Immortalized and tumorigenic adult human prostatic epithelial cell lines: characteristics and applications. Part I. Cell markers and immortalized nontumorigenic cell lines.

Several immortalized and malignant adult human prostatic epithelial cell lines have recently been developed. The three most widely used carcinoma cell lines, DU-145, PC-3, and LNCaP, developed between 1977 and 1980, have greatly contributed to our present understanding of prostate cancer. Before a cell line can be accepted as having prostatic epithelial origin, some basic characteristics must be established. Expression of specific cytokeratins, but absence of desmin and factor VIII, should be first determined to establish epithelial origin. Responsiveness to androgens and expression of androgen receptor and prostate specific antigen should be examined under stringent culture conditions to establish prostatic epithelial origin. Response to growth factors and expression of their receptors facilitates further characterization of cell behavior. Cell lines immortalized by human papillomaviruses (HPVs) are of special interest because HPVs are involved in a variety of anogenital cancers and may also play a role in prostate carcinogenesis. Malignant transformation of HPV-18 immortalized cells with the ras oncogene provides cell systems for investigating the multistep process of carcinogenesis. Each cell line has some unique characteristics, whether it arose directly from a carcinoma or resulted from immortalization with simian virus 40 (SV40) or HPV or was transformed in vitro by oncogenes. Comparisons of these characteristics should facilitate elucidation of the mechanisms involved in initiation, promotion, and progression of prostate cancer. These cell lines will further serve as useful models for investigating tumor progression, invasion, metastasis, new therapeutic strategies, drug resistance, and its reversal and chemoprevention. This review will be published in three parts and will summarize cell markers necessary for characterization, as well as the characteristics and some applications of the immortalized as well as malignant adult human prostatic epithelial cell lines. Part 1 deals with cell markers and the immortalized, nontumorigenic cell lines.

In vitro study of the comparative behaviour of a human acute lymphoblastic leukemia cell line and a reference normal cell line towards the effects of a mitogenic lectin.

An in vitro study of the behaviour of a human acute lymphoblastoid leukemia cell line (REH) towards the action of a mitogenic lectin of Robinia pseudoacacia was carried out. The results were compared with those a reference cell line (LHN13) established from normal human lymphocytes. In both cell lines, the lectin induces agglutination (measured by counting the number of aggregates as well as the number of cells in each aggregate) and decrease of growth (measured by counting the number of cells and the incorporation of tritiated thymidine into TCA-precipitable material per 10(6) cells). The agglutination and the decrease of growth are produced at the doses of 0.5 and 1 microgram/ml of culture medium and after 4 h of exposure of cells to the lectin, respectively. These effects increase progressively with higher doses of lectin and continues throughout the culture. However, the REH line is less sensitive than the LHN13 line to the effects of lectin. Both agglutination and growth decrease of REH as well as LHN13 cell lines by the lectin are reversible; this is confirmed by the fact that the monospecific anti-Robinia lectin serum suppresses these effects.

Patterns of chromosome segregation in Chinese hamster x mouse cell hybrids between permanent cell lines and thymus cells.

Hybrids between a tumorigenic Chinese hamster cell line (DC3F-aza) and normal mouse thymus cells very rapidly lost most of their mouse chromosomes, whereas hybrids between tumorigenic mouse cell lines (either Cl.1D of L cell line origin, or PCC4-aza 1 teratocarcinoma cells) and normal Chinese hamster thymus cells lost most of their hamster chromosomes. From three such fusion experiments, 20 cell lines were developed which all followed the same evolution, namely, the elimination of the majority of the chromosomes contributed by the normal thymus cell. In some hybrids, the elimination process resulted in the total absence of intact chromosomes contributed by the thymus cell parent. Such hybrids were distinguished from revertant parental cells growing in the selective medium by the presence of at least one enzyme in their cell extracts which displayed the electrophoretic mobility of the enzyme of the thymus cell parent. These observations, together with data from other reports, suggest that, as a rule, interspecific cell hybrids which develop upon fusion between normal diploid cells and tumorigenic cell lines maintain the chromosomes of the latter and eliminate preferentially many or most of the chromosomes contributed by the normal cell parents, independent of the respective species of the parental cells.

Isolation of melanized cell lines with stable phenotypes from a goldfish erythrophoroma cell line and cryopreservation of these cells by the use of autologous serum.

