Reliability of fat-free mass estimates derived from total-body electrical conductivity measurements as influenced by changes in extracellular fluid volume.

Our study addresses the concern that the relationship between total-body electrical conductivity (TOBEC) measurements and the fat-free mass (FFM) or total-body water (TBW) of an individual is altered if significant fluid and electrolyte changes occur. Body composition and TOBEC measurements were obtained from 11 healthy miniature piglets before and after an intraperitoneal injection of physiological saline. The procedure expanded the extracellular fluid (ECF) volume by 11.8-34.1%, which represented an average increase in TBW of 7.9%. The linear regressions that related the preinjection and postinjection estimates of TBW or FFM to the corresponding transformed TOBEC signals were the same. Thus, the prediction equations derived for the untreated piglets accurately predicted both TBW and FFM in the piglets whose volume was expanded. These data suggest that prediction equations derived from healthy subjects should be equally valid for subjects with altered fluid status.

Modifications of gastric compliance in dogs related to changes of extracellular fluid volume: a possible physiological role.

Gastric compliance was measured with an intragastric probe connected to a water manometer in pylorus-ligated anesthetized dogs. Measurements were carried out in the same animal at different levels of extracellular fluid (ECF) obtained by expansion, bleeding and reexpansion. Acute isotonic expansion of the ECF volume shifted gastric compliance curves upwards and to the left, indicating that the stomach retains less volume than the control at the same intraluminal pressure. Conversely, the acute loss of blood shifted gastric compliance curves downwards and to the right. The data suggest that the ECF volume contributes to the modulation of gastric compliance.

Kainic acid causes redox changes in cerebral cortex extracellular fluid: NMDA receptor activity increases ascorbic acid whereas seizure activity increases uric acid.

Kainic acid (KA) causes seizures and extensive brain damage in rats. To study the effects of KA on the redox state in cerebral cortex extracellular fluid (ECF), ascorbic and uric acid concentrations were measured in intracerebral microdialysis samples before and after systemic KA administration (ip). During seizures, concentrations of ascorbic and uric acid increased 500 and 100%, respectively. When midazolam was given with KA to prevent seizures, ascorbic acid still increased 400%, but uric acid increased only transiently. When the NMDA receptor antagonist aminophosphonovaleric acid (APV) was included in the microdialysis perfusion media, ascorbic acid levels decreased during baseline perfusion in a concentration-dependent manner. APV then suppressed the KA-induced increase in ascorbic acid levels, without blocking seizure activity. In summary, increased uric acid levels in brain ECF activity after KA administration are related to the induced seizure, but ascorbic acid levels are associated with NMDA receptor activity.

Changes in plasma volume and extracellular fluid volume and after addition of hydralazine to propranolol treatment in patients with hypertension.

In 16 patients with hypertension, BP could not be controlled satisfactorily by treatment with propranolol alone (mean dosage 325 mg/day). Plasma volume (PV) (T-1824) and extracellular fluid volume (ECV) (82Br-distribution space) were determined in these patients before and after the addition of hydralazine for three months (mean dosage 135 mg/day). After the addition of hydralazine, PV and ECV increased significantly, by 9% and 3%, respectively. Systolic and diastolic BPs decreased, by 15% and 13%. The mechanisms inducing fluid retention during treatment with hydralazine and the clinical significance of the problem are discussed. It is concluded that the addition of a diuretic to propranolol-hydralazine treatment is often well indicated.

Reactive oxidant species in piriform cortex extracellular fluid during seizures induced by systemic kainic acid in rats.

Kainic acid (KA) administered systemically to rats produces seizures and brain damage. We measured an increase in reactive oxidant species (ROS) during KA-induced seizures in the extracellular fluid (ECF) of the piriform cortex, a brain region known to be subsequently damaged. Intracerebral microdialysis samples were collected and assayed for isoluminol-dependent chemiluminescence before and after injection of KA (16 mg/kg, i.p.). Hydrogen peroxide (H2O2) concentrations were calculated from catalase-sensitive chemiluminescence, the difference between total and catalase-resistant chemiluminescence. During generalized tonic-clonic seizures, both total and catalase-resistant chemiluminescence increased significantly in samples from brain ECF. Catalase-resistant chemiluminescence, most likely produced by ascorbic acid, increased for a full hour during sustained seizure activity. H2O2 concentrations showed a trend towards elevation during seizures. Increased ROS suggest that oxidative stress occurs in brain ECF during sustained seizure activity.

Effect of extracellular fluid volume expansion on the interparabronchial septum of the avian lung.

Scanning electron microscopy was used to examine the interparabronchial septa of chickens as a potential site of lymphatic drainage in the avian lung. Anaesthetized chickens were subjected to extracellular fluid volume expansion to produce pulmonary oedema as a result of increased capillary fluid flux into the interstitial spaces of the lung. In normal (control) chickens, the adjacent parabronchi were separated by a minimal septal space. In the "volume-loaded" birds, however, the interparabronchial septal spaces were measurably thickened and engorged as a result of hydrostatic pulmonary oedema. These results, which were consistent with reports of the effect of hydrostatic pulmonary oedema in mammals, suggest that the interparabronchial septum is a potential route of lymphatic drainage in the avian lung.

Radiosulphate: its metabolism and use in measurement of extracellular fluid volume in calves.

Radiosulphate injected intravenously into calves under 14 days of age equilibrated, by 30 min, with a mean fluid volume of 311 ml/kg body weight. During the equilibration period 3-7 per cent of the injected dose was excreted in the urine. The volume of distribution of radiosulphate increased for at least 4-5 h after equilibration suggesting that it penetrated beyond the readily miscible water of the extracellular fluid.

Effects of extracellular fluid volume changes on renal response to low-dose dopamine infusion in normal women.

Dopamine (DA) i.v. infused in a low dose (0.1 microgram/kg/min) in healthy women during sustained hypotonic polyuria, produced different renal functional effects as variations in extracellular fluid volume occurred. (1) In hydro-saline retention (n = 23), induced by desoxycorticosterone acetate treatment, DA produced typical vasodilator and hydro-natriuretic effects (Goldberg, 1972). (2) In hydro-saline depletion (n = 19), induced by diuretic treatment and low dietary sodium intake, DA lost its vasodilator and natriuretic efficacy, manifesting, on the other hand, renal sodium conserving effects mainly dependent on the increase in distal sodium reabsorption and a trend towards afferent arteriolar vasoconstriction. (3) Treatment with prazosin (n = 9) or propranolol (n = 9) in hydro-saline depletion, was efficacious in partly restoring the typical vasodilator and natriuretic effects of DA. Thus, in hydro-saline depletion, DA produced sympathomimetic effects which were sufficiently intense to outweigh those due to activation of specific DA renal receptors.
