Type 1 deiodinase is stimulated by iodothyronines and involved in thyroid hormone metabolism in human somatomammotroph GX cells.

BACKGROUND: Local 5'-deiOdination of l-thyroxine (T4) to the active thyroid hormone, 3,3',5-tri-iodothyronine (T3) via two deiodinase isoenzymes (D1 and D2) has an important role for various T3-dependent functions in the anterior pituitary. However, no evidence has been presented yet for thyroid hormone inactivation via the 5-deiodinase (D3) in anterior pituitary models. METHODS: Using the human somatomammotroph cell line, GX, we analysed effects of T3 and its 5'-deiodination product, 3,5-di-iodothyronine (3,5-T2), on deiodinase activities, measuring release of iodide-125 (125I-) from phenolic-ring- or tyrosyl-ring-labelled substrates respectively. RESULTS: T3 and 3,5-T2 rapidly stimulated D1 activity in GX cells in the presence of serum in the culture medium, whereas D2 activity was not detectable under these conditions. However, when the cells were kept under serum-free conditions, specific activity of D2 reached levels similar to those of D1. With tyrosyl-ring labelled 3, 5-[125I]-,3'-T3 as substrate, a significant release of 125I- was observed in GX cell homogenates. This is comparable to the D1 activity of liver membranes, which preferentially catalyses 5'-deiodination, but to some extent also 5-deiodination, at the tyrosyl ring. CONCLUSIONS: D1 activity of human GX cells is increased by T3 and 3,5-T2. Inactivation of T3 in the anterior pituitary might occur by deiodination at the tyrosyl ring via D1, thus terminating the stimulatory thyroid hormone signal in human somatomammotroph cells.

[Counteraction effect of intermittent -Gx on changes of responsiveness of basilar artery in tail-suspended rats].

OBJECTIVE: To investigate counteracting effects of intermittent -Gx exposure on changes of responsiveness of basilar artery in tall-suspended rats. METHOD: Rats tail-suspended for 3 wk were daily subjected to 1 h 1.5 G or 2.6 G centrifugation, simulating -Gx exposure of various G values. Responses of perfused isolated basilar arterial rings to KCl was examined. RESULT: Maximal isometric contractile response to KCl were significantly enhanced in arterial rings isolated from 3 wk tail-suspended rats as compared with that from simultaneous control rats (P < 0. 05). However, maximal isometric contractile responses to KCl of arterial rings isolated from 3 wk tail-suspended plus daily -Gx exposure rats were no difference as compared with that from simultaneous control rats, and were significantly decreased (P < 0.05) as compared with that from 3 wk tail-suspended rats. CONCLUSION: Daily intermittent -Gx exposure as a countermeasure to prevent the effects of simulated weightless on changes in responsiveness of basilar artery from tail-suspended rats was valid.

[Effects of high +Gx during simulated spaceship emergency return on learning and memory in rats].

OBJECTIVE: To observe the effects of high +Gx during simulated spaceship emergency return on learning and memory in rats. METHOD: Thirty two male SD rats were randomly divided into control group, 7 d simulated weightlessness group, +15 Gx/180 s group and +15 Gx/180 s exposure after 7 d simulated weightlessness group, with 8 rats in each group. The changes of learning and memory in rats were measured after stresses by means of Y-maze test and step-through test. RESULT: In Y-maze test, as compared with control group, percentage of correct reactions decreased significantly (P<0.01) and reaction time increased significantly (P<0.01) in hypergravity after simulated weightlessness group at all time after stress; as compared with +15 Gx group or simulated weightlessness group, percentage of correct reactions decreased significantly (P< 0.05) and reaction time increased significantly (P< 0.05) immediately after stress. In step-through test, as compared with control group, total time increased significantly (P<0.01) in hypergravity after simulated weightlessness group at 1 d after stress; latent time decreased significantly (P<0.01) and number of errors increased significantly (P< 0.01) at all the time after stress. As compared with +15 Gx group, total time increased significantly (P<0.05) immediately, 1 d after stress. As compared with simulated weightlessness group, total time and number of errors increased significantly (P<0.05) immediately after stress. CONCLUSION: It is suggested that +15 Gx/180 s and simulated weightlessness may affect the ability of learning and memory of rats. Simulated weightlessness for 7 d can aggravate the effect of +Gx on learning and memory ability in rats.

[Pharmacologic profile of a new compound with antiinflammatory activity: 2-(4-dimethylvinylphenyl)propionic acid (Gx 258)].

