Ciliary body adenoma of non-pigmented epithelium.

Adenomas of the non-pigmented epithelium of the ciliary body are rare neoplasms and most of the studies are in the form of case reports. There are only 27 documented cases of acquired neoplasm of the non-pigmented ciliary epithelium (NPCE) reported in the English literature. In most reports, there was a clinical suspicion of melanoma and the diagnosis of NPCE adenoma was made on histopathological evaluation of the resected tissue. The entity has not been reported in the Pakistani population to date. This report describes a case of ciliary body adenoma of NPCE in a 27 year old Pakistani man. The histological and immunohistochemical profiles were typical of the adenomas described in the literature.

Identification of endothelin receptor subtypes in rat ciliary body using subtype-selective ligands.

The endothelins are important vasoactive ocular peptides and there is some evidence that they may modulate intraocular pressure. We investigated the existence and localization of endothelin receptor subtypes using subtype selective ligands in rat ciliary body. Scatchard transformation of saturation binding experiments revealed that the KD and Bmax for [125I]ET-1 and [125I]ET-3 to membranes from ciliary body were 41.7+/-9 pM and 236+/-20 fmol mg-1 protein and 37. 8+/-0.4 pM and 160+/-2.0 fmol mg-1 protein, respectively. Competitive experiments in the presence of cyclic pentapeptide BQ123 (selective for ETA receptors) and BQ3020 (selective for ETB receptors), demonstrated the existence of ETA and ETB receptors in a ratio of 35:65. Cross-linking of [125I]ET-1 and [125I]ET-3 to ciliary body membranes resulted in the labeling of two bands with apparent molecular masses of 52 and 34 kDa, suggesting that ETA and ETB receptors have similar molecular mass. The 34 Kda band is a proteolytic degradation product of the 52 Kda band. Autoradiographic results show that specific [125I]ET-1 binding sites, displaced by BQ123 and BQ3020, are localized to the ciliary epithelium, supporting the idea that ETA and ETB subtype receptors exist in this tissue.

Non-selenium glutathione peroxidase without glutathione S-transferase activity from bovine ciliary body.

A glutathione peroxidase was purified from bovine ciliary body by ammonium sulfate fractionation. Sephacryl S-300 gel filtration, diethylaminoethyl (DEAE)-cellulose chromatography and hydroxyapatite chromatography. The purified enzyme has an apparent mw of 112 kDa by gel filtration and 29 kDa by SDS-polyacrylamide gel electrophoresis. The enzyme therefore is composed of four identical subunits. The ciliary enzyme is active with H2O2 (25), cumene hydroperoxide (170), t-butyl hydroperoxide (22), triphenylcarbinyl hydroperoxide (12), linoleic hydroperoxide (34) and 5-phenylpentenyl hydroperoxide (22): the numbers after substrates are K'm in microM. Glutathione is essential for the reaction; L-cysteine, dithiothreitol and 2-mercaptoethanol are inactive. Mercaptosuccinate (10 microM) inhibits the enzyme competitively (Ki = 7 microM) when cumene hydroperoxide is substrate, and uncompetitively (Ki = 10 microM) when H2O2 is substrate. No selenium was found in the enzyme by the fluorometric assay with 2.3-diaminonaphthalene. The enzyme demonstrates no glutathione S-transferase activity when tested with 1-chloro-2,4-dinitrobenzene, and several other compounds. A partial sequence of the enzyme shows some similarities both to Se-glutathione peroxidases and a glutathione S-transferase isozyme.

Lacrimal gland choristoma of the ciliary body.

A 22-month-old girl was found to have a pigmented mass of the ciliary body of the right eye. Five months later the globe was enucleated because of enlargement of the tumor and the appearance of brown pigment on the adjacent epibulbar surface. Histopathologic examination disclosed a lacrimal gland choristoma of the ciliary body with involvement of the limbal conjunctival surface and superficial sclera. Choristomas of the lacrimal gland are rare and only eight cases have been reported previously to occur intraocularly. Most of these cases showed extrascleral involvement which suggests that the anlage for these choristomas may be entrapped intraocularly during the development of the globe. We recommend conservative management, either by observation or local resection if possible.

