[Studies of 15N-labeled lysine in colostomized laying hens. 6. 15N incorporation into basic amino acids of the liver, kidney, meat, bones and remaining body as well as 15N balance].

Three colostomised laying hybrids received 91.40 mg L-lysine-15N-excess (15N') each over a period of four days in a metabolism experiment with 15N-labelled lysine. After another four days, during which the hens received the same rations supplemented by commercial L-lysine, the animals were butchered and divided into individual fractions. After the hydrochloric hydrolysis of the organs and tissues the heavy nitrogen of lysine, histidine and arginine were separated, quantitatively evaluated, processed and measured with an emission spectrometer. Atom-% 15N' on an average amounted to 0.20 in the liver, 0.16 in the kidneys, 0.06 in the meat and 0.05 in the bones. Of the rediscovered 15N' applied, faeces contained 8.1%, urine 18.3%, the eggs 24.3%, the blood 4.9%, the meat 20.5%, the bones 5.2%, the gastro-intestinal tract with its contents 4.5%, the liver 3.5%, the kidneys 0.9%, the reproductive organs 3.7% and the rest 6.1%. The quota of rediscovery of the 15N' applied was 95.7%. 62% of the total 15N' was rediscovered in eggs, body and faeces as lysine 15N'. There was significantly more 15N' in the arginine in all fractions than in the histidine. The quota of the lysine-15N' of the total 15N' differed considerably in the fractions: less than 40% bones and blood; 48...56% gastro-intestinal tract, faeces, oviduct, kidneys; 62...63% remaining ovary, rest; 69...71% eggs, meat, liver. It could be proved that the alpha-amino group of lysine is to a large extent incorporated into other amino acids. Further proof that the amino acid metabolism proceeds in two phases was submitted, i.e. hight amounts of amino acids previously deposited in the body are used for egg synthesis.

Attempt to reconstruct body stature basing on circumferences of long bones in fossil body remains.

Studies were conducted on long bones from Middle Ages row cemetery in Milicz (Wrocław voivodship). Basing on measurements of trunc length in humerus, radius, ulna, femur, tibia and fibula as well as on measurements of circumferences of the truncs, simple correlation coefficients were calculated. The correlation coefficients were used to formulate regression equation permitting to reconstruct bone length from corresponding circumference in cases when epiphysis was destroyed. When applied appropriately, the method may be used to reconstruct length of long bones and the entire stature of extinct population members.

[Utilization of 15N-labeled urea in laying hens. 9. Bones, feathers and 15N-incorporation in the rest of the body].

For studying the incorporation of the 15N labelled urea into individual organs and tissues 3 colostomized laying hens were butchered after they had received 1% urea (96.06 atom-% 15N excess) with a high quality ration over a period of six days and after receiving conventional urea for another two days. Nitrogen and atom-% 15N excess (15N') were determined in the bones, the feathers and the remaining body (skin, lungs and windpipe, head with comb and wattle, lower leg without bones and with skin, pancreas and fatty tissue). In the remaining body the atom-% 15N' was determined in 15 amino acids. The labelling in the remaining body and the bones was approximately the same and averaged 0.37 atom-% 15N'. A significantly lower relative frequency could be detected in the feathers. The lysine of the remaining body contained only 0.04 atom-% 15N', tyrosine 0.06, histidine and arginine 0.07. The phenylalanine and proline molecules were labelled with 0.11 atom-% 15N'. Most 15N' was incorporated in serine and glutamic acid with over 0.30 atom-%. In the six non-essential amino acids out of the 15 amino acids studied, 48.6 of the non isotopic nitrogen of the total N of the remaining body and 70.7% of the isotopic nitrogen of total 15N' could be detected. Consequently the urea-N is mainly used for the synthesis of the non-essential amino acids, with its utilization being very low.

Body louse remains found in textiles excavated at Masada, Israel.

