Microbiological conditions of sheep carcasses from conventional or inverted dressing processes.

At a small abattoir, 25 sheep carcasses were dressed conventionally, with the carcass suspended by the rear legs, and 25 carcasses were dressed while inverted, with the carcasses suspended by the forelegs. Two swab samples were obtained from randomly selected sites on each carcass, and total aerobic, coliform, and Escherichia coli counts were enumerated for each sample. Each type of count was arranged in two sets of 25 counts for each type of dressing process, and a log mean number and/or log total number recovered was calculated for each set of counts. The log mean number of total aerobic counts for one set of counts from carcasses dressed while inverted was less than the corresponding log mean numbers for both sets from the conventionally dressed carcasses and the other set from the carcasses dressed while inverted, and differed from them by about 0.7 log units. The coliforms recovered from carcasses were largely E. coli. The log total numbers of coliform or E. coli counts recovered from carcasses dressed while inverted were about 1.5 log units less than the corresponding log total numbers recovered from conventionally dressed carcasses. Those data indicate that the substitution of inverted for conventional dressing might serve to reduce the numbers of E. coli on sheep carcasses by reducing the microbiological contamination of the hindquarters but that the general microbiological condition of the carcasses would be little improved unless some means of preventing or removing contamination of the forequarters was also used.

Evaluation of an automated clean-up system for the isotope-dilution high-resolution mass spectrometric analysis of PCB, PCDD, and PCDF in food.

An automated clean-up system was evaluated for the simultaneous analysis of polychiorinated dibenzo-p-dioxins (PCDD), dibenzofurans (PCDF), and biphenyls (PCB) in different foods. In addition to the seventeen 2,3,7,8-substituted PCDDIPCDF and four non-ortho PCB, by use of the clean-up system studied, it was possible to collect the eight mono-ortho and two di-ortho PCB and the seven indicator PCB in two separate fractions during the same clean-up run. The study was first performed using standard mixtures containing PCDD, PCDF and PCB, and a certified reference material. The recoveries of the 13C-labeled compounds ranged from 51 to 90%, indicating that the PCDD, PCDF, and PCB clean-up worked satisfactorily. Next, the automated system for PCDD, PCDF, and PCB analysis was evaluated for foods such as milk, egg, meat (beef, chicken, and pork), mussel, and olive oil. The recoveries of the 13C-labeled compounds ranged from 40 to 120% for PCB and from 57 to 113% for PCDD/ PCDF, meeting the requirements of well accepted methods. Thus, the automated clean-up system studied is a suitable alternative to conventional clean-up methods.

Foodhandler-associated Salmonella outbreak in a university hospital despite routine surveillance cultures of kitchen employees.

OBJECTIVE: To describe an outbreak of salmonella food poisoning that probably was due to contamination of mashed potatoes by a foodhandler, which occurred despite a policy for routine surveillance stool cultures of kitchen employees. DESIGN: A case control study of 223 individuals who ate the lunch meal on September 23, 1989, at the Jordan University Hospital (JUH) cafeteria. SETTING: Tertiary care university hospital in Amman, the capital of Jordan. PATIENTS: Individuals who developed loose stool or vomiting 6 to 72 hours after eating the lunch meal of September 23, 1989, at the JUH cafeteria. RESULTS: Of 619 individuals, 183 fit the case definition (attack rate, 19.6%); 150 were employees, 26 were inpatients, and seven were visitors. Twelve other employees became sick 4 to 6 days later and probably were infected secondarily. The incubation period ranged from 16 to 72 hours in 183 instances. Symptoms included diarrhea (88%), fever (71%), abdominal pain (74%), dehydration (34%), and bloody stool (5%). Eighty-four were hospitalized. Cultures of eight food items were negative, but stool culture on 90 of 180 patients and 11 of 61 kitchen employees yielded Salmonella enteritidis group D. A cohort study of 223 individuals revealed a food-specific attack rate of 72% for the steak and potato meal and 18% for the rice and meat meal (RR, 4; CI95, 2.62 to 6.24; P < 0.01). Stratified analysis of the steak and potato meal revealed that the potatoes were implicated most strongly (RR, 1.93; CI95, 1.42 to 2.64; P < 0.01). Cultures were obtained from all kitchen employees, and 11 of 61 grew Salmonella enteritidis group D. One asymptomatic, culture-positive employee prepared the mashed potatoes on September 23. All of these employees had negative stool cultures 3 months earlier. CONCLUSION: This outbreak probably was caused by massive contamination of mashed potatoes by the contaminated hands of the foodhandler. Routine stool culture of foodhandlers is not cost-effective and should not be used as a substitute for health education and proper hygienic practices.

