The delayed hypersensitivity (DH) response to picryl chloride was studied in Ehrlich ascites tumour-bearing and normal control mice. A significant depression of the DH response was found in the tumour-bearing mice, which was associated with a marked elevation of serum lactate dehydrogenase (LDH). Depression of DH was also observed in mice receiving cell-free ascitic fluid. These mice also showed an elevated serum LDH which is assumed to be associated with the lactate dehydrogenase virus. A method for assaying DH in vivo is described.

Inoculation of bone marrow and spleen cells of C3H/Sumice strain mice demonstrated that on the ninth day of growth of Gardner lymphosarcoma these tissues were invaded with tumor cells. The weights of mice with advanced tumors increased. Yet the deterioration of health status of mice is characterized by decreased food and water consumption, as well as by lower concentrations of proteins in mouse sera and plasma. The albumin fraction shows a remarkable drop. These changes could not be detected in tumor free mice following LDH-virus infection. The treatment with L-asparaginase influenced the manifestations of tumor growth, but not the effectivity of LDH-virus contaminating the tumor as demonstrated by the increased activity of lactate-dehydrogenase in the mouse sera.

Gel chromatography on Spheron P-500 of mouse serum yielded fractions elevating lactate dehydrogenase activity. The order of elution was viral activity, proteins and lactate dehydrogenase activity. Viral contamination occurred also in certain fractions subjected to repeated gel chromatography in which no proteins were detectable.
Flow cytometry showed that the enzyme lactate dehydrogenase (LDH-5) binds to peritoneal macrophages. The degree of binding appears to be genetically controlled and varies depending on the inbred strain of mouse. The rate at which LDH-5 is cleared from the circulation correlates with the degree of binding to macrophages. These findings suggest that the binding of LDH-5 to macrophages may serve as an in-vitro index of enzyme clearance.

Three porcine reproductive and respiratory syndrome virus (PRRSV) recombinants, generated by mutagenesis of an infectious cDNA clone of the Lelystad virus (LV) isolate, were tested for their safety and protective efficacy as potential PRRSV vaccines in pigs. Recombinant vABV688 contains two amino acid substitutions in the minor structural protein GP(2) resulting in improved growth on cell line CL2621; in recombinant vABV707 the region encoding the ectodomain of the major unglycosylated membrane protein M has been replaced by that of the murine lactate dehydrogenase-elevating arterivirus; recombinant vABV746 lacks the six C-terminal amino acids of the nucleocapsid protein N. First, we determined the safety of these recombinant viruses by monitoring the stability of the introduced mutations in 8-week-old pigs. We showed that the introduced genomic mutations were maintained throughout the viraemic period. Second, the protective efficacy of immunization with the recombinant viruses against challenge with a homologous and a heterologous PRRSV strain was determined in two pigs and compared with the efficacy of vABV437, a virus derived from the parental LV cDNA. The viraemia in pigs immunized with the recombinant viruses was reduced compared to pigs immunized with vABV437. In addition, the length of viraemia was reduced in the sentinel pigs that were introduced into the groups immunized with vABV746, vABV688, and vABV707, however, all of the sentinel pigs became infected. Pigs immunized with vABV707 and vABV437 were protected against challenge with homologous virus LV-Ter Huurne and transmission of the latter virus. None of the immunized pigs were protected against heterologous challenge with the virulent US isolate SDSU#73, but the vABV707- and vABV746-immunized pigs were protected against transmission of this virus from challenged pigs. In conclusion, the obtained viral recombinants are interesting candidates to be further explored for their use as vaccines against PRRSV.
Infectious lactic dehydrogenase-elevating virus propagated in primary cultures of mouse peritoneal macrophages in the presence of (3)H-uridine and isolated by isopycnic centrifugation was found to have a density of 1.12 g/cm(3). Ribonucleic acid extracted from the virus by treatment with sodium dodecyl sulfate was single stranded with a sedimentation coefficient of approximately 48S.
