Aminoglycoside antibiotics: thin-layer chromatography, bioautographic detection and quantitative assay.

A sensitive method is described for the identification of aminoglycoside antibiotics, for the detection of agents exhibiting biological activity in the course of fermentation, isolation and purification, and for following their chemical conversion. In the multi-component nebramycin complex the ratio of the factors can be established by the quantitative assay described.

[Effect of antibiotic resistance on the appearance of mutants in a culture of Streptomyces cremeus subsp. tobramycini producing an aminoglycoside complex].

Possible formation of auxotrophs and changing of the antibiotic production property connected with resistance to antibiotics of different modes of action were studied in Streptomyces cremeus subsp. tobramycini producing the nebramycin complex of 2-desoxystreptamine derivatives. Four hundred and five spontaneous and 1800 gamma-radiation induced antibiotic resistant mutants of the culture were studied. The frequency of the auxotrophs was shown to be increasing. Correlation between formation of strains producing monocomponent aminoglycosides and antibiotic resistance was observed. The frequency of mutants with preferable synthesis of the tobramycin component among strr-, rifr- and rubr-mutants was 3--10 times higher than among the sensitive portion of the population when total selection was used. Therefore, the spontaneous mutation of antibiotic resistance is selective with respect to both isolation of auxotrophs and strains producing separate aminoglycoside antibiotics.

[Antibiotic formation from a pyrimidine base group and an aminoglycoside group by a Streptomyces coeruleoaurantiacus sp. nov. culture].

An actinomycete strain 4009 was isolated from a soil sample collected in Volgograd. The strain showed a broad antibacterial spectrum. It produced antibiotics belonging to 2 different groups of chemical compounds, i.e. amycetin from the group of pyrimidine bases and nebramycin from the group of aminoglycosides. The antibiotic-producing organism is described as a type strain of Streptomyces coeruleoaurantiacus sp. nov. The new species is characterized by spiral spore chains, wart-like spore surface, blue aerial mycelium and dark yellow, orange-yellow or brown-orange-yellow substrate mycelium, the absence of soluble pigments on synthetic media and secretion of melanin pigments on the Tresner medium. Various conditions for submerged production of the antibiotic were studied.

Analysis of a ribosomal RNA methylase gene from Streptomyces tenebrarius which confers resistance to gentamicin.

Resistance to the aminoglycoside gentamicin in the nebramycin producer, Streptomyces tenebrarius, occurs at the level of the ribosome. A resistance determinant isolated from this actinomycete was previously shown to encode a methylase enzyme which modifies residue G-1405 of 16S ribosomal RNA. This gene (kgmB) has been sequenced and expressed in Escherichia coli using lacZ transcriptional signals since, like many other actinomycete genes, kgmB is not expressed in E. coli from its own promoter. The 5' end of the kgmB transcript has been mapped revealing a single promoter which does not obviously conform to the prokaryotic consensus.

Involvement of 16S ribosomal RNA in resistance of the aminoglycoside-producers Streptomyces tenjimariensis, Streptomyces tenebrarius and Micromonospora purpurea.

Resistance to aminoglycoside antibiotics in Micromonospora purpurea (the producer of gentamicin C complex), Streptomyces tenebrarius (the nebramycin producer) and Streptomyces tenjimariensis (which makes istamycin) occurs at the level of the ribosome. Reconstitution analysis has revealed, in each case, that 16S rRNA plays a critical role in determining such resistance.

In vitro susceptibility testing with tobramycin.

Tobramycin (nebramycin factor 6) is an aminoglycoside antibiotic active in vitro against many gram-negative species and Staphylococcus aureus. The susceptibility of 191 recently isolated pathogenic bacteria to tobramycin was measured by both a routine broth dilution procedure and the FDA standardized disc technique using a 10-mug disc. Twenty-five isolates each of Escherichia, Enterobacter, Klebsiella, indole-negative Proteus, Pseudomonas, Serratia, and S. aureus, and 16 isolates of group D enterococci were tested. The greatest activity was seen with S. aureus and Pseudomonas species; nearly all isolates of both were inhibited by 0.20 mug or less per ml. Tobramycin was slightly less active against Klebsiella and Enterobacter and moderately active against Escherichia and Proteus, with most isolates of these genera being inhibited by 1.56 mug/ml. Neither Serratia nor the enterococci were particularly susceptible. Correlations between zones of inhibition around the 10-mug disc and minimal inhibitory concentrations were determined, and a zone diameter of 16 mm was recommended as the critical point for prediction of susceptibility.

Pharmacokinetic studies of tobramycin and gentamicin.