Cell lines with stable melanized phenotypes were isolated from a goldfish erythrophoroma cell line. These cell lines include several interesting phenotypes: a) reversible dedifferentiation and redifferentiation in response to withdrawal and addition of fish serum; b) irreversible dedifferentiation in response to withdrawal of fish serum; c) independence on fish serum for melanization; d) dependence on fish serum for growth; and e) pigment aggregation in response to epinephrine. We also report that cryopreservation of all the melanized cell lines, but not any of the unmelanized cell lines, requires the presence of fish serum. This raises the possibility that there may be advantages to use autologous serum for the cryopreservation of sensitive cell lines.

Transient ectopic expression of PTEN in thyroid cancer cell lines induces cell cycle arrest and cell type-dependent cell death.

The tumour suppressor gene PTEN/MMAC1/TEP1 has been implicated in a variety of human cancers and several inherited hamartoma tumour syndromes, including Cowden syndrome, which has a high risk of breast and thyroid cancer. We have previously reported that overexpression of PTEN in MCF-7 breast cancer cells induces cell cycle arrest and apoptosis. In this study, we analysed PTEN status at both the structural and expression levels and explored PTEN's growth-suppressive effects on thyroid. We found that 1 of 10 thyroid cancer lines [follicular thyroid carcinoma FTC-133] had hemizygous deletion and a splice variant IVS4--19G-->A in the remaining allele. Four lines, including FTC-133, express PTEN mRNA at low levels. In general, PTEN protein levels correlated with mRNA levels, except for NPA87, which has low levels of transcript and relatively high levels of PTEN protein. Transient expression of PTEN in seven thyroid cancer cell lines resulted in G(1) arrest in two well differentiated papillary thyroid cancer lines (PTCs) and both G(1) arrest and cell death in the remaining five lines, including three FTCs, one poorly differentiated PTC and one undifferentiated thyroid cancer. The level of phosphorylated Akt was inversely correlated with the endogenous level of PTEN protein and overexpression of PTEN-blocked Akt phosphorylation in all cells analysed. Our results suggest that downregulation of PTEN expression at the mRNA level plays a role in PTEN inactivation in thyroid cancer and PTEN exerts its tumour-suppressive effect on thyroid cancer through the inhibition of cell cycle progression alone or both cell cycle progression and cell death.

Glycosyltransferase activities in leukemic cells from patients and human leukemic cell lines.

The cell membrane fraction from c-ALL, B-ALL, Ph' + ALL, B-CLL, T-CLL, AML, blastic-CML, normal leukocytes, PHA-stimulated lymphocytes and several T, B and myeloid human leukemic cell lines has been used in different cell types to demonstrate different patterns of glycosyltransferase activity. Both B- and T-CLL cell membranes have low fucosyltransferase B and A activity compared to acute leukemias; while sialyltransferase activity is higher in B- than in T-CLL. AML cell membranes and ML-1 human myeloblast cell line membranes have exceptionally high fucosyltransferase A activity compared to all other leukemic cells or cell lines. Human leukemic B cell lines expressed cell membrane sialyltransferase, fucosyltransferase B and probably fucosyltransferase A activity several times higher than T cell lines. Human myeloid cell lines ML-1 and HL-60 express 5- to 20-fold higher galactosyltransferase activity than human leukemic T and B cell lines. Both sialyltransferase and galactosyltransferase activity were higher in all leukemic cells than in normal leukocytes and PHA-stimulated normal lymphocytes. This is the first study carried out on glycosyltransferases using cells obtained from leukemic patients characterized immunologically. These results indicate that all glycosyltransferase activity, with the exception of fucosyltransferase activity in CLL, were higher in leukemic cells than in normal cells. Moreover, large differences in these enzymes, e.g. very high galactosyltransferase activity in myeloid cell lines compared to B and T cell lines, of fucosyltransferase A in AML and myeloblast cell lines compared to all other cells, and of sialyltransferase in B-CLL or B cell lines compared to T-CLL or T cell lines, could be useful in characterizing certain leukemias and hematopoietic cell lines.

Lanreotide-induced modulation of 5-fluorouracil or mitomycin C cytotoxicity in human colon cancer cell lines: a preclinical study.