Inhibition of peroxidase-catalyzed reactions by arylamines: mechanism for the anti-thyroid action of sulfamethazine.

Sulfonamide antibiotics, typified by sulfamethazine (SMZ), are widely used in veterinary practice. Sulfonamide residues in milk and meat products are of regulatory concern since SMZ is a thyroid carcinogen in rodents and sulfonamide-induced hypersensitivity reactions, including hypothyroidism, have been reported in humans. SMZ and other primary arylamines inhibited iodination reactions catalyzed by thyroid peroxidase (TPO) and the closely related lactoperoxidase (LPO). Inhibition of LPO-catalyzed triiodide ion formation by SMZ and other primary arylamines was complex as both apparent Km and Vmax values were affected, but consistent with a rapid equilibrium binding mechanism. The apparent Ki for SMZ inhibition of TPO- and LPO-catalyzed iodide ion oxidation was approximately 0.42 and 0.11 mM, respectively. The corresponding Ki values for a series of para-substituted anilines correlated with the ease of one-electron N-oxidation as measured by ionization potentials determined from semiempirical molecular orbital calculations. The aniline derivatives containing electron-donating substituents (e.g., p-CH3, p-OEt, p-Cl) were converted by LPO to colored products characteristic of one-electron oxidation. However, sulfonamides were not consumed in such reactions nor were any N-oxygenated derivatives formed in the absence of ascorbate (e.g., hydroxylamino, nitroso, nitro, azoxy). These observations suggest that the primary mechanism for sulfonamide-induced hypothyroidism is reversible inhibition of TPO-mediated thyroid hormone synthesis and not the formation and covalent binding of reactive N-oxygenated metabolites. These results are consistent with a hormonal mechanism for SMZ-induced thyroid carcinogenesis mediated by thyroid-stimulating hormone (TSH).(ABSTRACT TRUNCATED AT 250 WORDS)

Standardization of a simplified scintigraphic methodology for the assessment of gastric emptying in a multicenter setting.

OBJECTIVE: Scintigraphy remains the gold standard to study gastric emptying. The technique is onerous and normal values vary between centers. Standardized protocols, although desirable, are not presently available. We validated a simplified scintigraphic protocol in a multicenter setting. METHODS: In 69 healthy volunteers from seven Canadian institutions, gastric emptying of a standard meal (99mTc-labeled beef liver) was assessed by scintigraphy every 10 min for 1 h, then every 20 min for the next 2 h. Gastric retention was fitted to a power exponential model, Prop(t) = (-(kappat)beta) with Prop(t) = proportion of retention at time t, either using all 13 time intervals (conventional technique) or using measurements at 0, 1, 2, and 3 h (simplified technique). RESULTS: The power exponential model yielded identical emptying curves and T 1/2 values with both techniques. Gastric emptying was more rapid in men than in women < 35 yr (p<0.01) and in younger than in older men (p<0.005). Gastric emptying was slower in women from Québec than in women from Ontario (p<0.04). Gastric retention was similar at 1, 2, and 3 h among the seven centers. Gastric emptying of a beef liver meal was slower than that of a low fat egg substitute (p<0.03). CONCLUSIONS: A simpler scintigraphic approach, using four rather than 13 samples, provides results comparable to those of the conventional technique. This simpler approach provides an economical, yet accurate, alternative to the techniques presently used and is applicable to a multicenter setting.

Estimates of beef carcass intermuscular fat.

Three experienced persons evaluated 158 carcasses 24 h postmortem for USDA yield grade (YG) and quality grade factors, nine subcutaneous (SC) fat indicators, and four intermuscular (IM) fat indicators. Forty sides (YG 1.1 to 3.8) were selected for determination of chemical composition, two measures of cutability, and total IM fat from the round, loin, rib, and chuck. The IM fat estimates at the 12th rib, rib-plate juncture, and 5th rib were correlated with percentage of chemical fat (r = -.72, -.70, and -.55, respectively). Simultaneous consideration of YG factors accounted for 61% of the variation in chemical fat. Substituting the IM fat estimate at the 12th rib for adjusted fat thickness (AFT) in the equation explained 60% of the variation in percentage of chemical fat. An equation containing two IM fat estimates, marbling score and longissimus muscle area explained 68% of the variation in chemical fat. Simultaneous consideration of the YG factors accounted for 59% of the variation in boneless, closely trimmed (6 mm SC fat and no IM fat) retail cuts from the round, loin, rib, and chuck. Substituting the IM fat estimate at the 12th rib for AFT in the equation accounted for 65% of the variation. These data from a fairly uniform set of steer carcasses show that percentage of chemical fat and cutability can be reliably predicted from IM fat estimates and other traits that can be visually estimated on hot-fat trimmed carcasses.

