[Comparative effect of psychotropic agents on orienting-motor responses and cerebral Na,K-ATPase activity in vivo].

Levomepromazine, chlorpromazine (10 and 30 mg/kg), etaperaxine, haloperidol (3 and 10 mg/kg) inhibited the exploratory-motor reactions of rats and the brain Na, K-ATPase activity an hour after their administration. The effects of tranquilizers as well as of antidepressants on the exploratory reactions and on the enzyme activity were not found to stand in a clearcut relation to each other. The stimulating effect of amphethamine (3 mg/kg) was accompanied by activation and suppressive action (10 mg/kg)--by inhibition of the enzyme. It is suggested that the inhibition of the brain Na, K-ATPase activity by psychotropic drugs may play a role in the mechanism of their sedative action.

[A pharmacological analysis of the cholinergic system in animals with alloxan diabetes].

In the animal experiment the sensitivity to acetylcholine was shown to increase in alloxan-induced hypoinsulinemia. The blockade of M-cholinoreactive structure produced by administration of amizil caused a deterioration of the condition of the animals with alloxan-induced diabetes. The increased sensitivity to acetylcholine was accompanied by changes in the activity of acetylcholine esterase in the hypothalamus nuclei.

[An evaluation of methods for isolating antiparkinson preparations from cadaveric material].

Methods of amedine, amyzyl, and tropacine isolation from cadaveric material, blood and urine were developed. They ensure higher output of the given substances as compared with conventional methods of isolation of toxicologically significant substances. An extraction-photometric method was developed for estimation of amedine, amyzyl, and tropacine in the cadaveric material.

[Experience with amysil treatment of patients with vascular parkinsonism].

The results of treatment of 45 patients with vascular parkinsonism are reported. To the majority of the patients, daily amysil administration (1-2 mg 2-4 times a day) was prescribed. Most pronounced was the drug effect in patients with combined syndromes in which predominated bradikynesia (79.3%), rigidity (75%), and to a lesser extent, tremor (66.6%). In some patients, the sedative effect was evident. In most of the patients, the positive EMG changes were noticeable, especially in those with stage I of the disease.

Pharmacological study of cholinergic mechanisms of compensatory ovarian hypertrophy in rats.

Treatment of hemicastrated adult female rats with nicotine increased the compensatory ovarian hypertrophy (COH) while central n-cholinolytic IEM-506 decreased COH and prevented the estrogen-induced suppression of COH. Administration of L-DOPA abolished the effect of IEM-506, and disulfiram blocked gonadotropic action of nicotine. Elevation of the dose of arecoline decreased COH and the sensitivity of hypothalamo-gonadotropic complex to estrogen suppression. Treatment of rats with L-DOPA and disulfiram abolished the late effect of arecoline. Central m-cholinolytic metamizyl decreased COH, potentiated the effect of estrogen and prevented the gonadotropic effect of L-DOPA. The regulatory role of m-cholinergic systems in noradrenaline mediation of gonadotropic function and n-cholinergic system in dopamine ones are suggested.

[Use of acetylcholinesterase reactivators and central cholinolytics in the treatment of experimental tetanus poisoning].

Experiments were conducted on rabbits with tetanus intoxication induced by the intravenous injection of a lethal dose of the toxin; a study was made of the therapeutic efficacy of the acetylcholinesterase reactivators--dipyroxim and isonitrosine, and also of the central cholinolytics--amizyl and diphacyl. In dose of 25 mg/kg dipyroxim produced no therapeutic effect, and in doses of 30--40 mg/kg caused the animal death. Amizyl and diphacil in a dose of 3--4 mg/kg caused elimination of tonic convulsions for 1 1/2--2 hours. As to isonitrosin--it produced the same effect for 4--5 hours. In case of combined administration of reactivators and cholinolytics convulsions were eliminated for 4--5 hours.

[The GABA-ergic system and brain edema].

It has been shown in rats with experimental toxic and traumatic edemas that picrotoxin (1 mg/kg) removes the antiedematous action of diazepam, phenazepam, phenibut and amizyl and reduces the action of phentolamine. When the dose of picrotoxin is minimized to 0.5 mg/kg such an effect is not observed. Prolonged daily administration of picrotoxin in a dose of 1 mg/kg results in the development of brain edema. It is recommended that GABA-positive drugs be included into a complex of treatment measures for edema.

[Effect of neuroleptic, adreno- and cholinolytic agents on cerebral cortical function in edema].

Experiments on rats with experimental brain edema under water intoxication were made to study the effects of the neuroleptics aminazine and propazine, the central M-cholinoblocker amizyl and the alpha-adrenoblocker phentolamine on the time-course of the recovery of cortical electric activity after passing of the wave of Leao's spreading depression (SD). The drugs under study were demonstrated to have antiedematous properties under water intoxication and to make to a considerable degree for disorders in the processes of the recovery of cortical electrogenesis after SD. It is assumed that the action of the drugs on the electrophysiological parameters under study is associated with their effects on metabolic processes and with the presence of antiedematous properties.

