Synthesis and biological activities of some 7,8-dihydroimidazo[1,2-a]-1,3,5-triazine derivatives.

The alkylation of 7,8-dihydroimidazo[1,2-a]-1,3,5-triazine-2,4-(3H,6H)-dithione [I] with 2-chloro-N-arylacetamides gave 2-alkyltio derivatives [III]. Further reactions of 2-(alkyltio)-7,8-dihydroimidazo[1,2-a]-1,3,5-triazine-4(6H)- thiones [III] or [IV] with aryl isothiocyanates, sulfochlorides or 2-bromo-1-phenylethanone afforded corresponding B-substituted products [V,VI,VII].

Stable expression and coexpression of human cytochrome P450 oxidoreductase and cytochrome P450 1A2 in V79 Chinese hamster cells: sensitivity to quinones and biotransformation of 7-alkoxyresorufins and triazines.

V79 Chinese hamster cell lines were genetically engineered for the stable expression of human NADPH-cytochrome P450 oxidoreductase (CYPOR) alone or for the combined expression of CYPOR and human cytochrome P450 1A2 (CYP1A2). As determined by immunoblotting, the expression level of CYP1A2 in the latter cell line was found to be the same as in a previously constructed V79 cell line expressing CYP1A2 only. The heterologous expression of CYPOR in V79 cells resulted in increased sensitivity to quinone-type cytotoxins, e.g. duroquinone and menadione, that exert their toxicity primarily through the production of reactive oxygen species during redox cycling. The metabolic properties of the cell line expressing both CYPOR and CYP1A2 were characterized regarding dealkylation and deethylation of 7-alkoxyresorufins and sulfoxidation of the triazine derivatives ametryne and terbutryne, in comparison with the cell line expressing only CYP1A2. Increased CYPOR activity impaired the CYP1A2-dependent fluorometric resorufin assay, presumably by conversion of the 7-alkoxyresorufins and resorufin to their one-electron-reduced semiquinoneimine forms. The CYP1A2-dependent metabolism of the triazine derivatives ametryne and terbutryne was moderately enhanced by increased CYPOR activity. Interestingly, with CYPOR overexpression sulfoxidation was increased 2-3-fold, compared with N-deethylation, with a 1.3-1.9-fold increase. Thus, the level of CYPOR not only had an influence on CYP1A2 activity rates but also affected the relative proportions of metabolites in CYP1A2-specific metabolite profiles.

Synthesis and cytostatic properties of 5-substituted derivatives of 3-methylisoxazolo[5,4-d]1,2,3-triazine-4-ones and 3-methyl-5-triazene 4-isoxazolecarboxylic acid ethyl esters.

Six new 5-substituted derivatives of 3-methyl-isoxazolo[5,4-d]1,2,3-triazine-4-one and 3-methyl-5-triazene-4-isoxazolecarboxylic acid ethyl ester have been synthesized from 5-amino-3-methylisoxazole-4-carboxylic acid amides, and ethyl ester. They were examined for cytostatic activity in comparison with Dacarbazine. The 3-methyl-5-(4-chlorophenyl)isoxazolo[5,4-d]1,2,3-triazine-4-one showed better activity than Dacarbazine.

Synthesis and antiprotozoal activities of some new triazine derivatives including a new antitrypanosomal agent: SIPI-1029.

Two series of compounds, 1,2-dihydro-2,2-dimethyl-4, 6-diamino-1-(omega-haloalkyloxy)-s-triazines and O, O'-bis (4, 6-diamino-1, 2-dihydro-2, 2-disubstituted-s-triazin-l-yl) alkanediols were synthesized and tested against Plasmodium berghei and Trypanosoma evansi in mice. Most title compounds showed good antimalarial activity and compounds IIc-e showed good antitrypanosomal effect. After further studies on pharmacology, toxicology, pharmacokinetics and efficacy on infected cattles compound IIe (SIPI-1029, T-46) was shown to be a new highly active antitrypanosomal agent with low toxicity and long half life in plasma.

Effect of s-triazine compounds on testosterone metabolism in the rat prostate.

