[Enzyme histochemical examinations of the visual system of the adult rat following unilateral enucleation].

12 adult white lab-rats were enucleated and after a survival period of 1, 3, 7 and 30 days the activities of GABA-T, GDH, LDH, SDH and GPDH were demonstrated histochemically in the Tractus opticus (To), Corpus geniculatum laterale, pars dorsalis (CGLd) et ventralis (CGLv), Colliculus superior (Cs) and Nucleus olivaris praetectalis (Nop). Since the ipsi- and contralateral grisea are always in the same tissue section the enzyme activities can be quantitatively compared by visual impression without a greater mistake. In To enucleation caused a hypertrophy of astrocytes together with an increase of the activities of GABA-T, GDH, LDH, and GPDH in these cells. The reactions indicate a more intensive metabolism in connection with the myelin degradation. In CGLd, CGLv, Cs and Nop following enucleation there appeared contralaterally a graded loss in the activities of GDH, LDH, GPDH, and GABA-T; only SDH scarely changed its activity. The fastest and strongest reaction was found in Cs and Nop, while CGLd and CGLv reacted later and to a less degree. In CGLv the enzyme reaction was limited to the lateral part of the nucleus. As diminution of activity is caused by degeneration of the retinal terminals the effected enzymes must be localised in cytoplasma and mitochondria of these terminals. Taking into account findings from literature the following is concluded from the time patterns, the degree of diminution of the enzyme activities and the relation of retinal to extraretinal terminals in the individual nuclei: GABA probably acts as a transmitter in interneurons of CGLd, Cs, and Nop. Glutamate is a transmitter in Cs and Nop.

[Comparative microspectrophotometric study of the activity of oxidative enzymes in separate types of fibers of the skeletal muscle of rats in the normal state and in experimental ischemia].

Microspectrophotometrical investigation of the activity of oxidative enzymes (glutamatdehydrogenase, cytoplasmic and mitochondrial alpha-glycerol phosphate dehydrogenase, succinic acid dehydrogenase and glucose-6-phosphatedehydrogenase) in certain types of muscular fibres of m. soleus, gastronemius and quadriceps of rats during experimental stenosins of the abdominal part of the aorta has shown the existence of certain cyclic changes in the enzymic activity of different periods after graded constriction which may be associated with arising of adaptational reactions of metabolic systems in striated muscles in response to chronic hypoxia.

[Formation and interrelationship of the activity of several dehydrogenases during rat development].

The specific and general enzymic activity of some dehydrogenases of the liver, kidneys, heart, thymus and spleen was studied on rats in early postnatal period. Some regularities of the development of enzymes under study were established: ergontic correlations of the "maturing" enzymic organ systems and the coordination of the enzymic status of organs and the lymphocytes circulating in the blood. It may be supposed that the lymphocyte is a carrier of information about formation of metabolic processes of the developing organism. The mechanisms of appearance of coordination of the enzymic activity are discussed.

Enzyme activities in red and white muscles of guinea-pigs and rabbits indigenous to high altitude.

The activities of several enzymes functioning in different areas of fuel catabolism were measured under standardized conditions, using crude homogenates of sartorius and ventricular muscle from outbred guinea-pigs and rabbits indigenous to high or low altitude. The activities of sartorius and myocardium were found to reflect the metabolic patterns known to be associated with white and red muscle. Both species had right ventricular hypertrophy at high altitude. The enzyme activities in the high altitude guinea-pig were not significantly different from those in the sea level animals. In the high altitude rabbit, compared with the low altitude rabbit, the activities of glyceraldehyde-3-phosphate deydrogenase and phosphofructokinase were greater in both the sartorius and myocardium. In addition, mitochondrial glycerol-3-phosphate dehydrogenase activity was greater in the sartorius at high altitude, while aspartate aminotransferase and beta-hydroxyacylcoenzyme A dehydrogenase activities were greater in the myocardium at high altitude. Succinate dehydrogenase activity was comparable at the two altitudes for both tissues. There was a greater proportion of skeletal muscle type lactate dehydrogenase in the high altitude rabbit myocardium but no difference was found with the guinea-pig.

[Morphological and histochemical changes in the rabbit aorta during exogenous and endogenous hyperglycemia].

Alimentary hyperglycemia and insulin deficiency were accompanied in rats by atherosclerotic affection of the aorta whose degree increased in feeding of cholesterol to such animals. The detected morphological and histochemical changes in the wall of the aorta correlated with the biochemical atherogenic shifts in the blood serum of the corresponding animals groups established earlier.

[Enzymatic heterogeneity of the rat myocardium].

The histoenzymatic method was applied to the study of distribution of the activity of the redox enzymes in the myocardium of the ventricles in rats; distribution of the activity of lactic and malic dehydrogenase and of alpha-glycerophosphate proved to be the most manifest near the apex of the heart and was expressed in the presence of "spotty" areas of increased activity against the general homogeneous background of formazan deposits. The activity of mitochondrial upsilon-glycerophoric dehydrogenase was seen in all the portions of the ventricles and was characterized by an uneven distribution in the sarcoplasm with increase in the direction from the interdisc to the nucleus. Unevenness of distribution of the beta-oxybutyric dehydrogenase activity was detected in some of the animals and was pronounced in all the portions of the myocardium. The intensity of the reaction in detection of succinic dehydrogenase, NAD- and NADP-diaphorases varied but insignificantly.

