Combination chemotherapy of non-Hodgkin lymphoma.

A randomized therapeutic trial of three induction regimens for patients with lymphosarcoma and reticulum-cell sarcoma was conducted by the Cancer and Leukemia Group B (CALGB) (formerly Acute Leukemia Group B.) Addition of streptonigrin, but not cyclophosphamide, to the combination of vincristine and prednisone improved the complete remission rate achieved after a 42-day induction program. The duration of the subsequent remission and survival was not influenced by the induction combination assigned. Remission maintenance with cyclophosphamide was superior to maintenance with methotrexate. The results of this study are compared with others in which advantages for three or four drug treatment regimens have been reported.

Thymosin alpha(1) application augments immune response and down-regulates tumor weight and organ colonization in BALB/c-mice.

The immunomodulatory and antimetastatic/antitumor activity of thymosin alpha(1) (Talpha(1)) was evaluated in BALB/c-mice. Daily subcutaneous application (7 consecutive days, 0.01-10 microg of Talpha(1)/injection per mouse) upregulated the number of thymocytes and peripheral blood cells in tumor bearing mice. To check the influence of Talpha(1) treatment on growth of experimental metastases, RAW H10 lymphosarcoma cells or L-1 sarcoma cells were intravenously injected into BALB/c-mice to establish liver or lung metastases. Local tumor growth was induced by subcutaneous injection of L-1 sarcoma cells. Talpha(1) was subcutaneously administered daily for 7 consecutive days starting 24 h after tumor cell challenge. Organ colonization, as well as local tumor growth, were investigated on day 14 after tumor cell inoculation, and demonstrated a statistically significant (P<0.05) reduction of experimental liver and lung metastases and local tumor growth for Talpha(1) treated mice.

Application of standardized mistletoe extracts augment immune response and down regulates metastatic organ colonization in murine models.

The immunomodulatory and antimetastatic activity of standardized aqueous mistletoe extracts from plants grown on fir trees (ME-A) and pine trees (ME-P) were evaluated in BALB/c-mice. Regular subcutaneous (s.c.) and intraperitoneal (i.p.) applications (three times per week for 14 consecutive days; 5 and 50 microg per injection and mouse) upregulated thymus weight and peripheral blood leukocyte counts in tumor bearing mice. To check the influence of ME-A and ME-P treatment on growth of experimental metastases, RAW 117 H 10 lymphosarcoma cells and L-1 sarcoma cells were intravenously inoculated into BALB/c-mice to establish liver and lung colonization. ME-A and ME-P were regularly administered starting 24 h after tumor cell challenge. Organ colonization was investigated on day 14 after tumor cell inoculation and demonstrated statistically significant (P<0.05) reductions of experimental liver and lung metastases for ME-A and ME-P treated mice.

Soft tissue tumors in beagles injected with 241Am citrate.

The occurrence of soft tissue tumors has been studied in 117 beagles assigned to 8 dosage groups of between 2 and 26 animals each and injected with 0.07 to 104 kBq 241Am kg-1 as the citrate. In addition, 133 control beagles given no radioactivity were used as a comparison group. All 250 dogs were maintained under identical conditions and were observed for their entire lifespans. An important competing risk for the appearance of soft tissue tumors appeared to be the occurrence of skeletal malignancy, and at the highest injected activity (104 kBq kg-1), kidney and liver failure brought about the death of both of the two dogs in this group. Thyroid and liver were the only soft tissues that exhibited greater concentrations of 241Am than the skeleton. Liver tumors were associated with 241Am exposure (p < 0.001), but the thyroid tumor rate was not increased significantly in the irradiated animals (p > 0.10) as compared with the occurrence in controls. There was a greater relative occurrence of all vaginal tumors in control animals than in dogs given 241Am, a situation also found for all tumors of the pancreas, skin, testis, and mammary glands and for malignant ovarian tumors. All of these differences were statistically significant. The survival of animals given 0.07 to 0.59 kBq 241Am kg-1 could not be established (p > 0.10) as significantly different from controls, but the survival of all groups given 1.8 to 104 kBq kg-1 was decreased (p < 0.05). There was no indication in our studies of a positive association between relative exposure to 241Am and the occurrence of mammary tumors, mast cell sarcomas originating outside the liver, lymphosarcoma or tumors of marrow, including leukemia.