The influence of s-triazine compounds (atrazine, prometryne and deethylatrazine) on testosterone conversion and 5 alpha-dihydrotestosterone-receptor complex formation was studied in vitro and in vivo in the rat prostate. A marked in vitro influence of atrazine and prometryne (from 0.465 to 1.392 mumol) and their mixtures (in total concentration, 0.928 mumol) on 5 alpha-dihydrotestosterone formation was detected. 5 alpha-Dihydrotestosterone-specific receptor complex formation was inhibited in vitro by ca. 0.5 mumol of atrazine or deethylatrazine and only in vivo by 6 mg of atrazine 100 g-1 body wt. daily during 7 days in the prostate cytosol. The inhibition of the enzymic activities responsible for testosterone conversion and steroid hormone-receptor complex formation was non-competitive and reversible, and s-triazine compounds act as antiandrogens.

Hydrogen bonding and solvent effects on the lowest 1(n, pi*) excitations of triazines in water.

Our method for estimating solvent effects on electronic spectra in media with strong solute-solvent interactions is applied here to calculate the absorption and fluorescence solvatochromatic shifts of dilute triazines in water. First, the ab initio CASSCF method is used to estimate the gas-phase electronic excitation properties and state charge distributions; second, Monte Carlo simulations are performed to elucidate liquid structures around the ground and excited state solute; finally, the solvent shift is evaluated based on the gas-phase charge distributions and the explicit solvent structures. For the dilute triazine solutions, simulations predict one linear (different) hydrogen bond attached to each nitrogen atom. Upon the first (1)(n, pi*)electronic excitation one hydrogen bond is completely broken. For the absorption and fluorescence spectra, our calculations demonstrated that the specific solvent-solute interaction, in any electronic state, plays a critical role in the determination of solvent shifts.

The High-Resolution Infrared Spectrum of 1,2,4-Triazine Vapor between 550 and 1700 cm(-1).

The Fourier transform gas-phase IR spectrum of 1,2,4-triazine between 550 and 1700 cm(-1) was measured with a resolution of ca. 0.003 cm(-1). Comparing with the liquid-phase IR and Raman spectra and using ab initio predictions, most of the fundamental bands of 1,2,4-triazine below 1600 cm(-1) were assigned. From the high-resolution gas-phase spectra, 12 of the fundamental bands were analyzed by the Watson Hamiltonian model to yield upper state spectroscopic constants. A number of local resonances were identified and explained. From a simultaneous ground state combination difference analysis of four of the bands, a set of ground state rotational and centrifugal distortion constants were obtained. Air monitoring of aldehydes by use of hydrazine reagents with a triazine backbone.

Two hydrazine reagents, 4- N, N-dimethylamino-6-(4'-methoxy-1'-naphthyl)-1,3,5-triazine-2-hydrazine (DMNTH) and N-methyl-4- N', N'-dimethylamino-6-(4'-methoxy-1'-naphthyl)-1,3,5-triazine-2-hydrazine (MDMNTH) have been synthesized and used for the determination of aldehydes in air samples. Test tubes with the reagents coated on silica gel were prepared and used for monitoring of carbonyls in air. After elution with acetonitrile the hydrazones formed were separated by reversed-phase liquid chromatography. Detection was performed by UV-visible and fluorescence spectroscopy. The results were validated by use of standard atmospheres of the carbonyls and of nitrogen dioxide and ozone, as potential interferents. In comparison with established hydrazine reagents, e.g. 2,4-dinitrophenylhydrazine (DNPH), the results from use of MDMNTH correlate well; lower recoveries were obtained by use of DMNTH. The limits of detection for the new reagents are superior to those for DNPH, because of the possibility of fluorescence detection.

Conformation and structure of 2-amino-8-(beta-D-ribofuranosyl)imidazo [1,2-a]-s-triazin-4-one(5-aza-7-deaza guanosine), a potent antiviral nucleoside.

