Ampicillin or cefotaxime, alone or in combination with amikacin, were tested at levels achievable in CSF for bactericidal activity against eight clinical isolates of Haemophilus influenzae serotype b. Endotoxin release was determined by the limulus amoebocyte lysate test and by macrophage tumour necrosis factor production for each beta-lactam antibiotic, alone and in combination with amikacin. Accelerated killing was observed when amikacin was added to ampicillin or cefotaxime; however, the additional antibiotic-induced bacterial lysis observed after the addition of amikacin to beta-lactam antibiotics was not associated with an increase in endotoxin release.
The Compu-pet 100 diluter/dispenser is a versatile and accurate instrument which can be employed for a wide variety of microbiological tests and techniques. Time in performing tests is often more than halved when the diluter is used. Large numbers of tests can be performed with minimal fatigue.
In the course of screening for immunomodulators inhibiting the mixed lymphocyte culture reaction (MLCR), we found a novel immunosuppressant, dethymicin in mycelium of Amycolatopsis mediterranei MI710-51F6. From physico-chemical properties and biological activity it is different from immunosuppressants produced by microorganisms such as cyclosporins, FK506 and rapamycin. It inhibited immune responses in vitro and in vivo, and prolonged skin allograft in rats.
Laboratory and clinical studies on cefprozil (CFPZ, BMY-28100), a new cephem antibiotic, were carried out in the field of pediatrics. The results obtained are summarized as follows: 1. Serum concentrations, urinary concentrations and urinary recovery rates of CFPZ were determined upon oral administration of CFPZ after meal at doses of 4 mg/kg granules in a case, 7.5 mg/kg granules in 2 cases and 15 mg/kg granules in one. Peak serum levels of CFPZ were obtained at an hour in 3 cases and at 2 hours in 1 case after administration of the drug with a range of 2.7-8.6 micrograms/ml with half-lives of 0.69-0.95 hours. Urinary recovery rates in the first 6 hours after administration ranged from 59.4-71.3%. 2. MICs of CFPZ against 36 clinical isolates (Staphylococcus aureus 4 strains, Streptococcus pneumoniae 5, Streptococcus pyogenes 5, Escherichia coli 5, Haemophilus influenzae 12, Haemophilus parainfluenzae 4, and Branhamella catarrhalis 1) were compared with those of cefaclor (CCL) and ampicillin (ABPC). The antibacterial activity of CFPZ was superior to those of CCL against Gram-positive cocci, and to those of ABPC against E. coli, and was equal to those of CCL and inferior to those of ABPC against H. influenzae. 3. Thirty-seven pediatric patients with acute infectious diseases (pharyngitis/tonsillitis 17, bronchitis 7, pneumonia 3, skin and soft tissue infection 2, and urinary tract infection 8) were treated with CFPZ at daily doses of 10-47 mg/kg t.i.d. as a rule. The efficacy rates were 100% clinically and 56% bacteriologically. 4. Side effects or abnormal laboratory test values were not observed except for an increased platelet count in 1 case and elevated GOT, GPT values in 2 cases.
The 12 known serotypes of P. haemolytica and two biotypes A and T were found to occur in Kenya. Biotype T was more common in disease conditions than biotype A, which was more common in the nasal passages of healthy animals. Only biotypes A strains were recovered from cattle and the majority were serotypes 1 and 2, but serotypes 4 and 11 were also isolated. All serotypes were found to occur in sheep and goats, but serotypes 3, 4, 6, 8 and 10 were more commonly associated with pneumonia. It was observed that chickens could harbour both biotypes A and T in pathological conditions. Biotype A serotype 2 was isolated from an adult wildebeest, but the prevalence of P. haemolytica in wild animals needs furter investigation.

In this article a simple method of sensitivity testing of mycobacteria in semi-liquid serum-sauton-agar as AHSK-test is described and first results are reported. The sensitivity to INH, SM, EMB and RMP is tested. In comparative tests between the economical-test and the AHSK-test a total of 100 strains of mycobacteria of different sensitivity are investigated. The results largely agree with each other, from which the evaluation of the sensitivity test in the AHSK-test in 54% of the strains after 7 days, in 40% after 14 days and in 6% after 21 days follows. The simple composition and preparation of the culture medium as well as the rapid and characteristic growth of M. tuberculosis and M. bovis make the AHSK-test to a simple, economical and rapid method for sensitivity testing.
