Effect of cigarette-smoke condensate and norharman on the induction of SCEs by direct and indirect mutagens in CHO cells.

Cigarette-smoke condensate and norharman were investigated either alone or in combination with a number of direct or indirect mutagens for the induction of sister-chromatid exchanges (SCEs) in Chinese hamster ovary (CHO) cells. Cigarette-smoke condensate and norharman induced SCEs in these cells, only in the presence of a metabolic activation system. The number of SCEs induced by the direct-acting mutagens mitomycin C and N-methyl-N'-nitro-N-nitrosoguanidine was decreased in the presence of cigarette-smoke condensate or norharman. However, cigarette-smoke condensate and norharman showed synergistic effects in combination with the indirect mutagens 2-acetylaminofluorene, 2-aminofluorene, N-hydroxy-acetylaminofluorene, 2-aminoanthracene and benzo[a]pyrene. No synergism was observed when CHO cells were treated simultaneously with cigarette-smoke condensate or norharman and the indirect mutagen cyclophosphamide.

Mutagenicity testing of condensates of smoke from titanium dioxide/hexachloroethane and zinc/hexachloroethane pyrotechnic mixtures.

Condensates of smoke from titanium dioxide/hexachloroethane and zinc/hexachloroethane pyrotechnic mixtures were investigated for their potential to produce genetic damage in the tester strains TA98, TA100, TA1535 and TA1537 of Salmonella typhimurium and in the mouse bone marrow micronucleus assay. Both smoke condensates contained several chlorinated hydrocarbons among which tetrachloroethylene, hexachloroethane, hexachlorobutadiene and hexachlorobenzene were identified by GC/MS. Condensate of smoke from titanium dioxide/hexachloroethane showed a dose-related positive response in the Salmonella assay with strains TA98 and TA100 in the absence of metabolic activation from rat liver S9 fraction. Both smoke condensates were negative in the micronucleus assay but produced a small but significant depression of erythropoietic activity. The results indicate that smoke condensate from titanium dioxide/hexachloroethane mixtures contains unidentified compound(s) that may be considered mutagenic in the Salmonella assay.

Ion competition and micrococcal nuclease digestion studies of spermidine-condensed calf thymus DNA. Evidence for torus organization by circumferential DNA wrapping.

Spermidine-condensed calf thymus DNA structures have been studied by ion competition using a sedimentation assay and by micrococcal nuclease digestion. Competitor ions Mg2+, Ca2+ and putrescine2+ show specific ion effects; but all three appear to affect the DNA condensation-decondensation equilibrium caused by spermidine3+ in a qualitatively similar manner, suggesting the spermidine3+-DNA interaction is largely electrostatic. Our data show a hysteresis in condensation and decondensation transition directions. We interpret this in terms of a kinetic block in the condensation direction with decondensation representing the equilibrium state of the system. These results agree with results obtained from related systems using different measurement techniques. Micrococcal nuclease digestion of spermidine-condensed calf thymus DNA produces broad but discrete bands in gel electrophoresis experiments. At least two bands determined to be 760 +/- 87 bp and 1355 +/- 135 bp, possess the size ratio 1:1.8 +/- 0.4 consistent with their forming the monomer and dimer fragments of an arithmetic band series. We rationalize this result in terms of a localized micrococcal nuclease cleavage model of circumferentially-wrapped DNA toruses proposed previously by Marx, K.A. and Reynolds, T.C. (Proc. Natl. Acad. Sci. (1982) 79, 6484-6488). The arithmetic series monomer band (760 +/- 87 bp), corresponding to wrapping B DNA once circumferentially about the torus, is in agreement with the electron microscopic measurements of hydrated calf thymus DNA torus circumferences presented by Marx, K.A. and Ruben, G.C. (Nucleic Acids Res. (1983) 11, 1839-1853).

Clinical management of silicone intraocular lens condensation.

