Sutureless gingival grafting. A simplified procedure.

Suturing a gingival graft is often the most time-consuming stage of the grafting procedure, especially in regions of difficult access. Previously, it has been suggested that a combination of adhesive cellophane and cyanoacrylate may be used to immobilize the graft without suturing. The technique presented here attempts to simplify further sutureless grafting with the following modifications: - Bandage tape is used instead of adhesive cellophane. - The tape is not placed on the teeth but on the attached gingiva on either side of the graft site. - The use of cyanoacrylate is discarded. The success of the procedure depends on complete haemostasis as any moisture will prevent the tape from adhering to the gingiva. Inherent advantages of sutureless grafting accomplished with the use of bandage tape are: - The graft lies flat on the recipient site and is kept under slight pressure by the tape. - Minimal blood clot is present between the graft and the recipient site. - The "wick" effect of the sutures is eliminated. - The chairside time is greatly reduced. - A shorter appointment and no sutures to remove imply less discomfort for the patient. The technique is contraindicated should the patient present an allergic background to bandage tape.

Relationship between pinworm and urinary tract infections in young girls.

Urinary tract infection is particularly common in young girls and Enterobius vermicularis (pinworm) is one of the most prevalent worms found in children worldwide. Young girls, with or without urinary tract infection, were examined for pinworms in order to explore a possible relationship between these two problems. Of the 55 young girls with urinary tract infection, 20 (36.4%) had pinworm eggs in the perianal and/or perineal region monitored using the cellophane tape method, while 9 (16.4%) of 55 young girls who had never previously had a urinary tract infection were found to have Enterobius eggs in at least one of the cellophane tape tests, and the difference was found to be significant (p<0.05). These results suggested that urinary tract infections may be related to pinworms. When a urinary tract infection is diagnosed in young girls, cellulose tape should be applied to both the perianal and the perineal regions on at least three consecutive occasions.

Comparison of biomass dry weights and radial growth rates of fungal colonies on media solidified with different gelling compounds.

Penicillium commune, Aureobasidium pullulans, and Paecilomyces farinosus were grown on two different media solidified with agar, Pluronic F-127, Carrageenan X-4910, or Carrageenan X-4910 overlaid with cellophane. Growth on Carrageenan X-4910 was generally the same as that on agar, as was the visual appearance of the colonies, e.g., the pigmentation. The Carrageenan X-4910 gels had a melting point, depending on the medium, of 41 to 46(deg)C, and the dry weights of the colonies were readily determined at 60(deg)C. To determine the dry weights of the colonies grown on agar plates, the gels were boiled for 10 min to melt the agar. Comparison of these two procedures showed that the boiling procedure resulted in a 22% reduction of the biomass dry weight. Cellophane membranes did not affect the radial growth rate profoundly. The biomass density was almost halved for P. commune and P. farinosus grown with membranes, whereas the presence of the membrane did not affect the biomass density of A. pullulans. The biomass densities of the colonies grown on Pluronic F-127 were significantly reduced, while in most cases, the radial growth rates of colonies grown on Pluronic F-127 were significantly higher than those obtained on agar or Carrageenan X-4910. Furthermore, the morphology of the leading hyphae was altered, and the hyphal growth unit length was more than twice that obtained on agar and Carrageenan X-4910. Carrageenan X-4910 is a valuable gelling compound for the study of the growth of fungi, as the biomass dry weight is readily determined and growth is similar to that obtained on agar gels.

Expression of Magnaporthe grisea avirulence gene ACE1 is connected to the initiation of appressorium-mediated penetration.

Magnaporthe grisea is responsible for a devastating fungal disease of rice called blast. Current control of this disease relies on resistant rice cultivars that recognize M. grisea signals corresponding to specific secreted proteins encoded by avirulence genes. The M. grisea ACE1 avirulence gene differs from others, since it controls the biosynthesis of a secondary metabolite likely recognized by rice cultivars carrying the Pi33 resistance gene. Using a transcriptional fusion between ACE1 promoter and eGFP, we showed that ACE1 is only expressed in appressoria during fungal penetration into rice and barley leaves, onion skin, and cellophane membranes. ACE1 is almost not expressed in appressoria differentiated on Teflon and Mylar artificial membranes. ACE1 expression is not induced by cellophane and plant cell wall components, demonstrating that it does not require typical host plant compounds. Cyclic AMP (cAMP) signaling mutants delta cpkA and delta mac1 sum1-99 and tetraspanin mutant delta pls1::hph differentiate melanized appressoria with normal turgor but are unable to penetrate host plant leaves. ACE1 is normally expressed in these mutants, suggesting that it does not require cAMP signaling or a successful penetration event. ACE1 is not expressed in appressoria of the buf1::hph mutant defective for melanin biosynthesis and appressorial turgor. The addition of hyperosmotic solutes to buf1::hph appressoria restores appressorial development and ACE1 expression. Treatments of young wild-type appressoria with actin and tubulin inhibitors reduce both fungal penetration and ACE1 expression. These experiments suggest that ACE1 appressorium-specific expression does not depend on host plant signals but is connected to the onset of appressorium-mediated penetration.