[Preparation of derivatives of 3-(3,4-dimethoxyphenyl)propanic acid and a study of its biological activity].

From 3-(3,4-dimethoxyphenyl)propenic acid chloride and substituted amines and hydrazides, the appropriate amides and hydrazides (Table 1) were synthesized at 60-80 degrees C in the medium of benzene or toluene. The reaction of this chloride with benzaldehyde hydrazone at 70-80 degrees C yielded N,N'-bis[3-(3,4-dimethoxyphenyl)propenoyl]hydrazine (VIII). At ambient temperature the benzylidene hydrazide of 3-(3,4-dimethoxyphenyl)propenic acid (VII) and a small amount of compound VIII were isolated. In the reaction of 3-(3,4-dimethoxyphenyl)propenic acid chloride with benzylidene hydrazide (VII) at 70-80 degrees C, compound VIII was obtained (Scheme 1). Compounds I, VII, VIII and IX possessed higher indices of increase in fortnight tests of the first degree in the roosters of meat hybrides compared to the negative control, but the indices of conversion were unfavourable. The compounds did not reach the efficacy of the avoparcin standard. Derivatives II and VI possessed 67.5 and 63.5%, respectively, of anthelmintic activity of levamisol against Nippostrongylus brasiliensis. The prepared compounds were not antibacterially effective and they were not mutagenic in the tests following the method of Ames, either.

Characterization of H9N2 influenza A viruses isolated from chicken products imported into Japan from China.

We characterized eleven H9N2 influenza A viruses isolated from chicken products imported from China. Genetically they were classified into six distinct genotypes, including five already known genotypes and one novel genotype. This suggested that such multiple genotypes of the H9N2 virus have possibly already become widespread and endemic in China. Two isolates have amino-acid substitutions that confer resistance to amantadine in the M2 region, and this supported the evidence that this mutation might be a result of the wide application of amantadine for avian influenza treatment in China. These findings emphasize the importance of surveillance for avian influenza virus in this region, and of quarantining imported chicken products as potential sources for the introduction of influenza virus.

Epoxide hydrolase polymorphisms, cigarette smoking and risk of colorectal adenoma in the Nurses' Health Study and the Health Professionals Follow-up Study.

Microsomal epoxide hydrolase (mEH) is involved in the bioactivation and detoxification of polycyclic aromatic hydrocarbons derived from tobacco smoke and charred meat intake. Two coding region mEH variants located in exon 3 (Tyr113His) and exon 4 (His139Arg) have been described and may affect the enzyme's specific activity. We investigated these polymorphisms and tested interactions with smoking and charred meat intake in relation to risk of colorectal adenoma in two case-control studies nested in the Nurses' Health Study (NHS) and Health Professionals Follow-up Study (HPFS) cohorts. mEH exon 3 and exon 4 polymorphisms were not associated with overall risk of adenoma among 556 incident cases and 557 controls from the NHS or 376 prevalent cases and 725 controls from the HPFS. A statistically significant interaction was found between the exon 4 polymorphism and smoking for men (P = 0.03) and a borderline significant interaction was found between the exon 3 polymorphism and smoking for women (P = 0.06). Women having the exon 3 'rapid' Tyr/Tyr genotype were at increased risk when exposed to either > or =25 pack-years smoking [relative risk (RR) = 2.43, 95% confidence interval (CI) 1.47-4.01] or <25 pack-years of smoking (RR = 1.73, 95% CI 1.10-2.73) relative to non-smokers. Men with the exon 4 'slow' His/His genotype were at increased risk when exposed to > or =25 pack-years smoking (RR = 2.21, 95% CI 1.43, 3.41) or <25 pack-years smoking (RR = 1.71, 95% CI 1.13-2.59) relative to non-smokers. Charred meat intake was not associated with adenoma risk and there was no significant interaction with either mEH polymorphism. Our results indicate that individuals exposed to > or =25 pack-years smoking were at increased risk for colorectal adenoma and that risk is related to dose of tobacco carcinogens and mEH activity level, but the results were not consistent between men and women.

