[Effect of the antineoplastic substance brotheophine on the insertion of precursors into nucleic acids and proteins from Plisse lymphosarcoma].

The effect of antitumor medicine Brotheophine to insertion of the marked precursors into DNA (14[C]-timidine), RNA (3[H]-orotic acid) and proteins (14[C]-leucine) of tumor cells (Pliss lymphosarcoma) as the indices of biosynthetic processes was investigated. It has been shown that Brotheophine acts as an antimethabolite of purine exchange and inhibits the vertical genetic information transfer in tumor cell (DNA-RNA-protein). Namely, a significant inhibition of 14[C]-timide insertion to DNA has been observed. less distinct is the inhibition of 3[H]-orotic acid to RNA und 14[C]-leucine to proteins. The above revealed allows to assume that during Brotheophine treatment certain changes in DNA biosynthesis take place leading to synthesis of slightly transformed RNA with subsequent impairment of proteins synthesis.

[Active immunotherapy of acute leukemia and leukemic lymphosarcoma. Results of 10 years. Study of 200 cases].

The authors report a ten year study of active immunotherapy using BCG and irradiated allogeneic leukaemic cells in 200 patients. In acute lymphatic leukaemia, 57 out of 168 patients treated in this way remained in primary remission for 18 months to 10 years after active immunotherapy was begun, the relapse rate became low after 18 months and nil after 36 months. The results varied according to prognostic factors: the cytological type, active immunotherapy being above all effective in small cell (microlymphoblastic and prolymphocytic) types with a hope of cure in 50 to 60 p.cent of cases; malignant cellular volume; meningeal deposits. In microlymphoblastic forms the possibility of survival at the 5th year is greater than 90 p.cent. After relapse during active immunotherapy sensitivity to chemotherapy does not seem to be diminished. Trials of active immunotherapy in acute myeloid leukaemia are worthy of further pursuit. The results of active immunotherapy in leukaemic lymphosarcoma show that immunotherapy may be effective in preventing local recurrence, both of tumour as well as in the marrow. Four patients are in apparently complete remission for more than four years. On the basis of these results, trials of active immunotherapy for "residual disease" should be undertaken in the field of cancerology, going beyond the realm of leukaemias.

[Cyclophosphamide-induced apoptosis of murine lymphosarcoma cells in vivo].

The antitumor action of combination chemotherapy (cyclophosphamide + adriamycin + vincristin + prednisolone) for transplantable nitrosomethylurea-induced lymphosarcoma was studied in male CBA mice. Single injections of the mixture were followed by complete regression of tumors of up to 2 cm in diameter. The effect was shown to be caused by cyclophosphamide (CP) alone, by inducing apoptosis. The other components failed to potentiate the CD effect. Being useless, they are likely to cause harm by contributing to the overall toxic effect of therapy. The nature and duration of CP-induced remission appeared dose-dependent: on day 50 of the administration of 50, 100 and 150 mg/kg body, tumors were detected in 100, 55 and 0% of the animals, respectively. Such means of apoptosis induction as glycocorticoid treatment and ionizing radiation did not cause complete regression.

[Differential diagnostic and prognostic value of serum cytokines (soluble CD30, soluble interleukin-2 receptor, interleukin-10 and interleukin-6) in Hodgkin's disease and lymphosarcomas].

AIM: Determination of the importance of serum cytokines (sCD30, sIL-2R, IL-10, IL-6) for diagnosis, response to chemotherapy and remission in patients with lymphogranulomatosis and lymphosarcoma. MATERIAL AND METHODS: Cytokine concentrations were measured in 87 samples of serum (plasma) from 54 patients by ELISA. Diagnosis of Hodgkin's disease (HD) and non-Hodgkin's lymphoma (NHL) was made histologically in 24 and 30 patients, respectively. RESULTS: The threshold concentrations of sCD30 (and less specific sIL-2R) for HD and NHL patients allowed to estimate sensitivity to chemotherapy after the second course. The threshold concentration of IL-10 can distinguish HD from NHL. Changes in IL-6 concentrations were nonspecific. CONCLUSION: On the basis of the threshold concentrations of sCD30 and IL-10 we offer the scheme of lymphoma diagnosis and prediction of the disease sensitivity to chemotherapy which reduce the duration of lymphoma restaging.

