Identification, characterization, and spatial localization of two flagellin species in Helicobacter pylori flagella.

Flagellar filaments were isolated from Helicobacter pylori by shearing, and flagellar proteins were further purified by a variety of techniques, including CsCl density gradient ultracentrifugation, pH 2.0 acid disassociation-neutral pH reassociation, and differential ultracentrifugation followed by molecular sieving with a Sephacryl S-500 column or Mono Q anion-exchange column, and purified to homogeneity by preparative sodium dodecyl sulfate-polyacrylamide gel electrophoresis and transfer to an Immobilon membrane. Two flagellin species of pI 5.2 and with apparent subunit molecular weights (Mrs) of 57,000 and 56,000 were obtained. N-terminal amino acid analysis showed that the two H. pylori flagellin species were related to each other and shared sequence similarity with the N-terminal amino acid sequence of Campylobacter coli, Bacillus, Salmonella, and Caulobacter flagellins. Analysis of the amino acid composition of the predominant 56,000-Mr flagellin species isolated from two strains showed that it was comparable to the flagellins of other species. The minor 57,000-Mr flagellin species contained a higher content of proline. Immunoelectron microscopic studies with polyclonal monospecific H. pylori antiflagellin antiserum and monoclonal antibody (MAb) 72c showed that the two different-Mr flagellin species were located in different regions of the assembled flagellar filament. The minor 57,000-Mr species was located proximal to the hook, and the major 56,000-Mr flagellin composed the remainder of the filament. Western immunoblot analysis with polyclonal rabbit antisera raised against H. pylori or Campylobacter jejuni flagellins and MAb 72c showed that the 56,000-Mr flagellin carried sequences antigenetically cross-reactive with the 57,000-Mr H. pylori flagellin and the flagellins of Campylobacter species. This antigenic cross-reactivity did not extend to the flagellins of other gram-negative bacteria. The 56,000-Mr flagellin also carried H. pylori-specific sequences recognized by two additional MAbs. The epitopes for these MAbs were not surface exposed on the assembled inner flagellar filament of H. pylori but were readily detected by immunodot blot assay of sodium dodecyl sulfate-lysed cells of H. pylori, suggesting that this serological test could be a useful addition to those currently employed in the rapid identification of this important pathogen.

Separation discrimination with embedded targets.

Previous research has shown that separation discrimination thresholds are independent of the internal spatial scale (local spatial frequency) of the targets whose separation is being judged. The experiments reported here tested the generality of this conclusion for separation discrimination of targets that were embedded in an array of identical objects, where crowding could enhance the importance of the scale at which the individual target locations are encoded. No effect of the local spatial scale of the targets was found under these conditions.

[Localization disorders in squint amblyopia: horizontal line bisection and relative vertical localization].

We used two different procedures for the evaluation of spatial localization errors in strabismic and anisometropic amblyopes, strabismic alternators and normal control subjects: partitioning of horizontal lines and vertical alignment. We found defective spatial localization in the amblyopic eyes of squinters with both procedures. However, in the horizontal partitioning experiment, the control subjects also showed asymmetries between the two hemifields. Thus, there is no good baseline under these conditions for the results of the amblyopic eyes of squinters. Our results show that vertical alignment is the more appropriate procedure to quantify defective spatial localization in strabismic amblyopes.

Beyond benchmarking: an expert-guided consensus approach to spatially aware clustering.

Spatial omics technologies have revolutionized the study of tissue architecture and cellular heterogeneity by integrating molecular profiles with spatial localization. In spatially resolved transcriptomics, delineating higher-order anatomical structures is critical for understanding how cellular organization affects function. However, the reliability of current benchmarks of spatially aware clustering (SAC) methods is undermined by their narrow focus on Visium and brain tissue datasets and the incorrect interpretation of manual annotation as ground truth. Here we present SACCELERATOR, a community-driven, extensible framework that standardizes data formatting, method integration and metric evaluation, enabling rapid inclusion of new methods and datasets. Our analysis revealed substantial limitations in the generalizability and reproducibility of SAC methods and shows that anatomical labels commonly used as ground truths are often biased, error prone and unsuitable for benchmarking. Rather than ranking methods, we propose a consensus-guided workflow where descriptive spatial metrics highlight high-entropy regions of method disagreement, enabling targeted feedback for tissue experts. Applied to brain and cancer datasets, this approach uncovered biologically meaningful patterns overlooked by individual SAC methods and manual annotations, highlighting the need for iterative, expert-in-the-loop evaluation.