Identification of commercial fish species belonging to the orders pleuronectiformes and gadiformes: library of isoelectric focusing patterns.

Isoelectric focusing in polyacrylamide gel was used to establish an identification archive of fish species belonging to the orders Pleuronectiformes, or flat fish, and Gadiformes, or gadoid fish. The 2 orders include species of different commercial value and interest that are frequently requested in European fish markets, but are susceptible to substitution either because they are morphologically similar or because they arrive on the markets already filleted or sliced. The sarcoplasmic protein profiles are species-specific and reproducible. The use of densitometry and image analysis coupled with a simple computer program overcomes the subjective evaluation of the patterns, making it possible to identify species correctly.

Dietary exposure to polychlorinated biphenyls and dioxins from infancy until adulthood: A comparison between breast-feeding, toddler, and long-term exposure.

Food is the major source for polychlorinated biphenyl (PCB) and dioxin accumulation in the human body. Therefore, investigating food habits from early ages until reproductive age (25 years) is important in order to assess exposure risk for the next generation. The objective of this study was to assess the PCB/dioxin exposure and the relative contribution of different foods to total exposure during preschool age. Particularly, the importance of lactational PCB/dioxin exposure vs. dietary exposure until adulthood was investigated. A cohort of 207 children was studied from birth until preschool age. Based on 3 planar PCBs and 17 2,3,7,8-substituted dibenzo-para-dioxins (PCDDs) and dibenzofurans (PCDFs) measured in breast milk, a model was developed to calculate the cumulative toxic equivalent (TEQ) intake during breast-feeding (0-1 year). In 3. 5-year-old children, daily dietary intake of planar PCB-TEQ and dioxin-TEQ was measured with a validated food questionnaire. Cumulative TEQ intake from 1 to 5 years was estimated using the PCB- and dioxin-TEQ intake measured with the food questionnaire. Cumulative TEQ intake from 6 to 25 years was estimated using national food consumption and contamination data of PCB- and dioxin-TEQ intake. In toddlers, dairy products contributed 43% to PCB-TEQ and 50% to dioxin-TEQ intake. Meat and meat products contributed 14% and 19%, respectively, and processed foods 23% and 15%, respectively. Breast-feeding for 6 months contributed to the cumulative PCB/dioxin TEQ intake until 25 years of age, 12% in boys and 14% in girls. The daily TEQ intake per kilogram body weight is 50 times higher in breast-fed infants and three times higher in toddlers than in adults. Long-term dietary exposure to PCBs and dioxins in men and women is partly due to breast-feeding (12 and 14%, respectively). After weaning, dairy products, processed foods, and meat are major contributors of PCB and dioxin accumulation until reproductive age. Instead of discouraging breast-feeding, maternal transfer of PCBs and dioxins to the next generation must be avoided by enforcement of strict regulations for PCB and dioxin discharge and by reducing consumption of animal products and processed foods in all ages.

Functional polymorphism in porcine CYP2E1 gene: Its association with skatole levels.

Raising intact male pigs would have a significant economic impact on the pork industry. However, the presence of skatole (a major cause of boar taint) in meat from intact male pigs could be highly objectionable to consumer. The excessive accumulation of skatole in fat is a major cause of boar taint, and is associated with defective expression of cytochrome P4502E1 (CYP2E1). In pigs, it has been found that CYP2E1 is negatively correlated with accumulation of skatole. The searching for polymorphism of CYP2E1 and the relevant functional analysis would help develop a genetic marker for the selection of pigs with low skatole levels in fat. The aim of this study was to measure the expression pattern of CYP2E1 mRNA in various tissues of the pig, to identify genetic polymorphisms, and to evaluate the functional relevance of polymorphic sites with respect to the skatole level in fat. We show herein that a substitution of G --> A at base 1423 of the CYP2E1 gene in the liver causes a significant decrease in the expressed CYP2E1 level. Our data suggest that the G --> A substitute might be at least partially responsible for a high level of skatole in pigs. We believe that this is an important step toward the selection of genetic markers for boar taint by lowering fat levels of skatole in fat.

Simulated instrument augmentation of USDA yield grade application to beef carcasses.

