were observed in the proliferative response of the mast cells to interleukin 3 (IL-3) or in their apoptotic rate after IL-3 depletion. (Blood. 2000;95:1752-1757)

[The wasp-mosquito syndrome: extension of cross-allergenicity to the horsefly].

The crossed allergenicity between wasp venom and mosquito extract was shown during recent work based on clinical observations and correlation studies between different biological parameters, indicating an IgE-dependent biological mechanism. A common protein was identified by Immunoblot. From observation of one of our patients involved in this work, we examined the possibility of the extension of crossed reactivity between wasp, mosquito and horsefly. In effect, our patient presented an anaphylactic reaction with neurological complications from attack on the central grey nucleus, shown by IRM; the immunological study showed a common protein between wasp venom and the total extracts of mosquito and horsefly.

[Parameters of acquired immunity and cation proteins of neutrophilic granulocytes in pulmonary tuberculosis].

Clinical and immunological studies of 61 patients with pulmonary tuberculosis have indicated that the status of acquired and congenital (natural) immunity assessed by the intra- and extracellular levels of cation proteins of neutrophilic granulocytes is different in different clinical forms of pulmonary tuberculosis. The found correlations suggest that there is a relationship between the factors of acquired and congenital immunity and that cytokines are involved in the regulation of specific inflammation. More active degranulation of azurophilic granules of neutrophils occurs due to myeloperoxidase. Unlike other forms, fibrocavernous pulmonary tuberculosis shows a negative correlation between intra- and extracellular content of cation proteins. The findings make it possible to use the parameters of congenital immunity for more detailed characterization of an immune response in tuberculosis, which may be essential in developing new pathogenetic therapy regimens.

[Tissue basophils of the thyroid gland in separate and combined exposure to ionizing radiation and ethanol].

Experiments with male albino rats gave evidence that a single intraperitoneal injection of 15% ethanol at the doses of 0.28 and 2.25 g/kg resulted in enhanced elimination of biologically active substances by tissue basophils (TB) both due to intensified degranulation and greater number of TB in the thyroid gland. Single total exposure to ionizing radiation at the doses of 5, 10, and 20 Gy stimulated the functional activity of TB with simultaneous accumulation of biologically inactive substances. Radio modifying effect of ethanol with respect to thyroid TB consisted of an increase of their number and a decrease in the extent of degranulation which was not dependent on the sequence of radiation exposure and ethanol injection.

[Clinical and experimental study for allergic rhinitis with treatment of biminkang mixture].

OBJECTIVE: To observe the efficacy of Biminkang in treating allergic rhinitis. METHODS: One hundred and sixty-four cases of allergic rhinitis were treated with the Chinese medicine Biminkang mixture mainly consisted of the Kazak folk herb Artemisia rupestis L. and in 52 cases of allergic rhinitis were treated as the control with the patent medicine Biyankang. RESULTS: The total effective rates in both groups are 93.9% and 69.2% respectively. After stopping the medications, the recurrence rates were 46.8% and 87.1%. Biminkang was better than Biyankang in short or long term of effectiveness (P < 0.01). Pharmacological study demonstrated Biminkang played a significant role of resisting histamine, inhibiting allergic reaction I and stabilizing cell membrane of mastocyte. CONCLUSION: Biminkang mixture is an effective drug in treating allergic rhinitis.

Housewives with raw potato-induced bronchial asthma.

Allergy to white potato has rarely been described. We report two cases of atopic patients, housewives, in whom peeling raw potatoes precipitated rhinoconjunctivitis and asthmatic attacks, and, in one of them, contact urticaria. Type I hypersensitivity to raw potato antigens was demonstrated by means of immediate skin test reactivity, specific IgE determination by RAST, basophil degranulation, histamine release test and an immediate bronchial provocation test response to raw potato extract. The controls did not react to any of these tests. Potato allergenic constituent is currently being investigated but, as far as we know, it is heat-labile and has an MW of more than 10 Kd.

Swiss chard hypersensitivity: clinical and immunologic study.

Allergy to vegetables and fruits seems to be more prevalent in atopics, especially in birch pollen-sensitized individuals. We report a case of a grass pollen-sensitized woman, in whom the inhalation of vapor from boiling Swiss chard precipitated rhinoconjunctivitis and asthma. Type I hypersensitivity to Swiss chard was demonstrated by means of immediate skin test reactivity, specific IgE determination by RAST, basophil degranulation, histamine release test, and an immediate bronchial provocation test response to Swiss chard extract. The controls did not react to any of these tests. RAST inhibition assays suggest the presence of some cross-reactivity among Swiss chard and grass pollen antigens, as well as cross-reactivity between vegetables and weed pollens of the chenopod family.

Replication study concerning the effects of homeopathic dilutions of histamine on human basophil degranulation in vitro.

BACKGROUND: Various investigators have observed significant effects of highly diluted histamine on human basophil degranulation in vitro, compared to corresponding water controls. However, active and inactive dilution levels differed in most studies. OBJECTIVE: We aimed to reproduce former studies with flow-cytometry using rigorously controlled experimental conditions to minimise confounding factors. METHODS: In seven independent experiments, basophils of the same human donor were incubated with diluted histamine (up to 10(-34)M) or water controls and activated with anti-IgE antibodies. Basophil activation was determined by using bi-colour flow-cytometry. Experiments were blinded and performed with a randomised arrangement of the solutions on microtiter-plates. RESULTS: Histamine at the dilutions 10(-2)M and 10(-22)M was associated with a significant inhibition of basophil degranulation (p=0.018, Wilcoxon signed rank test) of 23.1% and 5.7%, respectively, if compared to "diluted" water treated in an identical manner. However, if all controls were pooled, only histamine 10(-2)M had a significant effect. Significant effects were seen for row numbers of the microtiter plates. CONCLUSION: We were not able to confirm the previously reported large effects of homeopathic histamine dilutions on basophil function of the examined donor. Seemingly, minor variables of the experimental set up can lead to significant differences of the results if not properly controlled.

IgG antibodies: in vitro blocking activity of IgE mediated reactions.

Sera from 177 atopic patients treated by immunotherapy were screened in order to detect IgD antibodies against a purified fraction of Lollium perenne (fractions C). IgD antibodies were found in 23% of the subjects. Eight of these sera were selected and their IgD isolated by means of immunoabsorption. The in vitro biological activity of the IgD antibodies against fraction C was tested by three methods: basophil degranulation, histamine release and RAST inhibition test. In all these assays, the data obtained indicated a possible blocking effect of the IgD antibodies upon the IgE in vitro mediated reactions.
