Characterization of dog repellent factor from cuticular secretion of female yellow dog tick, Haemaphysalis leachi.

During its natural life cycle, the yellow dog tick, Haemaphysalis leachi, has three hosts, and it has to spend enough time on each of them to complete a blood meal. When irritated, the females of this tick species produce a cuticular secretion that contains a dog-repelling allomone. This improves the tick's chances of survival by deterring the dog from biting the tick off its body. Employing response-guided isolation techniques in conjunction with gas chromatography-mass spectrometry, the defensive allomone of H. leachi was found to consist of the six homologous aliphatic aldehydes from hexanal to undecanal. A mixture of synthetic versions of these six aldehydes in quantities corresponding to those secreted by one tick elicited strong aversion reactions in the majority of dogs of various breeds.

Dysmyelination in Chow Chow dogs: further studies in older dogs.

The ultrastructure of myelin deficiency in Chow Chow dogs was studied in the spinal cord of a 15-month-old and a 3-year-old animal. It was found that myelination progresses with age in these dogs but is still deficient at the age of 3 years. The findings included axons with thin or uncompacted myelin sheaths, separated from each other by massive astrocytosis, and bizarre myelin formations. Normal numbers of morphologically normal oligodendrocytes were present in the myelin-deficient areas. The disease in these Chow Chow dogs consists of a strongly retarded myelination which is possibly due to a dysfunction or delay in glial maturation.

Lymphocyte 5'-nucleotidase activities in normal dogs and in dogs with malignant lymphoma.

Plasma membrane 5'-nucleotidase (5'NT) activity was assayed in lymph node lymphocytes from seven normal control dogs and in malignant lymphocytes from 25 dogs with lymphoma. The lymphoid tumors were classified according to the NCI Working Formulation into five histologic subtypes. When compared with normal controls significantly lower 5'NT activities were found in the lymphoblastic, small lymphocytic and diffuse large (noncleaved) subtypes while no significant differences were observed in the immunoblastic or small noncleaved groups. In addition, dogs with hypercalcemia or paraproteinemia had decreased 5'NT levels. However, no significant differences were found between prognostic groups or histologic subtypes of the NCI classification. In conclusion, canine lymphoma subtypes had generally decreased 5'NT activities which appeared to reflect the B or T cell lineage, degree of maturation and enzyme activity of the cell of origin.

The prevalence and epidemiology of cestodes in dogs in Clwyd, Wales. II. Hunting dogs.

Studies of a foxhound pack and gundog kennel in Clwyd, Wales showed 47 of 162 foxhounds (29%) and six of 25 gundogs (24%) to be infested with cestodes. Taeniia species were found in both groups, T. hydatigena being most common. Echinococcus granulosus and Dipylidium caninum were found in hounds only. The husbandry of the dogs was investigated and close relationships were found between diet, worming procedures and cestode infestation. These dogs play an important role in disseminating cestode eggs in the environment. Recommendations for control of the problem are made. Foxhounds and other hunting dogs play a particularly important role in the life-cycle of cestodes because their diet frequently includes uncooked carcasses and because they hunt over large areas of countryside, facilitating the widespread dissemination of infective eggs.

Determination of C-reactive protein in serum and plasma from healthy dogs and dogs with pneumonia by ELISA and slide reversed passive latex agglutination test.

The concentrations of C-reactive protein (CRP) in serum from dogs diagnosed as normal by clinical, haematological, and biochemical examination were determined by enzyme-linked immunosorbent assay (ELISA) and slide reversed passive latex agglutination (RPLA), using IgG antibody isolated from rabbit anti-canine CRP serum. The mean value of CRP in 66 normal dogs kept in private households was 8.4 +/- 4.9 micrograms/ml by ELISA and 8.5 +/- 6.3 micrograms/ml by RPLA. Thus, no significant difference was demonstrated between the values obtained by ELISA and RPLA. No significant age and sex-related differences were found in the CRP values. The mean concentration of CRP in 84 6-month-old Beagle dogs kept in kennels by breeders was 6.2 +/- 3.9 micrograms/ml by ELISA and 8.0 +/- 4.0 micrograms/ml by RPLA. Again no significant difference was found. The CRP values determined by ELISA and RPLA were closely correlated (r = 0.913). The serum and plasma concentrations of CRP measured by RPLA were also closely correlated (r = 0.994). This indicates that plasma can be used as well as serum to determine CRP concentrations by RPLA.