Biological and antigenic characteristics of HEL-12 virus.

The biological and antigenic properties of HEL-12 virus have been compared with gibbon ape lymphosarcoma virus (GALV) and simian sarcoma and simian sarcoma-associated viruses, SiSV and SSAV, respectively, HEL-12 virus did not transform human or marmoset fibroblasts but rescued SiSV focus-forming activity from non-productively transformed marmoset cells (HF/SiSV-NP). Like SSAV and GALV, HEL-12 virus induced syncytia with XC cells. In addition, HEL-12 cells which did not produce virus but which contained HEL-12 proviral DNA, rescued SiSV from HF/SiSV-NP cells in co-cultivation experiments. Results of neutralization and serum cytotoxicity tests utilizing SiSV rescued by HEL-12 [SiSV-(HEL-12)] indicated that HEL-12 virus envelope proteins are very closely related to those of SSAV but readily distinguished from those of GALV. Antigenic diversity of SiSV(SSAV), SiSV(GALV) and SiSV(HEL-12) envelope glycoproteins (gp70) was shown in competition radioimmunoassays (RIA) designed to detect minor antigenic differences using antiserum monospecific for SiSV(SSAV) gp70 or Friend murine leukaemia virus gp70. Antigenic differences between these gp70s were demonstrated in RIA using purified SSAV gp70 or HEL-12 gp70. These data indicate that HEL-12 virus has biological properties similar to those of SSAV and GALV, is distinguished from GALV in neutralization tests and has both distinct and SSAV-related gp70 antigenic determinants.

Thymic humoral factor-gamma 2 augments immune cell response and exerts antitumor activity in murine model systems.

The immunomodulatory and antimetastatic/antitumor activity of thymic humoral factor-gamma 2 (THF-gamma 2) was evaluated in BALB/c-mice. Daily subcutaneous applications (7 consecutive days, 20, 200 ng of THF-gamma 2 per injection/mouse) upregulated counts of thymocytes and peripheral blood cells in tumor bearing mice. To check the influence of THF-gamma 2 treatment on the growth of experimental metastases, RAW 117 H10 lymphosarcoma cells or L-1 sarcoma cells were intravenously inoculated into BALB/c-mice to establish liver or lung metastases, respectively. Local tumor growth was induced by subcutaneous injection of L-1 sarcoma cells. THF-gamma 2 was subcutaneously administered daily for 7 consecutive days starting 24 hrs after tumor cell challenge. Organ colonization as well as local tumor growth were investigated on day 14 after tumor cell inoculation and demonstrated a statistically significant (p < 0.05) reduction of experimental liver and lung metastases and local tumor growth for THF-gamma 2 treated mice.

Standardized mistletoe extract augments immune response and down-regulates local and metastatic tumor growth in murine models.

The immunomodulatory and antimetastatic activity of standardized aqueous mistletoe extract (sME) was evaluated in BALB/c-mice. Regular subcutaneous (s.c.) applications (three times per week for 14 consecutive days; 2, 20, 100 and 500 micrograms per injection and mouse) up-regulated thymocyte and peripheral blood leukocyte counts in tumor-bearing mice. Tumor weight and tumor volume were significantly down-regulated after application of sME doses greater than 20 micrograms per injection. To check the influence of sME treatment on growth of experimental metastases, RAW 117 H 10 lymphosarcoma cells and L-1 sarcoma cells were intravenously inoculated into BALB/c-mice to establish liver and lung colonization, respectively. sME was regularly administered starting 24 hours after tumor cell challenge. Organ colonization was investigated on day 14 after tumor cell inoculation and demonstrated statistically significant (p < 0.05) reductions of experimental liver and lung metastases for sME-treated mice.