The effect on growth of the long-acting somatostatin analogue lanreotide (LAN), alone or in combination with 5-fluorouracil (5-FU) and mitomycin C (MIT), was investigated in three human colon cancer lines. Cell survival inhibition induced by LAN alone, as evaluated by sulforhodamine B assay, ranged from 20% to 40% as a function of cell line and concentration. The IC50, the concentration inhibiting cell survival by 50%, was never reached. The antiproliferative effect produced by a 48 h exposure to 5-FU or MIT was synergistically enhanced in all cell lines by a subsequent 48 h exposure to LAN. The synergistic interaction was not related to specific cell cycle perturbations or to the somatostatin receptor 2 (sst2) mRNA abundance. In conclusion, our study seems to indicate that LAN is a potentially useful modulating agent for enhancing 5-FU and MIT activity in colorectal cancer patients.

Arsenic trioxide (As2O3) sensitivity of carcinoma cell lines and cancer cells from patients with carcinomatosis peritonei.

The effectiveness of As2O3 treatment was studied in 3 carcinoma cell lines, LoVo, OVCAR-3, and PA-1, and in cancer cells obtained from ascites fluids of 8 patients with carcinomatosis peritonei. LoVo, OVCAR-3, and PA-1 cell lines, and cancer cells from the patients were cultured in As2O3 gradient media; As2O3 sensitivity was evaluated by trypan blue dye exclusion and by morphologic examination after Wright staining. PA-1 was the most sensitive cell line to As2O3; OVCAR-3 and LoVo were resistant to As2O3. Cancer cells from 2 of 8 patients were sensitive to As2O3. The in vivo tumoricidal effect of As2O3 (100 microg/day, i.p.) was studied in 30 BALB/c nude mice following i.p. implantation of PA-1 tumor cells. The 17 As2O3-injected mice died of extensive intratumoral hemorrhage, necrosis, and hemorrhagic ascites within 48 hr after initial treatment. In 10 As2O3-untreated tumor-bearing control mice, only focal intratumoral hemorrhage and necrosis were noted. In summary, solid tumor cell lines and cancer cells from patients showed various As2O3 sensitivities in vitro, and As2O3 had a marked tumoricidal effect on PA-1 cells in vivo. These results suggest that As2O3 treatment might possibly be beneficial in patients with carcinomatosis peritonei who are resistant to conventional therapy and whose tumors show in vitro sensitivity to As2O3. However, to minimize the life-threatening tumor lysis effect, it would be better to administer As2O3 after removal of the peritoneal tumor masses.

Sister chromatid exchange in lymphoma line and lymphoblastoid cell lines before and after heterotransplantation into nude mice.

The sister chromatid exchange (SCE) rates of cell lines derived from patients with different malignant and premalignant disease were studied before and after intracerebral inoculation into nude mice and were compared to the SCE frequencies of cell lines from healthy controls. Lymphoma lines characterized by chromosomal markers as well as chromosomally normal lymphoblastoid cell lines were used. The SCE frequencies of nude mouse tumor-derived cell lines did not show a homogeneous pattern. They were either as high as, or higher or lower than, the SCE levels of the original cell line. But all cell lines, karyotypically abnormal as well as normal, originating from patients with malignant and premalignant diseases, showed significantly higher SCE rates than the control lines from healthy donors.

[Relationship between ataxia telangiectasia mutant(ATM) expression of HL-60 and SiHA cell lines and their cell cycle arrest after 60Co radiation].

BACKGROUND & OBJECTIVE: Ataxia telangiectasia is caused by ataxia telangiectasia mutant(ATM) gene and it is characterized by hypersensitivity to the radiation. So the ATM expression should be related to the radiation sensitivity. This study was designed to investigate the relationship between ATM expression in two kinds of tumor cell lines and their cell cycle arrest after irradiation of ionization radiation to explore their self-protection function. METHODS: ATM mRNA and protein expression of HL-60 and SiHA cell lines were measured by semi-quantitive reverse transcription polymerase chain reaction (RT-PCR) and flow cytometry. The cells were irradiated at the dose of 6, 10, and 15 Gy by (60)Co and the change of apoptosis and cell cycle arrest phenomenon at the time of 6, 12, 24, 48, and 60 hours after the radiation were observed. RESULTS: The ATM mean protein fluorescent intensity of HL-60 and SiHA cells were 14.11+/-2.38 and 27.74+/-1.16,respectively. The ATM protein abundance in SiHA was two times more than that in HL-60,and consistent with this, the ATM RNA relative expression of HL-60 and SiHA cells were 0.09 and 0.80, respectively. The ATM transcript levels in SiHA are 9 times more than that in HL-60. The G(2)/M phase arrest after the radiation was observed in both cell lines, whereas SiHA exhibits a much strong cell cycle arrest than HL-60. CONCLUSION: The cell cycle arrest of HL-60 and SiHA cell lines to the irradiation corresponds to their ATM expression levels. The lower ATM expression, the weaker response for cell cycle arrest induced by radiation.

