Effect of dietary supply of butters rich either in trans-10-18 : 1 or in trans-11-18 : 1 plus cis-9, trans-11-18 : 2 on rabbit adipose tissue and liver lipogenic activities.

Experimental butters with a high content of trans-18 : 1 fatty acids and/or cis-9,trans-11-18 : 2 (rumenic acid; RA) were fed to thirty-six New Zealand White rabbits to investigate their effects on adipose tissue (AT) and liver lipogenic activities. Animals received one of three atherogenic (0.2 % cholesterol) diets containing 12 % butter with either a standard fatty acid composition (rich in saturated fatty acids), rich in trans-10-18 : 1 (T10 diet) or in trans-11-18 : 1 plus RA (T11+ RA diet) for 6 or 12 weeks. The ingestion of butters rich in trans fatty acids and/or RA for 6 weeks had little or no effect on liver and AT lipogenesis. The ingestion for 12 weeks of butter rich in T11+ RA decreased perirenal AT weight, lipogenic enzyme and lipoprotein lipase activities, without affecting liver lipid concentration or lipogenic activities except for a decrease in glycerol-3-phosphate dehydrogenase activity. Similar trends, but of a lower magnitude, were observed in rabbits fed the T10 diet for 12 weeks. Ingestion of the T10 or T11+ RA diets for 6 or 12 weeks had no significant effect on plasma metabolites and hormones except for glucose which increased at 6 weeks in the T10 group. Plasma leptin concentration was positively correlated with AT weight but did not differ between the three diets. In conclusion, the supply of butters rich in either T10 or T11+ RA in an atherogenic diet for 12 weeks decreased rabbit AT lipogenesis, with a more marked effect of the T11+RA diet, but had no effect on liver lipogenesis.

[Histological and histochemical studies on testis, epididymis and ductus defrens of the rooster (Gallus domesticus)].

Testis, epididymis and ductus deferens of the adult domestic fowl and male gonads of juvenile roosters have been studied by means of histochemical and histological methods. Testicular interstitial cells: According to enzyme-histochemical results oxidative energy production seems to be of minor importance. An extraordinarily high activity of lysosomal enzymes (acid phosphatase and esterases in the adult, esterases only in the immature) is observed. Positive reactions of 3beta-steroid-dehydrogenase and enzymes of the pentose phosphate cycle indicate steroid hormone production; the pathways of the steroid synthesis, however, are probably different in adult and immature testes. A remarkable LAP content of juvenile Leydig cells is a parameter of an increased protein metabolism. Areas of reserve cells: Focal accumulation of these cell types are observed in testis and epididymis of the immature and in the epididymis of the adult fowl. Reserve cells reveal distinct activities of LAP (prospective growth ability) and 3beta-HstDH (reserve capacity for steriod synthesis). All other enzymes studied react weakly, thus pointing to a generally low metabolic activity. Seminiferous tubules: The strong reaction of alkaline phosphatase in the peritubular cells may play a part in energy disposition for contractions. Sertoli cells of adult animals are rich in lysosomal enzymes and enzymes of the glycolytic chain but oxidoreductases react weaker than in mammalian Sertoli cells. This indicates that nutritive interactions between germ and Sertoli cells in birds are different from those in mammals: The basally orientated germ cells of birds contain strong activities of diaphorases, LDH, SDH, Cyto-Ox and seem to be metabolically rather indipendent from Sertoli cells.

Oxidative enzymes in the development of Fasciola hepatica L. IV. The activity of oxidases and dehydrogenases in redia.

The object of the study was to investigate the occurrence and localization of oxidative enzymes in the redia -- the third larval stage of Fasciola hepatica L. The author detected cytochrome oxidase, peroxidase, NADH and NADPH tetrazolium reductases (diaphorases), as well as succinate, isocitrate, malate, lactate, alpha-glycerophosphate, glyceraldehyde phosphate, glucose-6-phosphate, 6-phosphogluconate, beta-hydroxybutyrate, L-glutamate, and alcohol dehydrogenases. The presence and localization of the enzymes in various periods of development of the redia were detected with histochemical methods. Out of the studied oxidases and dehydrogenases only cytochrome oxidase was found to be absent from the stages of young rediae. It was ascertained that the redia uses all three paths of release of energy i.e. the glycolytic, Krebs, and pentose cycles, glycolysis being presumably the principal mode of energy production.

Oxidative enzymes in the development of Fasciola hepatica L. V. Activity of oxidases and dehydrogenases in the Cercaria and Metacercaria.

