Effect of pressure on electrophoretic mobility of polystyrene latex particles.

A capillary electrophoresis system that can apply arbitrary helium gas pressures at both inlet and outlet reservoirs was constructed. The system was used to investigate the effect of pressure on electrophoretic behavior of polystyrene latex particles. The electrophoretic mobility of latex particles was increased with the application of pressure (< 3.0 kgf/cm2). The shrinkage of particle diameter under pressurization was observed using a microscope, however, the magnitude of shrinkage was not enough to explain the increase in electrophoretic mobility. Therefore, the application of pressure might increase the electric charge of the latex particle. Since methanol inhibited the enhancement in the electrophoretic mobility of the latex particles, water might play an important role in increasing mobility.

No programming required. Mobile PCs can help physicians work more efficiently, especially when the application is designed to fit the practice.

UNLABELLED: The Jacobson Medical Group San Antonio PROBLEM: Jacobson Medical Group (JMG) needed a way to effectively and efficiently coordinate referral information between their hospitalist physicians and specialists. SOLUTION: JMG decided to replace paper-based binders with something more convenient and easily updated. The organization chose to implement a mobile solution that would provide its physicians with convenient access to a database of information via a hand-held computer. RESULTS: The hand-held solution provides physicians with full demographic profiles of primary care givers for each area where the group operates. The database includes multiple profiles based on different healthcare plans, along with details about preferred and authorized specialists. KEYS TO SUCCESS: JMG adopted a user-friendly solution that the hospitalists and specialists would embrace and actually use.

XML-based synchronization of mobile medical devices.

Today, mobile computing provides enough resources to be used in medical applications. Patient treatment is a process that involves multiple partners. All those partners need access to common patient-data and need to make changes to the patients' health record. Therefore, data that the partners collect and change with mobile devices has to be synchronized on a central server to form the master patient record. Data conflicts resulting from the synchronization have to be solved automatically. Our project describes a solution for XML-based data replication and synchronization for mobile health applications.

Applicability of discovery science approach to determine biological effects of mobile phone radiation.

We argue that the use of high-throughput screening techniques, although expensive and laborious, is justified and necessary in studies that examine biological effects of mobile phone radiation. The "case of hsp27 protein" presented here suggests that even proteins with only modestly altered (by exposure to mobile phone radiation) expression and activity might have an impact on cell physiology. However, this short communication does not attempt to present the full scientific evidence that is far too large to be presented in a single article and that is being prepared for publication in three separate research articles. Examples of the experimental evidence presented here were designed to show the flow of experimental process demonstrating that the use of high-throughput screening techniques might help in rapid identification of the responding proteins. This, in turn, can help in speeding up of the process of determining whether these changes might affect human health.*

Proteolytic activity in intact sheets of polarized epithelial cells as determined by a cell-permeable fluorogenic substrate.

The purpose of the present investigation was to develop a system for continuous evaluation of extralysosomal proteolytic activity and its regulation in polarized epithelial cells. Filter inserts containing a tight monolayer of primary cultured pig thyrocytes were placed in a thermostated aluminium block. The cell-permeable, fluorogenic calpain and proteasome substrate succinyl-Leu-Leu-Val-Tyr-7-amino-4-methylcoumarin was added to the apical buffer and fluorescence changes were continuously measured via the fibre optics of a luminometer held at a fixed distance from the cell layer. Basal proteolytic activity was reduced by 60-70% by the proteasome inhibitor lactacystin. Proteolysis was increased within a few minutes after application of Ca(2+)-mobilizing agents (ionomycin, 4-bromo-A23187, thapsigargin and maitotoxin). Forskolin and staurosporine also enhanced the proteolytic activity. We conclude that Ca(2+)mobilization, and possibly also changes of protein kinase activity, rapidly increase non-lysosomal proteolysis in the intact thyroid epithelium.

Interaction of proteins of the adenylate cyclase complex: area-limited mobility or movement along the whole membrane? Analysis with the application of the percolation theory.

Extraction of the 'fluid' lipid fraction from rat reticulocyte plasma membranes by phospholipase/BSA treatment leads to a reduction in the percentage of beta-adrenoreceptors complexed with the regulatory N-proteins and to a decrease of adenylate cyclase stimulation with a beta-agonist. Analysis of the data obtained on the basis of the percolation theory suggests that proteins of the adenylate cyclase complex move along the whole membrane.

Strategy of analytical test kits.

The importance of test kits is increasing due to economical reasons. The different types of test kits allow qualitative, semiquantitative or quantitative results depending on the requirements. For applications where rapidity and mobility are absolutely necessary, they have advantages compared to laboratory methods. This is demonstrated by two typical examples. Besides the advantages, the limits of test kits are also mentioned. Test kits look very simple and are really easy to handle; nevertheless, not only the producers but also the users have to fulfil some basic demands.

Mutation in the Sp1 motif of the bovine leptin gene affects its expression.