The crystal and molecular structure of one imidazo[1,2-a]-s-triazine nucleoside and its antiviral activity are described. The crystal structure of 2-amino-8-(beta-D-ribofuranosyl)imidazo[1,2-a]-s-triazin-4-one monohydroate (C10H13N5O5.H2O) was solved by X-ray counter data. The compound crystallizes in the monoclinic space group P21 with cell dimensions a = 7.353 (1), b = 6.465 (1), c = 13.701 (1) A, B = 104.64 (1) degrees. The structure was solved by direct methods and refined by full matrix least-squares technique to a final value of the conventional R-factor of 0.049 using 1998 observed intensities. The orientation of the base relative to the sugar ring defined in terms of rotating about the C(1')-N(8) glycosyl bond is anti (47.8 degrees). The ribose moiety exhibits C(2')-endo, E conformation. The conformation around C(4')-C(5') is gauche-. Molecular packing is dominated by hydrogen bonds. Base stacking occurs long the b axis. 5-Aza-7-deazaguanosine has shown a marked antiviral activity in vitro against herpes simplex virus despite the fact that N(3) is effective as the hydrogen acceptor only.

Dissipation of s-triazines and thiocarbamates from soil as related to soil moisture content.

Half-lives (t1/2) of two soil incorporated s-triazines (atrazine and prometon) and two thiocarbamate (EPTC and triallate) herbicides were determined in relation to soil moisture content in two California soils. Treated soils were incubated at three moisture levels in aerated glass vials at 25 +/- 1 degree C and were analyzed at 0, 7, 16, 28, 56 and 112 day intervals. Loss of herbicides in all treatments followed first-order kinetics. The t1/2-values of all herbicides decreased with increasing soil moisture and followed an empirical equation, t1/2 = aM(-b) (where t1/2 is half-life; M the moisture content; and a and b are constants). Soil moisture had a greater effect on carbamates than on s-triazines . Prometon exhibited the longest half-life in both soils, whereas EPTC was least persistent in one soil and atrazine in another. The t1/2-values for atrazine, prometon, EPTC, and triallate with medium moisture levels and 10 microg/g concentration were 34.6, 43.2, 25.4 and 38.1 days in sandy loam and 26.5, 44.4, 44.1 and 25.9 days in loamy sand, respectively. Disappearance of 50% of the applied concentrations of most of the herbicide-soil combinations (except EPTC and triallate in one soil) took longer for lower initial concentrations (1 microg/g) than for higher concentrations (10 microg/g).

Analytical method using gas chromatography and ion trap tandem mass spectrometry for the determination of S-triazines and their metabolites in the atmosphere.

Gas chromatography-ion trap detector (GC-ITD) was used to detect atmospheric triazines and their degradation products in the gaseous and particulate phases. Because triazines and their metabolites are expected to be present at very low concentrations and enclosed in the complex atmospheric matrix, the analytical method used was both highly selective and sensitive. These two properties were obtained by associating chromatography with ion trap tandem mass spectrometry (GC-ITD (MS/MS)). To develop this method, a comparison between the two ionization modes (electron impact and positive-chemical-ionisation) in single-MS was first conducted to choose the parent ions of the five target analytes, i.e. atrazine, desethylatrazine, deisopropylatrazine, terbuthylazine and desethylterbuthylazine. Then, a MS/MS method was optimised by parameters such as the radio frequency storage level and the collision-induced dissociation excitation voltage. Finally, a last step enabled the development of a calibrating program based on the quantification of daughter ions. With this analytical procedure, the detection limits varied between 0.8 and 15 pg m(-3) depending on the compounds under study. This method was tested with success for four atmospheric samples collected in Strasbourg (France) in which four of the five target compounds were detected.

Generation and photoreactions of 2,4,6-trinitreno-1,3,5-triazine, a septet trinitrene.

We have studied the matrix photolysis of 2,4,6-triazido-1,3,5-triazine (cyanuric triazide, 1). Stepwise generation of the corresponding mononitrene, dinitrene, and trinitrene was observed by matrix IR and electron paramagnetic resonance (EPR) spectroscopy. The generated species were identified by comparison of their matrix IR spectra with density functional theory (DFT) computational results. The generation of 2,4,6-trinitreno-1,3,5-triazine with a septet ground state was confirmed for the first time by matrix EPR spectroscopy. The trinitrene readily decomposed into three NCN molecules upon further photoirradiation. This process was also confirmed by matrix EPR spectroscopy.

Photodegradation of triazine herbicides in aqueous solutions and natural waters.