The object of the study was the investigation of the occurrence and localization of oxidative enzymes in the 4th and 5th larval stages of the liver fluke, i. e. in the cercaria and metacercaria. The following enzymes were detected: cytochrome oxidase, peroxidase, NADH and NADPH tetrazolium reductases/diaphorases) as well as succinate, isocitrate, malate, lactate, alpha-glycerophosphate, glyceraldehyde phosphate, glucose-6-phosphate, 6-phosphogluconate, beta-hydroxybutyrate, L-glutamate and alcohol dehydrogenases. The occurrence and localization of the enzymes were investigated histochemically in the cercaria still unreleased from the snail tissues, in the free natatorial cercaria, and in the encysted specimen, i.e. metacercaria. Among the enzymes studied only peroxidase was found to be absent from the cercaria and metacercaria, the latter larva being deprived of alcohol and L-glutamate dehydrogenases as well. The aerobic path, i.e. the Krebs cycle, was ascertained as the principal mode of obtaining energy in the free natatorial cercaria and metacercaria, and glycolysis as the main energy path for the undetached larva. The analysis of the above-mentioned oxidative enzymes was the basis for the description, within the available range, of the function and metabolism of individual organs of cercaria in different periods of its development. In the recapitulation the author discusses the effect of the parasitic way of life of the larval forms of Fasciola hepatica on their energy metabolism.

[Cytoplasmic maturation of cow oocytes cultured in vitro and their dehydrogenase activity].

Approximately in 80% of cow oocytes (n = 632) ended cytoplasmatic and nucleus maturation to the state of metaphase II in the conditions of 24 hours in vitro cultivation. In 300 oocytes cytochemically we have determined the activity of enzymes--the succinate dehydrogenase (SDH, EC 1.3.99.1.), alpha-glycerophosphate dehydrogenase (GPDH, EC 1.1.1.8.) and glucose-6-phosphate dehydrogenase (G6PDH, EC 1.1.1.49.). The reaction intensity of the observed dehydrogenases increased in cow oocytes which were cultivated in vitro for 24 hours. Dehydrogenases are located in the mitochondria which are laid out regularly in the cytoplasm of oocytes. Part of oocytes showed polarization in the lay out of reaction and part of oocytes gave extramitochondrial reaction.

Histological and histochemical characteristics of the bovine notochord.

In 20 bovine embryos and fetuses 6-65 mm long (crown-rump length) and 23 to 60-70 days old, the structure and localization of acid and neutral mucopolysaccharides and glycogen in their notochord were investigated. Also, the localization in the notochord was examined of the activity of alkaline and acid phosphatases, alpha-glycerophosphate-, glucose-6-phosphate-, isocitrate-, glutamate-, lactate- and succinic- dehydrogenases, and nicotinamide-adenine-dinucleotide- and nicotinamide-adenine-dinucleotide-phosphate- diaphorases. It was found that the bovine notochord begins decomposing at the end of embryonal and the beginning of fetal development (45-50 days old) and that in the fetus aged 55-65 days it no longer represents an unbroken cord of notochordal cells. Secretory activity of notochordal cells which produce the notochordal sheath starts very early (in 10 mm-long embryos), and interruptedly increases up to the end of the embryonal developmental period when regression appears at the beginning of the fetal period. These findings agree with findings in the human embryo where, however, they relate to earlier developmental periods.

Histochemistry of oxidative enzymes in experimental allergic encephalomyelitis.

The histoenzymic pattern of oxidative enzymes (G3PD, IDH, SD, G6PD,HBD, NADPH: dehydrogenase) was investigated in experimental allergic encephalomyelitis (EAE) produced in rats according to PATERSON [13]. The results obtained lead to following conclusions: (1) The neuroglia, including the white matter oligodendroglia of immunized rats, exhibits increased oxidoreductase activities; (2) The neuroallergic reaction induces some stimulation of the oxidoreductive metabolism of oligoden-droglia; (3) The enzymatic hyperactivity in EAE does not show any relation to the morphological signs of alterations of the myelin sheath.

[Cytochemical parameters of lymphocytes in non-coronarogenic myocardial diseases].

Blood lymphocyte redox enzymes were examined in 102 patients with infectious allergic myocarditis and vegetative dyshormonal +myocardial dystrophy. Dehydrogenase activity was found shifted and cellular energy balance disordered, these factors being valuable for differential diagnosis. Estimation of the lymphocytic energy plastic coefficient permits a dynamic monitoring of patient's status and helps predict the outcome of rehabilitation process. The detected metabolic shifts prompted inclusion of ++pyridoxal phosphate, a new coenzymic drug, in complex of treatment of this patient population.

[Theca-like reactions in the stroma of cilioepithelial ovarian neoplasms].

