The analytical performance of an axial inductively-coupled-plasma time-of-flight mass spectrometer (ICP-TOFMS) as a detector for fast transient chromatographic signals resulting from the coupling to capillary gas chromatography (CGC) was investigated. A cryotrapping GC-ICP-TOFMS method for the determination of volatile metal(loid) compounds (VOMs) in gases was used and the suitability of the TOF mass analyzer for multielemental speciation analysis and multi-isotope ratio determinations was studied in terms of accuracy and precision. Isotope ratios 118Sn/120Sn and 121Sb/123Sb have been determined in in-house gas standard atmospheres in Tedlar bags at two different levels (100 pg and 1 ng) for different elemental species (SnH4, MeSnH3, Me2SnH2, Me3SnH, BuSnH3, SbH3, and MeSbH2). A limitation arising from counting statistics in both detection modes could be shown. A solution containing rhodium (10 ng mL(-1)) and cadmium (40 ng mL(-1)) was introduced simultaneously to the GC outlet. Rhodium acts as a continuous internal standard and Cd is used for mass-bias correction (by measuring the 111Cd/113Cd ratio). The detection system in both pulse counting and analog mode was examined. The best attainable precision was established for Me2SnH2 (analog mode, 12 replicates, 1 ng, RSD 0.34%, accuracy 0.31%) whereas most other species ranged between 0.4 and 0.5% RSD if higher concentrations were used. The limitations of the pulse counting system are clearly seen, with peak heights of more than 2000 counts reaching saturation (for an integration time of 100 ms), which reduces the accuracy of isotope ratio determinations. A dozen VOM could be detected in an aged landfill gas sample; several unidentified Sn compounds were present. Although their isotope ratios are within the confidence value of the standards, it is not yet clear if the acquired precision is good enough to identify isotopic fractionation of metal(loid)s through biovolatilization processes. With the precision achieved, the combination of cryotrapping GC and ICP-TOFMS is a powerful tool for monitoring volatile multi-element species in multi-tracer experiments and isotope dilution methodology.
Calibrated by both experimental data and high-level coupled-cluster calculations, the BAC-MP4 methodology was applied to 51 SbL(n) (L = H, CH(3), C(2)H(5), Cl, and OH, n = 1-5) molecules, providing calculated heats of formation and associated thermodynamic parameters. These data identify a linear variation in heats of formation with ligand substitution, trends in bond dissociation energies (BDEs) with ligand identity [BDE(Sb-C(2)H(5)) < BDE(Sb-CH(3)) < BDE(Sb-H) < BDE(Sb-Cl) < BDE(Sb-OH)], and a monotonic decrease in BDE upon successive ligand elimination. The linear variation in BDE is consistent with the behavior of other group V elements, in contrast to the characteristic high-low-high trend of adjacent group III (In) and group IV (Sn) elements. Additionally, these data complement those of previous studies of metal-organic species and provide a foundation of thermochemical data that can aid in the selection of CVD precursors and deposition conditions for the growth of antimony-containing materials.
Heating of dehydroepiandrosterone under nitrogen with a concentrated solution of SbCl3 in C6H5COCl (3 h at 90 degrees C) leads to a dark violet-coloured crystalline product of the composition C38H46Cl7O2Sb2 with antomony in a trivalent state. Treatment with hydrochloric acid yields an antomony-free brown-coloured product of the composition C38H40O2 with a molecular weight of 530 +/- 10%. Upon catalytic hydrogenation, it binds 6 moles of H2, yielding a yellow crystalline material which reacts already at room temperature with dilute SbCl3 to a bluish-violet solution. The structure of the substances in question is discussed.
Chronic hepatitis C virus (HCV) infection is a worldwide health problem causing serious complications, such as liver cirrhosis and hepatoma. Alpha interferon (IFN-alpha) or its polyethylene glycol-modified form combined with ribavirin is the only recommended therapy. However, an alternative therapy is needed due to the unsatisfactory cure rate of the IFN-based therapy. Using a modified reporter assay based on the HCV subgenomic-replicon system, we found that sodium stibogluconate (SSG), a compound used for leishmania treatment, suppressed HCV replication. We have previously reported that SSG is effective at inhibiting HCV replication in a cell line permissive for HCV infection/replication and in an ex vivo assay using fresh human liver slices obtained from patients infected with HCV (26). In this study, we show that the SSG 50% inhibitory dose for HCV replication is 0.2 to 0.3 mg/ml (equivalent to 345 to 517 microM of Sb) in the HCV subgenomic-replicon system. We also found that SSG and IFN-alpha exert a strong synergistic anti-HCV effect in both the traditional isobologram analysis and the median effect principle (CalcuSyn analysis). The combination of SSG and IFN-alpha could sustain the antiviral response better than SSG or IFN-alpha alone. The results suggest that SSG may be a good drug candidate for use in combination with other therapeutics, such as IFN-alpha and ribavirin, to treat HCV infection.
The indications, the contraindications, and the characteristics of the antimonial and nonantimonial drugs clinically available for the treatment of human schistosomiasis are outlined. Of the antimonial compounds, antimony potassium tartrate or antimony sodium tartrate, both given by the intravenous route, are effective against Schistosoma japonicum, S. mansoni, and S. hematobium, but the production of severe side effects limits their use outside the treatment of individuals. Sodium antimonyl gluconate is less effective against S. mansoni and S. hemotobium and is also given intravenously. Of those antimonial compounds given intramuscularly, antimony dimercaptosuccinate is the most effective against all three common human schistosomes. Four available nonmetallic schistosomicides are considered. Niridazole, orally administered, is effective against all three common species of schistosome infecting man, but activity is maximal against S. hematobium. Many minor side effects have been described, but the major and most important side effects, neuropsychiatric symptoms and signs, are fortunately rare. Lucanthone hydrochloride, of moderate efficiency when given orally for S. hematobium or S. mansoni infections, is probably best used as a suppressant in small doses. Troublesome gastrointestinal toxicity limits its therapeutic use. Metrifonate, a cholinesterase-inhibiting organophosphorus compound, is effective only against S. hematobium. Clinical tolerance is very good. Hycanthone mesylate is highly effective against S. mansoni and S. hematobium but ineffective against S. japonicum. It is given as a single intramuscular dose. Many contraindications to its use exist, and acute hepatotoxicity has occurred infrequently. Its association with mutagenicity in certain experimental test systems has stimulated numerous ongoing studies to clarify the implications of its use in humans.