A study on the differences in recovery effects of different types of nutritional supplements on competitive performance of esports athletes under mental fatigue.

BACKGROUND: To compare the effects of different nutritional supplements on the recovery of core competitive performance abilities in esports athletes following mental fatigue and to observe changes in the autonomic nervous system during recovery after nutritional supplementation by monitoring heart rate variability (HRV). METHODS: A randomized crossover within-subject controlled experimental design was adopted, including nutritional supplement type (caffeine, nitrate, Ginkgo biloba extract, catechins, placebo)&#x2009;&#xd7;&#x2009;mental fatigue state (initial, fatigued, post-supplementation). Twenty high-level first-person shooter (FPS) esports athletes were recruited. Mental fatigue was induced using a Stroop task. After ingesting the different supplements and resting for 60&#x2009;minutes, the participants completed assessments of shooting accuracy, shooting stability, spatial localization, and multitasking ability using the KovaaK's simulation trainer. HRV indices were also recorded to evaluate changes in autonomic regulation. RESULTS: For shooting accuracy, compared with the placebo condition, all four supplements significantly improved shooting accuracy scores following mental fatigue (all p&#x2009;<&#x2009;0.05); however, no significant differences were observed among the effects of the different supplements. For shooting stability, caffeine, nitrate, and catechins produced significant recovery effects on shooting stability (all p&#x2009; <&#x2009;0.05); however, no significant differences were observed among the effects of these three supplements. For spatial localization and multitasking ability, the improvements in these two abilities in the post-supplementation state may have resulted from natural recovery, and none of the four nutritional supplements demonstrated a significant recovery effect. The HRV results showed that indices including RMSSD and SDNN changed under some supplement conditions. CONCLUSIONS: Mental fatigue significantly reduced the competitive performance of esports athletes. Four types of nutritional supplements all promoted the recovery of shooting accuracy, while caffeine, nitrate, and catechins promoted the recovery of shooting stability. However, no additional recovery advantages of the nutritional supplements over placebo were identified for spatial localization or multitasking ability. Changes in HRV may reflect changes in autonomic regulation during recovery, but further research is still warranted.

Chromatic adaptation to natural and incandescent illuminants.

A color CRT image display system was used to present adapting backgrounds that were spatially and temporally varied. Three observers adjusted the chromaticity of test stimuli to produce an achromatic appearance under a variety of adapting conditions. The achromatic-appearing chromaticities were used as measures of the observers' states of chromatic adaptation. The spatial configuration of the adapting background was varied to measure the spatial extent of the mechanisms responsible for chromatic adaptation. The temporal configuration of the adapting background was varied to measure the time-course of these mechanisms. The results show that chromatic adaptation is spatially localized with a time-course on the order of 10 sec. Since the mechanisms were shown to be spatially localized, the observed temporal integration across eye movements is required to allow these mechanisms to adjust to the spatially integrated scene chromaticity.

The chemical landscape of plant surface metabolites: Acylsugars as models of ecological function and structural diversity.