[Action of some pyrazolone derivatives on thrombocyte aggregation].

The influence of four agents--pyrazolidinedione derivatives on the ADP-induced aggregation and activity of glycolytic enzymes--glyceraldehydrophosphate dehydrogenase and lactate dehydrogenase (GAPD, LDG) and of the anatomic oxidation enzyme--glucose-6-phosphate dehydrogenase (G-6-PD) was studied. The substances known by a conventional designation of MYaK-38, No-13 and No-2 are shown to display an antiaggregational action. Amidopyrine was found to depress the adhesive capacity of blood platelets, to activate GAPD and LDG and to lower the activity of G-6-PD.

In vitro influence of oxygenation on enzymes of rat adipose tissue.

Pre-incubation of rat adipose tissue from ad lib fed rats in the presence of 1.2 U/ml insulin and 2.8 x 10(-7) M prostaglandin F2 alpha for 16 hours in a gas atmosphere of 5% O2 led to an increase in glucose-6-phosphate dehydrogenase activity. No increase in glycerol-3-phosphate dehydrogenase or hexokinase activities were noted. Actinomycin D (50 micrograms/ml) and dehydroepiandrosterone (120 micrograms/ml) attenuated this increase and the increase in incorporation of [1-14C] glucose into carbon dioxide and tissue seen under these conditions. No increase in glucose-6-phosphate dehydrogenase was seen when pre-incubation was made in a gas atmosphere containing 20% O2. Although dehydroepiandrosterone inhibited both enzyme activity and isotope incorporation in a 20% O2 gas atmosphere, actinomycin D was without effect.

[Structural and functional characteristics of the spleen in systemic and local allogenic graft versus host reaction].

Morphological and histochemical characteristics of the spleen in the systemic and locally induced in the spleen transplant host reaction were compared. In both forms there were observed infiltration of the organ with macrophages and pyroninophilial blasts characterized by a high level of the activity of enzymes of lysosomas and a hexosemonophosphate cycle. In the process of interaction between genetically heterogenous mononuclear cells the disturbance of enzymatic processes in them and in the adjacent cellular elements took place, and susequently they underwent necrosis. Most frequently this process was observed in lymphoid follicles. The destructive component in the spleen tissue was more pronounced in the systemic transplant against host reaction.

Cardiac effects of 3,5-diiodothyropropionic acid, a thyroid hormone analog with inotropic selectivity.

Thyroid hormone exerts a strong positive inotropic action on the heart and induces alpha-myosin heavy chain (MHC) gene expression. 3,5-Diiodothyropropionic acid (DITPA), a carboxylic acid analog with low metabolic activity, was observed to induce alpha-MHC mRNA in heart cell culture with EC50 approximately 5 x 10(-7) M. To determine if the compound has positive inotropic actions, the effects of DITPA and L-thyroxine on heart rate, left ventricular pressures, left ventricular dP/dt, myosin isoenzymes and hepatic alpha-glycerolphosphate dehydrogenase activity were compared in hypothyroid rats. Binding affinities of DITPA and triiodothyronine for bacterially expressed alpha-1 and beta-1 thyroid hormone receptors (TRs) also were determined. Over the dosage range of 150 to 1500 micrograms/100 g, DITPA produced increases in left ventricular dP/dt comparable to those obtained with L-thyroxine at dosages of 1.5 to 15 micrograms/100 g, but with significantly less tachycardia. The increase in alpha-MHC mRNA was about the same with both compounds whereas alpha-MHC protein content and GPDH activity increased less with DITPA. These differences could not be explained by preferential binding of DITPA to TR subtypes. Because heart rate is a major determinant of myocardial oxygen consumption, DITPA is able to achieve increased cardiac performance at lower myocardial oxygen costs.

[Change in the glycolytic and glucose-6-phosphate dehydrogenase activity in experimental cirrhosis of the liver].

Development of cirrhosis of liver tissue did not influence the intensity of glycolysis, with glucose as a substrate, in supernatant fraction of liver homogenate in chronic intoxication with CCL4. In preparations of cirrhotic liver, as compared with liver from the intact animals, more distinct activation of glycolysis was caused by addition of ATP and NAD at the stage of 3-week intoxication and also by addition of hexokinase, glyceraldehydephosphate dehydrogenase and lactate dehydrogenase at the stage of distinct cirrhosis of liver (6 weeks of CCL4 intoxication). Km values for glucose-6-phosphate dehydrogenase increased over all the periods of intoxication.

[Infectious allergy. XIV. Histochemical changes in redox enzymes in cells of the small intestine smooth muscle during sensitization with diphtheria exotoxin].

Administration to guinea pigs of 1/Dlm of diphtheria exotoxin twice at an interval of 1 and 24 hours, or once in a dose of 25/100 Dlm caused changes in the activity of the redox enzymes. The character of their changes in the animals to which the whole dose of the exotoxin was administered once or repeatedly was directly reverse: in the first case the activity of the enzymes was uniformly increased, and in the second--ununiformly depressed; besides, there was a marked disturbance of coordination between the enzymes. An increased sensitivity of the infectious allergy type following injection of low doses of diphtheria exotoxin was not accompanied by immunity formation. Specific antibodies against diphtheria exotoxin formed only in the organism of animals to which 1/100 Dlm of diphtheria toxin was injected once a day for 15 days, but in this case as well no immunity against diphtheria exotoxin was observed.
