Duodenal rapeseed oil infusion in early and midlactation cows. 5. Milk fatty acids and adipose tissue lipogenic activities.

Lipogenic activities of perirenal adipose tissue were investigated in early (wk 3) and midlactation (wk 19 to 26) cows that received a duodenal rapeseed oil infusion (1.0 to 1.1 kg/d). In midlactation, oil infusion resulted in a decreased rate of fatty acid synthesis from acetate and a decreased rate of the activities of fatty acid synthetase and glucose-6-phosphate dehydrogenase, whereas lipoprotein lipase activity tended to increase. The rate of glucose incorporation into glyceride-glycerol and the activities of glycerol-3-phosphate dehydrogenase and malic enzyme were not significantly affected. Fatty acid C14:0 content of perirenal adipose tissue was decreased, and fatty acid C18:2 and C18:3 contents were increased in oil-infused cows. In early lactation, rates of acetate incorporation into fatty acids and activities of fatty acid synthetase and lipoprotein lipase were very low. Activities of glucose-6-phosphate dehydrogenase and glycerol-3-phosphate dehydrogenase were lower in the early than in the midlactation trial. Oil infusion did not change the measured parameters. In both trials, percentages and yields of milk fatty acids C18:1, C18:2, and C18:3 were increased, whereas those of C14:0 and C16:0 were decreased by oil. Calculated transfer rates of absorbed fatty acid C18:2 from oil to milk fat were 16 to 26%. Results suggested that oil fatty acids affected adipose and mammary de novo lipogenesis in a direct way without affecting fatty acid esterification in adipose tissue or total fat secretion in mammary tissue.

Some lipogenic enzyme activities in rat livers in which an excessive fat accumulation occurred due to feeding low-level amino acid mixture diets.

In liver homogenates of rats fed a low-level diet of Wheat-Rice- or Miyazaki-pattern amino acid mixture, some enzymes such as glucose-6-phosphate dehydrogenase, ATP citrate lyase, fatty acid-synthesizing enzymes, malic enzyme and L-alpha-glycerophosphate dehydrogenase, whose activities are indicators of lipogenesis have been determined from the viewpoint of the mechanisms producing fatty liver. In the early experimental period, malic enzyme activity increased more markedly in rats fed low amino acid mixture diets than in the control group, and L-alpha-glycerophosphate dehydrogenase activity in the liver increased slightly. Conversely, ATP citrate lyase and fatty acid-synthesizing enzyme activities remained almost at control levels, or glucose-6-phosphate dehydrogenase activity tended to decrease. These results suggest that some other associated factors, such as depression of the lipid transfer system in the liver rather than accelerated lipogenesis itself, may be the main cause of the fatty livers produced under these nutritional conditions.

Metabolic effects of exercise. I. Effect of exercise on serum lipids and lipogenesis in rats.

To evaluate the metabolic effects of exercise, three groups of exercised male rats were compared to their sedentary controls at the end of 10 wk of voluntary exercise in rodent activity cages. Exercises rats consumed more food than sedentary rats but had greater weight gain only at the higher levels of activity. Exercised rats had significanlty lower serum triglycerides and higher values of adipose tissue alpha-glycerophosphate dehydrogenase activity. At higher levels of physical activity, the exercised rats enhibited larger adrenal glands and lower values of hepatic glucose-6-phosphate dehydrogenase. It is concluded that specific levels of voluntary exercise are needed to achieve specific metabolic effects.

[Histoenzymatic characteristics of fibroblasts during organization of myocardial micronecroses].

The activity of dehydrogenase of succinate, lactate, glutamate, malate (NAD- and NADP dependent), alpha-glycerophosphate (NAD-dependent), glucoso-6-phosphate, NAD- and NADP-dyaphorase was studied in adult and old rats with the help of histoenzymatic methods in normal cardiac fibroblasts and under conditions of reparative regeneration developing in the places of foci of myocardial micronecroses caused by administration of adrenaline. Analysis of the activity of enzymes in normal fibroblasts revealed the predominance therein of an anaerobic way of metabolism. In the process of reparative regeneration the level of metabolism of fibroblasts rose with inclusion of the acitvity of pentose shunt and Krebs' cycle. No age differences in the activity of enzymes under study were revealed. The data obtained are discussed in connection with the problem of ascertaining inter-enzymatic relationships ensuring metabolism of the connective tissue in the process of its maturation.

[Histochemical study of tanycytes in connection with the adrenocorticotropic function of the hypophysis].