Plants produce a multifunctional assortment of specialized metabolites that play important roles in defense, environmental adaptation, and ecological interactions. Among these compounds, acylsugars, nonvolatile metabolites produced primarily in glandular trichomes of Solanaceae species, have emerged as informative model systems for understanding plant surface chemistry. Differences in acyl chain length, branching pattern, saturation, and attachment position generate extensive chemical diversity that influences herbivore deterrence, pathogen resistance, and the physicochemical properties of leaf surfaces. Recent advances in analytical chemistry, particularly liquid chromatography-ion mobility-tandem mass spectrometry (LC-IM-MS/MS), have greatly improved the ability to separate structurally related acylsugar isomers and characterize metabolite complexity at high resolution. When integrated with genomics, transcriptomics, and emerging spatial metabolomics approaches, these analytical tools provide new insights into acylsugar biosynthesis, pathway regulation, evolutionary diversification, and ecological function across plant species. This review positions acylsugars, particularly those of Solanum species, as model systems for understanding how structural diversity, spatial localization, and specialized metabolism shape ecological and physiological function at plant surfaces. We examine acylsugar structural diversity, biosynthetic pathways, ecological and physiological functions, and interactions with environmental and atmospheric processes. Major challenges, including extensive isomeric complexity, incomplete pathway characterization, and difficulties linking chemical structure to biological function, are discussed alongside emerging opportunities in integrative omics, crop improvement, sustainable pest management, and environmental monitoring. Overall, acylsugars provide a powerful model for linking molecular structure, spatial localization, and ecological function, offering broader insight into how specialized metabolism shapes plant adaptation, defense, and environmental interactions.

Noradrenaline depletion blocks behavioral sparing and alters cortical morphogenesis after neonatal frontal cortex damage in rats.

The possibility that cortical noradrenaline (NA) is necessary for sparing of function that occurs after neonatal frontal cortex damage was examined. Spatial localization by rats with frontal cortex damage on postnatal day 7 (P7) was better than that by rats with similar damage sustained as adults. The sparing was abolished in rats depleted of cortical NA by means of neonatal 6-hydroxydopamine (6HDA) administration. The blockade of sparing in the P7 frontal operates was associated with a smaller brain, thinner cortex, and reduced cortical dendritic branching relative to saline-treated P7 frontal operates. NA depletion alone in unoperated rats did not affect spatial learning but did reduce brain size and dendritic branching. Rats with frontal lesions on P4 did not show sparing of spatial localization, and 6HDA administration had no additional behavioral effect. Overall, these data are consistent with the notion that NA has some general function in maintaining some forms of plasticity in posterior cortex.

[Effect of various light sources on visual functions in the age aspect. I. Conscious reaction time and the manual spatial visual localization].

The authors present the results of investigations performed in 28 right-handed, normally seeing persons divided into 2 groups: 15 of them were in productive age and 13 in older age. The authors compared the dynamics of the behaviour of the visual functions in dependence on the age of the subjects and the stay in the glow and sodium light. No adverse influence of the sodium light on the hand localization could be detected, instead it seems that it negatively influenced the selective reaction in older productive persons in the sense of an increased number of faults.

Spatial frequency and light-spread descriptions of visual acuity and hyperacuity.

Resolution (visual acuity) and differential spatial localization (hyperacuity) targets were selected to allow rigorous psychophysical measurements as well as ready expression of both their spatial frequency spectrum and their retinal image light distribution. Thresholds were about 1 arc min for acuity and 4-6 arc sec for hyperacuity. As is consistent with the reciprocal relationship between the space and spatial frequency domains, the small locally restricted spatial differences between just distinguishable patterns are represented in the frequency domain by equally small differences, which are distributed over the entire spatial frequency spectrum. While they occur in many test situations, phase variations of spatial frequency components are not necessary for achieving optimum acuity and hyperacuity.

[Errors of monocular localization in strabismic amblyopia. Two-dimensional distortion].

Spatial distortions, i.e. spatial localization errors, and precision of localization were measured monocularly in the central visual field of 7 normal observers, 17 strabismic amblyopes and 2 anisometropic amblyopes. The task of the subjects was to construct circles of 2 degrees, 4 degrees and 6 degrees radius around a fixation point, using the dominant and the amblyopic eye in turn. Normal observers set distances on the vertical meridian smaller than distances on the horizontal meridian. Anisometropic amblyopes showed localization errors and variances similar to those of normal observers. The amblyopic eyes of strabismic observers with a large angle strabismus and deep amblyopia showed significant, individually different localization errors correlating with the depth of amblyopia. Strabismics with microstrabismus exhibited parallels between monocular localization and dichoptic retinal correspondence.

Spatial Transcriptomics Identifies Characteristic Immunological Niches in Atopic Dermatitis.

