Effect of ultrasound stimulation on metabolic modulation of pyruvic acid.

Pyruvic acid lay on the center position of Eremothecium ashbyii metabolic fluxes, during fermentation, a high performance liquid chromatography (HPLC) method is presented for the simultaneous determination of pyruvic acid in the Eremothecium ashbyii cell culture of both with ultrasound stimulation and the control. Dry weight and riboflavin are also mentored. The results of this study show that the ultrasound stimulation can give rise to pyruvic acid change, at the same time; change of pyruvic acid concentration is correlative to that of dry weight of mycelium and content of riboflavin.

Biotechnological production of pyruvic acid.

Pyruvic acid is an important organic acid widely used in the chemical and drug, as well as agrochemical, industries. Compared with the chemical method, biotechnological production of pyruvic acid is an alternative approach because of the low cost. An overview of biotechnological production of pyruvate, including direct fermentative production employing eukaryotic and prokaryotic microorganisms, production by a resting cell method and an enzymatic method as well as the recovery of pyruvate, is discussed. A multi-vitamin auxotrophic yeast strain, Torulopsis glabrata. has been used in the commercial production of pyruvate; emphasis is therefore placed on the mechanism and characteristics of pyruvate production by this strain.

Tests of the ability of two-cell mouse embryos to utilize selected precursors of phosphoenolpyruvic acid and pyruvic acid.

Phosphoenolpyruvic acid and pyruvic acid are among the few compounds that two-cell mouse embryos have been found capable of using as energy sources; most of the compounds in the glycolytic pathway and Krebs cycle that have been tested have been found unusable. Because 3-phosphoglyceric acid is the compound converted most directly to phosphoenolpyruvic acid in glycolysis, one objective of this study was to examine whether it could support development of two-cell embryos as phosphoenolpyruvic acid can. An additional objective was to examine whether alanine or serine, two amino acids that in later stages of development are converted to pyruvic acid for entry into the Krebs cycle, could support development of two-cell embryos as pyruvate does. Two-cell embryos were obtained from the oviducts of mice (C57BL x CBA) and were cultured in medium that contained 3-phosphoglyceric acid, alanine, or serine in lieu of the pyruvate, lactate, and glucose usually contained in the medium. The embryos failed to undergo cleavage but this was not attributable to only a single energy source having been provided; embryos developed in a medium that provided only pyruvate. The results suggest that mouse embryos at the two-cell stage of development are either unable to transport 3-phosphoglyceric acid, alanine, and/or serine or are unable to convert the compounds to chemically similar ones, phosphoenolpyruvic acid or pyruvic acid, which can be metabolized by two-cell mouse embryos.(ABSTRACT TRUNCATED AT 250 WORDS)

Plasma lactic acid and pyruvic acid levels in migraine and tension-type headache.

We examined the lactic and pyruvic acid levels in the plasma of 14 patients with migraine, 17 patients with tension-type headache, and 12 normal controls. The lactic and pyruvic acid levels in the plasma of the migraine patients were significantly higher than those of the normal controls (9.6 +/- 5.0 mg/dL and 0.51 +/- 0.30 mg/dL versus 3.3 +/- 1.9 mg/dL and 0.26 +/- 0.20 mg/dL, respectively). There were no significant differences in the levels between the patients with tension-type headache and normal controls. Our results suggest that migraine patients may have functional abnormalities in mitochondrial energy metabolism.

[Progress in biotechnological production of pyruvic acid].

Pyruvate, an important organic acid, is widely used in the industries of pharmaceuticals, chemicals, agrochemicals, food additives and so on. Compared with the chemical method, biotechnological production of pyruvic acid is an alternative approach because of the low cost and high product quality. In this article, biosynthesis of pyruvate, including direct fermentative production and resting cell method as well as enzymatic method, was discussed. Furthermore, a comparison of these different methods was proposed. Since, a multi-vitamin auxotrophic strain of Torulopsis glabrata is the most competitive strain for industrial production of pyruvate, emphasis was therefore placed on the development of strains screening and fermentation optimization. Finally, some suggestions were put forward to improve the research in this field in the near future.

Can determination of lactic acid and pyruvic acid in cerebrospinal fluid help in diagnosing central nervous system involvement in systemic lupus erythematosus?

The central nervous system (CNS) is frequently affected by systemic lupus erythematosus (SLE). Other causes, especially infection, may also originate CNS disease in SLE patients. Our findings indicate that measurement of lactic acid and pyruvic acid in cerebrospinal fluid (CSF) may be of help in separating inactive CNS-SLE from active CNS-SLE. Also it may be of help in differentiating sterile meningitis caused by SLE from bacterial meningitis. Because of the special risk on tuberculous and fungal infections in SLE patients, also some findings concerning these specific forms of meningitis are given.

Determination of lactic acid and pyruvic acid in serum and cerebrospinal fluid by ion-exclusion chromatography with a bulk acoustic wave detector.

A chromatographic method base don a combination of ion-exclusion chromatography separation and bulk acoustic wave series piezoelectric quartz crystal detector quantification for the determination of pyruvic acid and lactic acid in serum and cerebrospinal fluid (CSF) was developed. The separation was carried out using a Shim-pak SCR-102H ion-exclusion column with phosphoric acid solution as eluent. The method shows an acceptable detection limit and anti-interference ability. Serum and CSF from healthy individuals and patient were analysed successfully.
