In vivo selective monitoring of basal levels of cerebral dopamine using voltammetry with Nafion modified (NA-CRO) carbon fibre micro-electrodes.

The electrochemical technique of differential pulse voltammetry (DPV) with micro-biosensors has been used for a number of years to monitor in vivo and in situ changes in the extracellular concentration of cerebral ascorbic acid, as well as that of the metabolites of dopamine (DA) and serotonin (5-HT). We have recently prepared a carbon fibre micro-electrode (mCFE) which specifically pretreated and coated with Nafion (a negatively charged polymer which repels acids such as 3,4-dihydroxyphenylacetic acid (DOPAC)) allows the direct selective detection of the oxidation of DA and 5-HT in nanomolar concentration in vitro and that of extracellular basal levels of cerebral 5-HT in vivo (peak B at +240 mV). We describe here a modified version of this micro-biosensor now called NA-CRO mCFE as its active tip (30 microns in diameter) is coated with a 50/50 (v:v) mixture of Nafion and dibenzo-18-crown-6 (Aldrich). In vitro this newly reported electrode shows insensitivity to acids (e.g., DOPAC) up to 100 microns and sensitivity to 0.5-1 nM DA. In vivo, in the striatum of anaesthetised rats, a basal oxidation peak at +80 mV (peak A, on average 0.6 nA in height), which corresponds to the oxidation potential of DA in vitro, is consistently detectable with the NA-CRO mCFE (corresponding to an estimated concentration of 1.5 nM). Experiments performed in vivo in anaesthetised rats implanted in the striatum with uncoated (normal) mCFE to measure extracellular DOPAC or with NA-CRO mCFE have been performed in order to analyse the chemical nature of peak A in vivo. It is concluded that the addition of the crown-ether compound to the Nafion coat improves the sensitivity of the micro-biosensor for DA in vitro and allows the detection of its basal extracellular levels in vivo.

Microporous small diameter PVDF-TrFE vascular grafts fabricated by a spray phase inversion technique.

Microporous prostheses of 1.5 mm internal diameter were fabricated with a polyvinylidene fluoride-trifluoroethylene (PVDF-TrFE)n co-polymer by the spray phase inversion technique. Some of the grafts were made piezoelectric by poling under a high electrical field. Overall, 24 poled grafts (P) and 24 unpoled grafts (UP) (15-22 mm in length) were implanted in the infrarenal aorta of 48 adult rats. Patency rates in P were 100% (8/8) at 2 days, 100% (8/8) at 2 weeks, 75% (6/8) at 6 months, and 92% total (22 of 24). Patency rates in UP were 100% (8/8) at 2 days, 63% (5/8) at 2 weeks, 100% (8/8) at 6 months, and 88% total (21 of 24). Thus there was no significant difference in patency between the two types of grafts. Both showed similar macroscopic and microscopic findings. At 2 days, fibrin deposition was somewhat heavier on the poled grafts, but no difference in surface platelet deposition could be detected. Endothelialization was observed from both anastomoses at 2 weeks and was almost complete at 6 months. The excellent biocompatibility of PVDF-TrFE and the microporous structure of the grafts were probably the dominant factors in success with these grafts. Although piezoelectric activity in excised cleaned poled prostheses remained significantly higher than that in the control UP, the charges developed may have been too small to exert a biologic effect, either because of insufficient dipole orientation or inadequate mechanical deformation.

Galactosylated poly(vinylidene difluoride) hollow fiber bioreactor for hepatocyte culture.

To overcome the limitations of long-term expression of highly differentiated hepatocyte functions, we have developed a novel bioreactor in which hepatocytes are seeded in a ligand-immobilized hollow fiber cartridge. Galactosylated Pluronic polymer is immobilized on poly(vinylidene difluoride) (PVDF) hollow fiber surface through an adsorption scheme yielding a substrate with hepatocyte-specific ligand and a hydrophilic surface layer, which can resist nonspecific protein adsorption and facilitate cell binding to the galactose ligand. Interestingly, the galactosylated PVDF hollow fiber shows enhanced serum albumin diffusion across the membrane. Freshly isolated rat hepatocytes were seeded and cultured in the extralumenal space of the hollow fiber cartridge for 18 days in a continuously circulated system. Albumin secretion function of the seeded hepatocytes was monitored by analyzing circulating medium by enzyme-linked immunosorbent assay. Urea synthesis and P-450 function (7-ethoxycoumarin dealkylase activity) were measured periodically by doping the circulating medium with NH4Cl and 7-ethoxycoumarin, respectively. Hepatocytes cultured on galactosylated PVDF hollow fibers maintained better albumin secretion and P-450 functions than on unmodified and serum-coated PVDF hollow fibers when cultured in serum-containing medium. Morphological examination by scanning electron microscopy showed that hepatocytes cultured on galactosylated PVDF hollow fibers developed significant aggregation, in contrast to those cultured on unmodified PVDF fibers or on serum-coated PVDF fibers. Transmission electron microscopy images revealed that tight junctions and canaliculus-like structures formed in these aggregates. These results suggest the potential application of this galactosylated PVDF hollow fiber cartridge for the design of a bioartificial liver assist device.