Immunoglobulin superfamily receptor translocation associated 2 protein on lymphoma cell lines and hairy cell leukemia cells detected by novel monoclonal antibodies.

PURPOSE: The immunoglobulin superfamily receptor translocation associated 2 (IRTA2) gene encodes a cell surface receptor homologous to the family of Fc receptors. Because of the restricted expression of mRNA in B cell-lineage cells, IRTA2 is a new potential target for the immunotherapy of B cell malignancies. To study the expression of the IRTA2 gene product, we produced monoclonal antibodies (MAbs) specific to IRTA2. EXPERIMENTAL DESIGN: A mouse used for cell fusion was DNA-immunized with an expression plasmid encoding the IRTA2 cDNA. The reactivity of MAbs secreted from the hybridomas were characterized with recombinant proteins of IRTA family members in an enzyme immunoassay and a fluorescence-activated cell sorter (FACS). Nineteen human lymphoma cell lines and blood cells from five patients with hairy cell leukemia (HCL) were analyzed with IRTA2 expression using FACS. RESULTS: Three MAbs (F25, F56, and F119) were selected based on their specific reactivity with recombinant IRTA2 and lack of cross-reactivity with other IRTA family members. In a FACS analysis, MAbs F56 and F119 detected IRTA2 expression in six of seven B cell non-Hodgkin's lymphoma and one of six Burkitt's lymphoma cell lines. Reverse transcriptase-PCR experiments and Western blotting using MAb F25 confirmed the expression profile. We also found that HCL cells from five patients expressed IRTA2. CONCLUSIONS: Our results provide the first evidence that IRTA2 is expressed on the surface of human lymphoma cell lines and HCL cells. IRTA2 could be useful as a new target for immunotherapy.

Spontaneous establishment of a novel Japanese macaque cell line with epithelial cell phenotypes.

A novel macaque cell line (J3K) with epithelial phenotypes was spontaneously established from a kidney specimen of a Japanese macaque (Macaca fuscata). Its population doubling level reached more than 500, and it was regarded as an established permanent cell line. J3K cells have transformed cell phenotypes such as loss of contact inhibition and anchorage-independent cell growth. Unlike other monkey adherent cell lines, J3K had no SV40 large T antigen. After establishment, cells have constantly expressed telomerase activity, whereas telomere length has been maintained. No mutations in the coding regions of the p53 complementary deoxyribonucleic acid were detected in the late-passaged cells. J3K, a novel transformed epithelial cell line, will be useful material for the comparative study of human and other primate cellular aging as well as cancer cell biology.

S100 protein expression in human melanoma cells: comparison of levels of expression among different cell lines and individual cells in different phases of the cell cycle.

The synthesis of S100 protein in cultured human melanoma cells was examined using metabolic labeling with [35S]methionine, immunoprecipitation with anti-S100 protein antiserum, and polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. Six of seven cell lines derived from melanomas synthesized relatively large amounts of S100 protein, whereas three cell lines derived from normal melanocytes synthesized lesser amounts. Synthesis of S100 protein was not detected in 10 human cell lines of nonneuroectodermal origin. Analysis of poly(A+) RNA from one melanoma cell line by Northern blot hybridization with a probe specific for the beta subunit of rat S100 protein revealed a single mRNA species of 1.0 kb coding for the human protein. Flow cytometric analysis of individual cells of two melanoma cell lines and the rat glioma cell line C6 indicated that G0/G1 cells were heterogeneous with respect to S100 protein expression, while almost all the cells in S + G2 + M expressed S100 protein. These results suggest that expression of S100 protein in G0/G1 could be a prerequisite for progression of the cells through the cell cycle.

