[Endosteal hyperostosis with dominant transmission. Description of 8 cases in 3 generations of the same nuclear family].

The authors report on 8 patients with the dominant type of endosteal hyperostosis (Worth's disease), found in the same kindred over 3 generations. Three were males, five females, from 5 to 49 years of age. Major radiographic features were an increased density of bones of the skull and spine, with thickening of the diaphyseal cortex of tubular bones, without any change in remodeling. Seven patients were almost asymptomatic. A decompressive craniotomy had been performed in one, who showed a compression of the 2nd, 5th and 8th cranial nerves. Blood alkaline phosphatase was normal. The bone density increases with age, but in spite of this the clinical course if milder than in recessive type of generalized cortical hyperostosis (Van Buchem's disease, as reported in the 35 cases of endosteal hyperostosis (dominant type) previously described.

Toxicity of Cassia occidentalis in the horse.

Three Shetland ponies were given a single oral dose of ground Cassia occidentalis seeds in aqueous suspension. The clinical signs observed resembled those seen in naturally occurring and experimental cases in cattle. The syndrome was characterized by an afebrile course, incoordination, recumbency and death. Elevations of blood alkaline phosphatase, CPK, LDH, and SGOT were observed. Although muscle lesions were not seen grossly, microscopic lesions included segmental necrosis of skeletal muscle fibers. The findings were regarded as sufficiently characteristic of C. occidentalis poisoning to be useful in differential diagnosis. When Cassia poisoning is suspected, access of the affected horses to the plant should be demonstrated.

[Use of glycerin as a component of the solution in treating acute hemorrhage].

The multicomponent solution, containing 15% of glycerol, 4.5% of proteins, 0.9% of sodium chloride, 0.33% of potassium chloride and water for injections, was proposed. The ferments activity (aminotransferases, cholinesterase, aldolase, alkaline phosphatase), blood coagulating system state (the prothrombin level, plasma tolerance, her recalcification time), the mineral elements contents (potassium, sodium, calcium), the contents of protein and its fractions in blood before and after an acute blood loss compensation with the multicomponent solution, and also its influence on the animals organism in prolonged daily (during 30 days) intravenous injection were studied. The combination of components in the solution permit to store the studying indexes on level close to initial; if the loss of blood compensates in the first hours, high survival of animals is insured. Negative reactions of organism while prolonged intravenous injection of the multicomponent solution are not revealed.

[Alkaline phosphatase activity in blood serum, bile and urine of cattle and calves].

Mean values and the range of values for alkaline phosphatase activity in serum (expressed in units) were: - calves 909 (413-1445), bulls 325 (179-543) and cattle 131 (72 to 162). Differences between these groups were statistically significant. Values for bile were: - calves 416 (116-882), cattle 546 (145-1203), bulls 738 (108-1438) enzyme units; here there were no significant differences. Activity of the enzyme in urine was very low, mean values being 0.77 in calves, 0.39 in cattle and 1.69 in bulls; the differences between groups were statistically significant.

[The development of transaminase activity (SGOT and SGPT), lactate dehydrogenase and alkaline phosphatase in the blood plasma of calves up to the age of 4 months].

A trial was performed in 204 healthy calves (heifers) of the Bohemian Spotted breed in the post-natal period from birth to the age of four months. The activities of the following enzymes in blood plasma were determined: L-aspartate: 2-oxoglutarate: aminotransferase, EC.2.6.1.1. (GOT), L-alanine: oxoglutarate: aminotransferase, EC.2.6.1.2. (GPT), L-lactate: NAD oxidoreductase, EC.1.1.1.27 (LDH), and orthophosphoric acid monoester phosphohydrolase, EC.3.1.3.1. (alkaline phosphatase). The calves were divided into age categories according to the date of birth with an interval of one week. GOT activity in blood plasma increased significantly until the age of eight weeks (from the original value of 1.1708 +/- 0.2598 micronmol ml-1 to 1.8150 +/- 0.6362 micronmol ml-1, with the maximum of 2.0317 +/- 0.7777 micronmol ml-1 of plasma in the sixth week). In the subsequent period the GOT curve has not a characteristic course. While the activity of GOT increased in the first weeks after birth, the activity of GPT showed a significant drop (from the original level of 0.9000 +/- 0.3364 micronmol ml-1 to the minimum of 0.3675 +/- 0.1901 micronmol ml-1 of plasma in the seventh week); from the 10th week on the values rise so that at the end of the period of study they reach almost the same levels as in calves in the first postnatal week. The activity of LDH in blood plasma remains at almost the same level in the first five weeks after birth (between 43.4025 +/- 8.4893 micronmol ml-1 and 46.3792 +/- 14.8952 micronmol ml-1 of plasma); it was at a statistically significantly higher level only in a short period between the 7th and 10th week after birth. The highest values of alkaline phosphatase in blood plasma were recorded at the age of two or three weeks (maximum in the second week 23.9833 +/- 9.0945 micronmol ml-1 of plasma); from the fourth week on, the values of alkaline phosphatase are significantly lower until the end of the test period, ranging betweek 5.3133 +/- 1.6017 micronmol ml-1 and 7.5425 +/- 2.2437 micronmol ml-1 of plasma. Changes conditioned by postnatal development were observed in the development of all the enzymatic activities under study, the greatest changes being observed in alkaline phosphatase.