Broth dilution susceptibility tests of 100 isolates of Pseudomonas aeruginosa and 101 isolates of Staphylococcus aureus against tobramycin (formerly nebramycin factor 6) and gentamicin showed that tobramycin was more effective against P. aeruginosa and less effective against S. aureus. The minimal inhibitory concentration of tobramycin against the Pseudomonas sp. isolates that required 5 mug of gentamicin per ml for inhibition ranged from 0.63 to 0.31 mug/ml. Peak concentrations in the blood of 10 healthy adults after intramuscular injection of 80 and 40 mg of tobramycin averaged 3.7 +/- 0.62 and 2.4 +/- 0.27 mug/ml, and declined to 0.56 +/- 0.05 and 0.26 +/- 0.02 mug/ml, respectively, after 6 h. The urine recovery averaged 60%. The half-life was 1.6 h. During continuous intravenous infusion of tobramycin and gentamicin (infusion rate 6.6 mg per h), blood levels at steady state were 0.94 +/- 0.10 and 1.04 +/- 0.06 mug/ml, respectively. For both antibiotics, the calculated distribution volume ranged from 15 to 17 liters. The renal clearance to tobramycin averaged 76% and that of gentamicin averaged 85% of the total clearance, indicating that the drugs are primarily eliminated by the kidneys. The present results suggest that tobramycin may be more successful in the treatment of Pseudomonas infections than gentamicin at the same dosage (80 mg intramuscularly three to four times daily).

Tobramycin in bronchial secretions.

The penetration of tobramycin sulfate (nebramycin factor 6) into normal lower respiratory tract secretions was studied in a dog model. After a 1.7 mg/kg intravenous dose of tobramycin, the antibiotic was detected promptly in bronchial secretions, but the peak bronchial secretion concentration was not usually reached until 2 h after injection (mean level 1.15 mug/ml). The maximum tobramycin concentration in bronchial fluids was equal to the mean inhibitory concentrations that have been reported for about 80% of Pseudomonas isolates.

Streptoalloteichus, a new genus of the family Actinoplanaceae.

A new genus Streptoalloteichus is proposed in the family Actinoplanaceae to distinguish species of actinomycetes which form short or long spore-chains on aerial mycelium, bears oval sporangia with motile spores and has a characteristic cell-wall composition of strain C677-91 type. Strain C677-91 (ATCC 31217, FERM-P No. 4070) was named Streptoalloteichus hindustanus gen. nov. and sp. nov. The actinomycete strain C677-91 produces spore-chain clusters and sclerotia in the aerial mycelium which are morphologically similar to those found in some species of Streptomyces. The cultural characteristics of the strain on agar media also resemble those of Streptomyces species and the colonies have no distinct color. Strain C677-91 produces sporangia or sporangia-like vesicles which contain one to several spores in the vegetative mycelium. The sporangiospores possess a single long polar flagellum and are motile. The cell wall of strain C677-91 contains meso-alpha,epsilon-diaminopimelic acid, alanine, glutamic acid, galactose, mannose, rhamnose and glucosamine. Strain C677-91 has several important characteristics in common with Streptomyces tenebrarius including the production of nebramycin factors but the latter strain does not produce sporangia.

BMY-28190, a novel antiviral antibiotic complex.

BMY-28190, an antibiotic complex active against herpes simplex virus type 1 (HSV-1) was produced by the cultured broth of Streptoalloteichus hindustanus sp. nov., a producing strain of tallysomycins A and B. The antibiotic complex was recovered from the broth with Amberlite IRC-50 resin and separated from the coproduced tallysomycins and nebramycins by a series of chromatographies. BMY-28190 exhibited weak inhibitory activity toward Gram-positive and Gram-negative bacteria and strong inhibitory activity toward HSV-1. Structural studies disclosed that BMY-28190 is a novel complex of gamma-poly-D-alpha, gamma-diaminobutyric acids with an average MW of 5,130.

[Quantitative determination of the antibiotic tobramycin using high-performance liquid chromatography].

Separation of the main components of the nebramycin complex of apramycin, kanamycin B and tobramycin was achieved in the form of their 2,4-dinitrophenyl derivatives. A chromatograph SR 8000 of 'Spectra-Physics" and a column 'Sperisorb C6" (4.6 X 250 mm) with the granule size of 5 micrometers were used in the study with the mobile phase of acetone-trisbuffer, pH 7 in a ratio of 65 to 35, the flow rate of 1 ml/min, a temperature of 35 degrees C and detection at lambda 350 nm. Quantitative determination of the tobramycin purity level was performed with an equation developed for the given conditions. The purity levels of the reference tobramycin base and its sulfate were estimated. The results of the estimation corresponded to the data of biological titration.

Interaction of antibiotics with A- and P-site-specific bases in 16S ribosomal RNA.

