Flocculation of venereal disease research laboratory reagent by Helicobacter pylori.

Helicobacter pylori strains flocculated with Venereal Disease Research Laboratory (VDRL) reagent in a glass slide test. Other pathogenic bacterial and fungal strains were nonreactive. The specific VDRL reaction property of Helicobacter pylori indicates an affinity of the cells for lipoidal substances, and can be used as a diagnostic aid for species identification.

Continuous ethanol fermentation of lactose by a recombinant flocculating Saccharomyces cerevisiae strain.

Alcohol fermentation of lactose was investigated using a recombinant flocculating Saccharomyces cerevisiae, expressing the LAC4 (coding for beta-galactosidase) and LAC12 (coding for lactose permease) genes of Kluyveromyces marxianus. Data on yeast fermentation and growth on a medium containing lactose as the sole carbon source are presented. In the range of studied lactose concentrations, total lactose consumption was observed with a conversion yield of ethanol close to the expected theoretical value. For the continuously operating bioreactor, an ethanol productivity of 11 g L(-1) h(-1) (corresponding to a feed lactose concentration of 50 g L(-1) and a dilution rate of 0.55 h(-1)) was obtained, which is 7 times larger than the continuous conventional systems. The system stability was confirmed by keeping it in operation for 6 months.

Relationships between hydrodynamics and rheology of flocculating yeast suspensions in a high-cell-density airlift bioreactor.

In this article a hydrodynamic and rheological analysis of a continuous airlift bioreactor with high-cell-density system is presented. A highly flocculating recombinant strain of Sacharomyces cerevisiae containing genes for lactose transport (lactose permease) and hydrolysis (beta-galactosidase) was exploited to ferment lactose from cheese whey to ethanol. The magnetic particle-tracer method was used to assess the effect of operational conditions (air-flow rate, biomass concentration) on hydrodynamic behavior of an airlift bioreactor during the fermentation process. Measurements of liquid circulation velocity showed the existence of a critical value of biomass concentration at which a dramatic deceleration of net liquid flow appeared with increasing biomass quantity. Rheological analysis revealed exponential increase of viscosity of the yeast floc suspension at the same biomass concentration of about 73 g/dm3 corresponding to 42.8% v/v of solid fraction. These facts have a particular importance for the successful processing of a high-cell-density airlift bioreactor as only a circulated flow regime will be favorable to keep the solid particles in suspension state and evenly distributed throughout the bioreactor.

Physicochemical properties of glycyrrhizic acid in aqueous media II: Effect on flocculation-deflocculation behavior of suspensions of sulfathiazole and graphite.

The flocculation-deflocculation behavior of sulfathiazole and graphite in aqueous solutions of glycyrrhizic acid was studied by measuring the sedimentation volume and turbidity of supernates. The dispersing effect of glycyrrhizic acid on suspension of sulfathiazole showed a maximum in the pH 3-4 region, the same pH region where the zeta-potential of sulfathiazole particles showed a negative maximum. The results were explained by the variation of degrees of ionization of glycyrrhizic acid and sulfathiazole with pH. With graphite suspensions, the pH region where the dispersing effect of glycyrrhizic acid showed a maximum shifted to a higher pH compared with sulfathiazole. This result can be attributed to the fact that graphite is a nonpolar substance so the surface properties are not affected by a pH change. Hence, the adsorption of glycyrrhizic acid occurs even in a fairly high pH range.

Flocculation of Kluyveromyces marxianus is induced by a temperature upshift.

An upshift of the growth temperature from 26 to 40 degrees C in the presence of calcium leads to the aggregation of Kluyveromyces marxianus cells and to the formation of flocs. Analysis of cell wall proteins, either by sodium dodecyl sulphate-polyacrylamide gel electrophoresis of extractable mannoproteins or by immunolocalization, revealed an accumulation of a protein with Mr 37 kDa(p37), upon flocculation. Immunological studies confirmed the homology of this protein with the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH). When mRNA isolated from cells growing at 40 degrees C was translated in vitro, a 35 kDa newly labelled protein was synthesized and immunoprecipitation assays showed that this protein is recognized by p37-antiserum, suggesting that the 35 kDa polypeptide might be an unglycosylated precursor for of p37. The results indicated that the presence of this cell wall mannoprotein closely related to GAPDH is dependent on the growth temperature, suggesting its role as adhesin.

Molecular cloning and analysis of the yeast flocculation gene FLO1.

