[Modelling modification of chemical mutagenesis in human cells. III. Linear index of protection as the standardized criterion of modification].

The effect of substances with radioprotective activity, APAETP 2,3 (aminopropylaminoethylthiophosphoric acid 2,3), APAETP 3,3 and cystaphos, on chromosome aberrations, induced by thioTEPA in the culture of human lymphocytes was investigated. It is shown that the obtained curves "concentration -- effect" for thioTEPA can be described by equations rho = 1 -- e-(KC + alpha)2 and X = E -(KC + alpha)2 --1 for aberrant cells and for chromosome breaks in the presence of the investigated substances. On the basis of comparison of angle coefficients of regression the unificated characteristic of the efficiency of chemical mutagenesis is proposed: the linear protection index (LPI), with generalizes the effect of modificators in chemical mutagenesis.

Radioprotective and toxic effects of a mixture of AET and 5-methoxytryptamine in mice.

The authors studied the radioprotective efficacy and toxicity of an AET + 5-methoxytryptamine mixture and its effect on the course of respiratory exchange, using different weight ratios of the two components. The optomum was found to be 150 mg AET/kg b.w + 25 mg 5-MOT/kg b.w. This mixture gave very good protection in the supralethal exposure range and markedly prolonged its duration. The high toxicity of this mixture was abolished by the pre-administration of glutathione. Raised and longer protection was also manifested in the course of respiratory exchange, showing that an association exists between changes in energy metabolism and raised radioresistance of the organism.

[Radioprotective action of N-acetyl p. aminophenol in the mouse].

N-acetyl p. amino phenol given intraperitonealy to mice (20 or 40 mg kilo) before irradiation by 1000 or 500 rad has a radioprotective effect with a reduction in death rate and, after an initial fall, a quicker increase of the number of white blood cells in the treated group.

[Modifications by BCG of the mortality of the irradiated mouse].

Freeze-dried Bacillus Calmette Guerin (B.C.G.) of Institut Pasteur was given by intravenous route to mice at 1,2 and 4 mg/kg before and after gamma irradiation of animals by 1 000 rad. B.C.G. 1 mg/kg injected the day or the day after irradiation has a protective effect (mortality reduced from 77% for controls to 58% and 50% for treated mice). B.C.G. given before irradiation in single or double doses increased mortality.

[Medical aspects of superoxide dismutases].

The role of superoxide radicals and of superoxide dismutases in inflammation is described. Encapsulation of the enzyme in liposomes leads to an increased organ specificity on injection into animals. Preliminary results of medical application are presented.

[Pharamagnetic centers developing during irradiation of gamma-phos].

The ESR spectra's of X-irradiated radioprotector gamma-phos have been studied. Using the analysis of relaxation effects in this sample irradiated at room temperature three paramagnetic centers have been discovered. One of these centers is classed with the free radical approximately CH2--CH--CH2 approximately (R1)-G(R1) value decreases with a decrease of irradiation temperature. Free radicals R1 are not oxidized in the air. Two other of paramagnetic centers have not been identified.

[General characteristics and comparative evaluation of the radioprotective properties of aryl alkyl amine adrenomimetics in experiments on mice].

The radioprotective effect (RPE) of some arylalkylamines (AAAs) was studied in experiments on mice. Mesaton and its close analogues were injected subcutaneously 15 minutes prior to irradiation at a dose of 800 rad. The protective effect is exerted by AAAs in low doses (25--50 mumole/kg), the compounds show stable and high RPE (80--80% survival, dose reduction factor being 1.3--1.4) and low toxicity (LD50 = 4--8 mumole/kg). AAAs studied are not less effective than aminothiols. Their pharmacological spectrum--K = LD50/ED50 (200--500) is superior to that of known aminothiols and indolylalkylamines.

Studies on the effect of Fenchlorphos on postirradiation changes of serum proteins and nucleic acids content in rat liver.

The effect of Fenchlorphos on postirradiation changes of serum protein levels and nucleic acid content in rat liver were investigated. It was found that this compound significantly decreases postirradiation changes in serum proteins, properdin level and nucleic acid content in rat liver. An important role of these phenomena in radioprotective action of Fenchlorphos is suggested.

Cytogenetics analysis of meiotic chromosomes of irradiated mice and their progeny after treatment with streptomycin and dihydrodeoxystreptomycin.

The purpose of this investigation is to find out whether streptomycin and the related compound dihydrodeoxystreptomycin have any mutagenic effect and whether they both are capable of recovering X-ray induced chromosomal translocations in mouse spermatogonia of directly treated animals and their progeny of the first generation. The cytological findings show the absence of any mutagenic effect in animals nonirradiated and treated with streptomycin and dihydrodeoxystreptomycin. The frequency of chromosomal translocation after total irradiation was 9,07%; in animals treated with streptomycin following irradiation 5.13%, and in those irradiated and treated with dihyrodeoxystreptomycin, 3.70%. Male offsprings, originated from parents treated only with antibiotics show no chromosomal translocations. However, offsprings originated from irradiated and treated parents gave birth to the male offspring with chromosomal translocations.