Lymphosarcoma in an alpaca.

A 16-month-old, female alpaca presented with a 24-hour history of anorexia and depression. On necropsy, it was found that the liver was grossly enlarged and the cortices of both kidneys contained multifocal lesions. Histologic examination of these lesions and other tissues revealed infiltration with lymphocytic cells, a finding consistent with lymphosarcoma.

[Effect of olipifate on the wound healing proces and relapse after resection of Pliss lymphosarcoma].

The investigation was concerned with the influence of preliminary injections of Olipiphat, irrigation of operative wound and combination of both procedures on healing and relapse processes, following resection of Pliss lymphosarcoma at different stages after transplantation into rats. The physical condition of the animals after tumor resection on days 12, 10 or 7 of tumor growth was better than in controls, as a result of irrigation of the operative wound with Olipiphat or in combination with preliminary injections of the drug: they came out from anesthesia quicker, tidied themselves up and moved about the cage. Irrigation of the wound with Olipiphat or in combination with preliminary injections followed by longer survival after surgery performed at all stages of tumor growth. Moreover, one animal out of 16 in each of the 4 Olipiphat-treated groups survived 60 days recurrence-free. The drug proved more effective in stimulating the healing of larger wounds but contributed to healing by first intention in all cases.

Total body irradiation in lymphosarcoma.

Total body irradiation has shown encouraging result in the treatment of advanced lymphosarcomas. The response rate and remission rates are high. 66 percent (4/6) went in unmaintained complete remission. Good response was also observed in patients who had previous chemotherapy.

Lymphosarcoma of the right atrium. Angiographic and hemodynamic documentation of response to chemotherapy.

A patient had metastatic lymphosarcoma involving the right atrium and tricuspid valve in which functional tricuspid valve obstruction was relieved by chemotherapy. Patients with disseminated or mediastinal lymphoma in whom cardiopulmonary symptoms occur should be considered for catheterization of the right side of the heart and angiography. These procedures may be useful for determining the presence, location, and nature of cardiac disease and may aid in deciding the proper course of therapy.

[Value of determination of mononuclear blood cell proliferation in vitro in the diagnosis of lymphosarcoma].

The authors describe mitotic activity in non-stimulated cultures of mononuclear cells obtained from the peripheral blood of patients with lymphosarcoma. The investigations were carried out on 3 patients in stage 3rd and 4th of this disease and 5 patients in the stage of sarcomatous-cell leukaemia. During a period of 24-48 hours after setting up of culture a rise was observed in the number of blast cells and appearance of up to 10% of mitotic figures. This observation may be useful for differentiation of sarcomatous-cell leukaemia from chronic lymphocytic leukaemia in which cells obtained from the peripheral blood and cultured in vitro without addition of mitogen do not demonstrate mitotic activity.

[Apoptosis-inducing and anti-tumor effect of cyclophosphamide, cisplatin and adriamycin used separately or combined in murine lymphosarcoma LS].

The investigation deals with antitumor effect of cyclophosphamide, platin and adriamycin on transplantable mouse lymphosarcoma LS. It was found that the effect of the most potent drug cyclophosphamide is mediated mostly by apoptosis, while those of platin and adriamycin--by apoptosis and necrosis of tumor cells. When a combination of two or three drugs was used the necrotic effect increased proportionally, while the apoptotic one decreased. However, the overall antitumor effect did not exceed that of the one drug which induced apoptosis most effectively.

Inhibition of DNA synthesis in mouse lymphosarcoma LS/BL cells by fluorocitrate.

Fluorocitrate is a specific inhibitor of aconitase activity (EC 4.2.1.3). It causes an inhibition of citric acid cycle reactions and consequently reduces the oxygen consumption, as well as the total volume of oxidative phosphorylations. Experiments using 3H-thymidine incorporation into mouse lymphosarcoma LS/BL cells proved that the in vitro blocking of citric acid cycle by fluorocitrate (3.6 and 7.2 mM) was accompanied by a decrease in DNA synthesis by 40 to 60 percent, as compared to control cells. A similar inhibitory effect upon DNA synthesis was also found in cells cultured in the abdominal cavity of host mice injected intraperitoneally with 5 mg./kg. of sodium fluoroacetate. Following the injection, fluorocitrate is formed in vivo by way of Peters' lethal synthesis.