Antigen processing and presentation by EBV-carrying cell lines: cell-phenotype dependence and influence of the EBV-encoded LMP1.

Burkitt's-lymphoma (BL) lines which have maintained in vitro the tumor-cell phenotype (group-I BLs) are poor antigen-presenting cells (APC), in spite of a relatively high surface expression of MHC class II. In order to investigate the mechanism of this deficiency, we have compared group-I BL lines, their sub-lines which have progressed in vitro towards an LCL-like phenotype (group-III BLs), and EBV-transformed lymphoblastoid cell lines (LCLs), for their ability to bind and process tetanus toxoid (TT). The uptake and internalization of 125I-labelled TT was equivalent in the 3 cell types. Only LCLs and group-III BL lines were able to process the TT, as shown by the identification of discrete proteolytic products after separation of whole-cell extracts in tricine-SDS-polyacrylamide gels, and by the recovery of TCA-soluble radioactivity in the culture supernatant. Processing of TT was induced by expression of the EBV-encoded membrane protein LMP 1 in transfected group-1 BLs. The present findings suggest that the inability of group-1 BLs to act as APC is due to their failure to process exogenous antigens. This function appears to be related to phenotypic properties that can be modulated by the expression of LMP1.

Heterologous expression of mouse cytochrome P450 2e1 in V79 cells: construction and characterisation of the cell line and comparison with V79 cell lines stably expressing rat P450 2E1 and human P450 2E1.

A V79 Chinese hamster cell line was constructed for stable expression of mouse cytochrome P450 2e1 (Cyp2e1), as an addition to the existing cell battery consisting of cell lines stably expressing rat CYP2E1 and human CYP2E1 (V79 Cell Battery). The aim was to establish a cell battery that offers the in vitro possibility of investigating species-specific differences in the toxicity and metabolism of chemicals representing substrates for CYP2E1. The newly established cell line (V79m2E1) effectively expressed Cyp2e1 in the catalytically active form. The expression of catalytically active CYP2E1 in V79m2E1 cells was maintained over several months in culture, as demonstrated by Western Blotting and chlorzoxazone (CLX) 6-hydroxylase activity. The cells exhibited CLX 6-hydroxylase activity with a Km of 27.8 microM/l and Vmax of 40 pmol/mg protein/minute, compared with a Km of 28.2/28.6 microM/l and a Vmax of 130/60 pmol/mg protein/minute from V79r2E1/V79h2E1 cells. Furthermore, the CYP2E1-dependent mutagenicity of N-nitrosodimethylamine could be demonstrated in the V79m2E1 cells. Therefore, the new cell battery permits the interspecies comparison of CYP2E1-dependent toxicity and of metabolism of chemicals between humans and the two major rodent species--the rat and the mouse--that are usually used in classical toxicity studies.

[Studies on human cervical carcinoma cell line. II. Establishment of HGPRT- cell line CC-80IAR2 and its biological characteristics].

An HGPRT- cell line, CC-801AR2, was established from cloned human cervical carcinoma cell line CC-801 by using MNNG to induce cell mutation and 8-azaguanine (8-AG) to select the 8-AG resistant cells. The deficiency of HG-PRT in CC-801AR2 cells was proved by enzyme activity assay. This cell line was very sensitive to HAT medium. It was very similar in biological characteristics to CC-801, especially in that it retained its tumorigenicity when transplanted into nude mice. CC-801AR2 can therefore be used in somatic cell hybridization and gene transfer experiments.

Ultrastructural characterization of a mouse melanoma cell line, a mouse fibroblastic cell line, and hybrids between them.

This report describes a transmission electron microscopic study on hybrids between subhexaploid mouse melanoma cells and subtetraploid mouse fibroblasts. The melanoma cell line was heterogeneous in terms of pigment production, but all cells contained melanosomes, although in different stages of development. Characteristic features of the fibroblasts included the cytoskeleton, endocytic vesicles, and occurrence of dilated cisternae of granular endoplasmic reticulum. Despite the gene dosage in favour of the melanoma parent cell, the hybrids were devoid of melanosomes, and their phenotype was typically fibroblastic in character.