Gx 258, a new phenylalkanoic acid derivative, was shown in animal tests to possess potent antiinflammatory, analgesic and antipyretic activity. Its antiinflammatory activity was not mediated via the pituitary-adrenal axis, since it was also effective in adrenalectomized rats. Since large doses of Gx 258, 9-83 times greater than those showing antiinflammatory activity, were required to produce gastrointestinal lesions in rats, Gx 258 appears to have a very good therapeutic index. Its therapeutic index as an antiinflammatory and analgesic agent was 2.3-20.5 times greater than that of the parent compound ibuprofen. Gx 258, like ibuprofen, inhibited the biosynthesis of prostaglandins in vitro.

The elastolytic properties of subtilisin GX from alkalophilic Bacillus sp. strain 6644 provides a means of differentiation from other subtilisins.

A serine protease exhibiting high activity in alkaline media was purified from alkalophilic Bacillus strain GX6644. The enzyme, subtilisin GX, has a molecular weight of 25,000 and a pI greater than 9.5. The protease exhibited high elastolytic activity, but unlike most elastin hydrolyzing enzymes, elastin hydrolysis and binding were not inhibited by 0.1 M NaCl. The elastolytic properties of subtilisin GX together with its specificity toward amino acid phenyl esters functionally distinguishes this protease from other subtilisins. However, comparisons of the available amino-terminal sequence of subtilisin GX with subtilisins from alkalophilic and neutrophilic Bacillus species revealed extensive homology.

[Tolerance of +Gx by MIR 22 -- 27 main crew in space flights].

Data on +Gx tolerance by 16 cosmonauts-members of the Mir 22-27 main crews in missions from 8 up to 380 days in duration are reported. All cosmonauts showed good tolerance of g-loads during insertion into orbit. Owing to the use of anti-g suit Centaur and water-salt supplements prior to deorbiting, tolerance of +Gx in the course of descent was satisfactory in the majority of the cosmonauts and low only in one person. As compared with short missions (8 to 21 days), strain of the physiological systems exposed to the deorbit +Gx loads following an extended period in microgravity (186 to 380 days) was greater and manifested by more often arrhythmias, hard breathing and speech, vestibulo-autonomous reactions, petechial hematomas into the back tegmentum, and more severe sinus tachycardia. As a rule, there were no symptoms indicative of poor g-tolerance (visual disorders etc.) which speaks for effectiveness of the anti-g measures in most of the cosmonauts. However, prognostically bad cardiac rhythm disturbances (multitude of single, group and systemic polytope ventricular extrasystoles) registered in one cosmonaut on the deorbit phase mean that efforts should be redoubled to improve countermeasures against the deconditioning of organism by microgravity, particularly the cardiac rhythm disturbances.

[Correlation of the cosmonauts' physiological reactions to +Gx loads during deorbit with the hemodynamic shifts in the period of short-term microgravity].

Physiological reactions of four cosmonauts to +Gx during the descent after short-term (less than 30 d) space flights were correlated with the hemodynamic shifts developed previously during the LBNP test and occlusive plethysmography in microgravity. The cosmonauts who had exhibited a sharp decrease in cerebral circulation during the in-flight LBNP tests showed signs of deteriorated cerebral blood flow (vertigo, weakness, hyperhydration) at the attempt to assume the vertical posture and to climb out of the landing module. Also, the cosmonauts found to have the arterial and venous tone significantly dropped in microgravity, suffered petechial hemorrhages in the back integument during the exposure to +Gx. These data suggest a correlation between the physiological reactions of cosmonauts to +Gx at the end of short-term space flight and the shifts in hemodynamics earlier in microgravity.

[Resistance to +Gz and +Gx loads of subjects in older age groups, healthy and with initial signs of atherosclerosis].

Tolerance to +Gz and +Gx acceleration of healthy and atherosclerotic subjects, aged 40-49, was investigated during 256 centrifugation tests in Bulgaria and the USSR. As compared to the healthy people, the atherosclerotic subjects showed a lower tolerance: when exposed to 7 +Gz the tolerance threshold decreased from 5.93 +/- 0.57 g to 5.7 +/- 0.44 g (i. e. by 0.23 g). Visual disorders in the healthy and atherosclerotic subjects were recorded in 29 and 50%, respectively, and loss of consciousness in 9.56 and 15.15%, respectively (Bulgarian data). When exposed to 5 +Gz and 6 +Gx no subjects displayed visual disorders or loss of consciousness (Soviet data). However, lower tolerance of 5 +Gz due to cardiac arrhythmias was observed in 4.8% atherosclerotic patients and in none healthy subjects. Lower tolerance to 6 +Gx associated with cardiac arrhythmias was recorder in 23.8% atherosclerotic versus 11.8% healthy subjects (i. e. 12% more). Nevertheless, in most subjects, both healthy and atherosclerotic, acceleration tolerance was good. These findings indicate that individual assessment of acceleration tolerance is of great importance for people older than 40 years with health abnormalities.

