Epidemiological study on workers exposed to low dimethylformamide concentrations.

Reviewing medical and epidemiological reports, no definite clinical picture could be expected as a result of a low DMF exposure and experimental research on long term toxicity has always demonstrated some adverse effects but has not been sufficient to define a no-effect level in animals. This study was designed to assess the specificity of symptoms and the relevance of adverse effects as consequence of an exposure to airborne DMF concentration in the range of the present TLV (30 mg/m3 - 10 ppm). For this purpose 100 DMF-exposed workers, with homogeneous characteristics, were compared with 100 matched controls. Both groups were selected by a careful pair-matching. Mean DMF exposure was 22 mg/m3 (range 8-58 mg/m3). Exposed subjects and their matched controls were evaluated clinically and a questionnaire was used for the registration and the comparison of subjective complaints. A laboratory assessment was performed, including transaminase and gamma-glutamyl transpeptidase. Statistical analysis was based on McNemar Test procedure. The problem of dietary alcohol intake was particularly investigated. Among symptoms studied, headache, dyspepsia and digestive impairment of hepatic type could be specifically associated with chronic DMF exposure and increased levels of gamma-GT demonstrated minimal hepato-cellular damage, even without ethanol dietary intake. No chronic sickness was diagnosed and the disturbances observed are better considered as indicators of malaise and discomfort due to a toxic effect of DMF, whose consequences are discussed.

[Structural and ultrastructural changes in the rabbit myocardium exposed to dimethylformamide vapors].

The aim of this research is to show the alterations of alpha-actinin in rabbit myocardium after DMF treatment administered by forced inhalation. Z-lines observed with light and phase-contrast microscopy, appeared to be intact and they were clearly displayed by the indirect PAP-reaction. But if you consider that in normal muscle Z lines do not get coloured by the PAP-reaction without a previous light treatment with trypsin. It may be inferred that, in this case, The DMF has had the same effect as the trypsin, causing alteration to the protein structure. Besides the sarcomeric structure is undoubtedly altered by the DMF, causing alterations, in our opinion, similar to those found in rabbit skeletal muscle, under conditions of acute experimental ischemia.

[Effect of the intermittent and continuous action of dimethylformamide on adaptation processes].

Two series of experiments were carried out with 960 mg/kg and 3840 mg/kg DMF, applied dermally to albino rats every second, third day and daily as well. The treatment scheme quarantees equitoxic quantities of DMF with a dynamic observation by the 4th, 8th, 14th and 28th day from the beginning of the exposure and after 14 days of rehabilitation. The complex investigation with integral and specific clinical-laboratory tests revealed a cumulation of the toxic effect with progressing changes in the liver function, better manifested with the intermittent regimes. The dose of 3840 mg/kg induced distinct deviations in the lipid metabolism and serum enzymes. In contact with 960 mg/kg DMF, a phase-state was established during the first terms of observation with the development of adaptation under a monotonous effect and absence of restoration -- mainly in case of intermittent effect. Dermal toxicity of DMF should be given consideration in the assessment of the risk from exposure during production of polyacryl fibres.

[Gel filtration of protected synthetic peptides in dimethylformamide].

For large water-insoluble protected peptides, a novel method of analytical and preparative size exclusion liquid chromatography on Toyopearl HW-40, HW-50, HW-55, and HW-60 gels was proposed, with DMF as an eluent. The selectivity and molecular mass ranges of these gels were determined for separation of protected peptides in DMF. The molecular mass ranges of Toyopearl HW gels for protected peptides in DMF were found to differ significantly from those for peptides and proteins in aqueous buffer solutions.