A leg of the body louse, Pediculus humanus humanus L. (Phthiraptera: Pediculidae), was found in a storeroom at Masada build during the reign of King Herod the Great. The unearthed culture material clearly indicates that the room was occupied by the rebels during the first Jewish revolt against the Romans (AD 66-73/4). The context of the textiles associated with the louse and their nature suggest a rebel origin. An exuvium of a leg of the third nymphal stage of body louse was found. The first three parts of the leg (tarsus, including the tarsal claw, tibia and femur) were preserved entirely, while the fourth segment of the leg, the trochanter, was only partially conserved. The comparison of the tarsus of this specimen with the legs of present day third instar nymphs of body and head lice revealed that the leg found within the archaeological debris belongs to a body louse.

Exchange of nucleoside monophosphate-binding domains in adenylate kinase and UMP/CMP kinase.

Two types of active chimeric enzymes have been constructed by genetic engineering of chicken cytosolic adenylate kinase (AK) and porcine brain UMP/CMP kinase (UCK): one, designated as UAU, carries an AMP-binding domain of AK in the remaining body of UCK; and the other, designated as AUA, carries a UMP/CMP-binding domain of UCK in the remaining body of AK. Steady-state kinetic analysis of these chimeric enzymes revealed that UAU is 4-fold more active for AMP, 40-fold less active for UMP, and 4-fold less active for CMP than the parental UCK, although AUA has considerably lowered reactivity for both AMP and UMP. Circular dichroism spectra of the two chimeric enzymes suggest that UAU and AUA have similar folding structures to UCK and AK, respectively. Furthermore, proton NMR measurements of the UCK and UAU proteins indicate that significant differences in proton signals are limited to the aromatic region, where an imidazole C2H signal assigned to His31 shows a downfield shift upon conversion of UCK to UAU, and the signals assigned to Tyr49 and Tyr56 in the UMP/CMP-binding domain disappear in UAU. In contrast, AUA has a Tm value about 11 degreesC lower than AK, whereas UAU and UCK have similar Tm values. These results together show that the substrate specificity of nucleoside monophosphate (NMP) kinases can be engineered by the domain exchange, even though the base moiety of NMP appears to be recognized cooperatively by both the NMP-binding domain and the MgATP-binding core domain.

Technetium-99m labelled hydrazinonicotinamido human non-specific polyclonal immunoglobulin G for detection of infectious foci: a comparison with two other technetium-labelled immunoglobulin preparations.

Recently a new linker - hydrazinonicotinate (HYNIC) - was introduced for labelling of proteins and peptides with technetium-99m. HYNIC and other linkers have been used for labelling of human non-specific polyclonal immunoglobulin G (hIgG) with 99mTc for the detection of infections. In this study we compared the tissue distribution of three different 99mTc-hIgG preparations in groups of five Wistar rats with a focal intramuscular infection with Staphylococcus aureus. We compared 99mTc-HYNIC-hIgG with 99mTc-hIgG labelled via the so-called Schwarz method (reduction of disulphide bonds) and with the 99mTc-labelled commercially available Technescan-HIG. Unlike the HYNIC linker, in the two other labelling methods free sulph-hydryl groups are involved in the binding of 99mTc. High-performance liquid chromatography analysis of the labelled preparations and of plasma samples revealed aggregate or polymer formation in all three agents; this was least pronounced in the product labelled by means of the Schwarz method. The tested preparations did not show signs of degradation in vitro. The difference in linker chemistry was reflected in the tissue distribution. Thus the biodistribution of 99mTc-HYNIC-hIgG was significantly different from the distribution of the two other preparations: abscess (1.4%+/-0.2%ID/g), muscle, liver, spleen, plasma, lung, bone marrow, and small intestine concentrations were higher at 24 h p.i.; kidney uptake (1.19%+/-0.08%ID/g) was significantly lower. The abscess-to-plasma and the abscess-to-muscle ratios (0.5 and 11, respectively), however, were in the same range for the three preparations tested. Quantitative analysis of the scintigraphs revealed that the total body clearance of 99mTc-HYNIC-hIgG was significantly slower than for the other agents. The abscess uptake of 99mTc-HYNIC-hIgG as a percentage of the remaining body activity was significantly higher. Based on its high abscess uptake, its low uptake in the kidneys and the high percentage of its abscess uptake in relation to the remaining body activity, we conclude that 99mTc-HYNIC-hIgG seems superior to the two other preparations tested for the detection of infections.

