ToF-SIMS characterization of thermal modifications of bixin from Bixa orellana fruit.

Bixa orellana fruit extracts were studied by time-of-flight secondary ion mass spectrometry (ToF-SIMS). The intensity of the peak at m/z 396, assigned to the bixin molecular ion plus two hydrogen atoms (C25H(32)O(4+), decreased as the extract was heated and nearly disappeared with heating above 150 degrees C. Simultaneously, the formation of dimers at m/z 790, 804, and 818 was observed. The ToF-SIMS spectrum is characterized by a large amount of peaks generated by the principal ions and their multiple fragmentation patterns. To extract maximum information from the data set, multivariate statistical analysis was applied. Principal component analysis revealed important structural changes of the bixin molecule upon heating at different temperatures. This information can be used by the food industry as by controlling the temperature of the heating process the red/yellow balance of this colorant can be tuned.

Contact allergy to oak moss: search for sensitizing molecules using combined bioassay-guided chemical fractionation, GC-MS, and structure-activity relationship analysis.

In addition to pure synthetic fragrance materials several natural extracts are still in use in the perfume industry. Among them oak moss absolute, prepared from the lichen Evernia prunastri (L.) Arch., is considered a major contact sensitizer and is therefore included in the fragrance mix used for diagnosing perfume allergy. The process of preparing oak moss absolute has changed during recent years and, even though several potential sensitizers have been identified from former benzene extracts, its present constituents and their allergenic status are not clear. In the study reported here, we applied a method developed for the identification of contact allergens present in natural complex mixtures to oak moss absolute. The method is based on the combination of bioassay-guided chemical fractionation, gas chromatography-mass spectrometry analysis and structure-activity relationship studies. Our first results showed that atranol and chloroatranol, formed by transesterification and decarboxylation of the lichen depsides, atranorin and chloroatranorin, during the preparation of oak moss absolute, are strong elicitants in most patients sensitized to oak moss. Methyl-beta-orcinol carboxylate, a depside degradation product and the most important monoaryl derivative of oak moss from an olfactory standpoint, was also found to elicit a reaction in most patients.

[Experimental study on the mechanism of oilfield wastewater treatment by using hydrolysis-acidification with aerobic biological processes].

Hydrolysis-acidification + aerobic biological processes were conducted experimentally to treat oilfield wastewater pretreated with physical and chemical treatment in Xinjiang oilfield. The results showed that when the COD concentration in influent was 190-220 mg x L(-1), that in effluent reduced to 65-75 mg x L(-1) under HRT of 10h in both hydrolysis-acidification process and aerobic biological process, reaching the strictest requirement of Effluent Standards for Wastewater from Petroleum Development Industry (GB3550-83). Using GC/MS technology, the relative content of various organic pollutants was analyzed to discover the transfer and degradation law in the oilfield wastewater in biological treatment process. The system of DNA extraction technique, PCR and DGGE reacting systems were practical to analyze the microbial community in the hydrolysis-acidification and aerobic biological processes. The predominant sequences of several 16S rDNA DGGE fragments were determined and confirmed in comparison in GeneBank (NCBI).

Pressate from peat dewatering as a substrate for bacterial growth.

This study considered the possibility of using water expressed during the drying of fuel-grade peat as a substrate for microbial growth. Highly humified peat pressed for 2.5 min at 1.96 MPa produced water with a chemical oxygen demand of 690 mg/liter. Several biological compounds could be produced by using the organic matter in expressed peat water as a substrate. These included polymers such as chitosan, contained in the cell wall of Rhizopus arrhizus, and two extracellular polysaccharides, xanthan gum and pullulan, produced by Xanthomonas campestris and Aureobasidium pullulans, respectively. A very effective surfactant was produced by Bacillus subtilis grown in the expressed water. Small additions of nutrients to the peat pressate were necessary to obtain substantial yields of products. The addition of peptone, yeast extract, and glucose improved production of the various compounds. Biological treatment improved the quality of the expressed water to the extent that in an industrial process it could be returned to the environment.

Improved signals ratio resolution method by optimization of resolution function--simultaneous determination of Cu(II) and Cd(II) in water samples.