Chronic intussusception associated with ileocecal lymphosarcoma.

A 1011/12-year-old boy with chronic ileocolic intussusception associated with ileocecal lymphosarcoma was decribed. The patient had abdominal pain of 3 months' duration, vomiting, and a firm mass with smooth surface in the right lower abdomen. Barium enema showed intussusception at the distal part of the ascending colon, which was irreducible by hydrostatic pressure. Barium by mouth revealed markedly distended small intestines, which were displaced to the left abdomen. The tumor was inoperable, and the boy died.

Chemotherapy for canine lymphosarcoma.

Twenty dogs with naturally occurring lymphosarcoma were histologically diagnosed, clinically staged, and treated with combinations of drugs, including corticosteroids, alkylating agents, plant alkaloids and antimetabolities. Clinical response was categorized as complete, partial, or unsatisfactory. Of the 20 dogs, 17 responded to the chemotherapy completely or partially, with a mean objective remission duration of 104.8 days. The mean survival time for 19 dogs was 211.5 days, whereas the mean survival time for 16 dogs which responded completely or partially was 233.7 days.

Effect of continuous irradiation upon some biological properties of murine lymphosarcoma LS/BL.

Mouse lymphosarcoma LS/BL cells passaged in syngeneic hosts were continuously irradiated with 60Co gamma rays at an exposure of 1R/hour. Long-term irradiation did not substantially alter the proliferative rate of leukaemic cells. Radiation exposure in the duration of 60 and 70 weeks led to an increase in the mean survival time of the hosts and in the generation time of the cells. When compared to the changes observed in the hosts of unirradiated leukaemic cells, the decrease in erythrocyte counts was greater during the first few days after inoculation of irradiated tumour cells, whereas the increase in leucocyte counts was slower.

Light and electron microscopy of mouse lymphosarcoma LS/BL cells during the first hour after irradiation with x-rays.

Mouse lymphosarcoma LS/BL cells growing in tissue culture were irradiated with 100, 316 and 1000 R respectively and fixed 5, 15 or 60 minutes later. Semithin sections were studied under a light and ultrathin under an electron microscopes. Under the light microscope, differences between the morphology of individual nuclei were found in all groups, including controls; the rate of their occurrence was without any relation to the irradiation. They may reflect the differences in the state of LS/BL cells which were not completely adapted to cultivation in small glass tubes. On the other hand, the frequency of some ultrastructural changes seen under the electron microscope showed a relation to the irradiation. They were: undulation of the outer nuclear membrane and dilatation of the space between the outer and inner nuclear membranes, slots in karyosomes adjacent to the surface of the nuclei, enlargement of cisternae and vesicles of endoplasmic reticulum and Golgi apparatus. The importance of a careful examination of control groups in such experiments is stressed.

[Intracytoplasmic tubulareticular structures. Presence in human lymphosarcoma].

Intracytoplasmic tubuloreticular structures were found in the tumour cells of a human lymphosarcoma. Such structures have been described in a wide variety of conditions in man and in animals, as well as in cell cultures of various origin, but rarely in lymphoid tumours. The different hypotheses concerning their nature are discussed.

[Effect of amphotericin B and its combination with 5-fluorouracil on the development of transplantable rat lymphosarcoma].

The studies were carried out on rats with subcutaneously transplanted metastizing Pliss lymphosarcoma. The animals were treated with amphotericin B administered intravenously in complex with sodium desoxycholate in doses of 336 and 20 Units/kg for 7 days beginning from 24th hour after the tumor transplantation. The treatment resulted in a statistically reliable inhibition of the tumor metastasizing as compared to the control animals. When the animals with the tumors were subjected to combined therapy with 5-fluoruracyl and amphotericin B, the antitumor and antimetastatic effect of 5-fluoruracyl significantly increased. Determination of amphotericin B binding with the organ homogenates showed that the antibiotic bound with the homogenates or the organs of the rats with the tumors to a greater extent than with those of the intact animals. The levels of the antibiotics in the tumor metastases were higher than those in the primary tumors.