Production and characterization of Pseudomon as aeruginosa hemolysin.

Berk, Richard S. (Wayne State University, Detroit, Mich.). Production and characterization of Pseudomonas aeruginosa hemolysin. J. Bacteriol. 84:1041-1048. 1962.-Hemolysin from Pseudomonas aeruginosa was obtained by either extraction of blood agar media after bacterial growth or by saline washing of cellophane plate-grown cells, hemolytic activity being greatest in the latter preparations. Except for variations in heat stability, both types of preparations appeared to be similar if not identical. Complete hemolysis of red blood cells was observed over a wide pH range, although the optima appeared to be below 6.0. Acidification of agar extracts resulted in a coarse precipitate which appeared to be a hemolysin-agar complex. Partial purification of hemolysin present in cellophane washings was obtained by fractionation with ammonium sulfate or by gel filtration. Further characterization of hemolysin is described.

Methods of testing combined antibiotic bactericidal action and the significance of the results.

A DESCRIPTION IS GIVEN OF TWO METHODS OF MEASURING COMBINED ANTIBIOTIC BACTERICIDAL ACTION: a test in liquid medium with subculture and the cellophane transfer method. It is emphasized that information so obtained is necessary in order to predict the effect of combined treatment, particularly in bacterial endocarditis due to organisms not fully sensitive to penicillin. Eight case histories are given, in all of which such a prediction was fulfilled, one of failure and seven of success from the use of five different combinations. The cellophane transfer method was applied to the study of the nature of combined antibiotic action on multiple strains of several bacterial species. The results were rarely uniform for any given combination and species: the necessity for individual tests as a guide to treatment is thus confirmed.Modifications of the theory of combined action formulated by Jawetz are proposed.

Environmental blue light prevents stress in the fish Nile tilapia.

The present study aimed to test the effects of blue, green or white light on the stress response of the Nile tilapia, Oreochromis niloticus (L.). Each color was tested on two groups of isolated adult Nile tilapia (8 replicates each): one being subjected to confinement stress, and the other not (control). A different environmental color was imposed on each compartment by covering the light source with cellophane of the respective color (green or blue; no cellophane was used for white light). The intensity of green, white and blue lights was 250, 590 and 250 lux, respectively. Basal plasma cortisol levels were determined for each fish prior to the experimental procedures. The fish were confined by being displaced toward one side of the aquarium using an opaque partition for 1 h both in the morning and the afternoon of the two consecutive days of the test. At the end of this 48-h period, plasma cortisol levels were measured again. Basal cortisol levels (ng/ml) were similar for each group (ANOVA, F(2;42) = 0.77, P = 0.47). Thus, plasma cortisol levels were analyzed in terms of variation from their respective basal level. After confinement, plasma cortisol levels were not increased in fish submitted to a blue light environment. Thus, blue light prevents the confinement-induced cortisol response, an effect not necessarily related to light intensity.

Induction of nesidioblastosis will reverse diabetes in Syrian golden hamster.

Nesidioblastosis, which is the formation of new islets and the differentiation of cells within the islets, represents part of the spectrum of hyperfunctioning states of the islets of Langerhans at the clinical level. Nesidioblastosis in the Syrian golden hamster can be induced by wrapping the head of the pancreas with cellophane tape. Ligation of the duct is not involved, and acinar cell atrophy does not occur. The purpose of this study was to determine whether the induction of nesidioblastosis could be used as a means of reversing streptozocin-induced diabetes. Outbred hamsters (n = 32), 8 wk of age, were rendered diabetic by treatment with 40 mg/kg i.p. streptozocin, administered daily for 3 days. Four days later, 16 animals chosen at random underwent laparotomy with cellophane wrapping of the pancreas. Before surgery, the serum glucose and insulin levels (means +/- SE) in the unoperated control animals (389.0 +/- 18.6, 33.9 +/- 3.8) did not differ from those in the animals awaiting the operation (373.2 +/- 18.6, 37.9 +/- 3.8). After 7 wk, 50% of the operated animals had serum glucose and insulin levels that were normal, compared to only 12% of the unoperated control animals (chi2 = 5.53, P less than .05). Islets from normoglycemic operated animals were characterized by increased numbers, including many small islets, positive immunoreactive insulin staining, and minimal vacuolation of cells. Islets from hyperglycemic operated hamsters and from the unoperated control animals were decreased in number and generally larger in size, demonstrated little or no immunoreactive insulin staining, and exhibited marked vacuolation of cells.(ABSTRACT TRUNCATED AT 250 WORDS)

Role of the renin-angiotensin system in dogs with perinephritis hypertension.