The work deals with the study of the so-called theca-like reaction in the stroma of 283 cilioepithelial tumours of the ovary (50 simple, 28 proliferating cystadenomas, and 205 cancers developed from cilioepithelial cystadenomas). The composition of the interstitial tissue of neoplasms of this type (except simple cystadenomas) included cells morphologically undistinguished from the thecal elements of the ovary. They may contain cholesterolethers and sets of redox enzymes typical of the steroidproducing tissues. Manifestation of the theca-like reaction (findings of morphometry) depended upon the degree of anaplasia of the tumour epithelium: it was most pronounced in malignant highly differentiated tumours, and least pronounced in drastically anaplasic cancer.

The ammocoetes endostyle: its oxidative enzymes as an evidence of its homology with the thyroid of higher chordates.

Histochemical methods were used for the demonstration of activity of the following intracellular oxidative enzymes, unstudied hitherto, in the epithelial cells of the endostyle of the river lampre (Lampertr aluviatilis L.) ammocoetes: reduced NAD dehydrogenase (NADD), lactate dehydrogenase (LD), cytochrome oxidase (CO), succinate dehydrogenase (SD), alpha-glycerophosphate dehydrogenase (alphaGPD) and glucose-6-phosphate dehydrogenase (G6PD). The activities of NADD and LD in the iodophil and throidogenic cells of type 3, then of subtype 2c and partly types 4 and 5 of the endostylar epithelium and the hypobranchial duct-lining epithelium were particularly ithe larva proves the possibility of their participation in the formation of the thyroid gland in the period of metamorphosis. In type 1 cells of the ammocoetes, despite their fairly strong enzymatic reactivity, the oxidative activity does not change significantly during the ontogenetic stages examined. The data obtained make it possible to modify the present views on the genesis of the thyroid gland of the adult lamprey, namely, they indicate the participation of the type 6 cells of the hypobranchial duct-lining epithelium in the process of thyroidogenesis.

The activity of some intracellular oxidative enzymes in the thyroid follicular epithelium of Xenopus laevis Daud. in experimental long-term hypokinesia.

Histoenzymological methods were applied to examine the activities of previously little studied intracellular oxidative enzymes in the cells of the thyroid follicular epithelium of Xenopus laevis Daud. specimens kept in aquarium. Succinate, lactate, alpha-glycerophosphate, glucose-6-phosphate and reduced NAD and NADP dehydrogenases and cytochrome oxidase were studied. In comparison with other chordate species a very strong activity was revealed particularly by lactate dehydrogenase. The data obtained suggest that the metabolism in thyroid cells of the motionless Xenopus is based on glycolysis mainly.

A randomized clinical trial to investigate the usefulness of the addition of prednisolone to tamoxifen as adjuvants to mastectomy in primary breast cancer patients with a high risk of recurrence: a preliminary report.

The efficacy of the addition of prednisolone to tamoxifen as adjuvants to mastectomy in patients with primary breast cancer who were at a high risk of recurrence was investigated in a randomized trial. Primary carcinomas were collected from a series of 169 patients with loco-regional disease, undergoing mastectomy. The activities of alpha-glycerolphosphate dehydrogenase and 6-phosphogluconate dehydrogenase in the carcinomas were estimated biochemically and the ratio of the two enzymes was used to as the parameter to determine the risk of recurrence. 116 patients with a high risk of recurrence within five years of mastectomy were then randomized to either tamoxifen (2x20 mg/day) or tamoxifen+prednisolone (3x2.5 mg/per day) until recurrence. The patients are currently followed quarterly. The data were analysed at a median follow-up of 26 months (range 7-62 months). The probabilities of both disease-free and overall survival were not significantly different in either arm of the trial, indicating that there is no advantage in combining prednisolone with the antioestrogen. Recently, similar findings in terms of response have been reported for patients with metastatic disease treated with the same combination, raising doubts over the role of prednisolone in the management of patients with endocrine treatments.

[Effect of removal and stimulation of the superior cervical sympathetic ganglia on the activity of oxidation-reduction enzymes in the neurosecretory cells of the anterior hypothalamus in rabbits].

Experiments were conducted on rabbits. A study was made of the activity of the redox enzymes--glucose-6-phosphate dehydrogenase (G-6-PDH), lactate dehydrogenase (LDH), succinate dehydrogenase (SDH), malate dehydrogenase (MDH), NAD-and NADP-diaphorases, cytochromeoxidase (CCO), alpha-glycerophosphate dehydrogenase (alpha-GPDH) in the supraoptic and paraventricular nuclei of the hypothalamus and the posterior lobe of the hypophysis under conditions of stimulation and removal of the superior cervical sympathetic ganglia. There was revealed a correlation between the activity of the tissue respiration enzymes (SDH, MDH, NAD- and NADP-diaphorase, CCO) and the functional condition of the hypothalamo-neurohypophysial neurosecretory system. However, the enzymes of the pentose-phosphate (G-6-PDH) and glycerophosphate shunt (alpha-GPDH) and also of the anaerobic way of oxidation (LDH) reacted nonspecifically on the induced effects.