Chronic experiments were conducted on sexually mature rats; histochemical study of the activity of some redox enzymes (glutamic dehydrogenase, lactic dehydrogenase, glucoso-6-phosphoric dehydrogenase, glycerophosphoric dehydrogenase, and succinic dehydrogenase) was carried out in the ependymal cells of the floor of the third cerebral ventricle, the so called tanycytes, in case of an increased adrenocorticotrophic function of the hypophysis attained by bilaterial adrenalectomy, and in depression of this function as a result of chronic dexametasone administration. The activity of the enzymes under study decreased 2, 3 and 4 weeks after bilateral adrenalectomy. The activity of lactic dehydrogenase, glucerophosphoric dehydrogenase and succinic dehydrogenase increased in the tanycytes during administration of 5 gamma of dexametasone. Chronic administration of 100 gamma of dexametasone was accompanied by a toxic action of the preparation (a marked reduction in the weight of the adrenal glands, a negative body weight gain, and an aggravation of the animal's general condition). The results obtained pointed to the existence of a reverse correlation between the metabolic activity of tanicytes and the adrenocorticotrophic function of the hypophysis.

Ontogenetic development of energy-supplying enzymes in rat and guinea-pig heart.

The purpose of the present study was to compare the ontogenetic development of the activity of myocardial energy-supplying enzymes in two mammalian species, differing significantly in their level of maturation at birth. The animals were investigated during the late prenatal period and 2, 7, 14, 21, 25, 30, 63, 120 and 730 days after birth in the rat and 2, 21, 84 and 175 days in the guinea-pig. The following enzymes were assayed in the right and left ventricular myocardium: lactate dehydrogenase (LDH, lactate uptake and/or formation), triose phosphate dehydrogenase (TPDH, carbohydrate metabolism), glycerol phosphate dehydrogenase (GPDH, glycerol-P shuttle)), hexokinase (HK, glucose phosphorylation), malate dehydrogenase (MDH, tricarboxylic cycle), citrate synthase (CS, tricarboxylic cycle) and hydroxyacyl-CoA dehydrogenase (HOADH, fatty acid breakdown). The rat heart, highly immature at birth, exhibits three different developmental patterns of energy-supplying enzymes, identical in both ventricles: (i) two mitochondrial enzymes of aerobic metabolism (CS, HOADH) and GPDH have a relatively low activity at the end of prenatal life; thereafter their activity steadily increases, approaching the adult levels between the 3rd and 4th postnatal weeks. A significant decrease was observed between the 4th and 24th months. (ii) MDH and LDH: prenatal values were significantly higher as compared with the 2nd postnatal day; after this period the activities increased up to adulthood (4 months) and decreased during senescence. (iii) The activities of HK and TPDH are characterized by only moderate changes during development. HK differs from all other enzymes by the highest prenatal values, which exceed even adult values. In contradiction to the rat heart, the developmental differences in more mature guinea-pig heart were significantly less pronounced. The only ontogenetic differences observed were the lower activities of enzymes connected with aerobic metabolism at the end of the prenatal period. Our results point to possible differences in the development of adaptive metabolic pathways in animals with different levels of maturation at birth.

[Activity of the dehydrogenases of the myocardium and lymphocytes in experimental hypoxia].

The activity of succinatedehydrogenase, mitochondrial alpha-glycerophosphatedehydrogenase and NADH2-diaphorase was studied cytochemically and histochemically in the myocardium and lymphocytes of rats and mice after the effect of hypoxia in a hyperbaric chamber. A short-duration effect of hypoxia and step-wise adaptation during a month was shown to result in a growing activity of the enzymes both in the lymphocytes and in the myocardium. Hypoxia during a month and readaptation during the same period caused a monotonous decrease of the activity of dehydrogenases. The conducted analysis permitted to reveal a distinct correlation between the activity of dehydrogenases in the lymphocytes and in the myocardium. The authors emphasize the similarity of the changes in the activity of the enzymes in the lymphocytes and in the myocardium.

[Antipsoriatic effect of dithranol (anthralin). 1].

This review deals with the biochemical-pharmacological actions of anthralin (dithranol) and with the various possibilities of regulating the disturbed metabolism in psoriasis. A close relationship exists between enzyme inhibition, blocking of nucleic acid metabolism, and cytostatic-cytotoxic actions exerted by anthralin. Special reference is made of the inhibitory capacity on important enzymes of the pentose phosphate shunt the importance of which is briefly outlined. In vitro experiments with dithranol confirmed clinical observations such as the advantageous application of anthralin together with ultraviolet light or the decreased efficacy of anthralin incorporated in plain zinc paste.