Formation of acute pulmonary toxicants following thermal degradation of perfluorinated polymers: evidence for a critical atmospheric reaction.

In some small-scale tests of combustion-product toxicity, perfluorinated polymers (FP) have been shown to be much more toxic than other common materials. These studies were conducted to determine the conditions under which highly toxic decomposition products of FP are formed. A modification of the National Bureau of Standards exposure system (Levin et al. National Bureau of Standards 1982, NBS IR 82-2532) was used, in which materials could be heated either in a cup furnace or above a small methane flame. At 700 degrees C, the approximate lethal concentrations (ALCs) of the products formed from polytetrafluoroethylene (pTFE) and from hexafluoropropylene/tetrafluoroethylene copolymer (FEP) in the CH4 flame (applied for 1 min) are approximately 850 times higher than the ALCs of the products formed in the cup furnace. Analytically, the major products formed initially from pTFE at 700 degrees C under either condition (flame or cup furnace) are similar but they disappear rapidly in the presence of continuous heat. When the cup furnace is removed 1 min after pTFE is added (a procedure temporally similar to the use of the flame) the toxicity of the products is again low. However, when heat from either the cup furnace or from a small secondary flame is applied continuously (for up to 30 min) to the initial products formed from pTFE in the flame, high toxicity is observed. These observations are supported by pathological evaluation of the respiratory tracts of exposed rats. Thus, the inhalation toxicity of FP thermal decomposition products is related to a heat- and time-dependent atmospheric reaction. Such mechanisms should be considered in estimating the fire hazard of these materials in use.

Photocatalytic reactivities of Nafion-coated TiO2 for the degradation of charged organic compounds under UV or visible light.

Nafion (perfluorinated polymer with sulfonate groups)-coated TiO2 particles (Nf/TiO2) were prepared and their reactivities for the photocatalytic degradation (PCD) of charged organic substrates were investigated. The presence of Nafion adlayers drastically changed the positive TiO2 surface charge to a negative one over the entire pH range and significantly influenced the PCD kinetics and mechanisms. The UV-induced PCD of tetramethylammonium (TMA; cationic substrate) was greatly enhanced in the presence of Nafion adlayers on TiO2 because the ion-exchange sites within the Nafion can hold cationic substrates. On the other hand, despite the unfavorable electrostatic interaction between the Nf/TiO2 and anionic substrates, the PCD of dichloroacetate (DCA) and acid orange 7 (AO7) with Nf/TiO2 was not significantly inhibited. The visible-light-sensitized degradation of dyes was enhanced with Nf/TiO2 not only for cationic dyes (methylene blue (MB) and rhodamine B (RhB)) whose uptake on Nf/TiO2 is enhanced, but also for an anionic dye (AO7) that is less adsorbed on Nf/TiO2. The unexpected behavior in AO7 degradation seems to be related to the role of the Nafion layer in retarding the charge recombination. These observations indicate that Nf/TiO2 can enhance the PCD reactivity for cationic substrates without sacrificing the PCD reactivity for anionic substrates. In addition, it was found that the sensitized degradation of RhB followed a different path when the surface of TiO2 was coated with Nafion. The N-de-ethylation of RhB that leads to the generation of rhodamine-110 was a prevailing path with Nf/TiO2, whereas the cleavage of the chromophoric ring structure was dominant with pure TiO2. The effects of Nafion adlayers on the photoinduced electron transfer and PCD kinetics and mechanisms are discussed.

Fluoropolymer coated Dacron or polytetrafluoroethylene for femoropopliteal bypass grafting: a multicentre trial.

BACKGROUND: This trial was designed to compare graft patency between expanded polytetrafluoroethylene (PTFE) and fluoro-polymer coated Dacron for femoropopliteal bypass in patients in whom saphenous vein was unavailable. METHODS: A multicentre prospective trial randomized 129 patients (74 men, 55 women) who underwent femoropopliteal bypass using either a PTFE or fluoropolymer coated Dacron graft. The indication for operation was disabling claudication in 68 (52.7%) and critical limb ischaemia in 61 (47.3%) patients. Distal anastomosis was above the knee in 76 (58.9%) and below the knee in 53 (41.1%) patients. RESULTS: Primary patency at 6, 12 and 24 months was 71%, 56% and 47% for PTFE and 50%, 36% and 36% for fluoropolymer coated Dacron (P = 0.002), respectively. Secondary patency at 6, 12 and 24 months was 77%, 60% and 48% for PTFE and 66%, 49% and 46% for fluoropolymer coated Dacron (P = 0.13), respectively. The superior primary patency of PTFE over fluoropolymer coated Dacron was most evident in patients with poor prognostic indicators for graft survival: critical limb ischaemia (P = 0.001); below-knee anastomosis (P = 0.01); and smaller (6 mm) diameter grafts (P = 0.002). Graft thrombosis developed in the first month in 22 of 61 (36%) patients receiving fluoropolymer coated grafts compared to six of 68 (8.8%) patients receiving PTFE, which accounts for the difference in primary patency. Successful thrombectomy in 10 of the 22 fluoropolymer coated grafts resulted in similar secondary patency. CONCLUSION: Polytetrafluoroethylene has superior primary patency and similar secondary patency to fluoropolymer coated Dacron. These results support the preferential use of PTFE in patients with critical limb ischaemia, especially when a below-knee distal anastomosis and smaller diameter graft is required.