Because no instrument technology has been shown to predict beef carcass composition better than USDA yield grades, this study was conducted to determine whether an instrument could be used to augment and improve the accuracy of USDA yield grade placement. Adjusted preliminary yield grade (PYG), ribeye area (REA), estimated percentage of kidney, pelvic, and heart fat (KPH), hot carcass weight (HCW), and USDA yield grade (called and computed) were determined by five on-line USDA graders and two USDA grading supervisors for beef carcasses (n = 550) selected randomly in a commercial beef packing plant. Data were compared (2,737 comparisons) to Gold Standard yield grades and yield grade factors determined by an expert panel of carcass evaluators (unrestrained in access or time to evaluate carcasses). On-line USDA grader PYG were closely related (mean absolute error of .15 +/- .14 yield grade units; r = .91), and on-line REA and KPH were nominally related (mean absolute error of .51 +/- .35, .06 +/- .07 yield grade units and r = .48 and .66, respectively), to Gold Standard yield grade factors. On-line USDA graders determined adjusted PYG effectively, but they may require instrument assistance to evaluate carcass muscling traits and perform time-sensitive computations. To explain why instrument technology may not estimate beef carcass fatness as accurately as USDA yield grades, the absolute mean difference between Gold Standard measured PYG and adjusted PYG were compared. Only 5.6% of the sample population required no PYG adjustment, 94.4% required some adjustment, and 11.0% required over a .5 yield grade unit adjustment. Yield grades for beef carcasses, called by the USDA graders and supervisors at chain speeds, resulted in greater accuracy (absolute mean error of .24 +/- .43 yield grade units; r = .82) than when yield grades were computed for carcasses using the yield grade factors determined by on-line USDA graders and supervisors at chain speeds (absolute mean error of .52 +/- .41 yield grade units; r = .75). Gold Standard yield grade factors were sequentially substituted into the short-cut USDA yield grade equation for the yield grade factors determined at chain speeds by the USDA graders and supervisors. Results suggested that instrument augmentation would improve accuracy and precision of yield grade placement if on-line USDA graders determined PYG and an instrument determined REA and performed the necessary computations, incorporating KPH and actual HCW (P < .05).

Considerations in planning vegan diets: infants.

Appropriately planned vegan diets can satisfy nutrient needs of infants. The American Dietetic Association and The American Academy of Pediatrics state that vegan diets can promote normal infant growth. It is important for parents to provide appropriate foods for vegan infants, using guidelines like those in this article. Key considerations when working with vegan families include composition of breast milk from vegan women, appropriate breast milk substitutes, supplements, type and amount of dietary fat, and solid food introduction. Growth of vegan infants appears adequate with post-weaning growth related to dietary adequacy. Breast milk composition is similar to that of non-vegetarians except for fat composition. For the first 4 to 6 months, breast milk should be the sole food with soy-based infant formula as an alternative. Commercial soymilk should not be the primary beverage until after age 1 year. Breastfed vegan infants may need supplements of vitamin B-12 if maternal diet is inadequate; older infants may need zinc supplements and reliable sources of iron and vitamins D and B-12. Timing of solid food introduction is similar to that recommended for non-vegetarians. Tofu, dried beans, and meat analogs are introduced as protein sources around 7-8 months. Vegan diets can be planned to be nutritionally adequate and support growth for infants.

Polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/PCDF) in food samples collected in southern Mississippi, USA.

In 1994, we analyzed 43 foodstuff samples from local supermarkets in southern Mississippi, USA, for PCDD/PCDF. 2,3,7,8-Cl4DD could be quantified in 31 of these samples. On a lipid basis, levels in meat (0.53-1.10 pg I-TEQ/g) and dairy products (0.42-1.10 pg I-TEQ/g) were slightly lower than those reported from other industrialized countries. While levels in dairy samples from the United States and Europe are comparable, there is a difference in the contribution of individual congeners to the I-TEQ: for example, in milk samples from Germany approximately 40% of the I-TEQ is due to the presence of 2,3, 4,7,8-Cl5DF while in the Mississippi samples this congener only contributes 16%. The highest concentrations of PCDD/PCDF in our study were detected in the farm-raised catfish (10.2-27.8 pg I-TEQ/g). A unique finding was that in addition to the 2,3,7,8-substituted PCDD/PCDF the catfish samples contained many non-2,3,7,8-substituted congeners. This is unusual because vertebrate animals selectively eliminate or metabolize the non-2,3,7,8-substituted congeners.

Mutagenic specificity of 2-amino-3-methylimidazo[4,5-f]quinoline and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine in the supF shuttle vector system.