Stent recoil: comparison of the Wiktor-GX coil and the Palmaz-Schatz tubular coronary stent.

Wiktor-GX coil stents and Palmaz-Schatz 154A tubular stents were implanted in a consecutive series of 40 patients and 45 arteries undergoing successful primary stent placement in de novo lesions treated with single stents in order to compare the acute recoil of a coil stent to a tubular stent in native coronary arteries. Acute stent recoil was determined by measuring the difference between the maximum inflated diameter of the stent deployment balloon and the minimum lumen diameter after stent deployment. There was no difference between the two groups for reference vessel diameter, nominal balloon diameter, maximum inflated balloon diameter, maximum inflation pressure, or poststent vessel minimum lumen diameter. Acute recoil averaged 0.15 +/- 0.2 mm for the Wiktor-GX and 0.13 +/- 0.1 for the Palmaz-Schatz (P = 0.58). In this series of patients, there was no significant difference between the acute recoil of the coil stent compared to the tubular stent.

Studies of the orientation of the mitochondrial redox carriers. III. Orientation of the gx and gy axes of the hemes of cytochrome oxidase with respect to the plane of the membrane in oriented membrane multilayers.

The EPR absorption properties of the hemes of cytochrome oxidase and their liganded derivatives were examined in oriented multilayers from isolated oxidase, mitochondrial membranes and membrane fragments of a bacterium, Paracoccus denitrificans. The hemes of the oxidase in all the systems investigated were oriented normal to the plane of the multilayers. The directions of the g signals corresponding to the gx and gy axes of the g tensor were found to be different in low-spin ferric heme in fully oxidized oxidase and in half-reduced liganded oxidase. It is suggested that this different orientation of gx and gy in fully oxidized oxidase and half-reduced liganded oxidase arises because the respective EPR signals belong to two different hemes, those of cytochrome a and a3.

The Orbital Period of the Accreting Pulsar GX 1+4.

We report strong evidence for a approximately 304 day periodicity in the spin history of the accretion-powered pulsar GX 1+4 that is most probably associated with the orbital period of the system. We have used data from the Burst and Transient Source Experiment on the Compton Gamma-Ray Observatory to show a clear periodic modulation of the pulsar frequency from 1991 to date, in excellent agreement with the ephemeris proposed by Cutler, Dennis, & Dolan in 1986. Our results indicate that the orbital period of GX 1+4 is 303.8+/-1.1 days, making it the widest known low-mass X-ray binary system by more than 1 order of magnitude and putting this long-standing question to rest. A likely scenario for this system is an elliptical orbit in which the neutron star decreases its spin-down rate (or even exhibits a momentary spin-up behavior) at periastron passages due to the higher torque exerted by the accretion disk onto the magnetosphere of the neutron star. These results are not inconsistent with either the X-ray pulsed flux light curve measured by BATSE during the same epoch or the X-ray flux history from the All-Sky Monitor on board the Rossi X-Ray Timing Explorer.

Changes of pulmonary function in humans during exposure to +Gx acceleration after simulated and real microgravity.

An important goal of space medicine is preserving high tolerance and performance of cosmonauts an ring exposure to acceleration at the final flight stage given varying mission duration. Among physiological mechanisms limiting +Gx acceleration tolerance, an important role is played by disturbances of external respiration resulting from alterations of respiratory biomechanics, pulmonary gas exchange conditions, and arterial hypoxemia. However, at present data on external respiration changes during exposure to +Gx acceleration after simulated and real microgravity of varying duration, are extremely scanty.

[+Gx-tolerance by the Cosmonauts of ISS crews 1, 6-9 and visiting crews 1-7 aboard Soyuz vehicles].

Analysis of +Gx tolerance of 15 members of short-term ISS missions and 9 members of long-term ISS missions aboard the Soyuz vehicles showed good tolerance during insertion and satisfactory during descent provided the use of in-flight countermeasures and anti-g suit Kentaur inside the vehicle. Objective data about the tolerance of off-nominal +Gx (6.26 and 8.1 G) pointed to a more pronounced sinus tachycardia and tachypnea in a ballistic descent following long-duration weightlessness as compared with nominal. Physiological deviations were transient, functional by character. Cardiac arrhythmia was largely observed during return to Earth. Two cosmonauts were found to develop prognostically unfavorable ECG indications during off-nominal descent. Main factors in these deviations could be age (above 45) and existence of some individual specifics of cardiac rhythm regulation. These results emphasize the necessity of more careful screening of candidates older than 45 years, as weightlessness may impact their general condition and tolerance of g-loads during return to Earth.