Comparison of air- and bone-conducted brain stem auditory evoked responses in young dogs and dogs with bilateral ear canal obstruction.

Brain stem responses to air- and bone-conducted stimuli were analyzed in 11 young dogs, using an in-the-ear transducer and a vibrator designed for human hearing tests, respectively. The mean thresholds were 0 to 10 dB for air-conducted stimuli and 50 to 60 dB for bone-conducted stimuli. The wave forms and inter-peak latencies of the waves of the auditory evoked responses elicited by air-conducted and bone-conducted stimuli were similar. This indicated that the signals had the same origin and thus both the air-conducted and the bone-conducted responses could be considered to be auditory responses. Measurement of air-conducted and bone-conducted brain stem-evoked responses in five dogs with bilateral chronic obstructive ear disease revealed thresholds of 50 to 60 dB for air-conducted stimuli and 60 to 70 dB for bone-conducted stimuli. By comparison of these results with those in the 11 young dogs, it could be concluded that there was hearing loss other than that caused by obstruction of the ear canals.

Humoral and cell-mediated responses to Malassezia pachydermatis in healthy dogs and dogs with Malassezia dermatitis.

The in vitro proliferative responses of peripheral blood mononuclear cells from seven healthy basset hounds exposed to Malassezia pachydermatis antigen (500 micrograms/ml) exceeded (P < 0.05) those of seborrhoeic basset hounds with high populations of M pachydermatis and eight Irish setters with gluten-sensitive enteropathy. The stimulation indices in the latter two groups and in eight healthy beagles were comparable. The stimulation indices of the four groups after exposure to phytohaemaglutinin did not differ significantly. The serum titres of M pachydermatis-specific IgG and IgA measured by enzyme-linked immunosorbent assays (ELISA) in 21 seborrhoeic basset hounds and 11 affected dogs of various breeds exceeded those of 14 healthy basset hounds and eight healthy beagles (P < 0.01 for IgG, P < 0.05 for IgA). Total serum IgA concentrations measured by ELISA in the affected dogs were not lower than those of healthy dogs.

Comparison of the efficacy of a synthetic dog-appeasing pheromone with clomipramine for the treatment of separation-related disorders in dogs.

Sixty-seven dogs that showed signs of distress when separated from their owners (destructiveness, excessive vocalisation and house soiling) and hyperattachment were used in a randomised, blind trial to assess the potential value of a dog-appeasing pheromone in reducing the unacceptable behaviours. For ethical reasons, there was no placebo group and the effects of the pheromone were compared with the effects of clomipramine which is regularly used to treat this type of problem. The undesirable behaviours decreased in both groups, but the overall assessment by the owners indicated that there was no significant difference between the two treatments, although there were fewer undesirable events in the dogs treated with the pheromone, and the administration of the pheromone appeared to be more convenient.

"Dog pox": some clinical aspects of an eruptive condition of certain mucous surfaces in dogs.

An eruptive condition seen on certain mucous membranes of dogs occurs frequently but often asymptomatically. Lesions have been found to be present in over 60 percent of male dogs. The infection is probably due to a canine herpes virus and has been termed conveniently but inaccurately "dog pox". In many cases lesions are present without observable clinical signs but severe irritation, especially related to lesions in the lower bowel, can give rise to marked symptoms which may be confused with abnormality of the para-anal sacs, or to chronic diarrhoea of a type suggestive of colitis. The incidence, sites of infection and possible syndromes which may be ascribed to the condition are briefly discussed on a clinical basis only.