Leptin is expressed mainly by adipocytes and plays a crucial role in the regulation of energy expenditure, food intake, and adiposity. Using PCR-heteroduplex analysis and sequencing, we investigated a C/G substitution in the promoter region of the bovine leptin gene. Application of the electrophoretic mobility shift assay showed that the C-->G transversion decreased the leptin gene promoter binding capacity for nuclear proteins. With real-time PCR and Western blotting, we showed that the leptin expression level was higher in cattle with the CC than with the GG genotype.

Quantitative analysis of chemiluminescence signals using a cooled charge-coupled device camera.

Chemiluminescence has been considered an alternative to radioisotopic detection of materials in the life sciences. One application of this nonradioisotopic method is the electrophoretic mobility shift assay. The essential requirement for quantitative applications is that the chemiluminescence signal is linearly proportional to the concentration. However, the generation of chemiluminescence is a multi-step process, therefore linearity cannot be assumed. Therefore, it is important to verify linearity before applying the method as a quantitative tool. We used a commercial chemiluminescence generating system to evaluate the validity of quantitative measurements of biotin-labeled tRNA and single-stranded DNA. The results indicate that the relationship between the chemiluminescence signals and the quantity of biotin-labeled nucleic acids is hyperbolic rather than linear. However, it was found that with less than 50 fmol of biotin-labeled nucleic acid, which corresponds to 2.5 nM in 20 microl, linearity can be demonstrated within 5% error. Therefore, chemiluminescence-based quantitative measurements are a reliable method within these limitations.

Diagnostic accuracy of new handheld echocardiography with Doppler and harmonic imaging properties.

BACKGROUND: The first generation of handheld echocardiography devices was evaluated with divergent results because of inherent technical limitations. New handheld devices with continuous/pulsed wave Doppler and tissue harmonic imaging were introduced recently. In this study, comparisons were drawn among standard echocardiography, invasively measured systolic pulmonary artery pressure, and these new devices. OBJECTIVE: We sought to evaluate new handheld echocardiography and its diagnostic accuracy compared with standard echocardiography. METHODS: Two consecutive echocardiographic examinations were performed by experienced and independent examiners using handheld and standard echocardiography. Systolic pulmonary artery pressure was measured by Swan-Ganz catheter. RESULTS: In all, 177 (56.2%) patients had normal cardiac function; 138 (43.8%) had underlying cardiac pathology. Handheld echocardiography had an overall agreement of 94.8% and kappa of 0.89 to detect the main echocardiographic finding. Handheld echocardiography detected valve disease with an agreement of 96.7% and kappa of 0.93; global left ventricular function was assessed correctly in 85.6% of cases. Pericardial effusion was diagnosed with an agreement of 91.2%. Dyssynergy was found by handheld echocardiography with an agreement of 95.4% and kappa value of 0.88. Systolic pulmonary artery pressure measured by handheld echocardiography and Swan-Ganz catheter had a correlation of 0.97. CONCLUSION: This study demonstrates the high diagnostic accuracy of handheld devices with continuous/pulsed wave Doppler and harmonic imaging, and that these devices broaden the diagnostic spectrum while allowing for enhanced mobility in everyday clinical applications.

A lysimeter experiment to investigate the leaching of veterinary antibiotics through a clay soil and comparison with field data.

Pharmaceuticals used in livestock production may be present in manure and slurry as the parent compound and/or metabolites. The environment may therefore be exposed to these substances due to the application of organic fertilisers to agricultural land or deposition by grazing livestock. For other groups of substances that are applied to land (e.g. pesticides), preferential flow in clay soils has been identified as an extremely important mechanism by which surface water pollution can occur. This lysimeter study was therefore performed to investigate the fate of three antibiotics from the sulphonamide, tetracycline and macrolide groups in a clay soil. Only sulphachloropyridazine was detected in leachate and soil analysis at the end of the experiment showed that almost no antibiotic residues remained. These data were analysed alongside field data for the same compounds to show that soil tillage which breaks the connectivity of macropores formed over the summer months, prior to slurry application, significantly reduces chemical mobility.

Spinal manipulation postepidural injection for lumbar and cervical radiculopathy: a retrospective case series.

OBJECTIVE: To describe the safety and potential therapeutic benefit of spinal manipulation postepidural injection in the nonsurgical treatment of patients with cervical and lumbar radiculopathy. METHODS: The study design was a retrospective review of outcomes of 20 cervical and 60 lumbar radiculopathy patients who underwent spinal manipulation postepidural injection in a hospital setting. Patients received either fluoroscopically guided or computed tomography (CT)-guided epidural injection of a combination of lidocaine and Depo-Medrol. The manual therapy consisted of an immediate postepidural application of flexion distraction mobilization and then high-velocity, low-amplitude spinal manipulation to the affected spinal regions. Outcome criteria were empirically defined as significant improvement, temporary improvement, or no change. The minimum follow-up time for all patients was 1 year. RESULTS: There were no complications associated with spinal manipulation, whereas 3 complications associated with the epidural injection procedure were noted. Of lumbar spine patients, 36.67% (n = 22) noted significant improvement, 41.67% (n = 25) experienced temporary improvement, and 21.67% (n = 13) reported no change. Of the patients undergoing spinal manipulation after cervical epidural injection, 50% (n = 10) noted significant improvement, 30% (n = 6) experienced temporary improvement, whereas 20% (n = 4) exhibited no change. CONCLUSIONS: These data suggest that spinal manipulation postepidural injection is a safe nonsurgical procedure to use in the treatment of the patient with radiculopathy of spinal origin. This is also the first report of the use of spinal manipulation postepidural injection in the cervical spine.