We have studied the interactions of the antibiotics apramycin, kasugamycin, myomycin, neamine and pactamycin with 16S rRNA by chemical probing of drug-ribosome complexes. Kasugamycin and pactamycin, which are believed to affect translational initiation, protect bases in common with P-site-bound tRNA. While kasugamycin protects A794 and G926, and causes enhanced reactivity of C795, pactamycin protects G693 and C795. All four of these bases were previously shown to be protected by P-site tRNA or by edeine, another P-site inhibitor. Apramycin and neamine, which both induce miscoding and inhibit translocation, protect A1408, G1419 and G1494, as was also found earlier for neomycin, gentamicin, kanamycin and paromomycin. A1408 and G1494 were previously shown to be protected by A-site tRNA. Surprisingly, myomycin fails to give strong protection of any bases in 16S rRNA, in spite of having an apparently identical target site and mode of action to streptomycin, which protects several bases in the 915 region. Instead, myomycin gives only weak protection of A1408. These results suggest that the binding site(s) of streptomycin and myomycin have yet to be identified.

A genetic study of apramycin-resistant mutants of Escherichia coli.

The apramycin-resistant mutants of E. coli were isolated spontaneously. Mutations, aprA and aprB, conferring resistance to apramycin were located at 72 min on the E. coli genetic map where most of ribosomal genes were mapped. One of the mutants, carrying the aprB mutation, has an altered translational fidelity expressed as severe restriction of amber suppressor activity in vivo. The chromosome location and altered translational fidelity indicate that the apramycin-resistant phenotype could be a consequence of an alteration of the ribosomal structure.

Interspecies differences in the pharmacokinetics of kanamycin and apramycin.

Comparative studies on some selected pharmacokinetic parameters for kanamycin in sheep, goats, rabbits, chickens and pigeons, and for apramycin in sheep, rabbits, chickens and pigeons were carried out after intravenous administration of the two drugs at a dose of 10 mg/kg. The results revealed that a two-compartment open model was most suitable for kanamycin, while for apramycin a one-compartment open model was usually optimal. The log distribution rate constant (alpha) of kanamycin was significantly correlated to the log of the body mass (r = 0.919, n = 5, p < 0.05). Interspecies differences in the apparent volume of distribution (Vda) of kanamycin were small. These differences were larger for apramycin, as were the variations in the area under the serum concentration-time curve (AUC) and in the total body clearance (ClB) of both kanamycin and apramycin, both having almost a threefold difference depending on the species but without any correlation to body mass. The values of the log half-life of kanamycin in the mammals in this study and also those from data in the literature revealed a significant correlation with log body mass between animal species according to the equation: t1/2 beta = 38.47W0.21 (r = 0.7648, n = 10, p < 0.05).

Identification and functional analysis of dTDP-glucose-4,6-dehydratase gene and its linked gene cluster in an aminoglycoside antibiotics producer of Streptomyces tenebrarius H6.

Streptomyces tenebrarius H6 produces a variety of aminoglycoside antibiotics, such as apramycin, tobramycin, and kanamycin B. Primers were designed according to the highly conserved sequences of the dTDP-glucose-4,6-dehydratase genes, and a 0.6-kb PCR product was obtained from S. tenebrarius H6 genomic DNA. With the 0.6-kb PCR product as a probe, a BamHI 7.0-kb fragment was isolated. DNA sequence analysis of the 7.0-kb fragment revealed four ORFs and an incomplete ORF. In search of databases, the deduced product of one ORF (orfE) showed 62% identity to the dTDP-glucose-4,6-dehydratase, StrE of S. griseus. Three other ORFs (orfG1, orfG2, and orfGM) showed 55%, 62%, and 42% similarities, respectively, to glycosyltransferase from Clostridium acetobutylicum and mannosyltransferase from Xanthomonas axonopodis pv. citri str. 306 and glycosyltransferase from Pseudomonas putida KT2440. Upstream of the orfE was an incomplete ORF, and the deduced product showed 56% similarity to dTDP-4-dehydrorhamnose, StrL from S. griseus. The function of the orfE gene was studied by targeted gene disruption. The resulting mutant failed to produce tobramycin and kanamycin B, but still produced apramycin, suggesting that the orfE gene and linked gene cluster are essential for the biosynthesis of tobramycin and kanamycin B in S. tenebrarius H6.

Cloning and preliminary characterization of lh3 gene encoding a putative acetyltransferase from a rifamycin SV-producing strain Amycolatopsis mediterranei.

An ORF located immediately downstream of glnR gene was cloned from Amycolatopsis mediterranei U32 and was named lh3. Sequence analysis revealed that lh3 encodes a putative acetyltransferase, which shows high amino acid sequence similarities to the mycothiol synthase (MshD) from other actinomycetes. For functional analysis, mutation in lh3 gene was generated by gene replacement with an apramycin resistance gene through homologous recombination. Compared with the wild type strain, the resulting mutant was more sensitive to H2O2, apramycin and erythromycin by two- to three-fold. These results suggest that the lh3 gene plays an important role in the course of detoxification in A. mediterranei U32.