[Control of canine visceral leishmaniasis: comparison of results from a rapid elimination program of serum-reactive dogs using an immunoenzyme assay and slower elimination of serum-reactive dogs using filter paper elution indirect immunofluorescence].

The kala-azar control program, adopted by the Fundação Nacional de Saúde-FNS (National Health Foundation) has not been able to reduce to an acceptable level the incidence of human cases. The diagnostic method utilized is a blood eluate immunofluorescence. A dogs diagnosed as infected is eliminated a mean of eighty days after the blood collection. The low sensitivity of the test used and the continuing residence of the infected dog in the region due to the elimination delay may be critical in the lack of success of this program. In this study, the FNS standard canine control method is compared to a strategy based on ELISA identification of infected dog and elimination within 7 days. In both study areas the canine seroprevalence was noted ten months before and ten months after the intervention. In the routine FNS area a 9% decrease in seroprevalence was noted, compared to statistically significant greater 27%, reduction (p = 0.0015) in the ELISA intervention area.

Assessment of the use of RNA quality metrics for the screening of articular cartilage specimens from clinically normal dogs and dogs with osteoarthritis.

OBJECTIVE: To assess 2 methods of RNA purification by use of different quality metrics and identify the most useful metric for quality assessment of RNA extracted from articular cartilage from dogs with osteoarthritis. SAMPLE POPULATION: 40 articular cartilage specimens from the femoral heads of 3 clinically normal dogs and 37 dogs with osteoarthritis. PROCEDURES: RNA was extracted from articular cartilage by 2 purification methods. Quality metrics of each sample were determined and recorded by use of a UV spectrophotometer (Spec I; to determine the 260 to 280 nm absorbance ratio [A(260):A(280) ratio]), a second UV spectrophotometer (Spec II; to determine A(260):A(280) and A(260):A(230) absorbance ratios), and a microfluidic capillary electrophoresis analyzer (to determine the ribosomal peak ratio [RR], degradation factor [DF], and RNA integrity number [RIN]). The RNA was extracted from affected (osteoarthritic) articular cartilage and assessed with the same quality metrics. Metric results were compared with visual analysis of the electropherogram to determine the most useful RNA quality metric. RESULTS: No differences in methods of RNA purification were determined by use of quality metrics. The RNA extracted from unaffected (normal) cartilage was of higher quality than that extracted from affected (osteoarthritic) cartilage, as determined by the RIN and Spec II A(260):A(230) ratio. The RIN and RR were the most sensitive metrics for determining RNA quality, whereas the DF was most specific. A significant proportion (32%) of RNA extracted from osteoarthritic articular cartilage specimens was determined as being of low quality. CONCLUSIONS AND CLINICAL RELEVANCE: No single metric provided a completely sensitive and specific assessment of the quality of RNA recovered from articular cartilage.

The effect of ingredients in dry dog foods on the risk of gastric dilatation-volvulus in dogs.

Using dry dog food label information, the hypothesis was tested that the risk of gastric dilatation-volvulus (GDV) increases with an increasing number of soy and cereal ingredients and a decreasing number of animal-protein ingredients among the first four ingredients. A nested case-control study was conducted with 85 GDV cases and 194 controls consuming a single brand and variety of dry food. Neither an increasing number of animal-protein ingredients (P=0.79) nor an increasing number of soy and cereal ingredients (P=0.83) among the first four ingredients significantly influenced GDV risk. An unexpected finding was that dry foods containing an oil or fat ingredient (e.g., sunflower oil, animal fat) among the first four ingredients were associated with a significant (P=0.01), 2.4-fold increased risk of GDV. These findings suggest that the feeding of dry dog foods that list oils or fats among the first four label ingredients predispose a high-risk dog to GDV.

German shepherd dog pyoderma: a prospective study of 12 dogs.

Twelve German shepherd dogs, each diagnosed as having a recurrent or refractory deep pyoderma (i.e., German shepherd dog pyoderma [GSP]), were evaluated for several parameters over a six-year period. Results indicated that GSP could be associated with flea allergy dermatitis, atopic dermatitis, food allergy, cell-mediated immunodeficiency, or hypothyroidism, or could be an idiopathic disease. The combination of diseases present for a given dog varied from case to case. Adequate control of the pyoderma was achieved only after each identified underlying disease was treated specifically, along with aggressive concurrent medical therapy using systemic antibiotics and medicated baths.