Modified RP4 and Tn5-Mob derivatives for facilitated manipulation of large plasmids in Gram-negative bacteria.

We have constructed a set of RP4 (NmS/TcS) and Tn5-Mob derivatives which have applications in experiments involving mobilization of replicons in many Gram-negative organisms. The different selection markers of the RP4 and Tn5-Mob derivatives include streptomycin, chloramphenicol, gentamicin, and spectinomycin resistance as well as mercury resistance, and a constitutively expressed lacZ gene. This choice of markers allows the use of these derivatives in bacteria which are naturally resistant to many antibiotics, and in strains which contain pre-existing resistance plasmids, transposons, or antibiotic cassette insertions. In addition, a RP4 derivative carrying the sacB gene of Bacillus subtilis was constructed. This allows the selection for the loss of RP4 after it has been used to mobilize other plasmids. A Tn5-Mob-sacB derivative with a new marker (Gm) was also developed, as were vectors which take advantage of the sacB gene to facilitate replacement of existing Tn5 inserts with other Tn5 derivatives. As an example of the use of these tools, three Rhizobium leguminosarum bv. viciae VF39 plasmids which have been shown to be involved in symbiosis were differentially tagged and mobilized (individually and in various combinations) to the plasmid-free Agrobacterium tumefaciens strain UBAPF2. None of the resultant Agrobacterium strains was able to fix nitrogen in symbiosis with peas.

Application of photobleaching for measuring diffusion of prion proteins in cytosol of yeast cells.

Measurement of fluorescence recovery after photobleaching (FRAP) is a non-invasive technique for studying protein dynamics in real time in living cells. FRAP studies are carried out on proteins tagged with green fluorescent protein (GFP) or one of its spectral variants. Illumination with high intensity laser light irreversibly bleaches the GFP fluorescence but has no effect on protein function. By photobleaching a limited region of the cytoplasm, the rate of fluorescence recovery provides a measure of the rate of protein diffusion. A detailed description of the FRAP technique is given, including its application to measuring the mobility of GFP-tagged Sup35p in [psi(-)] and [PSI(+)] cells.

Enantiomeric resolution of a series of chiral sulfoxides by high-performance liquid chromatography on polysaccharide-based columns with multimodal elution.

The enantiomeric resolution of a series of 20 asymmetric sulfoxides was systematically investigated by HPLC using multimodal elution with amylose trisR(S)-1-phenylethylcarbamate], amylose tris(3,5-dimethoxyphenylcarbamate) and amylose and cellulose tris(3,5-dimethylphenylcarbamate) phases. The sulfoxide series was composed of aromatic, olefinic and ketosulfoxides, sulfinyl acids and esters. This work has shown that enantioselectivity and enantioresolution of the polysaccharide-based columns can be achieved by changing the type and composition of the mobile phase, widening the applicability of these chiral phases.

Conformation of surface bound polyelectrolytes: I. Implications for cell electrophoresis.

We present a self-consistent calculation for a layer of short-chain polyelectrolytes grafted to a planar interface. Individual self-avoiding chain configurations are generated on a tetrahedral lattice. By evaluating the complete ensemble of configurations in the presence of a self-consistent field we determine averages for the equilibrium layer height, segment and charge density profiles. These describe the response of the layer to changes in the surrounding medium, in particular to electrolyte concentration. This self-consistent model of a biological cell, which is known to have an extended layer of charge exterior to the membrane bilayer surface, is applied to the problem of cell electrophoretic mobility. We compare the electrophoretic mobility deduced from the application of this model with that for a constant profile of charge and segments and with the case of a constant segment profile with a plane of charge, usually assumed in the literature. We find that, within the region of concentrations studied here and with the chain lengths used, there is general agreement between the different descriptions.

Application of numerical taxonomy techniques to the choice of optimum mobile phase in high-performance thin-layer chromatography (HPTLC).

A total of eight solvent systems used for the separation of 1,4-benzodiazepine mixtures were investigated. The classification of solvent systems was carried out by the numerical taxonomy technique. The selection of optimum solvent system was accomplished on the basis of information quantity and objective function. The method was found to be a rapid and efficient tool in the choice of optimum system solvent.