2-Amino-3-methylimidazo[4,5-f]quinoline (IQ) and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) are heterocyclic amine mutagens/carcinogens formed from the cooking of meat. Here we used the pSP189 shuttle vector developed by Parris and Seidman (Gene, 117: 1-5, 1992) to study and compare the mutation spectra induced by these compounds. pSP189 was adducted by reaction with the N-acetoxy derivatives of IQ or PhIP. 32P-Postlabeling analysis was used to measure the C8-guanine adduct level and the total adduct levels formed in the plasmid. Plasmids were replicated and mutagenized in repair-proficient [GM0637(SV40)] or repair-deficient [XP12Be(SV40)] human fibroblasts. Resulting inactivation mutations in the supF gene were determined by the formation of white or light blue colonies on indicator bacteria (carrying a lacZ amber mutation) and cycle sequencing. With both compounds in either cell line, 85-93% of the mutations induced were base substitutions and the remainder of the mutations were base deletions. The majority of the substitution mutations involved a single base, and nearly all base substitution mutations (> 97%) were at guanine. This latter finding is consistent with the results from 32P-postlabeling showing that both compounds adduct to the guanine base with the major adduct being formed at the C8-guanine position. The predominant mutation found with IQ and PhIP in either cell line was G:C to T:A transversion, followed by G:C to A:T transition, and then G:C to C:G transversion; these mutations accounted for 59-72%, 19-27%, and 6-14% of total base substitution mutations, respectively. There was a preference seen with both compounds to induce mutations at a guanine base having a neighboring guanine or cytosine (i.e., GG and GC sites). However, despite the striking similarity in the kinds of base substitution mutations induced by IQ and PhIP, their mutation spectra were distinct. For example, in repair-proficient cells, 26% of the mutations induced with PhIP, but not with IQ, also involved a GA site, containing the 5-base pair sequence 5'-GCAGA-3'. Mutation spectra for IQ and PhIP were also different between repair-deficient and repair-proficient cells. The findings shown here may serve to be predictive of the kinds of mutations induced by the adducts of IQ and PhIP in oncogenes and tumor suppressor genes altered during heterocyclic amine-induced carcinogenesis.

[Carnitine--biochemistry and clinical aspects].

Carnitine was discovered in 1905. The human organism is able to synthetize carnitine from lysine and methionine. However the most important source of carnitine in human nutrition is meat. Carnitine plays a central role in the oxidation of long chain fatty acids, mediating their transport across the inner mitochondrial membrane. By means of the intramitochondrial reaction of acetyl-CoA with carnitine it favours an improved availability of CoA. This mechanism seems to be of major importance when pathological short chain fatty acids are accumulated within the mitochondrial matrix. Carnitine deficiency can be inborn or acquired. Thereby myopathy is always a prominent clinical symptom. Secondary carnitine deficiency occurs most frequently in uremic patients under intermittent hemodialysis. There is a positive inotropic effect of carnitine on the ischemic heart muscle. As a consequence of this effect, carnitine is on the way to become a substance with broadest clinical indications. If young infants are alimented parenterally with fat emulsions, carnitine should be substituted. This also happens to be the case in adult patients when total parenteral nutrition lasts longer than 20 days.

Duplex (thermotroph-psychrotroph) quadrant plates: convenient, error-avoiding tools for monitoring of HACCP-contained food lines and for epidemiological investigations under conditions of military or other constraints.

A set of two "two-tier" (thermotroph-psychrotroph) single quadrant plates (QPs) was developed previously to allow convenient enumeration of numbers of colony-forming units of most pertinent pathogenic bacteria or marker bacteria in foods. These include Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, thermotrophic and psychrotrophic Enterobacteriaceae, Clostridium spp., and Enterococcus spp. As the QPs had given excellent results in monitoring samples of marketed food products potentially involved in food-transmitted illnesses, the approach was tested for practicability under military deployment and other constraints. Three approaches were envisaged: (i) validating lapse-free adherence to meticulously codified good military catering practices; (ii) acceptance/rejection testing of locally procured foods or meals; and (iii) employing rapid culture in support of evidence obtained by microscopy in attempts to identify foods involved in infectious or toxic disease outbreaks occurring in the field. The method was found to be elegant, avoiding confusion when larger number of specimens were to be screened, as well as easy to teach to staff with little or no training in microbiology, and it provided entirely reliable results. For use outside the laboratory, preparation of food macerates by use of shake flasks containing glass or plastic beads and peptone saline as a substitute for stomaching was found acceptable, though the shake flask technique led to slightly diminished colony counts. Results obtained with incubation times shortened to ca. 12 h could be relied on only when the results were alarmingly positive, but not when the colony counts at the 12-h point did not yet indicate a reason for concern.