From an ecological and economical point of view, it is important to design analytical procedures for monitoring heavy metals in the environment and industrial processes in a way to minimize the use of hazardous reagents and reduce the analysis time. In this paper, a well-known dithizonate extraction-based method for the determination of many metal ions was improved by using chemometrical selectivity of the strongly overlapped spectra of copper and cadmium dithizonates in CCl4 for their simultaneous determination from a single extraction at pH 10. The individual absorption spectra, having absorption maxima difference of only 20 nm, were separated, and the metal ions were quantified by using an improved procedure for optimizing the resolving function in a recently proposed signals ratio method. The procedure consists of using many different resolving functions and plotting the difference of the mean of absolute and nonabsolute mean values of pseudosignals [PDMMV (PS)] against analyte concentrations obtained with each of the resolving functions, thus obtaining 2 straight lines having intersections that give a unique and reliable value of the unknown concentration of the individual analyte in mixture giving strongly overlapped spectra. In this way, the main drawback of the signals ratio resolution method, that is, the visual estimation of optimal resolving function, is eliminated. The proposed parameter, PDMMV (PS), was tested by using both simulated and experimental spectra. Copper was determined in the mixture with ca 20-fold excess of cadmium, and cadmium was determined in ca 10-fold excess of copper at submicromolar concentration levels.

Survey of cured meat products for volatile N-nitrosamines: comparison of two analytical methods.

A survey has been completed of 106 cured meat samples for 14 volatile N-nitrosamines. N-Nitrosopyrrolidine has been confirmed in fried bacon at levels ranging from 5 to 75 ppb. Unconfirmed trace levels of N-nitrosodimethylamine have been observed in a variety of cured meat products. The comparison of the multidetection gas-liquid chromatographic (GLC)-mass spectrometric method with the mineral oil distillation-thermal energy analyzer (TEA) method for the determination of volatile N-nitrosamines in foods shows good agreement between the analytical methods, especially at the 10 ppb level, and excellent agreement between the GLC and TEA analyses of an identical sample extracts.

[Diagnostic study of 5 cases of allergic asthma due to sensitization to silk allergens in textile workers from the Prato area].

Five cases of allergic asthma due to sensitivity to silk allergens have been studied. All the patients worked in the Prato textile industry. The presence of specific antibodies against extracts of silk has been shown by the direct RAST and confirmed by the indirect RAST. The onset of the symptoms, with regard to the various stages in the processing of silk, leads one to assume that the allergen involved is sericin.

Selected uses of enzymes with critical fluids in analytical chemistry.

The use of enzymes coupled with supercritical fluid (SF)-based analytical techniques, such as supercritical fluid extraction (SFE), provides a safer environment platform for the analytical chemist and reduces the use of organic solvents. Incorporation of such techniques not only reduces the use of solvent in analytical laboratories, but it can also lead to overall method simplification and time savings. In this review, some of the fundamental aspects of using enzymes in the presence of SF media are discussed, particularly the influence of extraction (reaction) pressure, temperature, and water content of the extracting fluid and/or the sample matrix. Screening of optimal conditions for conducting reactions in the presence of SF media can be readily accomplished with automated serial or parallel SFE instrumentation, including selection of the proper enzyme. Numerous examples are cited, many based on lipase-initiated conversions of lipid substrates, to form useful analytical derivatives for gas chromatography, liquid chromatography, or SF chromatography analysis. In certain cases, enzymatic-aided processing of samples can permit the coupling of the extraction, sample preparation, and final analysis steps. The derived methods/techniques find application in nutritional food analyses, assays of industrial products, and micro analyses of specific samples.

Surveillance of work-related cold injuries using workers' compensation claims.

The usefulness of workers' compensation claims for the surveillance of occupational injuries was evaluated by analyzing claims for cold injury. Five hundred ten claims filed in 1983 from 23 states participating in the Bureau of Labor Statistics Supplementary Data System and an additional 147 claims filed with the Division of Safety and Hygiene of the Industrial Commission of Ohio from Jan 1, 1984, to June 30, 1985, were examined. As expected, the number of injuries per winter day increased as temperature decreased and wind speed increased. The rate of injury began to increase when temperatures fell below 10 degrees F and wind speeds exceeded 10 mph. These weather conditions were milder than previously published cold hazard charts suggest. Frostbite injuries of the lower extremities occurred at milder temperatures, required more lost workdays, and were more costly than cold injuries to the head and face or to the upper extremities. Industries with the highest rates of injury included agriculture, oil and gas extraction, trucking and warehousing, protective services, and interurban transportation. Injuries during nonwinter months involved the processing, distribution, and preparation of food. Vehicle breakdown or contact with water, gasoline, alcohol, or cold water were noted as contributing factors on many of the compensation claims. This probe suggests that claims for workers' compensation adequately reflect the expected association of environmental factors and the occurrence of cold injury.

