Human skin biotransform naturally occurring steroid hormones. 4-14C progesterone skin fate might involve C-19 derivates formation. In vitro incubation of 4-14C-progesterone with defammation. human abdominal skin minces for 5 days periods, predominantly yield: 1) Metabolites reduced at C-20 and C-5 positions of 4-14C progesterone. 2) 4-pregnen-17 01-20-one (0.85%). 3) 4-Androsten-17-01-3-one isolated as an acetylated derivative (0.29%). 4) A metabolite that behaves like 4-androsten-3, 17-diol. The metabolites identity was established through repeated cocrystallizations with known purified carriers in different organic solvents 17 -hydroxy-progesterone was found at greater concentrations in the group of polar metabolites. These metabolites are of biochemical interest but can not be assessed until the effects of these compounds on human skin are known.

On the basis of data from literature and by the use of commercial accessories radioimmunologic method of the determination od 17-alpha-hydroxiprogesteron (17-OHPG) serum level was determined in two variants: a) addition to the previous extraction, b) without extraction. It was initiated in the clinical work for diagnostic of congenital adrenal hyperplasia (CAH) induced by the enzymatic block of C21-hydroxilasis. 17-OHPG level was determined in a group of 15 children aged 2-12 of both sexes not suffering from endocrinous diseases. Mean values od X = 3.2 +/- 2.7 nmol/L and X = 4.3 +/- 3.3 nmol/L were obtained under a) and b), respectively. Also, 17-OHPG concentration was determined in 15 women without any endocrinous diseases by the method under b) and mean value of X = 2.5 +/- 1.7 nmol/L was obtained. 17-OHPG level was determined in a group of 7 children in whom CAH had been verified 3 days after the interruption of the substitutional therapy. According to the results of the investigation it was pointed out that there was a marked increase in 17-OHPG serum level (particularly by the use of b) method) which was far outside the range of reference values. In cases both with 17-OHPG determination and the daily pregnantriol excretion it was noticed that the increase in 17-OHPG was almost always accompanied by the increase in pregnantriol as well. Cortisolemia ranged from normal up to very lowered values.(ABSTRACT TRUNCATED AT 250 WORDS)
Oxyprogesterone capronate (OPC) was administered preoperatively to 46 patients with endometrial tumors and tamoxifen (T) + OPC--to 72 patients. Complex treatment was followed by more pronounced pathomorphosis in tumor. Both schedules inhibited gonadotropin secretion and lowered colpocytoscopic indexes. Dual effect of complex treatment (T + OPC) on the level of cytoplasmic receptors to progesterone was observed. Initially low levels rose while high ones dropped by the end of a 20-day course. Progestin treatment in endometrial carcinoma patients with relatively low estrogen levels should be complemented with tamoxifen to stimulate tumor sensitivity to progestagens.

The paper is devoted to an experimental study of the hormonal properties of two new derivatives of 17 alpha-oxyprogesterone--mepregenol monoacetate and mepregenol diacetate--for their possible use as minipills. Both compounds showed a high progestational activity which was much higher than that of progesterone and the activity of alpha-norgestrel in peroral administration. The above compounds made no inhibitory effect on hypophyseal gonadotropic function; on the contrary, they stimulated follitropine secretion. Besides, different dose-related influence of the steroids on a progesterone level was revealed: the use of small doses resulted in enhanced secretion and larger doses brought about the suppression of secretion. Thus, the new compounds of the progesterone series--mepregenol monoacetate and mepregenol diacetate--appear promising compounds for monohormonal contraception and for the management of conditions with lowered function of the corpus luteum.
Blood spot 17OH-P concentrations were determined in 14 healthy premature (mean birthweight 1439 g, mean gestational age 30 weeks) and full-term newborn infants (mean birthweight 3532 g, mean gestational age 39.2 weeks) during the first five weeks of life to provide reference data for infants with various gestational and postnatal ages. It was demonstrated that with advancing age there was an abrupt fall in 17OH-P from 296.2 +/- 84.1 nmol/l on the first day to 101.2 +/- 19.5 nmol/l on the 7th day (p less than 0.001) and 75.7 +/- 8.7 nmol/l (p less than 0.05) on the 14th day in premature infants. In full-term neonates its initial value is much lower (90.1 +/- 12.5 nmol/l) and its fall during the first week is much less pronounced (51.5 +/- 6.5 nmol/l, p less than 0.01). Comparing the postnatal changes in 17OH-P in the two groups it proved to be significantly higher in premature than in full-term infants at all ages except for the 4th week. When blood spot 17OH-P values were studied as a function of gestational age at the age of 5 days a significant inverse relationship was found between the two parameters. It is assumed that in addition to placental 17OH-P production and perinatal stress, renal salt wasting may also account for the long lasting elevation of 17OH-P plasma level seen in premature infants.