[Effect of gravitational loads (+Gx) of the training schedule on the structure of the main arteries of dogs].

In dogs subjected to gravitational overloadings (+Gx) according to a special training schedule the wall structure of the main arteries (carotid, humeral, femoral and aorta) was studied by histological methods. The amount of catecholamines (adrenaline and noradrenaline) in the venous blood plasma and in the adrenal tissue was the index of the sympathoadrenal system condition. It was demonstrated that in trained animals the tolerance threshold to continuously increasing overloadings (+Gx) rose. It was accompanied by an elevated amount of catecholamines in blood and the adrenals In the walls of the main arteries studied a moderate hyperelastosis and a slight collagenization of the adventitia was noted. All parts of the microcirculatory bed were moderately dilated, which favoured a better blood outflow. The data obtained demonstrate adaptational changes, which can be interpreted as indices of training and reliability of the organism.

[Tolerance of +Gx-loads by females before and after a 120-day hypokinesia].

The goal of the work was to study female tolerance of g-loads before and after simulated weightlessness, and evaluate the effectiveness of countermeasures and anti-g means proposed for space flight. Centrifuge with a 7.25 m arm was used for 29 runs of 8 female subjects. The subjects were exposed to the transverse loads (+GX) at 8.2 g on a special regimen with the total run time of 9 min to simulate the ballistic descent from orbit in a SOYUZ-TM vehicle. Baseline tolerance of the females to transverse loads up to +8GX was good and satisfactory. Following 120 days of bed rest without the countermeasures or the anti-g suit g-tolerance was significantly lowered as evidenced by visual disorders in 67% of cases and exaggerated, if compared with the pre-hypokinesia levels, heart and respiration rates, i.e. 20% and 23.5%, respectively. The countermeasures and anti-g suit Centaur maintained g-tolerance at essentially the baseline level and precluded visual disorders. Results of the study speak in favor of the countermeasures and anti-g means used by the females in simulated long-duration weightlessness which can be recommended for the final phase of extended space mission.

[Role of age in +Gz and +Gx tolerance by female candidates to astronauts].

Analyzed was the role of age in the tolerance of +Gz (up to 5 units) and +Gx (up to 8 units) by female candidates to cosmonauts on the stage of initial centrifuge selection in the years of 1979 through to 1993. Selection was made of 26 healthy non-flying women aged 23 to 39 who were divided into 4 groups with 5-year intervals. Female tolerance of g-loads and shifts in a number of physiological parameters during centrifugation were discovered to be age-dependent. The poorest tolerance of +Gz and +Gx loads was typical of women below 25 years, the best, at the age of 31 to 40 years. It was stated that tolerance of these demanding g-loads by women, especially below 30, was generally worse compared with men. There were also some distinctions in the cardiovascular response in women and men. These results point to the significance of centrifugation in selection female candidates to cosmonauts irrespective of their age.

[Human tolerance of +Gx loads in anti-G suits during deorbiting of the "Soiuz" space vehicles].

Reported are results of the study of tolerance of the "thorax-back" g-loads (+Gx) during deorbiting of 83 total suited and unsuited cosmonauts in the period of 1997-1997. G-loads averaged 3.7 +/- 0.058 units (from 3.1 up to 5.1 units). Pneumatic gradient G-suits (PGS) Karkas and Centaur had a positive effect on the +Gx tolerance of humans following microgravity. As compared with unsuited crews returning from missions of similar duration, wearing PGS has proven to improve the general health of cosmonauts and to reduce the strain of the leg and abdominal muscles. Tight fitting of PGS abolished visual disorders due to the longitudinal component of G-loads that were registered in 6.6% of observations of unsuited cosmonauts. Sinus tachycardia was significantly weaker (P < 0.05). The authors recommend the use of the standard antiblackout Centaur suit as the efficacy of this PGS was equal to that of Karkas while its ergonomic characteristics were superior.

Marine Vibrio Biocatalysts as Unique Green Transformation (GX) Tools at the Time to Sustainable Development Goals (SDGs).

Vibrios have sustained various types of ocean ecosystems, being key players in marine mineral cycles and essential partners in specific groups of marine life. Observed genome plasticity and metabolic versatility are some of the unique biological features of vibrios, and these traits could contribute in expanding their ecological niche in marine environments. Vibrios are now recognized as ecophysiologically essential microbial species for our planet. At the time to "Sustainable Development Goals" (SDGs), their genome plasticity and metabolic versatility have also been studied with the aim of solving global issues such as energy production and plastic pollution by creating new microbial biocatalysts. Here, we introduce recent progress on the application of vibrios aiming towards green transformation (GX).
