[Biopharmaceutical aspects of the medicinal forms of cephalexin].

The rate of cephalexin release from various dosage forms such as tablets, capsules and suspensions manufactured by several firms was studied. In vivo studies showed that pharmacokinetic indices of the absorption depended on the rate of cephalexin release from the dosage forms. The proposed procedure for in vitro estimation of the cephalexin release rate reflects bioavailability of the antibiotic dosage forms and may be used in quality control of finished dosage forms.

Biopharmaceutical investigation of fenoprofen.

The salts of fenoprofen formed with different metals have shown various crystal forms and solubility. The calcium salt has proved the most suitable characteristics for tablet and capsule production. Dissolution and absorption parameters of this substance were studied using in vitro and in vivo methods. The absorption rate and the correlation between pH and membrane diffusion rate constant were investigated in vitro using the "Sartorius" apparatus. The dissolution rate--depending on pH--was investigated by the oscillometric method. The in vivo disintegration of an experimental sample was compared with a commercial preparation. The comparison has been documented by endoscopic photography.

Applications of column-switching technique in biopharmaceutical analysis. I. High-performance liquid chromatographic determination of amitriptyline and its metabolites in human plasma.

A new high-performance liquid chromatographic method for the determination of amitriptyline and its metabolites, nortriptyline, 10-hydroxynortriptyline and 10-hydroxyamitriptyline, in plasma is described which uses direct injection and a column-switching valve. The method is based on the enrichment of drugs on a reversed-phase concentration column, packed with Corasil RP. The enriched drugs were then separated, using back-flush mode on a bonded-phase CN column using an isocratic acetonitrile-acetate buffer (60:40, v/v) mobile phase. The validation of the method showed excellent sensitivity, precision and reproducibility. The limit of detection, using a 250-microliter direct injection of plasma, was between 5 and 10 ng/ml for each of the four drugs. The mean coefficient of variation for intra- and inter-assay was better than 5%. The method showed obvious advantages over conventional extraction procedures in terms of speed and ease of sample handling. The method has been successfully applied to the samples from patients receiving oral doses of amitriptyline.

Biopharmaceutical studies of oral formulations containing propyphenazone.

Studies were performed with oral dosage formulations of propyphenazon, i.e. aqueous suspension (1%), instant suspension (1%) and tablets (100 mg) for pediatric use. The influence of the pharmaceutical adjuvants of the release of the drug from the model preparations was established. The biological availability determined from rabbits showed that the formulated preparations are biologically equivalent.