Polyvinylidene fluoride monofilament sutures: can they be used safely for long-term anastomoses in the thoracic aorta?

Polyvinylidene fluoride (PVDF) represents an attractive alternative to polypropylene as a monofilament vascular suture because of its satisfactory physicochemical properties, it ease of handling, and its good biocompatibility. However, the polymer's ability to remain mechanically and chemically stable when exposed to a mild hydrolytic environment over the long term has yet to be demonstrated. One in vitro study involved the comparison of the long-term relative resistance of PVDF and polypropylene sutures to hydrolysis for a period of 9 years. The PVDF suture showed major molecular rearrangements from the original ratio of three crystalline structures to the single beta crystalline phase. The observation of some surface oxidation and water inhibition did not significantly modify the tensile strength of the PVDF suture, which retained 92.5% of its original value. In contrast, the polypropylene sample did not undergo any recrystallization but was associated with more oxidation byproducts and more water molecules near the surface, which contributed to a 46.6% loss in initial tensile strength. An in vivo study confirmed that PVDF sutures are biocompatible and are able to maintain satisfactory biostability when used to anastomose thoracic aortic allografts for a period of 6 months in the dog. The cellular reaction of fresh allografts as well as the control autografts to PVDF sutures was minimal. In other allografts that had been preserved in a supplemented medium for 1 week prior to implantation, the PVDF sutures healed satisfactorily with the formation of neocollagen and few macrophages surrounding the monofilament. No evidence of instability at the allograft-host artery junction was observed, confirming that the PVDF sutures were able to ensure a secure anastomosis in the thoracic aorta. PVDF sutures have demonstrated superior long-term biostability in vitro and minimal tissue response in vivo. These are two essential requirements when evaluating the use of a suture for vascular surgery in general and thoracic aortic surgery in particular.

Tissue response to expanded polytetrafluoroethylene and silicone implants in a rabbit model.

BACKGROUND: Expanded polytetrafluoroethylene (ePTFE) and silicone are safe and relatively biocompatible materials. OBJECTIVE: To compare, using multiple histologic parameters, the tissue response to a standard silicone soft tissue implant with the response to a modified ePTFE implant. The modified form of ePTFE is reinforced with fluorinated ethylene propylene (FEPRePTFE), which provides increased pliability and material integrity. METHODS: The implants were placed into a subperiosteal pocket over the skull of adult New Zealand white rabbits. At 7, 30, and 90 days after implantation, en bloc tissue specimens, including skin, implants, surrounding soft tissue, and underlying bone were harvested for gross and histologic evaluation. OUTCOME MEASURES: The tissue response to the implants was assessed with respect to the number of foreign body giant cells present, the thickness of the fibrous capsule, and the general inflammatory response (n = 6 for each implant at each evaluation period). RESULTS: There were no cases of rejection, extrusion, or infection. The silicone implants elicited a significantly thicker capsule and less neovascularization (P<.05). CONCLUSION: The FEPRePTFE demonstrated a favorable tissue response when compared with silicone, particularly in regard to capsule thickness and vascular ingrowth.

An adjustable, butterfly-design, titanium-expanded polytetrafluoroethylene implant for nasal valve dysfunction: a pilot study.

OBJECTIVE: To investigate the design of a simple, adjustable, biocompatible nasal implant that consistently corrects nasal valvular dysfunction. METHODS: This study presents data on an adjustable titanium-expanded polytetrafluoroethylene (ePTFE) implant designed as a spanning butterfly graft. Each patient was measured for implant effectiveness subjectively by patient questionnaire and objectively with static and dynamic photographs as well as acoustic rhinomanometry data. RESULTS: Rhinomanometry studies, photographic evidence, and patient questionnaires revealed that a great improvement in the nasal airway can consistently be achieved at the level of the nasal valve using the titanium-ePTFE butterfly design implant. CONCLUSION: The titanium-ePTFE butterfly-design implant provides a consistent and adjustable correction of the dysfunctional nasal valve.