Desmopressin enhances the binding of plasma von Willebrand factor to collagen in plasmas from normal dogs and dogs with type I von Willebrand's disease.

A new in vitro von Willebrand factor-collagen binding activity (vWF:CBA) assay was used to assess qualitative changes in vWF in normal dogs and dogs with Type I von Willebrand's disease (vWD) following treatment with desmopressin acetate (DDAVP). Although DDAVP induced increases in vWF antigen concentrations at 1 hour postinfusion in both normal and vWD dogs (57% and 60% increases, respectively), there were disproportionately greater increases in vWF:CBA (96% and 103% increases). These results support the hypothesis that the enhanced hemostatic activity induced by DDAVP is, at least in part, due to the selective release of more functionally active vWF multimers. The assay, as described, provides a convenient means of simultaneously assessing vWF quantity and function before and after DDAVP administration.

[Perineal hernia in dogs--colopexy, vasopexy, cystopexy and castration as elective therapies in 32 dogs].

In 32 male dogs colopexy, vasopexy, cystopexy and castration was performed for the treatment of perineal hernia. Recurrence rate in this study is 22%, what is comparable to other studies using different methods. The degree of severity and the number of complications is lower with this technique than with others. Enlargement of the prostate was evident in 59% and bladder retroflexion in 22% of the dogs. A celiotomy allows to recognize, assess, reduce and fix displaced organs which is not possible by using other methods. The aim is to regain the tubular structure of the ampulla recti and to fix prostate and bladder cranioventrally to the pelvic entrance. The castration performed at the same time causes the prostate gland to atrophy within 2-3 weeks, what makes the pelvic entrance even wider and the dogs return to normal defecation.

Erythrocyte and plasma fatty acid patterns in dogs with atopic dermatitis and healthy dogs in the same household.

Recent studies have indicated that dogs with canine atopic dermatitis (CAD) may have a disorder of fatty acid metabolism: possibly low or absent activity of delta6-desaturase or delta5-desaturase, or both. To clarify this possibility, we examined the erythrocyte and plasma fatty acid patterns of 8 dogs with CAD and their 8 healthy housemates. Atopic dermatitis was diagnosed according to the criteria proposed by Willemse; other causes of dermatitis were excluded clinically and by appropriate tests. Erythrocyte ghosts were prepared from blood samples. Membrane lipids were extracted and separated by thin-layer chromatography. From plasma and lipid fractions, fatty acid content was determined by gas chromatography. In erythrocytes, but not in plasma, we observed significant differences in the fatty acid pattern that suggested a reduction in the n6 fatty acid products of the delta6- and delta5-desaturases in dogs with atopic dermatitis when compared with healthy housemates.

Urban dog rabies endemicity and dog physiology.

The frequency of dog-bite cases receiving anti rabic vaccine was studied for five years. Their monthwise frequency was studied. Six opportunities of contact in any calendar year which might lead to rabies transmission were identified to be provided by dog's physiological behaviour. A prediction on this basis of the frequency of months when dog rabies would appear, was brought out. This was compared with observed frequency. These are discussed.

Ehrlichiosis in wolves, dogs, and wolf-dog crosses.

Canine ehrlichiosis was diagnosed in wolves, dogs, and wolf-dog crosses at a small zoo in northcentral Florida. Five of 9 adult canids and all 8 pups confined to a common kennel died as a result of the infection. The epizootic was associated with a massive Rhipicephalus sanguineus infestation. Hematologic and pathologic findings in an adult wolf-dog cross that died were characteristic of canine ehrlichiosis. That animal and four other canids were seropositive for Ehrlichia canis. No deaths occurred following the oral treatment of remaining canids with tetracycline hydrochloride and the treatment of canids and premises with acaricides.