Cellophane perinephritis hypertension was produced in four dogs, while five additional dogs served as normotensive controls. A competitive antagonist of angiotensin II, 1-sarcosine-8-alanine angiotensin II, was infused iv into these conscious dogs at a rate of 6 mug/min/kg of body weight for 45 min. Arterial pressure averaged 170 +/- 11 (SEM) mm Hg in the dogs with perinephritic hypertension, and was not altered significantly during infusion of the angiotensin antagonist. In the normal dogs the arterial pressure averaged 100 +/- 10 mm Hg and likewise, did not change during administration of the angiotensin analog. Plasma renin activity values were essentially the same in these two groups of dogs and did not change during infusion of the angiotensin antagonist. These studies provide strong evidence that the renin-angiotensin system is not involved in maintaining the elevated arterial pressure in dogs with chronic hypertension produced by cellophane perinephritis.

[Screening Examination and Management of Dermatophytosis by Trichophyton tonsurans in the Judo Club of a University].

Thirty-one members of the Judo Club of a certain university (age: 18~23) underwent a screening examination for dermatophytosis by Trichophyton tonsurans. Test items were: age, sex, height, weight, living mode, exercise duration, number of judo contestants, presence of foreign contestants, occurrence, if any, of dermatophytosis past or present according to a subject's answers to a questionnaire, medical examinations and mycological examinations (KOH, cellophane tape culture, and hairbrush culture). Twenty-four subjects (77%) replied that they had suffered from dermatophytosis in the past, and 8 subjects (26%) had had head eruption in the past. Eleven subjects (35%) had suspicious dermatophytosis at the time of screening; 3 of them were found positive by direct microscopy, 2 of them were positive by cellophane tape culture. Eleven subjects (35%) were found positive by the hairbrush culture, but only 2 had eruption-like folliculitis. The remaining 9 subjects were free from clinical symptoms and were judged to be asymptomatic carriers. As countermeasures, we recommended cleaning and the use of shampoo containing miconazole nitrate. Subjects with suspicious tinea corporis were treated with antimycotic ointment. The 7 subjects who showed more than 5 colonies by the hairbrush culture were treated with 1-week pulse therapy of 400 mg itraconazole, and 3 of these who took a total dose of a pulse became negative through one pulse therapy.

Evidence for alteration in chemical and physical properties of water and modulation of its biological functions by sunlight transmitted through color ranges of the visible spectrum-a novel study.

We investigated the changes in the properties of water when exposed to sunlight for 40 days. We hypothesize and prove that solar irradiation to water entraps electromagnetic radiation as potential energy, which becomes kinetic energy in various systems. It is postulated that photochemically-induced energy transfers, associated with individual spectral emission of visible spectrum of solar light, exert diverse influences on biological systems. Bottles of distilled water, individually wrapped in spectral-colored cellophane were exposed to sunlight and compared to an unwrapped bottle to determine chemical and physical changes as well as modifications of biological properties. Each bottle of water was named according to the color of cellophane paper with letter E (stands for exposed) as a prefix with (E-violet, E-indigo, E-blue, E-green, E-yellow, E-orange, and E-red). E-control (without wrap) was exposed to polychromatic sunlight. This study addresses two main issues viz., the chemical and physical changes in E-water and its effect on biological activities. Chemical and physical composition analysis using inductively coupled plasma atomic emission spectrometry; physical conductance by a Wheatstone Bridge type conductivity meter; osmolarity by a vapor pressure osmometer; and, salt solubility profile of 10% sodium bicarbonate were determined. Furthermore, testing the effect of E-waters on human lymphocyte proliferation, mosquito larvae hatching and seed germination determined the functional role of solar radiation through specific spectrum/s of visible light on various biological processes. We found that water exposed to visible spectral emissions of sunlight had an altered elemental composition, electrical conductance, osmolarity and salt-solubility, as well as differences in bio-modulatory effects. A gradual increase in leaching of Boron from E-violet to E-red was noted. E-indigo showed maximal increase in electrical conductance and maximal salt solubility of sodium bicarbonate. E-blue inhibited phyto-hemagglutinin-induced immune cell proliferation and mosquito larvae hatching. E-orange stimulated root elongation in seed germination. We conclude that 40-day exposure of water to specific solar spectrum changes chemical and physical properties and influences on biological activity.

Dose measurement of 90Sr source for radiation chemical reaction.

