Malignant lymphomas of follicular center cell origin in man. I. Immunologic studies.

Lymphomas with histologic features indicating a follicular center cell (FCC) origin were analyzed from 26 patients of a group of 45 consecutive non-Hodgkin's lymphoma patinets whose tumors were studied for B- and T-cell characteristics. They were compared with benign, reactive lymphoid tissue from 14 patients. Cell suspensions from biopsy material, blood, or bone marrow were examined for surface Ig and for rosette formation with sheep erythrocytes (E rosettes). Of the 26 patients with FCC lymphomas, 22 had 40% or more Ig-bearing cells; all patients with FCC lymphoma tissues had 25% or less E rosette-forming cells. Cells from most FCC lymphomas of the cleaved type had surfac IgM; those from several FCC lymphomas had both IgM and IgD. Cells from lymphomas of noncleaved cell type had surface IgG or IgA. Light-chain analysis showed that cells from FCC lymphomas bore a predominant light-chain type, which indicated their monoclonal nature. Neoplastic cells from several FCC lymphomas synthesized the surface Ig which they bore. Reactive tissues usually contained fewer Ig-bearing and more E rosette-forming cells than FCC lymphomas; the Ig-bearing cells, with one exception, had a polyclonal distribution. Correlation of histologic and immunologic observations indicates that most lymphomas identified as FCC in origin by light micorscopic criteria mark as B cells with the use of immunologic techniques and that FCC lymphomas are the most common type of non-Hodgkin's lymphoma.

B-Lymphocytes

Among naive precursor cell subpopulations only progenitors of memory B cells originate germinal centers.

Immunization leads to the generation of both antibody-forming cells (AFC) and memory B cells which are thought to arise in germinal centers within lymphoid follicles. The findings that the precursors to memory B cells reside in the J11Dlo subpopulation of the spleens in non-immune mice and that this subpopulation is distinct from conventional AFC precursors, including CD5+ B cells, suggest that the precursors of germinal centers might also reside in the J11Dlo subpopulation. To test this hypothesis, SCID mice were repopulated with CD4+ carrier-primed T cells and T-depleted J11Dlo, J11Dhi or CD5+ B cells and immunized with a hapten-carrier conjugate. Only the J11Dlo population was enriched for cells that produced germinal centers. Thus, the subpopulation of precursors that generates memory B cells also originates germinal centers.

Malignant lymphomas of follicular center cell origin in man. II. Ultrastructural and cytochemical studies.

Tissues from malignant lymphomas with both nodular and diffuse growth patterns, thought by light microscopy to be composed of cells of follicular center cell (FCC) origin, Were examined by electron microscopy; the tumor cells were similar to lymphoid cells found in reactive follicular centers. Tumor cells from neoplasms thought to be composed of cleaved FCC often had more pronounced nuclear folding than did cleaved FCC of reactive follicles, whereas cells in tumors of noncleaved FCC type were indistinguishable from their presumed counterparts in reactive follicles. Large cell noeplasms, previously classified as "histiocytic" lymphomas were composed of cells with ultrastructural characteristics of transformed lymphocytes; they showed neither ultrastructural nor cytochemical features of mononuclear phagocytes. These findings support the concept that a major group of lymphomas arises from lymphocytes of follicular centers.

B-Lymphocytes

Measles virus from a long-term persistently infected human T lymphoblastoid cell line, in contrast to the cytocidal parental virus, establishes an immediate persistence in the original cell line.

To investigate the mechanisms of measles virus (MV) establishment and maintenance of persistence in lymphoid cells, we have established a long-term persistent infection with MV, Edmonston strain, in the human T lymphoblastoid cell line MOLT4, which has been in continuous culture for over 8 years. In this culture, designated MOMP1, more than 98% of cells display viral antigens. The MOMP1 culture is immune to superinfection with MV and is not cured by anti-MV antibodies. No evidence of defective interfering particles was obtained. The persistently infected culture releases an infectious virus showing a miniplaque and thermoresistant modified phenotype that, unlike the parental virus Edmonston strain which produces a lytic infection with extensive cell fusion, establishes an immediate persistence in MOLT4 cells with neither significant loss of cell viability nor cell fusion. This suggests that the modification in the virus suffices to maintain the state of persistence without requiring a coevolution of the host cell during the infection, as has been reported in other persistent virus infections.

Cerebrospinal fluid cytomorphology identification of benign cells originating in the central nervous system.

The chief cellular consituents of CSF are the "free cells" of the pia-arachnoid, ependyma, and choroid. These cells are of diverse types, but have their mesenchymal origin in common. Usually they offer no difficulty in identification since they are predominantly histiocytic. However, these "free cells" may be morphologically changed in reaction to an appropriate stimulus, and may therefore be a source of error in diagnostic material. Cells having mesothelial characteristics are not found in the CSF compartment and therefore the term pia-arachnoid mesothelial cells appears inappropriate. The only neuroectodermal derivative that was easily identifiable was the choroidal cell. It is distinctive and unequivocal. We were unable to recognize ependymal cells in our present study. Glial cells (protoplasmic and fibrous astrocytes) and neurons can be recognized when neural tissue is penetrated in the process of collecting CSF.