Identification of arterial and venous segments of blood vessels using alkaline phosphatase staining of ink/gelatin injected tissues.

The present study describes a method for discriminating between the arterial and venous segments of blood vessels in mouse tissues and organs using alkaline phosphatase (ALPase) staining of ink/gelatin injected tissues. Anesthetized mice were injected through the left ventricle with blue ink/gelatin, and various organs and tissues were removed from the body and fixed by immersion in 10% formalin. Sections 50-100 microm thick were incubated for ALPase in a medium containing naphthol AS-BI phosphate and fast red TR by the azocoupling method. In such specimens as the brain and skeletal muscles, ALPase activity was found in arterioles and capillaries on the arterial side, whereas it was absent in capillaries on the venous side and in venules. In the liver, only branches of the hepatic artery were positive. ALPase activity was absent in the vessels of the lung except for a positive reaction in branches of the bronchial arteries. These findings indicate that the ALPase activity is confined to the arterioles and arterial segments of the capillaries in the systemic circuit. Thus, ALPase staining of ink/gelatin injected specimens is a useful method for differentiating the arterial and venous segments of the micro-vascular bed in various organs and tissues in mice.

[Thermostable alkaline phosphatase in the blood of pregnant women].

In 310 women with normal pregnancies the exponential character of increase of serum thermostable alkaline phosphatase was assessed in the course of pregnancy; depending on the BDP of the course of pregnancy; depending on the BDP of the foetus according to the equation y (mean) = 0.0207.1.0415x (for x = 21-99 mm) and relation to the stage of pregnancy according to formula y (mean) = 0.0152.1.1176x (for x = 15-41 weeks). The menstruation age of pregnancy was in all instances verified by biometric findings of the first sonographic screening during pregnancy. The values of themostable alkaline phosphatase in serum of 15 pregnant women with hypotrophic foetuses (48 measurements) were only 45.7 +/- 9.9% (mean + s) of the mean physiological levels.

ABO blood groups, intestinal alkaline phosphatase and haptoglobin types in patients with serum hepatitis.

A series of 150 patients with serum hepatitis were examined for the incidence of the Australia antigen (HBsAg) and associations with ABO blood groups, haptoglobin types and occurrence of intestinal serum alkaline phosphatase. Among the patients studied 11.3% were positive for HBsAg. When compared to controls patients with blood group O showed a significantly increased risk for serum hepatitis (p less than 0.05), while those with group B showed a decreased risk (p less than 0.01). The presence of the intestinal fraction of alkaline phosphatase showed a negative association with serum hepatitis (p less than 0.01) and there was no significant association between alkaline phosphatase types and ABO groups among the patients. The frequency of the Hp1 gene was significantly increased (p less than 0.01) among the patients as compared to controls.

Dephosphorylation of human fibrinogen, previously phosphorylated in vitro by protein kinase C, by whole blood or intestinal alkaline phosphatase. Effects on thrombin-induced gelation of in vitro dephosphorylated human fibrinogen.

Human fibrinogen, a phosphoprotein, was either left untreated or phosphorylated by protein kinase C. Then both were dephosphorylated by calf intestinal alkaline phosphatase. The dephosphorylated fibrinogen gave an increased fibre thickness during thrombin-induced gelation. Whole blood anticoagulated by heparin, EDTA or sodium citrate, contained dephosphorylating activity against 32P-labeled fibrinogen, although there were significant differences in activity among the three anticoagulants.

Correlation of alkaline phosphatase with submucosal blood flow and morbid histology of the ischemic colon.

In an ischemic Thiry-Vella colonic segment which was constructed to facilitate the serial measurement of submucosal blood flow by radioxenon clearance, we found that, after producing ischemia, the submucosal xenon clearance fell from 68+/-3 to 21+/-4 percent. In the succeeding two hours, a further fall in xenon clearance to 16+/-3 per cent occurred as a result of vasoconstriction in the peripheral mesenteric bed. From 24 hours onward, the submucosal flow increased. However, throughout the 96 hours, no healing of the ischemic necrosis was seen in the mucosa, and submucosa. Serial alkaline phosphatase levels showed a great rise from 24 hours onward, and began to fall as submucosal blood flow increased. This rise in the alkaline phosphatase value appears a useful marker of the degree of ischemia of the intestinal wall.

Inhibition of tumor cell line alkaline phosphatase by human peripheral blood monocytes. Correlation with growth inhibitory activity.

Alkaline phosphatase (AP) activity in intact Ag8 cells was measured by a colorimetric assay in which spectrophotometer readings were obtained directly through wells of microtiter plates using a Multiskan ELISA reader. The wide difference between enzymatic activity observed in cells of this cell line and human peripheral blood monocytes permitted an assessment of inhibition of cell line growth in the presence of plastic-adherent peripheral blood cells. Comparison at 24 h of incubation of enzymatic measurements with those obtained using 3H-thymidine uptake showed comparable results, although values obtained with the enzyme were generally lower. Similarly, direct comparison of results obtained by the enzyme-based method showed good correlation with the number of viable tumor cells remaining in culture. Inhibition of AP activity was observed earlier than inhibition of 3H-thymidine incorporation, reaching its peak after 10 h of incubation. This technique has potential application for other enzyme systems and provides a new tool for analyzing mechanisms whereby effector cells are able to control proliferation of transformed cells.