The photodegradation of three triazines, atrazine, simazine, and prometryn, in aqueous solutions and natural waters using UV radiation (lambda > 290 nm) has been studied. Experimental results showed that the dark reactions were negligible. The rate of photodecomposition in aqueous solutions depends on the nature of the triazines and follows first-order kinetics. In the case of the use of hydrogen peroxide and UV radiation, a synergistic effect was observed. The number of photodegradation products detected, using FIA/MS and FIA/MS/MS techniques, suggests the existence of various degradation routes resulting in complex and interconnected pathways.

Chemical reduction of 2,4,6-tricyano-1,3,5-triazine and 1,3,5-tricyanobenzene. Formation of novel 4,4',6,6'-Tetracyano-2,2'-bitriazine and its radical anion.

Chemical reduction of 2,4,6-tricyano-1,3,5-triazine, TCT, results in the formation of an unstable radical anion that undergoes immediate dimerization at a ring carbon to form [C(12)N(12)](2-), [TCT](2)(2-), characterized by a long 1.570 (4) A central C[bond]C. [TCT](2)(2-) can decompose into the radical anion of 4,4',6,6'-tetracyano-2,2'-bitriazine, [TCBT]*-, the one-electron reduced form of planar (D(2h)) TCBT, which is also structurally characterized as the [TMPD][TCBT] charge-transfer complex (TMPD = N,N,N',N'-tetramethyl-p-phenylenediamine) with a 1.492 (2) A central sp(2)[bond]sp(2) C[bond]C. Although crystals could not be obtained for the radical anion [TCBT]*-, the electrochemistry (E degrees = +0.03 V), EPR (g = 2.003, (2)A((14)N) = 3.347 G, and (4)A((14)N) = 0.765 G and a line width of 0.24 G), and theoretical calculations support the formation of [TCBT]*-. In addition, thermolysis of [TCT](2)(2-) yields [TCBT]*-. Chemical reduction of 2,4,6-tricyanobenzene, TCB, forms an unstable radical anion that immediately undergoes dimerization at a ring carbon to form [C(12)H(6)N(6)](2-), [TCB](2)(2-), which has a long 1.560 (5) A central C[bond]C. Reaction of TCT with tetrathiafulvalene (TTF) forms structurally characterized [TTF][TCT], and in the presence of water, TCT hydrolyzes to 2,4-dicyano-6-hydroxy-s-triazine, DCTOH. In contrast, the reaction of TCT with TMPD forms [TMPD][TCT], which in the presence of water forms structurally characterized [HTMPD](+)[DCTO](-).

Dienophilicity of imidazole in inverse electron demand Diels-Alder reactions. 4. Intermolecular reactions with 1,2,4-triazines.

Intermolecular inverse electron demand cycloadditions of 2-substituted imidazoles with various 1,2,4-triazines produced both imidazo[4,5-c]pyridines (3-deazapurines) and pyrido[3,2-d]pyrimid-4-ones (8-deazapteridines). The product distribution was controlled by reactant substituents and influenced by reaction temperature. A regioselective method for the preparation of 6-unsubstituted 1,2,4-triazines was also developed. By using this route to 8-deazapteridines, a new 8-deazafolate analogue was prepared.

Mechanism of an Exceptional Class of photostabilizers: a seam of conical intersection parallel to excited state intramolecular proton transfer (ESIPT) in o-hydroxyphenyl-(1,3,5)-triazine.

We present a detailed CASSCF study of the mechanism of excited-state intramolecular proton transfer (ESIPT) in the o-hydroxyphenyl triazine class of photostabilizers. The valence-bond analysis of the ground state and the two pipi* excited states permits a simple chemical interpretation of the mechanistic information. Our results show that the barrier to enol-keto tautomerism on the ground-state adiabatic surface is high. Following photoexcitation to the charge-transfer state, the ESIPT is predicted to take place without a barrier. Radiationless decay to the ground state is associated with an extended seam of conical intersection, with a sloped topology lying parallel to the ESIPT path, which can be accessed at any point along the reaction path. Our results show that the triazine class of photostabilizers has the photochemical and photophysical qualities associated with exceptional photostability.