The irradiation characteristics for a plate source of 90 Sr 222GBq (6 Ci) were examined with the blue cellophane dosimeter and the thermoluminescence dosimeter. The 90Sr source used has the active area of 35x20mm2 and the surface concentration of 31.5 GBq (0.85 Ci)/cm2. The high dose rate of 1.1 kGy (1.1x10(5) rad)/h was obtained at the distance of 5 cm from this source. At that time, the isodose curve determined by the blue cellophane was similar to a circle. The peak in depth dose curve was fairly displaced to the shallow direction in comparison with that obtained from the irradiation with an electron beam accelerator (2 MeV). The calculated dose rate attains to 800 kGy (i.0x10(7) rad)/h at the distance of 5 cm from the source when the infinite plate source with the maximum surface concentration (1.51 TBq(40.7Ci)/cm2) is placed on a plane.

The influence of thymus extracts on the chemotaxis of polymorphonuclear neutrophils (PMN) from patients with insulin-dependent diabetes mellitus (IDD).

The main goal of this study was to evaluate the possible influence of thymus extracts on the chemotaxis of PMN isolated from diabetic patients. The analyzed group consisted of fifty patients with insulin-dependent diabetes mellitus (IDD), and twenty healthy adults were taken as controls. The results showed, that PMN isolated from patients with IDD manifested the impaired chemotaxis toward zymosan-activated plasma (ZAP), plasma incubated with cellophane and supernate from E. coli culture. Both thymus extracts did not change the directed migration toward control plasma and toward ZAP. THYMEX-L did improve the migration of diabetic PMN toward plasma incubated with cellophane, whereas TFX-THYMOMODULIN ameliorated the impaired chemotaxis toward supernate from E. coli culture. Both thymus extracts can attract PMN obtained from healthy donors, but not the cells isolated from diabetic patients.

Prevalence of intestinal parasitosis in children with gastrointestinal symptoms associated with socio-economic conditions in Manisa region.

Intestinal parasitosis is still an important public health problem. The aim of this study was to determine the prevalence of intestinal parasitosis in children with gastrointestinal symptoms, and to evaluate its association with socio-economic and environmental factors. Stool samples of 3,216 children were examined by the saline-iodine method and trichrome staining. The cellophane tape method was also performed on 2,160 children. According to the educational levels and the economic status of families, the patients were classified as coming from underdeveloped, developing and developed areas. In 770 (23.9%) of 3,216 stool samples, various parasites were detected by the saline-iodine method and trichrome staining. The most common parasite was Giardia intestinalis (40.1%), followed by Entamoeba coli (10.2%). Enterobius vermicularis eggs were detected by the cellophane tape method in 221 (10.3%) out of 2,160 patients. The positive cases were evaluated according to the socio-economic and the environmental criteria; and most of them were found to have come from underdeveloped and developing areas. Health care and governmental officers should cooperate in order to improve the living conditions, and also people should be informed about the signs, symptoms and prevention methods of the parasitic diseases.

Aortic wraps a modification to aortic grafting.

Aortic wrapping has been attempted on occasion when the standard replacement of a diseased lower third of the aorta by a trouser graft is not feasible. Cellophane has been the most common agent employed, but these wraps have met with minimal success and on occasion has even been passed rectally. Kartchner and Lovett have reviewed the techniques and results of the later procedures replacing the cellophane wraps. In most instances a Dacron sheet was utilized. Six cases are reviewed from a solo private practice in which a standard trouser graft replacement was varied by using an alternative form of wrapping.

Problems of calibration and stabilization of tcPO2 electrodes.

The stirring effect factor phi, used to correct tcPO2 readings for gradient of PO2 induced in skin by electrode O2 consumption, was measured with 20 micrometer cathode electrodes at 44 degree C for various membrane, spacer and electrolyte combinations. Two in vitro models closely resemble skin phi values: (1) 50% ethylenge glycol/water equilibrated with air at 44 degree C, compared to air above this liquid; (2) a mock skin consisting of 25 micrometer Telfon on foam rubber, in air, dry. phi varied from 1.01 with 22 micrometer polypropylene over a 12 micrometer cellophane spacer, with either aqeous or non-aqueous, chloride-free alkaline electrolyte, to more than 1.10 when 25 micrometer Telfon was tested without a spacer. phi may be predicted: phi = 1 + 0.44 i/(M + 10C + 20) where i is pA/mmHg electrode sensitivity, and M and C are membrane and cellophane thickness, micrometer. Pressure sensitivity, tested agains foam rubber, was minimized by use of a spacer, and often increased with age of Telfon membrane. Drift may result from water vapour movements across membranes if calibration media have P(H2O) either too high or too low. Miminum drift on skin is also caused by a slow effect of PCO2 on reference electrode potential, and this is minimized by buffering electrolyte with K2B4O7.