A xanthomatous lesion resembling balloon cell melanoma of the iris and ciliary body of a dog.

A slow-growing xanthomatous tumour which diffusely infiltrated the iris and ciliary body of a dog was composed of large, pale, vacuolated cells which contained scattered fine pigment granules. Electron microscopy revealed coalescing vacuoles, lamellar membranous structures, and small groups of melanosomes in the cytoplasm, suggesting melanocytic origin. The lesion resembled balloon cell melanoma of the ciliary body of man. The behaviour of the tumour was benign. This is a clinical and pathological entity which has not been reported previously in the dog.

[The action of instilled physostigmine on the acetylcholine content of the bovine ciliary body (author's transl)].

The authors have compared the content of acetylcholine (ACh) of the two ciliary bodies (150 microgram of tissue) after the instillation of a stimulating dose of a 1% aqueous solution of physostigmine (10 applications of three drops at intervals of three minutes) into the conjunctival fornix of one eye. Kymographic measurements on guinea-pig ileum demonstrated an average elevation of the ACh content in the instilled ciliary bodies (0.48 microgram) against the controls (0.33 micron). The eyes were enucleated one hour after the last application.

Adenocarcinoma arising from the epithelium of the iris and ciliary body.

Our discussion of adenocarcinomas arising from the ciliary and iris epithelium, based upon a case report and a review of the literature, attempts summarize what little is known about these tumors. In most instances, the tumors arise from previously healthy eyes in middle-aged people. There appears to be no known way to make the diagnosis preoperatively. As long as the tumor has not extended outside the globe at the time of removal, the prognosis is excellent. The morphology of the tumors is virtually identical to those arising from the retinal pigment epithelium, with anaplastic epithelioid cells often forming tubes, cords, and acinuslike structures. In a few cases, tumors with these traits have extended from the globe and have eventually resulted in the death of the patients; one of these cases unquestionably arose from the iris and ciliary body epithelium. Knowledge of these tumors is in a primitive stage, and much clinical and pathological information is needed in order to determine their casue, diagnostic clinical features, and proper treatment.

[The tension of zonule and aging changes of ciliary bodies].

The zonular tension was studied in thirty fixed autopsy eyes (7 eyes under 30 years old, 15 eyes seventh decade and 8 eyes over 85 years old) by pulling the lens backward, and the surface structure of the ciliary body was studied histopathologically following the removal of zonular fibers. The tension for breaking zonules was 49.1 +/- 8.0 gm in younger age group, and 35.3 +/- 8.2 gm in older age group. The zonules in the younger age group was statistically stronger than those in the older age group (p less than 0.01). This breaking occurred at the region adjacent to the ciliary body in the younger age group, but at the region adjacent to the lens in the older age group. The internal surface of the pars plicata and the anterior pars plana was characterized by regular meridional ridges and grooves in the younger age group but was irregular in older age group. In the older age group, grooves among the ridges were wide. In the posterior pars plana, spine-like structure was observed learning parallel toward the lens along the course of zonular fibers. These ageing changes of zonules and ciliary body may depend on the load for the accommodation.

Results of the treatment of neovascular glaucoma by diathermy of the dissected ciliary body.

The most usual treatment of neovascular glaucoma is suppression of the production of aqueous by means of coagulation of the ciliary body. In addition to cryocoagulation, diathermic coagulation of the exposed ciliary body by Benedikt and Hiti's method has been applied recently. The results which we obtained in 21 cases with this method are encouraging. In most cases the pain disappeared and the intraocular pressure decreased to below 30 mmHg. In half of the cases cryocoagulation was performed as well as diathermy.

Tissue culture of rabbit ciliary body epithelial cells on permeable supports.