BACKGROUND: Atopic dermatitis (AD) is primarily driven by a Type 2 immune response, with T helper (TH2) cells producing IL-4 and IL-13, thereby promoting inflammation, itch, and a compromised skin barrier. Yet, the spatial organization of pathogenic immune cells and their interactions with stromal and epithelial compartments in human AD skin remain incompletely understood. METHODS: We performed 10&#xd7; Genomics Visium spatial transcriptomics on FFPE skin biopsies from patients with AD (n&#x2009;=&#x2009;6), psoriasis (n&#x2009;=&#x2009;2), and healthy controls (n&#x2009;=&#x2009;5). Data were integrated with AD single-cell RNA sequencing (scRNA-seq) datasets and complemented by imaging mass cytometry (IMC) and multiplex immunofluorescence (IF) to validate the spatial localization of immune cells. Cell-cell communication analysis revealed putative signaling interactions within immune niches. RESULTS: Spatial clustering resolved tissue compartments and demonstrated transcriptional dysregulation in keratinocytes in AD and psoriasis. AD lesions showed a conserved spatial organization of immune aggregates within the superficial dermis. Integration of scRNA-seq signatures revealed spatially organized co-localization of T cells and mature migratory dendritic cells (mmDCs). We developed a ring-based neighborhood analysis to characterize the cellular organization of the immune-stromal niches, revealing T cell-enriched regions surrounded by inflammatory fibroblasts and activated keratinocytes. Intercellular communication analysis further identified putative signaling within mmDC-T cell niches that may promote pathogenic T cell recruitment and activation. Application of tertiary lymphoid structure (TLS) signatures indicated the presence of TLS-like regions. IMC and IF validated the close spatial proximity between activated TH2 cells and mmDCs. CONCLUSION: AD lesions contain spatially organized TLS-like immune niches at the dermal-epidermal junction, characterized by the close association of T cells and mmDCs and coordinated interactions with surrounding stromal and epithelial compartments. These mmDC-T cell niches may represent potential targets for future therapeutic strategies aimed at disrupting persistent local inflammatory pathways and improving long-term disease control.

Auditory localization under conditions of unilateral fitting of different hearing aid systems.

The spatial localization function of hearing-impaired listeners, usually fitted bilaterally with BTE, ITE or ITC devices, was tested under conditions of unilateral fitting of each of their own hearing aids, and unilateral fitting of stock versions of each of the other types. For the BTE wearers average localization accuracy and individual variability were not greatly changed when wearing only their left ear device, compared with bilateral aided performance. In ITE wearers, unilateral fitting in either ear led to somewhat poorer performance than bilateral. In the ITC wearers unilateral fitting produced inconsistent outcomes. Both BTE and ITE wearers fared poorly when fitted unilaterally with stock forms of devices 'foreign' to them, whereas ITC wearers did not show such a contrasting outcome. A group of non-impaired listeners showed severe disruption of localization under unilateral BTE and ITC hearing aid conditions, and to a lesser extent with ITEs. Results for the hearing-impaired listeners are interpreted in terms of adaptation to different usage patterns, with BTE wearers suggested as having adapted to their own systems unilaterally as well as bilaterally.

Localized in vivo high-resolution NMR imaging using gradient subencoding technique.

A new spatial localization technique for in vivo high-resolution imaging is presented here. In contrast to other localization techniques that use a series of rf pulses to define a volume of interest, only one rf pulse is utilized in the proposed method for selection of a region to be imaged. Instead of rf pulses for region selection, subencoding gradient pulses are used for the localization together with a convolution process on each phase-encoding gradient by a set of additional gradients (e.g., y direction). Then the 2-D localization is completed by restricting the bandwidth in the readout direction (e.g., x direction). The latter is simply achieved by using a low-pass filter in the receiver system. By applying this technique on a human body, localized in vivo high-resolution images are obtained for the knee with much improved resolution. 100 x 100 microns in-plane (x,y plane) resolution images obtained from the human knee demonstrate that localized in vivo high-resolution imaging for both human and animals is possible with an in-plane resolution of below 100 microns.