[Effect of N- and M-cholinergic-potentiating and cholinergic-blocking agents on the epileptogenesis of a penicillin focus in the dorsal hippocampus].

Chronic experiments on rabbits with penicillin epilepsy have shown that intravenous injections of the ACHE inhibitor galanthamine (1 mg/kg), and N-cholinergic-blockers, ganglerone (3 mg/kg) and eterofen (8 mg/kg), decreased or suppressed completely epileptogenesis manifestations. Combination of galanthamine with one of the N-cholinergic-blockers markedly increased their anticonvulsive actions. On the contrary, combination of galanthamine with the M-cholinergic-blocker metamizil (0.5 mg/kg) enhanced epileptogenesis activity. It is postulated that both M- and N-cholinergic mechanisms take part in the genesis of penicillin epilepsy. The use of N-cholinergic-blockers and their combinations with M-cholinomimetics as anticonvulsants seems expedient.

Influence of beta adrenergic blockers and their combinations with anticholinergic drugs on epileptiform activity in rabbit hippocampus.

The influence of alpha- and beta-receptor antagonists combined with drugs which act as antagonists at muscarinic and nicotinic receptor sites on epileptic focus produced by topic penicillin application was investigated. The beta-receptor blocking agent caused a faster and more intense activity of epileptogenic focus in comparison with activity in control animals, but the number of seizures decreased relatively for 25%. N-receptor antagonists diminished the facilitating effect of the beta-receptor antagonist on focal activity. Combination of m- and beta-receptor antagonists did not influence the spike activity of the focus. Treatment with alpha-receptor antagonists enhanced spikes and number of seizures only insignificantly. The combination of alpha- and n-receptor blocking substances does not influence the development of the epileptic process. The alpha-receptor antagonist together with the m-receptor blocking drug induces a facilitation of spike activity.

[Effect of M and N-cholinoblockers on the excitability of the dorsal hippocampus in acute alcoholic intoxication].

It has been established in chronic experiments on rabbits that acute alcohol intoxication increased the after-discharge thresholds in response to electrical stimulation of dorsal hippocampus. It has been shown that neurotropic agents selectively blocking M- or N-cholinoceptors exerted various effects. M-cholino-blocker methamizol (1 mg/kg, i. p.) decreased the excitability of dorsal hippocampus, potentiated EEG and behavioural effects of alcohol intoxication. N-cholinoblocker etherophen (IEM-506, 20 mg/kg, i. p.), on the contrary, increased the excitability of dorsal hippocampus and reduced behavioural effects of alcohol administration.

[Effect of amizil on the carbohydrase activity in intact animals and animals subjected to stress exposure].

Effect of a central cholinolytic drug amizyl on the activity of some digestive enzymes involved in carbohydrate hydrolysis was studied in experiments on control rats and animals exposed to immobilization stress. The experiments showed that amizyl administration modifies the activity of carbohydrases that could affect disintegration and assimilation of carbohydrases. Preliminary administration of the drug was shown to prevent substantially the changes in the enzyme activity occurring during stress situation.

Adrenocorticotropic Hormone↗

[Effect of amizil and the ACTH4-10 fragment on the ability of mice to extrapolate the direction of movement].

Mice possessing the ability to extrapolate the direction of movement (100% of correct choices) were injected i.p. with different doses of m-cholinolytic amizil 2 hours before experiment. Doses of 2-5 mg/kg reduced the percentage of correct choices, the adequate solving strategy being replaced by stereotyped unidirectional reactions or stereotyped alternating responses. Doses of 8-12 mg/kg induced "refusals" to solve extrapolation problem. When amizil treated mice were intraventricularly injected with 1 mg/kg of ACTH4-10, their extrapolation ability was restored. This compensatory action of peptide could be mediated by its influence on cholinergic as well as on other neurotransmitter systems.

[Effect of amizil on the activity of enzymes participating in protein hydrolysis in intact animals and animals subjected to stress exposure].

The effect of amizil on the activity of digestive enzymes involved in the initial and final stages of protein hydrolysis was studied on intact and stress exposed rats. The experiments showed that amizil administration produced varying changes in the activity of proteases and dipeptidase. At the same time a preliminary administration of the drug was found to prevent the changes in the enzyme activity observed during stress situations.

[Shifts in spatial synchronization of rabbit cortical potentials evoked by applying acetylcholine and amizil to the visual area].

Acetylcholine solution application to the visual cortex of walking unrestrained rabbit led approximately in 5 minutes to a certain increase of the spatial synchronization of the cortical potentials at the small conditions, the the cortex, 24 EEG channels being used. Under the same conditions, the application of muscarinic antagonist amizil had an opposite more distinct effect. Endogenous cholinergic input increasing the spatial synchronization is supposed to play an important role in the formation of the spatiotemporal cortical potentials' organization necessary for the occurrence of complex behavioural acts.