Histiocytic lymphoma of B cell origin.

A histiocytic lymphoma with a terminal leukemic phase occurred in a 50-year-old man. Immunologic studies indicated that neoplastic cells from the peripheral blood sample, bone marrow aspirate, and lymph node carried surface immunoglobulin. Cytochemical and immunohistochemical analysis of this histiocytic lymphoma and its subsequent leukemic phase displayed a profile consistent with a lymphocytic rather than a histiocytic origin of the neoplastic cell.

The maturation of reticulocytes. II. Life-span of red cells originating from stress reticulocytes.

Life-spans of macroreticuloytes and macrocytic red cells were studied. Rabbits were made anemic by injecting phenylhydrazine. Peripheral blood rich in reticulocytes was drawn, labeled with 3H-amino acids, and injected back into the anemic animal. Autoradiographic observation on circulating red cells revealed that macroreticulocytes matured at nearly the same time as normal-sized reticulocytes but that the macrocytic red cells had a short life-span compared to normal-sized red cells.

Sertoli cell origin of testicular androgen-binding protein (ABP).

In this report it is suggested that the specific adrogen-binding protein (ABP), previously shown to originate in the testis of rat and other species, is produced by the Sertoli cells. This suggestion is based upon the following experimental findings: 1) ABP was found in high concentrations in testicular efferent duct fluid but only in trace amounts in inter-tubular lymph. i) ABP could be recovered from crude preparations of testis tubules, but not from Leydig cells from the same testes. 3) Testes whose germinal epithelium had been severly damaged by gamma irradiation showed no decrease in ABP content. The transport of ABP to epididymis was also preserved as judged from the levels of ABP in caput epididymis. 4) Testes that were completely devoid of germ cells following prenatal gamma irradiation showed high levels of ABP, These high levels approached zero following hypophysectomy, but could be restored by FSH administration to the hypophysectomized animals. ABP has been well characterized and now provides a valuable experimental tool as an indicator of Sertoli cell function.

Lymphoma cutis of apparent B cell origin.

A nodular, poorly differentiated lymphocytic lymphoma involving lymph nodes, spleen, and skin is reported in a 60-year-old man. The clinical course of this disorder strongly implied cutaneous involvement as the initial site of disease. Immunologic studies indicated that the neoplastic cells carried surface immunoglobulin (IgMgamma), thus suggesting a B lymphocyte origin for the lymphatic neoplasia.

Lack of evidence for glial cells originating from the external granular layer in mouse cerebellum.

In 25 day mice cerebella, a quantitative electron microscopic analysis showed that glial cells were not seen among 749 cells counted in the molecular layer. Likewise, a light microscopic autoradiographic study showed that labelled oligodendroglia and/or astroglia in the cerebellum were not derived from the external granular layer (EGL). Previous claims, that these cells derived from the EGL, may have arisen because other cell types, i.e., endothelial cells, pericytes, microglia, and other ectopic granule cells may have been misidentified as oligodendroglia and/or astroglia. It seems likely that the EGL is a unique germinal cell layer in the mammalian nervous system because it gives rise only to neurons, whereas cerebellar astrocytes and oligodendrocytes are derived from the subventricular layer of the fourth ventricle, as first suggested by Cajal in 1911.

Coculture of in vitro fertilized bovine embryos with oviductal epithelial cells originating from different stages of the estrous cycle.

Bovine embryos derived from in vitro fertilization procedures were cocultured in vitro with oviductal cells obtained from heifers between d 4 and 6 or d 14 and 16 of the estrous cycle. In addition, proteins secreted by oviductal cells isolated between d 4 and 6 or d 14 and 16 of the cycle were monitored. Embryos (2- to 4-cell) were incubated in Tissue Culture Medium-199 with 10% fetal bovine serum with or without oviductal cells at 39 degrees C for 10 d following in vitro insemination. There were more morulae, blastocysts, and hatched blastocysts following coculture with oviductal cells than with culture in medium alone. However, no differences were noted in embryo development following coculture with oviductal cells obtained between d 4 and 6 or d 14 and 16 of the estrous cycle. Also, no differences were detected in the amount of [35S]methionine-labeled proteins secreted by oviductal cells isolated from different days of the estrous cycle. These results indicate that oviductal epithelial cells isolated from early and late luteal phases of the estrous cycle will effectively support early embryonic development following prolonged in vitro culture.

Melanin containing neurofibroma: case report with evidence of Schwann cell origin of melanin.

This case report describes a melanin-containing neurofibroma involving a spinal nerve root. Electron microscopy of the tumor shows that neoplastic Schwann cells are capable of melanogenesis. Although this capability is suggested in the literature, few reports provide ultrastructural confirmation. The likely identity between reported "cellular blue nevi of spinal nerve roots" and nerve sheath tumors is discussed. It is possible that pigmented nerve sheath tumors behave more aggressively than nonpigmented ones, although it is debatable.