The aqueous humor is produced by the epithelium of the ciliary body, a complex structure encircling the anterior segment of the eye. Aqueous humor production occurs by active transport, but the mechanism of this process is not understood. To produce a preparation in which active transport can be investigated, we have attempted to prepare cultures suitable for measurements of ion and water flux. We have grown rabbit ciliary body epithelial cells on permeable supports, coated with several extracellular matrix proteins. We then examined the ability of these proteins to promote the growth of a differentiated layer of epithelial cells. Non-pigmented and pigmented cells formed sheets of contiguous cells when grown on a variety of support media. The most successful substrate was a permeable support produced by Falcon/Becton Dickinson coated with a mixture of collagen IV, laminin and heparan sulfate. Under these conditions, cultures could be maintained for several months, but pigmented cell cultures did not develop a measurable transepithelial resistance (TER), and the TER of non-pigmented cell cultures was typically only 20-30 omega cm2. Much higher TERs exceeding 200 omega cm2 could be measured from non-pigmented cell cultures 3-5 days after plating, but these high values were unstable. Examination of the cultures with electron microscopy revealed that the cells were partially differentiated with the formation of a basal lamina and intercellular junctions. Labelling with a specific monoclonal antibody marker for tight junction protein (ZO-1) suggested that non-pigmented cell cultures showed extensive tight junction formation. The low TER of the non-pigmented cell cultures appears therefore not to be due to the lack of tight junctions but rather to the presence of spaces between cells.

Choroidal and ciliary body malignant melanomas.

53 cases of choroidal and ciliary body melanomas have been followed for a minimum of 5 years. 26 patients had enucleation after an initial period of observation, mainly because of growth. Survival rates of these patients compared to other series treated by initial enucleation are unaffected. Small melanomas which did not grow did not cause death in this series. The results of this series thus far indicate that patient survival is not jeopardized by initial observation and that useful vision is preserved for many patients.

Partial purification of cathepsin B in the bovine ciliary body and iris.

Cathepsin B in the bovine ciliary body and iris was studied biochemically using alpha-N-benzoyl-D,L-arginine-2-naphthylamide as substrate. The enzyme was purified to 210-fold from the autolyzed extract. The partially purified enzyme had a pH optimum at 6.0 and a molecular weight of 27,000. The apparent Km value for the substrate was 1.6 mM. The enzyme was activated by disodium ethylenediamine tetraacetate, cysteine, and dithiothreitol. The enzyme activity was inhibited strongly by leupeptin and partially by hyaluronate and chondroitin sulfate A.

The fine surface architecture of the primate ciliary body with special regard to the pars plana.

The fine surface architecture of the pars plana of the ciliary body was studied by scanning electron microscopy (SEM) in young and adult primate eyes following the removal of zonular fibers by hydrolysis with hydrochloric acid. In the young eye, the internal surface of the pars plana was characterized by a series of meridional ridges and grooves and, in the posterior region, by conspicuous surface invaginations on the ridges. In the adult eye, the meridional ridges in the anterior pars plana were smaller than in the young eyes and in a varying configuration, and the meridional grooves among the ridges were wide. In the mid-pars plana, spine-like architectures were observed leaning parallel to each other toward the lens, along the course of the zonular fibers. In the posterior pars plana, the internal surface exhibited a reticularly multilayered appearance with interstices of various sizes. In incompletely digested specimens, the zonular bundles were found to dissociate into fan-shaped off-shoots, and were observed entering the meridional grooves and the surface invaginations on the ridges. The procedure used in the present study was considered to be highly useful for SEM observation of the surface morphology of the ciliary body.

Endolaser treatment of the ciliary body for severe glaucoma.

Endolaser photocoagulation of the ciliary body was used to treat 42 eyes with severe glaucoma that could not be managed successfully by medical therapy and conventional glaucoma surgery. Follow-up ranged from 6 to 36 months (mean, 13 months). The preoperative intraocular pressure (IOP) ranged from 13 to 76 mmHg (mean, 37 mmHg). The postoperative IOP ranged from 1 to 44 mmHg (mean, 17 mmHg), representing an absolute decrease of 20 mmHg, and a mean decrease of 48%. After one or two (7 eyes) treatment sessions, 11 eyes (26%) had an IOP less than 21 mmHg without medications; 21 eyes (50%) had an IOP less than 21 mmHg with medications; 5 eyes (12%) had an IOP of 21 to 25 mmHg with or without medications; and 5 eyes (12%) had an IOP greater than 25 mmHg. Postoperatively, the number of antiglaucoma medications required was reduced from a mean of 2.8 +/- 0.13 to a mean of 1.4 +/- 0.19. Twenty-three (72%) of 32 patients were able to discontinue carbonic anhydrase inhibitors.