PURPOSE: To report a novel technique in the management of silicone intraocular lens condensation after gas-fluid exchange. DESIGN: Interventional case report. METHODS: Clinical management of an obstacle to treatment. setting: Private retina practice. patient population: One patient with silicone posterior chamber intraocular lens implant and posterior capsulotomy who subsequently experienced complicated retinal detachment. intervention: Application of a heating pad to clear intraocular lens condensation. main outcome measure: Visibility of the fundus for laser treatment. RESULTS: Application of a heating pad to the closed eyelid significantly reduced the intraocular lens condensation and provided a clear view for the surgeon to apply laser retinopexy. CONCLUSIONS: Treatment of retinal detachment is complicated by lens condensation after gas-fluid exchange in patients who have undergone silicone posterior chamber intraocular lens implantation and posterior capsulotomy. We report a simple, inexpensive, and effective method to reduce lens condensation and enable in-office examination and laser treatment.

Cross-condensation and particle growth in aqueous silane mixtures at low concentration.

We have observed the condensation of a mixture of gamma-glycidoxypropylmethyldiethoxysilane and 3-aminopropyltriethoxysilane in dilute aqueous solutions. NMR and IR spectroscopy have allowed to follow the condensation process in the mixture, which is noticeably enhanced and proceeds faster than for each silane on its own. Cross-condensation between the two silanes was evidenced. When the hydrophilic aminosilane is in excess, the condensation, as evidenced by dynamic light scattering, proceeds toward gel formation because the oligomers formed are essentially hydrophilic. When the more hydrophobic epoxysilane is in excess, oligomer growth proceeds slowly and results in a destabilization of the solution: due to their hydrophobic character, the oligomers formed coalesce suddenly into 200-nm-diameter aggregates. Coatings deposited from such solutions with high epoxysilane content can be used to strengthen glass. We show that the progress of condensation in solution results in a wetting transition during deposition of the silane film on glass by dip coating. The production of increasingly hydrophobic oligomers as the reaction time increases results in adsorption of more hydrophobic aggregates at the surface, which eventually leads to dewetting of the film: in the absence of film, glass strengthening disappears.

Line of charges in electrolyte solution near a half-space I. Counterion condensation.

The effect of a half-space on counterion condensation around a line of charges in electrolyte solution is examined in the framework of Debye-Hückel electrostatics. The half-space substrate is allowed to be a conductor, a dielectric, or a semiconductor. Counterions are predicted to be released completely as the line of charges approaches a conducting substrate. When it approaches a dielectric substrate, depending on the ratio of solvent to substrate dielectric constant, there are three possibilities: (1) epsilon(sol)/epsilon(sub) < 1; the counterions are partially (or completely) released; (2) epsilon(sol)/epsilon(sub) = 1; the amount of condensation remains unchanged; and (3) epsilon(sol)/epsilon(sub) > 1; more counterions condense. Depending on the relative magnitude of screening lengths in the semiconductor and in the solution, its effect on condensation follows either that of a metal or that of a dielectric. For the case of a moderately doped silicon substrate, condensation is predicted to be similar to that for a dielectric.

Is phenol condensation one of the major pathways in the formation of polychlorinated dibenzofurans in municipal waste incinerators?: Model prediction vs. field observation.

The role of phenol condensation pathways in the formation of polychlorinated dibenzofurans (PCDF) in a municipal waste incinerator was assessed by comparing predicted PCDF homologue and isomer patterns with those obtained from the incinerator. A two-phenol condensation model, dependent only on the distribution of phenols, was used to predict the distributions of PCDF congeners in the incinerator. Complete distributions of phenols and PCDF congeners were obtained from the incinerator. To quantify the degree of agreement between obtained isomer distributions and those predicted by the model, R-squared values from linear correlations were calculated for the dichlorinated- through hexachlorinated-isomers. They ranged from 0.001 to 0.1. Agreement between obtained and predicted PCDF isomers was very poor for all homologues, suggesting that phenol condensation pathways are unlikely to be the primary route in the formation of PCDF in the incinerator. However, dibenzofuran (DF) is likely to be produced from a condensation of two phenols. This paper shows the use of PCDF homologue and isomer patterns calculated by the two-phenol condensation model for testing PCDF formation mechanism attribution in a municipal waste incinerator.