Relationship of brain blood flow and oxygen consumption to perfusion flow rate during profoundly hypothermic cardiopulmonary bypass. An experimental study.

A study was made of the relation of brain blood flow and oxygen consumption to changes in perfusion flow rate during cardiopulmonary bypass at 20 degrees C in nine cynomolgus monkeys. Four perfusion flow rates varying from 0.25 to 1.75 L X min-1 X m-2 were randomly instituted, each for a 10 minute period. At the end of each period, brain arteriovenous oxygen content difference was measured and 15 mu radioactive microspheres were injected into the arterial perfusion line. The brain was then removed and section into anatomic regions and radioactivity was counted. Regional and total brain blood flows were calculated, as was whole brain oxygen consumption. Brain perfusion continued in all areas at all perfusion flow rates. Whole brain blood flow decreased (p less than 0.0001) as perfusion flow rate was reduced (45 +/- 6.5, 41 +/- 7.9, and 23 +/- 2.8 ml X min-1 X 100 gm-1 at 1.5, 1.0, and 0.5 L X min-1 X m-2, respectively). The proportion of the total perfusion delivered to the brain increased (p = 0.003) with decreasing perfusion flow rates (5.4% +/- 0.78%, 7.1% +/- 1.24%, and 8.2% +/- 1.11% at 1.5, 1.0, and 0.5 L X min-1 X m-2, respectively). Brain blood flow resistance remained unchanged (p = 0.4) while that of the remaining body increased (p less than 0.0001). There was a greater reduction of blood flow in the cortical white matter (p = 0.01) than in other regions of the brain. Brain oxygen consumption was the same (p = 0.5) at all perfusion flow rates, related to an increasing percent oxygen extraction with decreasing perfusion flow rate (p less than 0.0001). The data indicate that all areas of the brain remain perfused, even at low perfusion flow rates, during profoundly hypothermic cardiopulmonary bypass, and that brain oxygen consumption is maintained in part by increased oxygen extraction and in part by redistribution of the perfusate from the remaining body to the brain.

The osteology of the African elephant (Loxodonta africana): vertebral column, ribs and sternum.

The vertebral column, sternum and ribs of the African elephant were studied and illustrated. In the cervical series, the vertebrae are characterized by very short (compressed) vertebral bodies and short spinous processes. There are 20-21 thoracic vertebrae that carry ribs, and three lumbar vertebrae. The neural arches of the five sacral vertebrae fuse with each other as well as with the wings of the ilium, while the intervertebral discs do not ossify and the vertebral bodies remain separate. There are 19-21 caudal vertebrae. In the latter, the neural arches of only the first five to six vertebrae fuse dorsally, the vertebral foramens of the other vertebrae as well as the vertebral canal remain open dorsally. The body of the first rib is greatly expanded while that of the last three to four ribs are reduced. The cartilages of the first six ribs articulate with the sternum, the last five to six ribs do not bear costal cartilages and are not attached to the costal arch. The sternum consists of five sternabrae that form three approximately equal, but separate, segments. The first segment is formed by the first sternabra, the second segment is formed by the second to fourth sternabrae and the last segment is formed by the fifth sternabra. The first and second sternabrae articulate with each other by means of a synovial joint, the second to fourth sternabrae are fused to each other and the fourth and fifth sternabrae are loosely attached to each other by connective tissue.