Protein phosphatases 1 and 2A in rabbit ciliary epithelium and iris-ciliary body.

A significant dephosphorylation of exogenous phosphorylase a by serine/threonine protein phosphatases in tissue extracts of rabbit ciliary epithelium and iris-ciliary body was observed. Okadaic acid caused a concentration-dependent inhibition of this dephosphorylation. In a series of diluted samples, selective inhibitions of protein phosphatase 2A by 1 nM okadaic acid and protein phosphatase 1 by 1 microM okadaic acid were determined. In the preparation of ciliary epithelium, the ratio of protein phosphatase 1 to protein phosphatase 2A activity was estimated to be 1:2. Dephosphorylation by other phosphatases was little. In the preparation of iris-ciliary body, the ratio of protein phosphatase 1 to protein phosphatase 2A activity was approximately 2.5:1. Other phosphatases were also present.

Purification and properties of glutathione reductases from bovine ciliary body.

Two glutathione reductases, designated GR-I and GR-II, were purified from bovine ciliary body to apparent homogeneity by ion-exchange chromatography on DEAE-agarose and gel filtration on Sephacryl S-200. The two enzymes were isolated whether protease inhibitors were included or not in the medium during tissue homogenization. GR-I was found to have a molecular weight of about 140,000 by gel filtration and be composed of two identical subunits. On the other hand, GR-II was shown to exist as aggregates with molecular weights larger than 670,000. The subunit molecular weight of GR-II was estimated to be about 45,000 by SDS-polyacrylamide gel electrophoresis. The high molecular aggregate dissociated into smaller species by treatment with mercaptoethanol. The isoelectric points of GR-I and GR-II were determined to be 5.8 and 6.4, respectively. The K'm values for NADPH and oxidized glutathione (GSSG) were 22- and 84 microM for GR-I and 11- and 98 microM for GR-II. Coenzyme A-glutathione disulfide served as a substrate, though it was much less reactive than oxidized glutathione, for both enzymes. S-alkyl glutathiones were inactive as substrates. Nitrofurantoin was a non-competitive inhibitor of the enzymes. 1,3-bis(2 chloroethyl)-1-nitrosourea (BCNU) inhibited GR-I and GR-II irreversibly. The two enzymes are similar, if not identical, in their amino-acid composition. The two enzymes are also very similar on the basis of the relative position of tryptic peptides separated in two-dimensional peptide maps. From the tissue content and turnover numbers of these enzymes it is concluded that bovine ciliary body has a sufficient capability for rapid reduction of oxidized glutathione produced in the tissue during glutathione-dependent peroxide detoxification.

Effects of melanin on lysosomal enzymes in bovine ciliary body and iris in vitro.

We studied biochemically the effects of melanin on activities of lysosomal enzymes prepared from the bovine ciliary body and iris in vitro. Melanin was prepared from the bovine ciliary body and iris by acid treatment. Acid phosphatase, N-acetyl-beta-D-glucosaminidase, alpha-D-mannosidase, alpha-L-fucosidase, and beta-D-glucuronidase in the ammonium sulfate fraction were used as lysosomal marker enzymes. After the enzyme solution was incubated with melanin, enzyme activity was reduced and protein content in the supernatant was decreased. Each enzyme showed a different activity. The data suggested that 1) the decreased activity may depend on the affinity of lysosomal enzyme protein for melanin and 2) each enzyme may have a different affinity for melanin. These findings were discussed with respect to biochemical interaction between melanin and lysosomal enzymes in the ocular tissue in vivo.