Relationship between non-return rate and chromatin condensation of deep frozen bull spermatozoa.

This study analyzes the relationship between chromatin condensation and field fertility, expressed as 90 days non-return rate (NRR), of bulls actively used by AI studs. Frozen-thawed semen from five bulls (six ejaculates per bull, three straws per ejaculate), that showed a non-return rate between 60 and 80%, were analyzed to assess sperm chromatin condensation and stability. The chromatin condensation was determined by flow cytometry using propidium iodide as fluorochrome, and the chromatin stability was evaluated by inducing its decondensation with SDS and EDTA. Coefficient of variation among replicates was less than 7% and 5% for chromatin condensation and stability, respectively. No correlation was present between chromatin condensation and NRR. However, significant correlation was found between chromatin stability and NRR. Chromatin stability was higher (P < 0.05) in those bulls that showed higher fertility. The results obtained in this study conclude that assessment of stability could be a valuable tool for routine evaluation and identification of ejaculates with high levels of sperm chromatin abnormalities and to detect animals of higher reproductive potential.

Implications of the reorganizational behavior of condensed phosphates for mitochondrial phosphorylation.

The composition of phosphorus-32 labeled polyphosphoric acid (32PPA) prepared from, (i) H3(32)PO4 and (82 percent w/w) P2O5 and, (ii) by incorporation of radioactivity from H3(32)PO4 into unlabeled polyphosphoric acid (PPA) was examined and found to be the same. The identity of these condensed phosphorate mixtures, as established, argues strongly in favor of a rapid and complete equilibration and provides direct evidence in support of Van Wazer's reorganization theory of condensed phosphates. No evidence for metaphosphates or label exchange in an aqueous environment was found. 32PPA was found to exist in metastable equilibrium with 32PPA as a crystalline solid of different composition. Agreement of hydrolytic rate data from the mixtures of condensed phosphates with literature values for individual species suggests that the hydrolysis of the pyro- and tripoly-phosphate species proceeds independently in the presence of other homologs. The rate of hydrolysis of the condensed phosphates (at pH5.0 and 65 degrees C) was found to be proportional to chain length, increasing about four-fold between pyro- and hexapoly-phosphate. The same correlation is also manifest in the adenosine 5' polyphosphates. A slight enhancement in the rate of hydrolysis of adenosine diphosphate, increasingly more pronounced in the case of adenosine tri- and tetra-phosphate, was also noted. This effect is attributed to the presence of the adenosine moiety. The most interesting aspect of the rate data is to be found in the similarity of the rates of hydrolysis of the adenosine 5' polyphosphates, relative to their inorganic polyphosphate counterparts. The significance of these findings is discussed and the viewpoint is presented that the properties of condensed phosphates are compatible with the requisite characteristics that must be predicated of any in situ phosphoryl precursor in mitochondrial phosphorylation.

Use of mandelic acid condensation polymer (SAMMA), a new antimicrobial contraceptive agent, for vaginal prophylaxis.

OBJECTIVE: To assess the contraceptive properties, antimicrobial activity, and safety of mandelic acid condensation polymer (SAMMA). DESIGN: Experimental study of SAMMA's in vitro and in vivo properties. SETTING: Academic research laboratories. PATIENT(S): Healthy volunteers for semen donation in an academic research environment. INTERVENTION(S): Inhibition of sperm function indicators, conception, sexually transmitted infection-causing pathogens (including HIV), and lactobacilli was evaluated. Safety indicators were studied. MAIN OUTCOME MEASURE(S): Quantitation of SAMMA's effect on microbial infectivity or multiplication and on sperm function in vitro; evaluation of contraceptive efficacy in vivo; assessment of safety in vitro and in vivo. RESULT(S): Mandelic acid condensation polymer is not cytotoxic toward lactobacilli, microbial host cells, and spermatozoa. The compound inhibits hyaluronidase and acrosin, induces sperm acrosomal loss, and is contraceptive in the rabbit model. Mandelic acid condensation polymer prevents infectivity of HIV and herpesviruses 1 and 2 and, to a lesser extent, of Chlamydia trachomatis. It inhibits the multiplication of Neisseria gonorrhoeae. Mandelic acid condensation polymer is not mutagenic, has low acute oral toxicity, and is safe in the rabbit vaginal irritation assay. CONCLUSION(S): Mandelic acid condensation polymer inhibits sperm function, is contraceptive, has broad-spectrum antimicrobial activity, and is highly safe. Further development as a microbicide is warranted.