Analytical and biological analyses of test materials from the synthetic fuel technologies. I. Mutagenicity of crude oils determined by the Salmonella typhimurium/microsomal activation system.

We have assayed the mutagenicity of crude industrial products and effluents with the Salmonella/microsomal activation system. Test materials (crude products from coal-conversion processes and natural crude oils) were initially fractionated into primary classes by liquid--liquid extraction and then further fractionated by column chromatography. Prescreening was accomplished over a wide concentration range with the Ames tester strains. Active fractions (mainly the neutral fractions containing polycyclic aromatic hydrocarbons and certain basic fractions) can be identified, and dose--response relationships can be established. Standard values are expressed as revertants/mg of the test material assayed with frameshift strain TA98 including metabolic activation with rat-liver preparations. Total mutagenic activity of synthetic fuel samples was consistently higher than that of natural crude "controls." Activities of subfractions are roughly additive and presumably reflect the mutagenic potential of the whole test material. These results are being extended to other genetic assays. Chemical identification is carried out along with the bioassays.

Production of biodegradable plastics from activated sludge generated from a food processing industrial wastewater treatment plant.

Most of the excess sludge from a wastewater treatment plant (60%) is disposed by landfill. As a resource utilization of excess sludge, the production of biodegradable plastics using the sludge has been proposed. Storage polymers in bacterial cells can be extracted and used as biodegradable plastics. However, widespread applications have been limited by high production cost. In the present study, activated sludge bacteria in a conventional wastewater treatment system were induced, by controlling the carbon: nitrogen ratio to accumulate storage polymers. Polymer yield increased to a maximum 33% of biomass (w/w) when the C/N ratio was increased from 24 to 144, where as specific growth yield decreased with increasing C/N ratio. The conditions which are required for the maximum polymer accumulation were optimized and are discussed.

Activity and elution profile of laccase during biological decolorization and dephenolization of olive mill wastewater.

The performance and enzymatic strategy exhibited by basidiomycete Euc-1, a laccase producing strain, was investigated during the biodegradation of olive mill wastewater (OMW). This strain yielded better decolorization of solidified OMW than Phanerochaete chrysosporium and removed 90% of phenols (initial concentration=800 mg l(-1)), 73% of color (initial A465=4.4), and 45% of chemical oxygen demand in batch cultures containing OMW. Since partial phenol removal occurred before the detection of enzymatic activity, no plausible correlation could be established between them. In contrast, decolorization occurred only after the detection of laccase activity and coincided with its production over time. Two laccase fractions (Lac1 and Lac2) were separated by chromatography. OMW strongly induced Lac2 that was almost absent in defined liquid medium. Furthermore, Lac2 was the main laccase fraction in the presence of OMW. This study pointed out that basidiomycete Euc-1 and its ligninolytic system could be a useful tool for the bioremediation of wastewater generated in the process of olive oil extraction.

Extraction of cashew (Anacardium occidentale) nut shell liquid using supercritical carbon dioxide.

This work investigated the extraction of cashew nut shell liquid (CNSL) using supercritical carbon dioxide (SC-CO(2)). Effects of process parameters such as extraction pressure, temperature and flow rate of SC-CO(2) were investigated. The yield of CNSL increased with increase in pressure, temperature and mass flow rate of SC-CO(2). However, under different operating conditions, the composition of CNSL varied. The study of physical properties and chemical composition of the oil obtained through super critical fluid extraction (SCFE) showed better quality as compared to the CNSL obtained through thermal route. Experimental results were compared with diffusion based mass transfer model. Based on this simple model, extraction time was optimized.

Mercury contamination in chile: a chronicle of a problem foretold.