The DNA sequence of the flocculation gene FLO1 of Saccharomyces cerevisiae, which is located on chromosome I (Watari et al., 1989) was determined. The sequence contains a large open reading frame (ORF) of 2586 bp and codes for a protein of 862 amino acids. However, further study (genomic Southern and polymerase chain reaction analyses) indicated that the gene we cloned was not the intact FLO1 gene but a form with an approximately 2 kb deletion in the ORF region. The intact FLO1 gene was then cloned and its nucleotide sequence determined. The sequence revealed that the ORF of the intact gene is composed of 4611 bp which code for a protein of 1537 amino acids. A remarkable feature of the putative Flo1 protein is that it contains four families of repeated sequences composed of 18, 2, 3 and 3 repeats and that it has a large number of serines and threonines. In the deleted FLO1 form, a large part of these repeated sequences was missing. The N- and C-terminal regions are hydrophobic and both contain a potential membrane-spanning region, suggesting that the Flo1 protein is an integral membrane protein and a cell wall component.

Ethanol production by a mixed culture of flocculent strains of Zymomonas mobilis and Saccharomyces sp.

Pure and mixed cultures of Zymomonas mobilis and Saccharomyces sp. were tested for the production of ethanol using sucrose as the carbon source. Both strains, isolated from spontaneously fermenting sugar-cane juice, are flocculent and alcohol-tolerant. The best results were obtained using a mixed culture, with a yield of 0.5 g ethanol/g sugar consumed and a volumetric productivity of 1.5 g ethanol l-1 h-1. No levan was produced even if a sucrose-based medium was used.

The quantitative estimation of diphtheria and tetanus toxoids. 1. The flocculation test and the Lf-unit.

Considerable confusion exist in the quantitative estimation of diphtheria and tetanus toxoids. The present paper is the first in a series of papers comparing various quantitative methods. It describes the flocculation test, gives a mathematical model for the three-dimensional reaction surface, and suggests different designs which facilitate the unbiased calculation of the end-point.

Chitosan as flocculant for concentrating Euglena gracilis cultures.

The practical criteria for the usefulness of an algal separation process for laboratory routine being effectiveness and time consumption, we tested the feasibility of a flocculation procedure to harvest large volumes of Euglena gracilis in culture. This procedure turned out to be a technically viable system which avoided tedious centrifugation and preserved E. gracilis flagellar apparatus integrity. E. gracilis cultures were treated with chitosan, a by-product derived from chitin from the exoskeleton of crustaceans. Since this polymer carries a positive charge, it functions as a polycationic coagulating agent by adsorbing onto particles in suspension and by bridging together into agglomerates, or flocs. A 96-98% reduction of suspended cells in cultures with 200 mg/l of chitosan, at pH 7.5, was obtained.

Colour removal from landfill leachate by coagulation and flocculation processes.

A study was conducted to investigate the efficiency of coagulation and flocculation processes for removing colour from a semi-aerobic landfill leachate from one of the landfill sites in Malaysia. Four types of coagulant namely aluminium (III) sulphate (alum), ferric (III) chloride, ferrous (II) sulphate and ferric (III) sulphate were studied using standard jar test apparatus. Results indicated that ferric chloride was superior to the other coagulants and removed 94% of colour at an optimum dose of 800 mg/l at pH 4. The effect of coagulant dosages on colour removal showed similar trend as for COD, turbidity and suspended solids. This suggested that colour in landfill leachate was mainly contributed by organic matters with some insoluble forms that exhibited turbidity and suspended solids readings. The results from this study suggested that ferric chloride could be a viable coagulant in managing colour problems associated with landfill leachate.

Flocculation in brewing yeasts: a computer simulation study.

A new simulator, INDISIM-FLOC, based on the individual-based simulator INDISIM, is used to examine the predictions of two different models of yeast flocculation. The first, proposed by Calleja is known as the "addition" model. The second, proposed by Stratford is known as the "cascade" model. The simulations show that the latter exhibits a better qualitative agreement with available experimental data.

Removal of cyanobacterial blooms in Taihu Lake using local soils. I. Equilibrium and kinetic screening on the flocculation of Microcystis aeruginosa using commercially available clays and minerals.

Algal removal abilities of 26 clays/minerals were classified into three categories according to the 8-h equilibrium removal efficiency (Q8h) and removal rate at a clay loading of 0.7 g/L. Type I clays (sepiolite, talc, ferric oxide, and kaolinite) had a Q8h > 90%, a t50 (time needed to remove 50% of the algae) < 15 min, and a t80 < 2.5 h. Type II clays (6 clays) had a Q8h 50-90%, a t50 < 2.5 h, and a t80 > 2.5 h. Type III clays (14 clays) with Q8h < 50%, t50 > 8 h and t80 > 14 h had no practical value in removal of algal blooms. When the clay loading was reduced to 0.2 g/L, Q8h for all the 25 materials decreased to below 60%, except for sepiolite whose Q8h remained about 97%. The high efficiency for sepiolite to flocculate M. aeruginosa cells in freshwaters was due to the mechanism of netting and bridging effect.