Activation of acyl condensation reaction of monomeric 6-hydroxymellein synthase, a multifunctional polyketide biosynthetic enzyme, by free coenzyme A.

6-Hydroxymellein (6HM) synthase is a multifunctional polyketide enzyme induced in carrot cells, whose fully active homodimer catalyzes condensation of acyl-CoAs and the NADPH-dependent ketoreduction of the enzyme-bound intermediate. 6HM-forming activity of the synthase was markedly decreased when the reaction mixture pH was adjusted from 7.5 to 6.0. However, under these slightly acidic conditions, the acyl condensation catalyzed by the dissociated monomer enzyme was appreciably stimulated by addition of free coenzyme A (CoA). In contrast, the condensation reaction at pH 6.0 was significantly inhibited in the presence of CoA when the reaction was carried out with the NADPH-omitted dimer synthase. Among the kinetic parameters of the acyl condensation, velocity of the monomer-catalyzing reaction at the acidic pH was appreciably increased upon addition of CoA while K(m)s did not show any significant change in the presence and absence of the compound. These results suggest that CoA associates with a specific site in the dissociated monomeric form of 6HM synthase, and the velocity of the acyl condensation reaction catalyzed by the CoA-synthase complex appreciably increases in acidic conditions.

Endodontic retreatment of Thermafil versus laterally condensed gutta-percha.

The Thermafil device is becoming a popular obturating material. Because endodontic failures inevitably occur with all techniques, questions of retreatment of Thermafil-obturated teeth will also arise. The purpose of this study was to compare retreatment of Thermafil and laterally condensed gutta-percha. Thirty-five mandibular incisors were prepared with a stepback flare technique and divided into two groups. One group (n = 20) was obturated using Thermafil, the other group (n = 15) was obturated with laterally condensed gutta-percha. The teeth were stored in a humidor for 3 months to allow sealer to set. Retreatment of both groups was accomplished using a combination of heat and chloroform solvent. The time for retreatment was recorded. The teeth were split longitudinally and the amount of gutta-percha remaining in the root canal was measured and analyzed statistically. The results showed that in the coronal one-third of the canal, use of the Thermafil device resulted in significantly more remaining gutta-percha than use of laterally condensed gutta-percha. However, in the apical and middle one-third of the canal the difference in remaining gutta-percha between Thermafil and laterally condensed gutta-percha was not significant. The metal carrier was easily removed and the mean time for retreatment was 6.3 min for Thermafil compared with 5.7 min in the lateral condensation group.

Condensation on the posterior surface of silicone intraocular lenses during fluid-air exchange.

PURPOSE: Posterior chamber foldable silicone intraocular lenses (IOLs) are becoming increasingly prevalent in patients undergoing a pars plana vitrectomy with fluid-air exchange. The authors report an important limitation of foldable silicone IOLs during fluid-air exchanges in pars plana vitrectomies. METHODS: The charts of 18 pseudophakic patients with foldable silicone IOLs who underwent vitrectomy with fluid-air exchange by the authors were reviewed. RESULTS: There was a statistically significant difference in the occurrence of condensation during fluid-air exchange between the group of patients with a capsulotomy versus those that did not have a capsulotomy (P = 0.003). Condensation limiting the view of the retina occurred during fluid-air exchange in 11 of 11 of the patients with foldable silicone lenses and a capsulotomy. Attempts to remove the condensation with a soft-tipped aspiration cannula resulted in limited view of the retina for 1 to 2 minutes in 6 of 11 patients. Use of a thin film of silicone oil restored the view in one patient. In the presence of an intact posterior capsule, condensation did not occur on identical foldable silicone IOLs in seven of seven patients. CONCLUSION: Recognition of the presence of a foldable silicone lens is important when an air-fluid exchange is anticipated. If a capsulotomy is present, the surgeon must be aware that condensation may limit the view of the retina severely during and after surgery. Intraoperatively, the view of the retina usually can be restored in short surgeries by wiping the posterior lens surface with a soft-tipped cannula, and in more complex surgeries by applying a thin film of silicone oil on the posterior surface of the lens.