This review analyzes the effects of environmental mercury contamination in Chile. This contamination generates one of the most important environmental conflicts in the country in that it affects air, ground, and water (rivers and oceans), which are fundamental in maintaining natural biotic equilibrium and at the same time important for the nation's economy. Chile possesses extraordinarily wealthy mining resources between Regions I and IV that have developed into an extraction industry essential for the economy of the country. However, waste discharges from this production have created an environmental problem in that the majority of the mines are located in the Andes mountain range, or areas close by, and the water used in the extraction process is deposited into the rivers, significantly increasing the amount of chemical contamination. Therefore, the cities and downstream waters used in agriculture suffer the negative consequences of a natural resource that is becoming more and more scarce. In addition, minerals released from mills into the atmosphere are deposited onto the soil, drastically affecting the biological resources of these areas. One of these affected areas is the Metropolitan region, where one of the highest contamination levels of mercury in the country was found in one of its affluents due to industrial and domestic waste discharge. In a country that is only 200 km in width, the gathering of all these contaminants in the rivers results in a rapid flow to the ocean, thereby contaminating coastal waters and the biota. In general, this contamination has been detected in semiclosed bodies of water (bays). Between Regions VII and IX, the principal sources of mercury contamination are related to cellulose industrial sites (Regions VII and VIII) and, until the 1980s, the bleach-soda industry. The most important industrial and fishing activity is also found in this area. In San Vicente Bay, waste discharges released into the ocean include sewage, industrial residues, residues from fishing and mining industries, hydrocarbons, petrochemical derivatives, oils, and detergents. This combination of chemical assault makes the San Vicente Bay the most contaminated in the country and the area where the majority of mercury contamination studies have been carried out. Between Regions X and XII, mercury contamination is reduced due to decreased release of domestic residues, especially batteries and sanitary waste. Beginning with the decade of the 1990s, Chile made a great effort to decrease contamination through governmental organizations (CONAMA, SERNAGEOMIN, DGA, ECOMIN, SONAMI), nongovernmental organizations (NGOs), universities, government mining industries (CODELCO, ENAMI), and private mining industries (El Indio, La Escondida, La Candelaria, Fachinal, etc). These reduction efforts within the last 10 years exceed $900 million, and in the private mining sector alone more than 1,100 monitoring stations have been installed and more than 100,000 environmental measurements have been carried out each year. Furthermore, an important educational program on the use of mercury has been implemented in the small mining area to decrease contamination to the air, water, and soil. However, the consequences of mercury accumulation are seen in their damaging effects to the rivers that deliver water to crops and cities, in the bays where food is extracted, and in the air of some cities where there exist mills that release chemical substances into the atmosphere.

[Soft tissue filling with hyaluronic acid].

PHYSIOLOGY AND PHARMACOLOGY: Hyaluronan was discovered by Karl Meyer in 1934 in the vitreous humor of cattle eye. Hyaluronic acid, (AH), is a natural polysaccharide and a ubiquitous component of the extra cellular matrix. It is largely biocompatible and has a short half life. In the early 1990's, preceded by the use of bovine collagen, AH started to be employed in the fields of Dermatology and Plastic Surgery; currently it is a major intervention product in both soft tissue augmentation and facial volume loss treatments. Often well tolerated, AH, has high water retention properties and is an effective tissue volumizer. Industry first attempted to extract AH from rooster comb, then through bacterial fermentation and succeeded in increasing its lifespan by cross linking. However, industrially produced AH can contain residues from the manufacturing process. It thus appears critical to us to be informed of the process used in manufacturing AH including the reticulant utilized. Manufacturers should be legally required to publish this data as well as to conduct physiochemical follow-up studies over the short, mid and long terms. LEGAL CONSIDERATIONS: In Europe, CE marking is a prerequisite to market injectable products. However, a CE marking does not necessarily imply that the product's efficacy and side effects have been assessed objectively in clinical studies. However, this marking is expected to fall into line with the US legislation, where the marketing of any medicinal product is subject to FDA approval, based on comprehensive animal and clinical studies and on more systematic and better centralised side effect reporting. We have examined most of the products used in Europe and internationally in 2004, whether of animal or bacterial origin, reticulated or non-reticulated, and used to restore, increase volume or as a product vector. Before injecting these products, it is essential to have a thorough understanding of their absolute and relative contraindications and anaesthetic requirements, differences between types of wrinkles to fill in and indicated techniques, differences between concerned regions or tissues, and the impact of associated cosmetic treatments. These resorbable injectable products have mild and, more importantly, short-lived side effects. However, in patients with a relative contraindication, a double test is justified, as allergic reactions are known to occur in 1 to 3% of patients. It is also advisable not to inject HA in a site previously injected with a non-resorbable product. More precise statistics on results and side effects based on double-blind randomised studies are still wanting, due to the lack of legal requirement and effective centralisation of data. Therefore, all side effects must be reported to improve our knowledge as well as the safety of injections. CONCLUSION: Although their duration of effect is limited, HA products are the most commonly used fillers, before collagens. Many questions have yet to be answered, but they produce very significant results in filling procedures. Both clinicians and patients praise these products for their tolerance, resorbable nature, and limited side effects.