Acute toxicity removal in textile finishing wastewater by Fenton's oxidation, ozone and coagulation-flocculation processes.

This study evaluates the effectiveness of Fenton's oxidation (FO) process and ozone (O3) oxidation compared with a coagulation-flocculation (CF) process to remove effluent toxicity as well as colour and COD from a textile industry wastewater. Daphnia magna was used to test acute toxicity in raw and pre-treated wastewater. The operational parameters for each process were determined on the basis of complete toxicity removal. The FO process removed COD at a higher rate (59%) than O3 (33%) while colour removal was similar (89% and 91%, respectively). The CF process removed both COD and colour at rates similar to the FO process. A colour range of 150-250 platin-cobalt (Pt-Co) unit was assessed for toxicity.

Modified inorganic polymer flocculant-PFSi: its preparation, characterization and coagulation behavior.

As a new kind of water and wastewater treatment reagents, inorganic polymer flocculants (IPFs) are in a trend of rapid development. Among them, iron-based IPF without any toxic problems is greatly expected. In this paper, a new protocol for preparation of iron-based IPF is investigated. Three kinds of silica, named silicaA, silicaB and silicaC, are prepared and used as modifiers to tailor-make polyferric silicate (PFSi), denoted PFSiA. PFSiB and PFSiC, respectively, in accordance to the above silica. Based on several direct and indirect speciation methods, the species distribution and characteristic of three kinds of PFSi are investigated in detail. The experiment results show that the species distribution of PFSi is mainly decided by the kinds of silica introduced and Si/Fe ratio adopted. SilicaA and silicaB function as precipitation preventing reagents during the hydrolysis of Fe(III). At certain amount of basicity, Fe(a) (monomers) increases with increase of Si, Fe ratio, while Fe(c) (colloidal species) decreases markedly. Although the formation of Fe(c) is inhibited, the stabilization of Fe(b) (oligomers or polymers) can still not be achieved. SilicaC on the other hand exhibits little effect on the species distribution of modified Fe(III) solutions. The coagulation behavior of above PFSi is also examined. The different effect of silica is then discussed with respect to the chemical species.

Network strength and dewaterability of flocculated activated sludge.

Works in literature proposed the use of the rheological properties of wastewater sludges as an index for conditioner assessment and control. We demonstrated in this work, on the contrary, that one could not justify the consistency of the commonly used rheological characteristics of the sewage sludge samples taken from the same site but at different dates. A physically relevant index was proposed instead for describing the total network strength, which was hypothesized to correlate the dewatering efficiency of flocculated sludge. Based on this index, the network of a sludge was demonstrated to be largely destroyed after shearing, while the relaxation in an unbound environment or mild pouring over action could partially reinstall the structure. Moreover, a uniform shear rate field could produce network of greater strength when compared with that conditioned in a stirred tank.

Nutrient removal and sludge production in the coagulation-flocculation process.

Nutrient removal and sludge production in the coagulation-flocculation process, applied to a slaughterhouse effluent, have been studied. Fe2(SO4)3, Al2(SO4)3 and polyaluminium chloride were used as coagulants. Inorganic products were used as coagulant aids: activated silica, powdered activated carbon and precipitated calcium carbonate and synthetic polyelectrolytes: cationic polyacrylamide, polyacrilic acid, anionic polyacrylamide and polyvinyl alcohol. Performances were measured under optimum conditions for the products used. They were found after studying the different variables which influence the process. Phosphorus removal is very high (approximately 100% for the orthophosphate and between 98.93% and 99.90% for the total phosphorus). Ammonia nitrogen removal is very low although appreciable performances are observed for albuminoid nitrogen (73.9-88.77%). The use of coagulant aids reduces the volume of the sludge produced up to 41.6%.

Optimisation of the coagulation-flocculation process of raw water by optimal design method.

In the present paper, a systematic study of the influence of raw water quality and operating conditions on the effectiveness of the coagulation-flocculation process using aluminium sulphate (AS) is presented. The influence of raw water composition is studied at two levels, fractional factorial designs and a Doehlert uniform shell design to determine the influence of the operating conditions. The analysis of the experimental responses (residual turbidity and minimally AS dose are denoted as (Tu(m)) and (AS(m)), respectively) shows that the parameters which have the greatest influence change depending on the response. It is not always possible, for a given water composition, to obtain simultaneously low residual turbidity and minimally AS dose. However, under the conditions of this study, the search for a criterion of optimisation must bear the minimisation of residual turbidity, because it remains the principal criterion of effectiveness of the process, even, if in a few cases, it results in an increase in the cost of treatment.