Mutagenicity of smoke condensates from cigarettes, cigars and pipe tobacco.

Smoke condensates from cigarettes, cigars and pipe tobacco were mutagenic on Salmonella typhimurium TA100 and TA98 when activated with rat liver microsomal system. Mutagenicity of a unit weight of smoke condensate was rather high in cigars, low in pipe tobacco and intermediate in cigarettes. Specific mutagenic activity was almost comparable among smoke condensates from low- to high-tar cigarettes, although some variations were observed depending upon the country producing the cigarettes. Marked mutagenicity of cigarette smoke condensate could not be explained by the benzo (a) pyrene or nitroso compounds it actually contained, suggesting the presence of other very potent mutagens in tobacco smoke condensates.

Mutagenic activity of marihuana smoke condensates.

Smoke condensates prepared from marihuana cigaretts were mutagenic in strain TA98 of the Ames Salmonella/microsome test, a short-term bioassay which estimates the mutagenic and carcinogenic potential of some chemicals. The mutagens in marihuana smoke condensates required liver enzymes to be activated. The specific mutagenic activity of marihuana smoke condensates were similar to that of tobacco smoke condensates prepared from American cigarettes. Fractionation studies of the marihuana smoke condensates showed that basic components accounted for 76% of the recovered mutagenic activity.

Removal of an airborne low volatility metal under fuel-rich and fuel lean conditions through condensation onto soot and/or sorbent particles.

The removal by a sorbent of a low volatility heavy metal, barium, from a combustor exhaust stream was investigated experimentally. The barium was introduced into the gas phase through a combustion process simulating incineration of waste materials. Sorbent- and metal-feed-rate, temperature and air intake were varied to study the impact on the gas-to-particle conversion and the metal condensation process. Peak temperature and subsequent temperature drop were found to be the dominant factors affecting the condensation process. Fuel-rich and fuel-lean conditions result in lower peak temperatures and a more rapid temperature drop relative to stoichiometric conditions. Since the metal vapor saturation pressure falls exponentially with temperature a larger condensation rate was observed, as long as nucleation was suppressed. Fuel-rich conditions promote the formation of soot particles. These particles function as sites for subsequent metal-species condensation. Chlorine was injected into the flow, to study the effect on metal vaporization. Chlorine promoted the volatilization of barium and therefore the subsequent condensation capture was made more effective.

Aberrant replication timing induces defective chromosome condensation in Drosophila ORC2 mutants.

BACKGROUND: The accurate duplication and packaging of the genome is an absolute prerequisite to the segregation of chromosomes in mitosis. To understand the process of cell-cycle chromosome dynamics further, we have performed the first detailed characterization of a mutation affecting mitotic chromosome condensation in a metazoan. Our combined genetic and cytological approaches in Drosophila complement and extend existing work employing yeast genetics and Xenopus in vitro extract systems to characterize higher-order chromosome structure and function. RESULTS: Two alleles of the ORC2 gene were found to cause death late in larval development, with defects in cell-cycle progression (delays in S-phase entry and metaphase exit) and chromosome condensation in mitosis. During S-phase progression in wild-type cells, euchromatin replicates early and heterochromatin replicates late. Both alleles disrupted the normal pattern of chromosomal replication, with some euchromatic regions replicating even later than heterochromatin. Mitotic chromosomes were irregularly condensed, with the abnormally late replicating regions of euchromatin exhibiting the greatest problems in mitotic condensation. CONCLUSIONS: The results not only reveal novel functions for ORC2 in chromosome architecture in metazoans, they also suggest that the correct timing of DNA replication may be essential for the assembly of chromatin that is fully competent to undergo mitotic condensation.