[Studies of the activity of various dehydrogenases in the desert rat (Meriones unguiculati) nephrons].

Researches about the Activity of some Dehydrogenases in the Nephrons of Desert Rats (Meriones unguiculati).--Our desert rats Meriones unguiculati (Mongolian Gerbils) show--also when held under the same experimental conditions as our other laboratory rodents (Wistar rats and white mice)--a somewhat different enzyme histchemical behaviour of these nephrons. The essential difference in the Meriones is that their distal convolutions display on the whole stronger enzyme activities compared as well with the proximal convolutions of the same kidneys, as with the distal convolutions of Wistar rats.

Kinetic studies of bovine liver fructose-1,6-bisphosphatase.

Initial rate kinetic studies with bovine liver fructose-1,6-bisphosphatase were carried out in both directions of the reaction to determine the sequence of product release from the enzyme. Product inhibition by fructose-6-P was found to be S-linear, I-linear noncompetitive relative to fructose-1,6-bisphosphate, whereas inorganic orthophosphate was determined to be linear competitive with respect to the substrate. The kinetics of the reverse reaction were studied by coupling the phosphatase reaction to the aldolase, triosephosphate isomerase, and glycerolphosphate dehydrogenase reactions. The kinetic results were found to be in harmony with the Uni Bi ordered and random sequential mechanisms as well as a Uni Bi ping-pong mechanism. The nomenclature is that of Cleland (Cleland, W.W. (1963) Biochim. Biophys. Acta 67, 104-137). However, nonkinetic considerations, when taken together with the kinetic results, suggest that the steady state ordered Uni Bi mechanism is the most likely possibility. There is evidence that isomerization of the binary complex of enzyme and phosphate occurs in the kinetic mechanism. Although magnesium is required for the reverse reaction, there is no evidence to suggest that the enzyme discriminates between the magnesium-associated or divalent cation-free forms of the substrates.

[Effect of oxythiamine on pyruvate and lactate metabolism in rat tissues].

The ratio of two substrates (pyruvate and lactate), activity of enzymes, responsible for their turnover in liver tissue, sceletal muscles and blood, were studied simultaneously in experiments on the effect of hydroxythiamin, which was administered at doses, causing a state of hypovitaminosis B1. Hydroxythiamin was shown to inhibit specifically the activity of thiamin-dependent enzymes (transketolase and pyruvate dehydrogenase). It affected also the metabolism of triose phosphates, pyruvate and lactate. At the same time, hydroxythiamin was responsible for changes in activities of other enzymes, participating in turnover of the substrates studied.

[Use of electric stimulation to prevent the development of muscle changes during hypokinesia].

An attempt was made to use electric stimulation of antigravity muscles of hind limbs of rats exposed to a 15 and 30 day hypokinesia as a method for preventing structural and metabolic disorders that develop in the muscular tissue during a diminished motor activity. The electric stimulation was carried out using the Bion device with a frequency-amplitude modulation. The impulses of an amplitude of 0.7-1 msec and a frequency of 100 to 130 Hz were modulated with a frequency of 0.5 Hz. During the first experimental days the electric stimulation was 5 to 7 min and by the end of the first week it reached 30 min. Throughout the first week the voltage grew from 6-7 to 20 V. Electric stimulation was applied 5 times a week. The favourable effect of the electric stimulation was demonstrated in 4 out of 12 animals during every experimental run. It is suggested that a higher percentage of the protective effect of the method can be achieved with the stimulation ensuring a more intensive influence on the deeply located muscles.

[The significance of histochemical demonstration of oxidative enzymes in the interpretation of local and remote effects of experimental brain lesions].

In cortex, habenular- or hypothalamic regions of white rats electrocoagulation was carried out. Animals were killed from 6 days to 5 months after the operation. Serial slides of the brain were investigated by the aid of a reaction of SDH, alfa-GPDH, LDH, G6PDH, TPND. Group of neurons around the lesion may show a decreased, normal and increased activity of the enzymes investigated. As for the far effect of the lesion in case of the injury of the postcomissural fornix changes could have to be revealed in the corpus mamillare. Activization of neuroglia is marked by the increase of the reaction of G6PDH, LDH and TPNDH. Enzyme activity of cells of microglial origin at the beginning seems to be significant, later on, when the activity of phagocytes comes to the end it decreases to the minimum. Regeneration of cells of the leptomeninges and their role in demarcation of the process is marked by the increase of enzyme activity.
